1. Inhibitory effects of wnt inhibitor IWR-1 on proliferation and migration of osteosarcoma MG-63 cells

Objective: To investigate the effects of Wnt inhibitor IWR-1-endo (IWR-1) on the proliferation and migration of human osteosarcoma MG-63 cells, and to explore the possible mechanism. Methods: MG-63 cells were treated with different concentrations of IWR-1 (2, 4, 8 and 16 μmol/L). Then the growth inhibitory rates of MG-63 cells was determined by CCK-8 assay. The cell cycle and apoptosis rate of MG-63 cells were detected by FCM method. The migration ability of MG-63 cells was abserved by scratch wound healing assay. The expression levels of β-catenin and cyclin D1 mRNAs were detected by real-time fluorescent quantitative PCR. The expression levels of Axis inhibition protein 1 (AXIN1), β-catenin, phospho-β-catenin (p-β-catenin) and cyclin D1 proteins were detected by Western blotting. Results: The proliferation inhibitory rate of MG-63 cells after IWR-1 treatment increased in a time-and concentration-dependent manner (all P < 0.01). The proportion of MG-63 cells treated with IWR-1 in G0/G1 phase were increased (P < 0.05), and the apoptosis rate of MG-63 cells increased significantly with the increase of IWR-1 concentration (P < 0.01). IWR-1 reduced significantly the scratch healing rate of MG-63 cells (P < 0.01), decreased the mRNA (both P < 0.01) and protein (both P < 0.05) expression levels of β-catenin and cyclin D1, and increased the expression levels of AXIN 1 and p-β-catenin proteins (both P < 0.05). Conclusion: IWR-1 can inhibit the proliferation and migration of MG-63 cells by blocking Wnt/β-catenin signaling pathway.
2.Application of transesophageal echocardiography in perioperative period of pediatric patients with congenital heart disease

Objective To evaluate the role of transesophageal echocardiography (TEE) in perioperative period of pediatric patients with congenital heart disease (CHD).Methods From July 2011 to December 2012,TEE was used in 393 pediatric patients(≤ 14 years) with congenital heart disease in perioperative period.We make a retrospective review with the clinical data of these cases.Results Operative schemes or therapeutic schedules of 60 patients(15.3%) were altered according to TEE.By preoperative TEE,the diagnosis of transthoracic echocardiography (TTE) of 4 cases(1.0%) were amended,4 cases (1.0%) were complemented,and 4 cases (1.0%) got the auxiliary diagnosis,among which operative schemes of 11 cases (2.8%)were altered.During the operate,29 cases(7.4%) found residual problems,11 cases(2.8%) got the auxiliary diagnosis.By postoperative TEE,7 cases(1.8%) found residual problems,1 case(0.3%) got the auxiliary diagnosis.Complications occurred in 9 cases(2.3%) of the 393 patients.Oral and pharyngeal mucous membrane bleeding occurred in 7 cases (1.8%),inadvertent tracheal extubation in 2 cases(0.5%).Conclusion TEE plays an important role in confirming preoperative diagnoses,formulating surgical plans,evaluating immediate operative results,identifying patients with residual defects and guiding the therapeutic schedule in perioperative period of pediatric patients with congenital heart disease.
3.Key words-based analysis of subjects in Chinese papers on Kashin-Beck disease

Objective To reveal the main interests in studies of Kashin-Beck disease by analyzing the key words in Chinese papers on Kashin-Beck disease .Methods Chinese papers on Kashin-Beck disease were retrieved from CNKI, CBM, VIP, and Wanfang digital journal full-text database.A co-word matrix was generated by abstracting their key words which were analyzed by gCLUTO clustering .Results The studies on Kashin-Beck disease were centered on the role of fusarium metabolic toxins and fulvic acid in pathology of Kashin-Beck disease, etiology of Kashin-Beck disease, functional change in Kashin-Beck disease patients, relation between Kashin-Beck disease and geological envi-ronment, monitoring and X-ray diagnosis of Kashin-Beck disease, surgical treatment of Kashin-Beck disease, preven-tion and treatment efficiency analysis of Kashin-Beck disease.Social network figure showed that etiology of Kashin-Beck disease, relation between Kashin-Beck disease and geological environment, monitoring and X-ray diagnosis of Kashin-Beck disease, surgical treatment of Kashin-Beck disease, prevention and treatment efficiency analysis of Kashin-Beck disease were the hot spots in studies of Kashin-Beck disease at present.Conclusion The research subjects determined by co-word clustering analysis are consistent with the interests in studies of Kashin-Beck disease in China at present.
4.Analysis of the hemodynamic changes of transplanted liver with acute rejection using color Doppler flow imaging: a preliminary study

Objective To investigate the viability of analysis of hemodynamic changes with color Doppler flow imaging in the prediction of acute rejection in transplanted liver.Methods Sixty-eight patients enrolled in the study were categorized into three groups:transplanted liver without acute rejection [rejection activity index(RAI) 0-3,n =24],transplanted liver with mild acute rejection (RAI 4-5,n =23),transplanted liver with moderate and severe acute rejection (RAI 6-9,n =21).All the patients were confirmed by pathology.The color Doppler flow imaging were performed in all the patients within 24 hours and 1 week after biopsy.Results In transplanted liver with moderate and severe acute rejection,the peak systolic velocity of portal vein (PV-PSV) was (31.4 ± 14.1)cm/s,significantly lower than that in transplanted liver without acute rejection,which was (45.1 ± 17.7)cm/s (P <0.05).A week later after steroid therapy,the PV-PSV in transplanted liver with moderate and severe rejection was increased to (46.7 ± 21.8)cm/s(P <0.05).Patients with acute rejection were associated with the decrease of the PVPSV (P <0.05).Conclusions The decrease of PV-PSV may have some clinical value in evaluation the acute rejection in transplanted liver.
5.Research competence of authors in papers on Kashin-Beck disease and characteristics of their cooperation

Objective To provide the evidence for cooperation in research on Kashin-Beck disease in China by ana-lyzing the research competence of authors in papers on Kashin-Beck disease and their cooperation .Methods Papers on Kashin-Beck disease were retrieved from 4 commonly used Chinese perodical databases .Key words Information was collected and organized using BICOMS to produce a co-words matrix,atlas of co-words was plotted using NetDraw, the data were analyzed using SPSS 17.0.Results Of the 3454 authors engaged in research of Kashin-Beck disease from 1957 to 2012,81 published 15 or more than 15 papers.Cluster analysis showed that these 81 authors were from Institute of Endemic Diseases,Xi'an Jiaotong University Medical School; Institute of Kashin-Beck Disease,Chinese Center for Endemic Disease Prevention and Control;Jilin No.2 Center for Endemic Disease Prevention and Control;Centers for Disease Prevention and Control of China Academy of Sciences , Henan Province and Gansu Province .Conclusion A large number of researchers are engaged in research of Kashin-Beck disease .Their research interest and charac-teristics are clear.However, the cooperation between different institutions is weak and should be strengthened.
6.Progress in Wnt signaling pathway and tumorigenesis

The Wnt/beta-catenin signaling pathway plays an important role in tumorigenesis and development,and is a key pathway mediating carcinogenesis. Therefore,further studies of Wnt/beta-catenin signaling pathway may provide powerful help for exploring the pathogenesis of tumors and seeking effective diagnosis and therapy. This article reviews the research progress of Wnt/be-ta-catenin signaling pathway and tumorigenesis.
