1.Effect of transplanted mesenchymal stem cells transfected with adenoviral ventor mediated HGF gene on snrvival volume of grain fat grafts

ObjectiveTo investigate the effect of mesenchymal stem cells (MSCs) transfected with hepatic growth factor (HGF) gene on the survival volume of free grain fat grafts.Methods MSCs of male Wistar rats were transfected with Ad-GFP or Ad-HGF.The transfection infectivity of Ad-GFP to MSCs,the expression of HGF were measured using ELISA assay.150 rats were randomly divided into 5 groups:control group (group A),MSCs group (group B),Ad HGF group (group C),MSCs transplanted with Ad-GFP group (group D),and MSCs transplanted with Ad-HGF (MSCsHGF) group (group E).Then,the same volume of frain fat graft,mixed with DMEM LG andMSCs,Ad HGF,MSCs-GFP,MSCs HGF respectively,were transplanted to rats back,but control group were only mixed with DMEM-LG.Fat graft was obtained on days 3,5,7,14,28,and 60 after implantation.The volume of fat graft was measured by messcylinder,and the expression of HGF and CD34 in transplanted fat tissue were detected by immunohistochemistry.ResultsThe transfection infectivity of Ad-GFP to MSCs was 89.6 % at 100 MOI,the expression of HGF in MSCs culture medium reached to the level after being transfected with Ad-HGF for 48 h.Compared with other 4 groups,at days 3,5,7,and 14 post-transplantation,the expression of HGF in E group transplanted fat of group E had statistics significance (P<0.05).The persentage of survival volume of fat graft in group E was significantly higher than that of other group ( P<0.05) at days 28,and 60 post transplantation.ConclusionsMSCs transplanted with Ad-HGF could secrete HGF and increase the survival volume of fat grafts.
2.Application of multimodal analgesia of ropivacaine combined with dezocine in cheiloplasty of children and evaluation on its postoperative analgesia efficacy

Objective: To discuss the postoperative analgesia efficacy of multimodal analgesia of ropivacaine combined with dezocine, and to illuminate the feasibility of multimodal analgesia in the children undergoing cheiloplasty.Methods:In the randomized, controlled and double blind study, sixty children scheduled for cheiloplasty were randomly divided into ropivacaine group,dezocine group and multimodal analgesia group (n=20). The children in ropivacaine group and multimodal analgesia group were treated with infraobital nerve blockade (1.5 mL 0.25% ropivacaine)before skin incision.The children in dezocine group received the same volume of normal saline. The patients in dezocine group and the multimodal analgesia group received dezocine (0.15 mg·kg-1 )20 min before the end of operation, and the children in ropivacaine group received the same volume of normal saline.The children’s ages and weights,duration of anesthesia and operation, reviving and extubation time,agitation score and incidence,laryngospasm or bronchospasm,CRIES scores at 2,4,6,8,12, and 24 h after operation and adverse reactions were all recorded.Results:There were no significant differences in the age,weight,the duration of anesthesia and operation of the children between three groups (P >0.05).Compared with ropivacaine group,the reviving and extubation time of the children in dezocine group and multimodal analgesia group were increased (P <0.05).There were no significant differences in the scores and the incidence of agitation between three groups (P >0.05).There were no laryngospasm or bronchospasm occured in all groups.The CRIES score at 2 h after operation of the children in multimodal analgesia group was the lowest and there were significant differences compared with other two groups (P <0.05).The CRIES score at 4 h after operation of dezocine group was increased compared with 2 h after operation and was higher than other two groups (P < 0.05).The CRIES score at 6 h after operation of the children in multimodal analgesia group was yet the lowest, and there were significant differences compared with other two groups (P <0.05).The CRIES score at 6 h after operation of the children in ropivacaine group was also increased,but there was no significant difference compared with dezocine group (P >0.05).There were no significant differences in the CRIES scores at 8,12,and 24 h after operation between three groups (P > 0.05).Compared with other two groups,the incidence of tachycardia and the cases using analgesic in multimodal analgesia group were the lowest,and there were significant differences compared with other two groups (P < 0.05 ).There was no respiratory inhibition in all groups.Conclusion:The multimodal analgesia of ropivacaine combined with dezocine can effectively prolong the postoperative analgesia duration and reduce adverse reactions, and it can be safely used in the postoperative analgesia in the children undergoing cheiloplasty.
3.Study of Inhibition Fibrosis of MicroRNA-25 on Diabetic Nephropathy by Regulating Expression of MAP2 K4

Objective To determine the expression level of microRNA-25 in animal models of diabetic nephropathy and human renal tubular epithelial cells (HK-2) cultured in different conditions, and to explore its regulating effect on the fibrosis in diabetic nephropathy. Methods The expression of microRNA-25 was detected by real-time PCR. The downstream target protein of microRNA-25 was verified by bioinformatic prediction, transient transfection of cells and Western blotting. Results MicroRNA-25 was down-regulated in animal models of diabetic nephropathy and HK-2 cells which were cultured in high-glucose medium (P<0. 01). MAP2K4 might be the downstream target protein of microRNA-25. Overexpression of microRNA-25 reduced the protein expression of MAP2K4 and α-SMA (P<0. 01). Conclusion MicroRNA-25 inhibits the fibrosis in diabetic nephropathy by regulating the expression of MAP2K4.
4.Effect of Curcumin on Non-Small Cell Lung Cancer A549 Cells

Objective To study the inhibitory effect of curcumin on the proliferation,migration and invasion of non-small cell lung cancer cell A549,and to discuss further if it is closely related to the expression of c-Jun N-terminal kinase (JNK) and relative protein p38.Methods A549 cells were cultured by conventional method,and then treated with different concentration of curcumin (10,20,40,80 μmol · L-1).The proliferation,migration and invasion of A549 cells were measured by real-time cellular analysis (RTCA).The expression levels of JNK,p-JNK,p38 and P-p38 were detected by real-time PCR and Western blotting.Results Curcumin showed an antiproliferation effect against A549 cells with IC50 =40 μmol · L-1,and curcumin exhibited obviously inhibitory effect on the migration and invasion of A549 cells.Additionally,compared with control group,curcumin suppressed the expression of JNK and p38 at the gene level,and significantly inhibited the expression of JNK,P-JNK,p38 and p38 (P<0.05) at the protein level.Conclusion These results demonstrated that curcumin can inhibit the proliferation,migration and invasion of A549 cells via reducing the level of JNK,p38 phosphorylation,and blocking JNK signal transduction pathway.
5.Influence of periodontal therapy on serum interleukin-6 and carotid metalloproteinases in animals with chronic periodontitis associated with atherosclerosis

Objective To investigate the effect of periodontal mechanical treatment on serum interleukin-6 (IL-6) and carotid artery matrix metalloproteinase(MMP)-2 and MMP-9 expression in chronic periodontitis(CP) SD rats with atherosclerosis (As).Methods Forty-four six-week-old male SD rats were randomly divided into three groups:control group(group A),As group (group B),As + CP group (group C).According to different periodontal interventions,group C was randomly subdivided into four groups:natural process group (C 1),the periodontal mechanical treatment group (C2),the periodontal mechanical treatment + local drugs group(C3),and the periodontal mechanical treatment + local and system drugs group(C4).Each group received the appropriate treatment and periodontal interventions.Serum IL-6 levels were determined by enzyme linked immunosorrbent assay (ELISA).MMP-2 and MMP-9 levels in the proximal aorta were examined by immunohistochemistry.Results The gray value of MMP-2 and MMP-9 was basically the same in all groups.Compared with group A,the gray value of MMP-2 and MMP-9 of group B and C were decreased.C1 group showed the formation of atherosclerotic plaque and fibrous cap.Compared with group B (126.4 ± 2.0,124.8 ± 2.8),the gray value of group C1 (101.3 ± 2.4,101.2 ± 4.1) was significantly weaker(P < 0.05).The staining depth of MMP-2 and MMP-9 of groups C1,C2,C3 and C4 were sequentially decreased,and the differences of gray value were statistically significant(P < 0.05).The levels of serum IL-6 in groups B and C1 increased gradually with time and became significantly higher than that of group A(P < 0.01).The levels of serum IL-6 in groups C2,C3,and C4 increased gradually and reached the peak 5 weeks after the establishment of model (P < 0.001).After that,the levels of serum IL-6 decreased gradually and was lower than baseline.The levels of serum IL-6 in groups C3 and C4 were significantly lower than that in group C2 7 weeks after the establishment of model(P < 0.01).Conclusions In rats with periodontitis and cardiovascular diseases,chronic periodontal inflammation may significantly increase the severity of As and promote the formation of atherosclerotic plaque.Mechanical periodontal therapy may cause short-term systemic inflammation and then reduce vascular inflammation in long term.With supplement use of local and systemic antibiotics,the mechanical periodontal therapy may get the vascular disease and systemic inflammation improved.
6.Effect of oral intervention on matrix metalloproteinase-2,9 expression in carotid arteries and serum interleukin-6 in rats with chronic periodontitis

Objective To establish chronic periodontitis model in SD rats,and to investigate the effect of oral intervention on atherosclerosis.Methods Fifty male SD rats were randomly divided into three groups,group A(normal control),group B(atherosclerosis,As) and group C(chronic periodontitis,CP).Group C was further divided into group C1(natural process),group C2(simple mechanical treatment),group C3 (systemic antibiotics),group C4-1(teeth extraction) and group C4-2(teeth extraction+systemic antibiotics),each group consisted of 7 rats.Every group received oral intervention.Serum interleukin(IL)-6 levels were detected in five different time points(1,3,5,7,9 weeks after a successful modeling) by enzyme linked immunosorbent assay.All animals were killed after 24 weeks.Matrix metalloproteinase(MMP)-2,9 in the proximal aorta was detected by immuno histochemistry.Results The levels of serum IL-6 in groups B and C1 increased gradually with time and became significantly higher than that in group A(P<0.01).Levels of serum IL-6 were increased gradually in each intervention group(C2,C3,C4-1,C4-2) and reached its peak at 5 weeks after modeling[C2:(62.3 ± 14.3) ng/L,C3:(58.2±8.7) ng/L,C4-1:(127.0±29.9) ng/L,C4-2:(120.6± 23.1) ng/L].Compared with group B,group C4-1 and C4-2 increased most significantly(P<0.01).Levels of serum IL-6 decreased gradually.Eventually,group C2[(28.6± 8.1) ng/L],C3[(40.8 ± 15.1) ng/L] and C4-2 [(32.7± 11.1) ng/L] were significantly lower than group B(P<0.05),and in group C2 IL-6 was the lowest.Although levels of serum of IL-6 significantly decreased in group C4-1[(72.8± 16.4) ng/L],but remained the highest.Immunohistochemistry showed that MMP-2,9 were expressed in group B,C 1 and C4-1,and significantly higher than in group A(183.0±2.0,181.3 ± 2.0),the gray value differences were statistically significant(P<0.01).Group C4-1 (123.1 ±2.9,121.0± 3.2) was the strongest,group B (126.4±2.0,124.8 ±2.8)and C1 (140.0±2.2,139.7±3.2) were decreased (P<0.01).While group C2(169.3±3.4,169.7±2.3),C3 (149.0±1.7,145.1±2.5) and C4-2(157.7± 1.2,155.8±2.7) were significantly lower than group C1(P<0.01),and group C2 was close to normal.Conclusions Periodontitis could increase the risk of atherosclerosis in rats with chronic periodontitis.Periodontal mechanical treatment and teeth extraction may increase the risk of As in the short time.However,the risk would gradually reduce in a long time.
7.Improving Effect of Curcumin Analogue H8 on Glucose and Lipid Metabolism Disorder in db/db Mice

Objective To investigate the influence of curcumin and its analogue H8 on glucose and lipid metabolism disorder in db/db mice. Methods The type 2 diabetes mouse model (db/db mice) was intragastrically administrated with curcumin and analogue H8 for 8 weeks.The blood biochemical indexes were measured.The expression of PEPCK and G6Pase mRNA was detected by real-time PCR in liver tissues.The expression of PEPCK and G6Pase protein was detected by Western blotting. Results Curcumin analogue H8 reduced blood glucose and lipids in db/db mice (P<0.01) and improved liver function related enzymes significantly.The levels of PEPCK and G6Pase mRNA in db/db mice were significantly decreased (P<0.01) and the expression levels of PEPCK and G6Pase protein were significantly decreased(P<0.01). Conclusion Curcumin analogue H8 improves the glucose and lipid metabolism disorder in db/db mice,and it is related to inhibiting the expression of PEPCK and G6Pase gene and protein.
8.Clinical Study on Si-Zi Powder for Hot Compress To Improve Living Quality of Hemodialysis Patients

This study was aimed to observe the clinical effect and quality of life (QOL) of Si-Zi (SZ) powder for hot compress to improve gastrointestinal functions of hemodialysis patients.A total of 60 hemodialysis patients were randomly divided into the treatment group and the control group.Gastrointestinal Symptom Rating Scale (GSRS) and QOL of both groups before and after the treatment of SZ powder were compared continuously.The results showed that after intervention,the total relieve rate was 96.7% in treatment group,and 53.3% in the control group.The effect of SZ powder group was to obviously improve the gastrointestinal symptoms and QOL (P < 0.05).It was concluded that SZ powder for hot compress can improve gastrointestinal functions and increase QOL of hemodialysis patients.
9.Construction of nursing quality sensitive indicator system for ovarian neoplasms patients

Objective:To construct a scientific and practical ovarian tumor nursing quality sensitive indicator system so as to provide a reference for evaluating the quality of nursing care for patients with ovarian tumors.Methods:Based on Donabedian's three-dimensional theoretical model of structure-process-outcome quality management, we used literature review and Delphi expert correspondence consultation to construct a nursing quality sensitive indicator system for ovarian tumor patients. From February to April 2019, we selected 20 experts from 16 ClassⅢ Grade A general hospitals and 2 higher nursing institutions from 7 provinces/municipalities in Shandong Province, Jiangsu Province, Beijing, Jilin Province, Shanghai, Guangdong Province and Sichuan Province for consultation.Results:Among two rounds of consultation, valid recovery rates were 90.00% and 94.44% respectively; authority coefficients were all 0.92; familiarity coefficients were 0.89 and 0.91 respectively; judgment coefficients were 0.94 and 0.92 respectively; Kendall harmony coefficients were 0.204 and 0.426 respectively; the differences were all statistically significant ( P<0.05) . The final nursing quality sensitive indicator system for ovarian tumor patients included 3 first-level indicators, 12 second-level indicators and 23 third-level indicators. Conclusions:The nursing quality sensitive indicator system for ovarian tumor patients is highly scientific and practical which can be used to standardize clinical nursing care for patients with ovarian tumors by gynecological nurses and improve the nursing quality.
10.Current situation analysis and quality evaluation of ERAS guidelines/consensus in obstetrics and gynecology surgery

Objective To analyze the current status and quality of existing obstetrics and gynecology enhanced recovery after surgery(ERAS)guidelines/consensus,understand the evidence-based practices in this field,and provide a basis for clinical decision-making.Methods A systematic search of PubMed,CNKI,and other databases,as well as NGC,SIGN,and websites of anesthesia societies domestically and internationally was conducted to obtain relevant guidelines and consensus on ERAS for gynecological surgery.The search was conducted from inception to July 1,2023,and the AGREE Ⅱ and RIGHT tools were used to analyze the current status and quality of the included guidelines/consensus,as well as to conduct a summary of the recommendations.Results A total of 5 guidelines and 10 consensus statements were included.The publication years were from 2018 to 2023,originating from the United States,China,Canada,and the United Kingdom.In the 6 areas evaluated using AGREEⅡ,clarity(66.06%)scored highest,followed by scope and purpose(56.67%),involvement of stakeholders(42.02%),and editorial independence(37.64%)with scores above 30%.The scores for rigor of development(26.88%)and applicability(4.17%)were relatively low.The same items among different evaluators indicated good consistency.The results of the scores in each area were as follows:A-level 2,B-level 13,and no C-level.Two high-quality guidelines were selected.The average RIGHT reporting rate was 44.38%.Subgroup analysis showed that guidelines were higher than consensus statements in the areas of rigor of development and applicability(P<0.05),and that using GRADE was higher than not using(P<0.05).The areas of rigor of development and clarity were higher in foreign guidelines than domestic guidelines(P<0.05),and using evidence grading tools was higher than not using(P<0.05).The summary of the recommendations showed that the quality of the evidence supporting the recommendations was uneven,and some recommendations were not specific enough.Conclusion The quality of the 15 included ERAS guidelines/consensus in obstetrics and gynecology is uneven,and 2 of them can be used as high-quality guidelines for clinical reference.The quality of evidence-based guidelines in foreign countries is higher,and the use rate of GRADE in foreign guidelines is high,most of which can provide clear recommendation strength and evidence level.Chinese guidelines/consensus are of lower quality compared to overseas,and methodology and report quality need further improvement.Overall,it is still necessary to strengthen the formulation of methods and reporting standards,improve the quality of evidence,and refine specific recommendations.
