2.Optimization of Freeze-drying Technology of Albendazole Nanosuspension

OBJECTIVE:To optimize freeze-drying technology of albendazole nanosuspension so as to prepare albendazole nanometer powder. METHODS:By adopting freeze-drying method,with particle size and Zeta potential as the indexes,single fac-tor test and verification were made on pre-freezing temperature and the type,ratio and mass fraction of cryoprotectants,and then the albendazole nanosuspension prepared by liquid phase precipitation method was made into albendazole nanometer powder. RE-SULTS:When the pre-freezing temperature was-20℃and the cryoprotectant was 4%glucose-mannitol(3∶7),the average parti-cle size of the prepared nanometer powder was (208.03 ± 2.13) nm,and average Zeta potential was (-15.53 ± 0.18) mV. CON-CLUSIONS:Albendazole nanometer powder with better particle size and potential can be prepared by freeze-drying technology.
3.Analysis of relationship between hyperuricemia and ischemic stroke in elderly patients with hypertension

Objective To investigate the relationship between blood uric acid and ischemic stroke in the elderly with hypertension.Methods Totally 100 elderly patients with hypertension were divided into study group (blood uric acid≥417 μmol/L,n=49) and control group (blood uric acid<417 μmol/L,n=51).The relationship between the level of uric acid and ischemic stroke were analyzed in two groups.Results There were not significant differences in age,body mass index (BMI),smoking and drinking,diabetes(P>0.05)between the groups,but there were differences in uric acid levels,smoking history,and stroke incidence at admission into hospital(P<0.05).In study group,uric acid and stroke severity score were significantly related in different times(P<0.05).After adjusting the related risk factors,logistic multiple regression analysis showed that the odd ratio of ischemic stroke was greater in study group than in control group (β=1.059,OR =2.884,P=0.03).
4.Research on Optical Parameter along Puncture Path in Spinal Surgery Navigation Based on Near Infrared Spectroscopy.

Accurate placement of pedicle screws is a key factor of spinal surgery. Investigation of a new real-time intra-operative monitoring method is an important area of clinical application research which makes a contribution to planting pedicle screw accurately. Porcine spines were chosen as experimental objects. The changes of reduced scattering coefficient (μ'(s)) along normal puncture path, medial perforation path and lateral perforation path were measured and studied. A conclusion is drawn that there are two distinct peaks throughout the puncture process, appearing at the junction of cancellous bone and cortical bone, at the beginning and at the end, respectively. The reduced scattering coefficient is proved to be a good monitoring factor which can identify whether the screw is about to reach the critical position of the spine puncture. Moreover, the variation provides an important reference for spinal surgical navigation process.

5.The role of HPV type 16 E6 variant in persistent infection and recurrence of cervical HSILs after conization

Objective To explore the role of humall papillomavims (HPV) type 16 E6 variants in the persistent infection and recurrence of the cervical high-grade squamous intraepithelial lesion (HSIL) after conization. Motheds During May to June 2012, 100 HSIL patients with HPV16 positive but negative at the margin after Loop electrosurgical excision procedure (LEEP) were selected from Shanghai People′s Hospital of Pudong District. The gene sequencing was done to the E6. The patients were followed up for 2 years. The E6 gene sequencing was done again to detect the HPV16 variants among those with persistent infections and recurrence. Results Among the 100 cases, E6 variants were detected, 81% of them HPV E6 variants Asian prototype, 14% European variant; 2% European prototype (EP) and 3% African variant 1. LEEP for Asian prototype was susceptible to the clearance of HPV16 (P < 0.001). The European variants of T350G, and A442C were correlated with persistent infection and recurrence (P < 0.001). Conclusion The HPV16 variant is closely related to the HSIL incidence. HSIL caused by HPV16 in Pudong New Area of Shanghai belong to the Asian type variant. The European variants of E6 , T350G/A442C were likely related to the recurrence of HPV16.
6.Etiological characteristics and molecular typing of food poisoning incidents by Vibrio parahaemolyticus in Fuqing City, Fujian Province from 2021 to 2023

Abstract: Objective To analyze the etiological characteristics of Vibrio parahaemolyticus (VP) isolated from food-borne outbreaks in Fuqing City between 2021 and 2023, and to explore its distribution characteristics and epidemic types. Methods The serotypes, virulence genes (tlh, tdh, trh), and drug resistance profiles of 23 VP strains isolated from 4 food-borne disease outbreaks were tested by agglutination, multiple real-time PCR, and broth microdilution methods. Pulsed-field gel electrophoresis (PFGE) was used for molecular typing to analyze homology. Results From 2021 to 2023, four food-borne disease outbreaks occurred in Fuqing City, leading to the isolation and identification of 23 VP strains. Among these, 19 clinical strains were divided into two serotypes: O10:K4 type, comprising 13 strains (68.4%), and O4:KUT type, comprising 6 strains (31.6%); the tdh gene carrying rate was 100.0%, and no trh gene was detected. The serotypes of the four food isolates were O3∶K4, O10:K24, O11∶KUT, and O3∶K37, respectively, all differing from the clinical strains, with two strains coming from the same food item without tdh and trh genes detected. The resistance rate of all isolates to cefazolin was the highest (87.0%), with two strains (8.7%) from different sources showing multidrug resistance. The PFGE genotypes of 19 case isolates were clustered and correlated with their serotypes and drug resistance profiles; strains isolated from the same outbreak event showed high similarity, whereas food strains exhibited polymorphism in PFGE genotypes. Conclusions The predominant serotype causing the four outbreaks was O10:K4, with some strains showing multidrug resistance; all clinical strains carried the tdh gene. The high correlation between strains from the same outbreak event suggests they are homologous. Special attention should be paid to the risk of outbreaks and pandemics caused by the O10:K4 type strain again. Additionally, food safety risk monitoring and assessment should be strengthened to provide an effective basis for epidemic prevention and control and early identification and warning.
