1.Extraction and Determination of Polysaccharide from Discarded Fi brous Roots of Radix Panacis Quingueforlii

Objective:To establish a feasible method for extracti ng polysaccharide from the discarded fibrous roots of Radix Panacis Quingueforli i afterpanaquilon had been extracted, and determine the polysaccharide content. Methods:Enzymolysis technique and alcohol was applied for decolorization, and ph enol-sulfuric-acid method was used to determine the active polysaccharide conte nt.The content of trace element and heavy mental was measured by element-analyze r and atomic fluorescence photometer respectively. Results: The yield of polysac charide from the fibrous roots was close (about 11.7%) to the main root.And the content of heavy metal can match the national standard.Conclusion:I t is valuable to extract the polysaccharide from the discarded fibrous root of R adix Panacis Quingueforlii.

3.Effect of pulse electro-magnetic fields on the repair of osteo chondral fracture in rabbits

Objective:To study the effects of pulse electro-magnetic fields (PEMFs) on the healing of tibial condylar osteochondral in rabbits.Methods:Sixteen rabbits were divided into two groups,and osteochondral fractures of left tibia condyle were created,internal fixation was performed with steel wire.Group 1 was constantly treated with PEMFs six hours per day;group 2 was not treated.The results were studied by gross morphology,light microscopy and elect ro-microscopy.Results:75% and 13% fractures were repaired by hyalin-like cartilage in group 1 and in group 2 respectively.Conclusion:PEMFs are essential to facilitation of the cartilage repair.
4.Multi-groups controled study on the association of Helicobacter pylori infection with gastric cancer and stomach diseases

Objective: This multi-group case control study was performed to determine whether Helicobacter pylori infection was associated with gastric cancer and stomach diseases. Methods: Gastric cancer and stomach disease patients that were diagnosed pathologically between 1994 and 1996 years were divided into several case groups and control group, all histological specimens collected in gastric cancer high risk region of Shandong Province were used to measure exposure history. Results: After variables concerning age, sex and education were adjusted, odds ratios of chronic superficial gastritis in antrum, peptic ulcer disease and active degree of inflammation associated with H. pylori infection remained significant (OR was 2.072, 2.980, 2.086 respectively). Correlation analyses showed negative relationship between degree of stomach diseases and H. pylori infection(r=-0.217). Conclusion: The results support the hypothesis of a relationship between H. pylori infection and the development of chronic superficial gastritis in antrum, peptic ulcer disease and active degree of inflammation.
5.Significance and expression of E-cadherin and focal adhesion kinase in rectal cancer

Objective To investigate the significance and expression of E-cadherin(E-CD),focal adhesion kinase(FAK)and FAK py397 in rectal cancer.Methods Thirty paraffin specimens of rectal cancer and adjacent tissues collected from 2001 to 2002 and 35 fresh specimens of rectal cancer and adjacent tissues resected from 2006 to 2007 at People's Hospital of Wuhan University were adopted in this study.The expression of E-CD,FAK and FAK py397 was detected by immunohistochemistry and Western blot.All data were analyzed using chi-square test.Results E-CD was positively expressed in the membranes of epithelial cells in normal rectal mucosa,while no E-CD expression was found in the membranes of epithelial cells in rectal cancer tissues.There was a significant correlation between the expression of E-CD,FAK,FAK py397 and tumor difierentiation,depth of invasion and lymph node metastasis(χ~2=7.099,18.358,25.612;12.316,28.823,23.168;8.927,18.122,22.620,P<0.05),while no relationship between the expression of E-CD,FAK,FAK py397 and age,sex was detected(χ~2=0.439,1.899,3.676;0.541,4.051,1.135,P>0.05).The expression levels of FAK and FAK py397 were 83% (54/65)and 68% (44/65)in rectal cancer tissues,and were 31%(20/65)and 26% (17/65)in adjacent tissues,with significant difference(χ~2=33.707,34.163,20.897,P<0.05).Conclusion Lost expression of E.CD in the membrane of rectal cancer tissue and elevated expression of FAK and FAK py397 in the rectal cancer tissue might be related to the invasion and metastasis of rectal cancer,which may predict the biological behavior of rectal cancer.
7.Effecting of inhibiting ubiquitin-proteasome pathway on proliferation and apoptosis of gastric carcinoma cells

Objective To investigate the effects of inhibiting ubiquitin proteasome pathway(UPP) on proliferation of gastric carcinoma cells and the possible mechanism was discussed. Methods The gastric carcinoma cell strain SGC 7901 was treated with MG 132 to inhibit its UPP specially. The effect of growth suppression on cells was evaluated with MTT assay. Cell cycle and apoptosis were detected by flow cytometry(FCM). DNA fragment analysis was used for confirming the presence of apoptosis. The activity of telomerase was examined by TRAP PCR ELISA. Expression of p27kip1 was detected by immunocytochemical technique. Results MG 132 had great inhibitory effect on the growth of SGC 7901 cells. 8.Study on expression of SENP1 and c-myb gene in patients with acute lymphoblastic leukemia

Objective To investigate the SENP1 and c-myb gene expression and their correlations in bone marrow specimens in the patients with acute lymphoblastic leukemia (ALL ) to provide the basis for expounding the role ,mechanism and prognosis of SENP1 and c-myb in ALL .Methods 31 patients diagnosed with ALL (22 cases of B-ALL ,1 case of T-ALL and 8 cases of uncate-gorized ALL ;6 cases in the low/medium risk group ,25 cases in the high risk group) and 31 patients with proliferative bone marrow and hyperplastic anemia diagnosed by the morphology were taken as the control group .The real-time PCR and immunocytochemical staining(SP method) were adopted to detect the mRNA and protein expressions of SENP1 and c-myb in the bone marrow specimens of the ALL patients and the control group .Results The expression of SENP1 and c-myb were both increased in the bone marrow specimens and smears of ALL patients ,which showed the statistical difference compared with the control group (P< 0 .05) ,the Pearson correlation analysis found that the high expression of SENP1 and c-myb had correlation .The expression of SENP1and c-myb in the low/medium risk group were lower than that in the high risk group ,but the difference had no statistical significance . Conclusion The high expression of SENP1 and c-myb exists in the bone marrow specimens of the ALL patients ,SENP1 and c-myb could possibly have the correlation with the occurrence and development of ALL ;but now the differences of SENP1 and c-myb ex-pression among different risk groups of ALL patients are yet to be proven .
9.Effects of combination of octreotide and wortmannin on proliferation of gastric carcinoma cell BGC-823

Objective To investigate the effect and mechanism of octreotide with or without wortmannin on the growth of gastric carcinoma cell BGC-823.Methods From June to August of 2007,in Remin Hospital of Wuhan University,cells were exposed to octreotide for four dilution(10-5、10-4、10-3、10-2g/L)with or without wortmannin(a special inhibitor of PI3'K/Akt pathway,the dilution is 40 nmol/L).And then the cytotoxicity was assessed by determining cell survival with MTT after 24 hours,with also were exposed to octreotide for the dilution of 10-3 g/L with or without Wortmannin in 12 h,24 h,36 h,48 h,then the cell survivral rate was accounted and the results were formulated in a table,and the cell cycle was checked-out with Flow Cytometer(FCM);the expression level of p27 gene and protein was determinated with RT-PCR and Western-bloting Test.Results The five groups within one reagent treating were compared with control group,and the results had difference,but the combination of two reagents group had significant difference(P
