1.Survival analysis of patients with high risk refractory lymphoma treated by autologous hematopoietic stem cell transplantation

Objective To investigate the prognosis and survival situation of the patients with high risk refractory lymphoma treated by autologous hematopoietic stem cell transplantation .Methods The follow up data in 110 patients with high risk refractory lymphoma treated by autologous hematopoietic stem cell transplantation in our hospital were retrospectively analyzed .The survival and prognosis factors of the patients after autologous hematopoietic stem cell transplantation were analyzed by using the Kaplan-Meier survival analysis and Cox proportional hazard regression analysis .Results The median survival time was 39 .4 months in 110 cases ,the 3‐year overall survival rate (OS) and progression free survival rate (PFS) were 80 .9% and 76 .4% respectively .The pa‐tients achieved CR status before transplantation(P=0 .016) and consolidation therapy after transplantation (P=0 .006) were the favorable prognostic factors of the patients undergoing transplantation .The prognosis in the patients with high LDH values ,IPI score>2 and bone marrow infiltration and HBV infection were poor(P<0 .05) .Conclusion Autologous hematopoietic stem cell transplantation can improve the long‐term survival rate in the patients with high risk refractory lymphoma .
2.Peroxisome proliferator-actived receptor-? ligand troglitazone induces apoptosis in renal cell carcinoma

Objective: To investigate the expression of peroxisome proliferator actived receptor ? (PPAR ?)and the inducement of apoptosis by PPAR ? ligand in renal cell carcinoma(RCC) derived cell lines.Methods:RT-PCR and Western blot analysis were performed to determined the expression of PPAR ? mRNA and protein in two RCC derived cell lines(786 O and A498) and two normal kidney(NK) derived cell lines(HK 2 and HMCC). Two RCC cell lines were treated with 50 ?mol/L troglitazoned for and evaluated for the effects of antidiabetic thiazolidinediones (TZDs) on the cells apoptosis by fluorescence microscopy and DNA ladder assay.The mutative expressions of Bcl 2 and Bax before and after TZDs treatment were also performed by western blot analysis. Results: The expression of PPAR ? was observed to be stronger in 786 O and A498 cells than in HK 2 and HMCC cells by RT-PCR and Western blot analysis. Treated with 50 ?mol/L troglitazone (for 48 h) it induced typical apoatosis in 786 O and A498 cells. After treatment, a decrease in Bcl 2 expression in RCC cells was observed by Western blot analysis,and the expression of Bax,however,was up regulated.Conclusion: The results reveal that troglitazone has the tumor suppressive effect on RCC cells. High affinity PPAR ? ligands (TZDs) may be the candidates for a novel approach to the treatment of this refractory neoplasm.
3.Anesthetic efficacy comparison of fentanyl and sufentanil in vocal cord tumors excision in elderly patients

Objective To compare the anesthetic effect of fentanyl and sufentanil in vocal cord tumors excision in elderly patients.Methods Sixty elderly patients from the First Hospital of Shanxi Medical University scheduled for elective excision of vocal cord tumors were randomly divided into fentanyl group (group F) and sufentanil group(group SF) according to the random number table method, and each group had 30 cases. The dose was determined by its efficiency ratio of analgesic (10:1) followed by anesthesia induction with compound etomidate and cisatracurium. Besides, injection remifentanil and propofol with pump was used for anesthesia maintenance during the operation. Haemodynamics indicators of group F and group SF were recorded at different time point, including before (T0), after induction of anesthesia (T1), immediately after intubation (T2), during the resection of tumor (T3), before extubation (T4) and after extubation (T5). Time to resuscitation, time to extubation, the cooperation at extubation, the sedation scores of 10 minutes after extubation (according to Ramsay sedation level score), and the complications of anesthesia were recorded respectively. Results In the group SF, T0pressure: (135±6) mmHg / (82±6) mmHg (1 mmHg = 0.133 kPa), heart rate:(80±7)/min;in the group F,T0pressure:(129±7)mmHg/(79±6)mmHg,heart rate:(80±7)/min.In the group SF,T1pressure:(102±5)mmHg/(61±4)mmHg,heart rate:(64±4)/min;in the group F,T1pressure:(100±6) mmHg / (61±4) mmHg, heart rate: (61±3) /min. T3heart rate: (89±5) /min (group SF), (92±5) /min (group F). Compared with T0, the pressure and heart rate of T1were decreased in both groups, and the heart rate of T3was increased (P < 0.05). The time to resuscitation and time to extubation in the group F was shorter than that in the group SF[(5.5±1.8)min vs.(10.4±3.1)min,(8.6±2.4)min vs.(15.4±2.6)min,both P<0.05].Ramsay score in the group F was lower than that in the group SF[(2.4±0.3) scores vs. (3.2±0.5) scores, P<0.05]. There were no significant differences in the cooperation at extubation, incidence rate of vomiting and nausea between the two groups (all P > 0.05). Intraoperative awareness and postoperative irritability were not found in both groups.Conclusions The equivalent dose of fentanyl and sufentanil induction are safe and effective for elderly patients with excision of vocal cord tumors. The patients can get more rapid revival and higher revival quality after fentanyl induction.
4. Brain derived neurotrophic factor enhances the role of mesenchymal stem cells in inhibiting follicular helper T cells

Objective:
To investigate the effect of brain derived neurotrophic factor (BDNF) on mesenchymal stem cells (MSC) inhibiting follicular helper T cells (Tfh cells).
Methods:
The contents of indoleamine 2,3-dioxygenase (IDO), IL-10, TGF-β and IL-21 in MSC culture supernatant were detected by ELISA; The peripheral blood of healthy volunteers were collected, and lymphocyte in peripheral blood was separated by human lymphocyte separation solution; Co-cultures of MSC and lymphocyte were performed by Transwell chamber, and the proportion of CD4+CXCR5+ Tfh cells and their subtypes were detected by flow cytometry.
Results:

5.Expression of P57(kip2) and cyslinE proteins in human pancreatic cancer.

OBJECTIVETo investigate the effects of p57(kip2) and cyclinE proteins on the genesis and progression of human pancreatic cancer.
METHODSThe expression of p57(kip2) and cyclinE proteins in tumor tissues and adjacent tissues of pancreatic cancer in 32 patients was detected by SP immunohistochemical technique.
RESULTSThe p57(kip2) protein positive-expression rate in tumor tissues of pancreatic cancer was 46.9%, which was lower than that in adjacent pancreatic tissue (P < 0.05). The p57(kip2) protein positive-expression correlated significantly with tumor cell differentiation (P < 0.05) and did not correlate significantly with lymph node metastasis (P > 0.05). The cyclinE positive-expression rate in tumor tissues was 68.8%, which was higher than that in adjacent pancreatic tissues (P < 0.05). The cyclinE positive-expression also correlated significantly with tumor cell differentiation and lymph node metastasis (P < 0.05). The cyclinE protein positive-expression rate in the tumor tissues of the p57(kip2) protein positive-expression group was lower than that in the p57(kip2) protein negative-expression group, and there were no significant correlation between the two groups (r = -0.112, P > 0.05).
CONCLUSIONDecreased expression of the p57(kip2) protein and/or over-expression of the cyclinE protein may play an important role in the genesis and progression of human pancreatic cancer.

6.Effect of Dendrobii Caulis Mixture on Cell Inflammatory Response and Apoptosis in Diabetic Rats with Nonalcoholic Fatty Liver Disease Based on NF-κB/NLRP3/IL-1β Signaling Pathway

ObjectiveTo investigate the effect of Dendrobii Caulis mixture on cell inflammatory response and apoptosis in diabetic rat with non-alcoholic fatty liver disease(NAFLD) and its possible mechanism. MethodForty male SD rats were randomly divided into blank group and model group of type 2 diabetes mellitus(T2DM) with NAFLD according to body weight. The model was established by high-sugar and high-fat diet combined with intraperitoneal injection of streptozotocin(STZ), which was randomly divided into the model group, Dendrobii Caulis mixture group(16.67 g·kg-1·d-1) and metformin group(100 mg·kg-1·d-1) according to blood glucose and body weight, and 10 rats in each group. Rats in each group were administered by continuous gavage for 4 weeks, the blank and model groups were given saline by gavage at 10 mL·kg-1·d-1. Fasting blood glucose(FBG), serum insulin(INS), glycosylated serum protein(GSP), triglyceride(TG), total cholesterol(TC), high density lipoprotein(HDL-C), low density lipoprotein(LDL-C), alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were detected in each group of rats. The liver tissues of rats in each group were stained with hematoxylin-eosin(HE) to observe the pathological changes, and the positive expressions of nuclear transcription factor(NF)-κB, NOD-like receptor heat protein structural domain-related protein 3(NLRP3), interleukin(IL)-1β and tumor necrosis factor(TNF)-α were observed by immunohistochemistry. Western blot and fluorescence quantitative polymerase chain reaction(Real-time PCR) were used to detect the protein and mRNA expression of B-cell lymphoma-2(Bcl-2), Bcl-2 associated X protein(Bax), Caspase-3 and NF-κB p65, NLRP3, IL-1β, TNF-α in liver tissue of rats in each group. ResultCompared with the blank group, FBG, GSP, TC, TG, LDL-C, AST and ALT levels were increased, INS and HDL-C levels were decreased, Bax, Caspase-3, NLRP3, IL-1β, TNF-α protein and mRNA expression were increased in model group, the ratio of p-NF-κB/NF-κB protein increased, the expression of Bcl-2 protein and mRNA decreased, and the positive immunohistochemical expression of NF-κB, NLRP3, IL-1β and TNF-α increased, and the differences were statistically significant(P<0.01). The morphological structure of the liver was disrupted, and obvious fat vacuoles were seen. Compared with the model group, FBG, GSP, TC, TG, LDL-C, AST and ALT levels of Dendrobii Caulis mixture group and metformin group were decreased, INS and HDL-C levels were increased, and protein and mRNA expressions of Bax, Caspase-3, NLRP3, IL-1β and TNF-α were decreased, the protein ratio of p-NF-κB/NF-κB decreased, the expression of Bcl-2 protein and mRNA increased, and the positive immunohistochemical expressions of NF-κB, NLRP3, IL-1β and TNF-α decreased, and the differences were statistically significant(P<0.01). The liver morphology and structure were relatively complete, and the fat vacuoles were reduced. ConclusionDendrobii Caulis mixture can inhibit cell apoptosis, reduce inflammatory response and alleviate liver injury in rats with T2DM and NAFLD, the mechanism may be related to inhibiting the activation of NF-κB pathway, blocking the activation of NLRP3 inflammatory vesicles, reducing IL-1β secretion, attenuating Caspase-3 activity and reducing the ratio of Bcl-2/Bax.
7.Study on the correlation between color and inner quality during the processing of Prunus mume carbon and its processing end point

OBJECTIVE To study the correlation between color and inner quality during the processing of Prunus mume carbon, and provide reference for the determination of processing end point of P. mume carbon. METHODS The chromaticity value of P. mume carbon powder was measured by colorimeter, and the inner quality of P. mume carbon was measured by selecting the contents of water, water-soluble extract, citric acid and tannin. The dynamic change trend of the chromaticity value, water, water- soluble extract, the contents of citric acid and tannin in P. mume carbon under different processing time was analyzed. The correlation between color and the above indexe contents was analyzed, and the regression equation of inner quality-chromaticity value was established. Combined with principal component analysis (PCA), hierarchical cluster analysis (CA) and partial least squares discriminant analysis (PLS-DA), the difference of P. mume carbon at different processing times was analyzed to determine the processing end point. RESULTS With the extension of processing time, the sample color gradually deepened; the chromaticity values L* and E* of the samples increased at first and then decreased, the chromaticity values a* and b* decreased, and finally all tended to be stable. The content of water-soluble extract, citric acid and tannin in the sample increased at first and then decreased, the water content of the sample decreased with time and finally stabilized. Correlation analysis showed that water, water-soluble extract, citric acid and tannin were positively correlated with L*, a*, b* and E*(P<0.001). PCA and HCA showed that P. mume carbon under different processing time could be clustered into two categories: the processed samples of 0-30 min and those of 40-60 min. PLS-DA showed that water and water-soluble extract were important quality indexes and b* was an important chrominance index in the processing of P. mume carbon. The chromaticity value of the samples processed for 50 min and 60 min were not significantly different. The contents of water, water- soluble extract, citric acid and tannin in the samples processed for 60 min were less than those processed for 50 min. CONCLUSIONS There is a certain correlation between the color and the inner quality of P. mume carbon. The processing time of P. mume carbon should be 40-50 min.
