1.Study on the effects of estrogen receptor alpha gene polymorphism on the efficacy of ursodeoxycholic acid in primary biliary cirrhosis treatment

Objective To investigate the effects of estrogen receptor alpha (Erα) gene polymorphism on the efficacy of ursodeoxycholic acid (UDCA) in female primary biliary cirrhosis (PBC) treatment. Methods The polymorphism of Xba Ⅰ and Pvu Ⅱ sites in No. 1 Intron of Erαgene was detected by polymerase chain reaction-restriction fragment length polymorphism in 65 female patients with PBC. The patients were treated with UDCA tablets daily (13-15 mg·kg-1 · d- 1,divided for 3 times oral taking). The variations of patients' serum total bilirubin (Tbil), alanine aminotransferase (ALT), alkaline phosphatase (ALP), γ-glutamyle transpeptidase (GGT) were followed up totally for 24 months. Results Totally the complete follow-up data had been obtained in 60 patients. Four indicators of XX or PP type patients slowly declined till the end of follow-up point still had not reached the effective treatment standard. Four indicators of Xx type patients were also slowly declined, while the effective treatment standard was achieved till the end of follow-up. Four indicators of Pp/pp or xx type patients were rapid declined to the effective treatment standard. The effective rate of UDCA in PBC treatment was 75.0% (45/60). The distribution difference of Pvu Ⅱ between the UDCA treating effective group and the ineffective group in PP, Pp, pp types patients and XX、Xx、xx types patients was statistic significant (x2 = 12.13、 P = 0.003 and x2 = 9.95 、 P = 0.007 respectively). The distribution difference of Xba Ⅰ between the effective group and the ineffective group with UDCA treatment in XX, Xx and xx types patients was also significant (P <0.05). The effective rate of Pp, pp, Xx, xx types patients [82.61% (19/23)、80.65% (25/31) 、9/14、83. 33% (35/42) respectively] was higher than PP and XX types patients (1/6 and 1/4 respectively).Conclusion Erα gene polymorphism influenced the efficacy of ursodeoxycholic acid in treating primary biliary cirrhosis.

Logistic regression analysis was applied to analyze the influence of six exogenous pathogenic factors and the changes of serum level of IgE, eosinophil (EOS) counting, contents of third component of complement (C3) and C-reactive protein (CRP) were observed. 3.Endovascular embolization treatment of intracranial aneurysms with detachable coils in the cerebral vasospasm time

Objective To evaluate methods and effect of the endovascular embolization of intracranial aneurysms. Methods From 2004,3 to 2004,8,14 patients with intracranial aneurysms, underwent intravascular embolization was reviewed. Clinic presentation: 6 cases males, 8 cases females, age range 45-78 years. 1 case ICA aneurysms, 8 cases PCA aneurysms, 5 cases ACA aneurysms . All patients were treated with DCS/MATRIX embolization. Results 11 cases aneurysms were 100% occluded, 2 cases aneurysms were 70-90% occluded,1 case aneurysm was 50% occluded; 6 patients found cerebral vasospasm.Conclusions In the Cerebral vasospasm time, endovascular embolization of intracranial aneurysms is a safe, minimal traumatic and effective method.
4.Briefly on Long-storaging Method for Chinese Materia Medica

In Qin Han periods,the long-storaging method for Chinese materia medica(CMM) has been used in traditional Chinese medicine.People have named the Prescription with "chen" in the Song Dynasty,such as "Er Chen Tang".With the traditional Chinese medicine storing and treating with different methods in appropriate period,medicine becomes old medicine from the new,there by,reaching property of a medicine of reducing the poisonous or over-strong property,strengthening the medicine function and processing effect,getting rid of stench taste,increasing fragrant purpose,so as to further consistent with the demands of clinical treatment.By now,this traditional and distinct processing technique is on the verge of extinct.In order to fully reflect the superiority of long-storaged medicine by therapy,it is imminent to conserve,investigate and develop such medicine and such method.We should reinforce to research the changing of its effective component and pharmacology effect before and after processing drugs,discuss the contribution of the long-storaging method on the quality of CMM,so as to make this method applying and developing further.
5.Changes of reactive astrocytosis after blockage of heat shock protein 70

Objective To study the changes of reactive astrocytosis after heat shock protein 70 (HSPT0) was blocked by anti-HSP70 antibody. Methods We established cell model of scratch inju-ries by in situ culture and prurification of rat astrocytcs. Anti-HSP70 antibody was added into the nutrient medium at once after injury for intervention (intervention group). Then, immunocytochemical staining of glial fibrillary acidic protein (GFAP) was done at different time points in control group and intervention group to observe astrocytosis and morphologic changes, mRNA expression of GFAP was observed by means of reverse transcriptase-polymerase chain reaction (RT-PCR). Results Compared with the con-trol group, average cell area, average dentritic length and number of dentrities of astrocytes were signifi-cantly reduced in the intervention group(P < 0.05 or P < 0.01), with down-regulated mRNA expression of GFAP (P < 0.05). Conclusion HSP70 plays a facilitative role in reactive astrocytosis after injury of astrocytes. Reactive astrocytosis can be controlled to some extent by blocking HSP70 with anti-HSP70 antibody.
6.Changes of Oscillatory Potentials of Electroretinogram after Methanol Intoxication in Rats

Objective To study the characters of oscillatory potentials (OPs) of electroretinogram (ERG) after methanol intoxication in rats. Method The SD rat models of methanol intoxication were established and divided into control group, 3-day intoxication group, 7-day intoxication group. The changes of OPs of ERG were recorded in a dark room. Results The total amplitudes of 3-day and 7-day intoxication groups decreased approximately 50% compared with that of the control group, while the schedule de-layed approximately 16% and 61%, respectively. Conclusion The characters of methanol intoxication in rats included delay in schedule and decline in the total amplitude of OPs.
7.Expression and Mechanism of αB-crystallin in Retina and Extraocular Tissues and Organs

αB-crystallin is the structural protein of vertebrate lens, w hich is w idely expressed in non-lens tissue. As one of the heat shock protein fam ily m em bers,αB-crystallin possesses biological proper-ties of m olecular chaperones and anti-apoptotic effects. Multi-factor injuries, such as retinopathy, inflam-m ation and nervous system diseases, have a closely relationship w ith αB-crystallin. T his paper review s the research progress of the expression and m echanism ofαB-crystallin in retina and extraocular tissues and organs.
8.Distribution of Formic Acid after Methanol Intoxication in Rats

Objective To investigate concentration and distribution in blood and tissues of form icacid after m ethanol intoxication in rats. Methods The Sprague-Dawley rats were divided into groups for con-trol group and 3-day and 7-day intoxication treatm ent groups. The experim ental groups were adm inis-tered m ethanol by gavage with the initial dose of 8 m L/kg and followed with 4 m L/kg supplem ental dose 24 h later. After 3 days and 7 days later, rats were killed by decapitation. Then sam ples of cardiac blood, liver, kidney, brain, heart and stom ach of each group were collected. Form icacid concentrations were detected by high perform ance liquid chrom atography. Results Form icacid concentrations in tissues were higher than in blood. Com pared with 3-day intoxication group, there was an increase form icacid of concentration in brain and stom ach in 7-day intoxication group, while a decrease in liver and kidney (P<0.05). Conclusion H igh perform ance liquid chrom atography could be used to accurately detect form icacid. As the m etabolite of m ethanol, form icacid accum ulates in rat blood and tissues after intoxication and the concentrations in organs and tissues are obviously higher than in blood.
9.Evaluation of Carotid Artery Structure and Function of the Brachial Artery in Children with Coarctation of Aorta Using High-frequency Ultrasound

Purpose To evaluate the carotid artery structure and diastolic function of the brachial artery in children with coarctation of aorta (CoA) using high-frequency ultrasound. Materials and Methods Twenty-four cases of children with CoA were selected, among them seven cases were simple CoA and 17 cases were combined with other cardiac malformations, and 20 cases of simple ventricular septal defect (VSD) children and 20 healthy children over the same period were selected as the control groups. Carotid artery intima-media thickness (IMT) and brachial artery flow-mediated vasodilation (FMD) in all subjects were evaluated using high-frequency ultrasound, vascular structure and functional changes were compared before CoA correction and VSD repair treatment. Results Carotid artery IMT of CoA group was (0.47±0.12) mm, which was significantly larger than the VSD group (0.41±0.10) mm and the control group (0.40±0.09) mm, and the difference was statistically significant (F=4.275, P<0.05); brachial artery FMD [(5.48±1.66)%] of CoA group was significantly lower than that of the VSD group [(6.69± 1.45)%] and the control group [(6.59±1.84)%], and the difference was also statistically significant (F=4.318, P<0.05), but there was no significant difference (q=0.400, 0.191;P>0.05) between the VSD group and the control group for carotid artery IMT and brachial artery FMD. Conclusion In children with CoA, besides the structure abnormalities of the narrowing vascular, there is also the combination of structural and fuctional changes such as carotid artery intima-media complex thickening and brachial artery flow-mediated dilation function decrease.
10.In vitro study on influence of residual methylene blue after virus inactivation plasma on immune function of human PBMC cells

Objective To study the influence of residual methylene blue after plasma viral inactivation on the human immune cell function by using the peripheral blood mononuclear cell(PBMC) .Methods PBMC were isolated by adopting the Ficoll‐Hypaque density gradient centrifugation method and co‐cultured for 72 h in presence of specific T cell stimulating factors(Anti‐CD3/28 and Anti‐CD28) ,with or without different concentration of methylene blue .The culture supernatant was collected and detected the cyto‐kines secretion situation by ELISA .After 66 h culture ,CCK‐8 dye was added and continueously cultured for 4-6 h ,the prolifera‐tion was determined at A450 .Results The high‐concentration doses of methylene blue (1 .25 ,2 .5 ,5 μmol/L groups) had signifi‐cantly inhibiting effect on the proliferation of PBMC stimulated by Anti‐CD3/28(P< 0 .01) ,its OD value was decreased from 0 .897 ± 0 .385 to 0 .632 ± 0 .334 ,0 .524 ± 0 .254 and 0 .445 ± 0 .287 respectively ,showing certain dose dependent effect .The high concentrations of methylene blue (1 .25 ,2 .5 ,5 μmol/L groups) could down‐regulate interleukin(IL)‐17a ,IL‐10 and interferon (IFN)‐γ secreted by anti‐CD28 induced PBMC ,moreover showing a dose dependent effect .1 .25 ,2 .5 ,5 μmol/L methylene blue af‐fected the IL‐17a level secreted by PBMC from (406 ± 57)pg/mL descending to (276 ± 38) ,(192 ± 31) ,(134 ± 24)pg/mL respec‐tively ;affected PBMC to secrete IL‐10 ,its level was reduced from (184 ± 15) pg/mL to (132 ± 13) ,(110 ± 12) ,(42 ± 8)pg/mL ;af‐fected PBMC to secrete IFN‐γ,its level was deduced from (4 512 ± 187)pg/mL to (2 876 ± 143) ,(2 234 ± 153) ,(1 988 ± 112)pg/mL respectively .Conclusion High concentrations of methylene blue (≥1 .25 μmol/L ) has the significant inhibiting effect on the proliferation and cytokine secretion functions of PBMC .In other words ,the residual methylene blue concentration in viral inactiva‐tion plasma (≤0 .33 μmol/L) has no obvious effect on the immune function of PBMC ,but whether this concentration of methylene blue having the effect on human pure T cell immune function needs to be further evaluated and studied .
