The epidemic tendency of dental caries of school students over the past 15 years in China was analyzed in order to provide bases for prevention of dental caries. Data sets of boys and girls at the age of 7, 9, 12 years (deciduous caries) and 7, 9, 12, 14, 17 years (permanent caries) were collected from the series of Chinese National Surveillance on Students' Constitution and Health (CNSSCH) between 1991 to 2005, a survey that covers 30 provinces of and autonomous regions of China, with Tibet Autonomous Region and Taiwan Province excluded owing to unavailability of related data. According to their socioeconomic status, three urban regions and three rural regions (province capital must be included) were selected in each province, where its socioeconomic status and sports exercise can represent the general level of the locality. All subjects were Han students. Prevalence of deciduous and permanent caries was measured. In 2005, the prevalence of deciduous caries at 7, 9, 12 years was 52.87%, 52.04% and 9.80% in urban boys, 59.37%, 58.68% and 12.18% in rural boys, and that of permanent caries in boys at 7, 9, 12, 14, and 17 years was 2.35%, 5.63%, 13.94%, 19.05% and 21.29% in urban boys, 3.42%, 7.26%, 13.35%, 16.16%, 18.55% in rural boys, while in girls at the same age groups, the prevalence of deciduous caries was 53.53%, 49.48%, 7.92% in urban girls, 59.95%, 56.13%, 8.85% in rural girls; the prevalence of permanent caries was 3.61%, 8.99%, 19.32%, 24.75%, 28.62% in urban girls, and 3.47%, 10.65%, 17.66%, 22.52%, 24.27% in rural girls. The epidemic manifested a gradient distribution in age groups, which was closely related to region of the study population. A dramatic decreasing trend was witnessed among all sex-age subgroups in the rural groups between 1990 and 2005, and such a trend was stronger in deciduous caries than in permanent caries, however, this kind of decreasing trend was not steady, because there was bounce during some year. Although the morbidity of dental caries in China is lower than the standard set by the World Health Organization, and the prevalence of dental caries decreased from 1991 to 2005, the prospect of epidemic dental caries in China is in no way optimistic. The total population with dental caries is still very large and the prevalence decreased unstably. Therefore, financial support on school oral health care should be increased, and a national school oral health service network should be set up.

OBJECTIVETo observe the effect of Zhenqing Recipe (ZQR) on non-alcoholic fatty liver (NAFL), and the expression of hepatic salt-inducible kinase 1 (SIK1) and sterol-regulatory element binding protein-ic (SREBP-lc) in type 2 diabetes rats.
METHODSA rat model of type 2 diabetes was established by high fat/sucrose diet combined with intraperitoneal injection of small dose streptozotocin (STZ) . Modeled rats were randomly divided into the model group, the ZQR group, and the metformin group, 8 in each group. Eight rats were recruited as a normal control group. ZQR at the daily dose of 12 g crude drugs/kg was administered to rats in the ZQR group by gastrogavage. Metformin suspension at the daily dose of 150 mg/kg was administered to rats in the metformin group by gastrogavage. Equal volume of distilled water was administered to rats in the normal control group and the model group. All medication lasted for 12 weeks. The levels of fasting blood glucose (FBG), free fatty acid (FFA), serum triglyceride (TG), serum total cholesterol (TC), serum alanine aminotransferase (ALT), and aspartate aminotransferase (AST) were detected. The body weight and wet liver weight were weighed, and the liver weight index calculated. The liver TG content was measured. The pathological changes of liver and the expression of SIK1 were observed by HE staining and immunohistochemistry. The mRNA and protein expression of SIK1 and SREBP-1c were detected using RT-PCR and Western blot.
RESULTSCompared with the normal control group, FBG, FFA, TG, TC, ALT, AST, liver weight index, and liver TG contents significantly increased (P < 0.01); liver steatosis was severe, the mRNA and protein expression of SIK1 obviously decreased (P < 0.01); mRNA and protein expression of SREBP-1c increased (P < 0.01). After drug therapy, compared with the model group, FBG, FFA, TG, TC, ALT, AST, and liver weight index significantly decreased, liver TG contents significantly decreased, the mRNA and protein expression of SIK1 obviously increased, while mRNA and protein expression of SREBP-1c obviously decreased (P < 0.05, P < 0.01) in the ZQR group and the metformin group (P < 0.05, P < 0.01); and the pathological changes were also improved. All the indices were improved more in the ZQR group (all P < 0.05).
CONCLUSIONIn this experiment, we found that the expression of SIK1 decreased in NAFL rats with type 2 diabetes. ZQR could alleviate lesion of NAFL type 2 diabetes rats possibly by up-regulating hepatic SIK1 expression at mRNA and protein levels.

Objective To investigate the mutations of CLCNKB gene in a family with classic Bartter syndrome. Methods Genetic DNA was extracted from peripheral blood leucocytes of family members. The coding exons and intron exon junctions of CLCNKB gene were amplyfied by PCR and sequenced directly.Fifty unrelated healthy subjects were selected to exclude the possibility of polymorphism. Results A heterozygous(missense)mutation(482T>G,L161R)was detected in the exon 4 of patients. The hetemzygous mutation(L161R)was found in the mother, while no mutation was found in the father of this family.L161R had not been reported and was a novel mutation when referring to literatures and human genomic database home and abroad. Conclusion A new CLCNKB gene mutation(L161R)is identified for the first time.

Aim To observe the changes of diaphragm contractility and cytoskeletal proteins titin, nebulin and sarcoplasmic reticulum Ca~(2+)-ATPase gene expressions in adriamycin-induced cytotoxicity in rats. Methods The animal models of diaphragm damage were duplicated by injecting adriamycin into abdominal cavity one time.Forty male sprague-dawley rats were randomly divided into four groups(n=10):Three groups received adriamycin in low, middle and high dosage(10,20 and 40 mg·kg~(-1))respectively.Meanwhile,the normal saline was given to rats in control groups.Three days later, these rats were killed, and the diaphragm was removed by thoracotomy. The diaphragm contractility was assessed in isolated diaphragm strips perfusion by these paramemters including peak twitch tension(Pt),maximum tetanic tension(Po),time to peak contraction(CT),half relaxaion time(1/2RT),maximal rates of contraction(+dt/dt_(max))and maximal rates of relaxation(-dt/dt_(max)).The expressions of titin, nebulin and sarcoplasmic reticulum Ca~(2+)-ATPase(SERCA)at mRNA level were detected by RT-PCR analysis. Results In contrast to those in control group,Po,Pt,±dt/dt_(max) in the adriamycin group were lower(P<0.01);CT,1/2RT in the adriamycin group increased significantly(P<0.01).The levels of titin, nebulin and SERCA gene expressions in middle-dose group were lower than those in control group(P<0.01).Conclusions The mRNA levels of titin, nebulin and SERCA of diaphragm are down-regulated in adriamycin-induced cytotoxicity in rats.It may be associated with the decline of diaphragm contractility.
