1.Expression of YAP in human papillary thyroid cancer and its influence on cell growth

Objective To investigate the expression of Yes‐associated protein(YAP) in human papillary thyroid cancer and its influence on cell growth .Methods The samples in 57 cases of papillary thyroid cancer treated by operation resection in the gen‐eral surgery department of this hospital and the matched tumor‐adjacent tissues were collected .All the cases were definitely diag‐nosed by the pathology examination .The expression of YAP protein in the cancer tissue and corresponding tumor‐adjacent tissue were determined by the immunohistochemistry(IHC)staining .The relationship between the YAP protein and the clinicopathological data was statistically analyzed .siRNA was used to silence the expression of YAP in B‐CPAP cells ,MTT and flow cytometry were used to measure the proliferation and apoptosis changes .Results The expression level of YAP was markedly higher in papillary thyroid cancer tissues than in tumor adjacent tissues (P<0 .05);moreover the expression of YAP protein was positively correlated with the tumor size and TNM stage (P<0 .05) .Silencing YAP gene could significantly inhibit the cell proliferation ability and pro‐mote cell apoptosis(P<0 .05) .Conclusion YAP is highly expressed in papillary thyroid cancer tissues and is related with its ad‐verse clinicopathological characteristics ,down‐regulating YAP gene can significantly inhibit the cell growth .
2.Efficacy of Cefazolin vs. Cefuroxime in Preventioning Cesarean Section Wound Infection

OBJECTIVE:To compare the efficacy of cefazolin and cefuroxime prevention of perioperative cesarean section wound infection. METHODS:291 cases of elective cesarean section surgery,according to the use of medicines are divided into group A(n=140)and group B(n=151),were retrospectively analyzed. Patients in group A,preoperative 30 min intravenous infusion of cefazolin 2.0 g;after surgery,2.0 g ivgtt,q12 h,continue to use 2~5 days. Patients in group B,preoperative 30 min intravenous infusion of cefuroxime 2.0 g;after surgery,2.0 g ivgtt,q12 h,continue to use 2~5 days. Comparison of the clinical effect of 2 groups patients. RESULTS:WBC,N%,Hb,complications,body temperature,wound healing had no statistical difference in postoperative between 2 groups. All patients were not the emergence of drug adverse reactions. CONCLUSION:cefazolin and cefuroxime has the same efficacy of prevention of cesarean section wound infection.
3.Comparison of Preventive Effect of Maternal-Infant Hepatitis B Virus Vertical Transmission by Symphysial Immunoprophylaxis

Objective To investigate the efficacy by inoculated with hepatitis B immunoglobulin(HBIG) and hepatitis B vaccine in dif-ferent doses and time points to prevent maternal-infant vertical transmission of hepatitis B virus(HBV).Methods Gravidas positive for HBV were selected and measured HBV DNA,according to difference HBV DNA degrees divided into A,B,C groups.Every group was randomly divided into 4 groups using a variety of combined immunity methods to compare its efficacies.Results There were no significant differences in the positive rate of HBV and HBsAb in A,B groups by statistics analysis.There were significant differences in the positive rate of HBV and HBsAb in group C by statistics analysis.Conclusion According to the different HBV DNA take different combined immunodeficiency approach to prevent maternal-infant vertical transmission.
4.Expression of CCL5 and S100A4 protein in breast cancer tissues and their relationship with clinicopathological features and prognosis

Objective To investigate the relationship between the expression of CC chemokine ligand 5(CCL5)and S100 calcium binding protein A4 (S100A4)protein in breast cancer tissues with clinicopathological features and prognosis.Methods The expression of CCL5 and S100A4 in 40 cases of normal breast tissues and 120 cases of breast cancer were detected by immunohistochemistry,and the relationship between the degree of expres-sion and the clinicopathological features and prognosis of breast cancer was analyzed.Results The expression posi-tive rates of CCL5 and S100A4 in breast cancer tissues were 56.67% and 62.50% respectively,which were not expressed in normal breast tissues,and the differences were statistically significant (χ2CCL5 =39.403,P <0.01;χ2S100A4 =47.062,P <0.01).The expression of CCL5 in breast cancer tissues was statistically correlated with clinical staging(χ2 =10.141,P <0.01 ),pathological type (χ2 =5.769,P =0.017)and lymph node metastasis (χ2 =34.178,P <0.01),but not correlated with patients'age,tumor size,pathological grading(all P >0.05 ).And the expression of S100A4 was statistically correlated with clinical staging(χ2 =44.311,P <0.01)and lymph node metas-tasis(χ2 =14.843,P <0.01 ),but not correlated with patients'age,tumor size,pathological type and pathological grading(all P >0.05).The expression of CCL5 and S100A4 in breast cancer was positively correlated(r =0.301, P <0.01).CCL5 was positively correlated with the recurrence of breast cancer(OR =6.270,P <0.01),and S100A4 was not correlated with the recurrence of breast cancer(OR =1.103,P =0.768).Survival analysis showed that the disease -free survival time of patients with positive CCL5 expression was significantly shorter than the patients with negative CCL5 expression(χ2 =11.851,P <0.01 ),and the disease -free survival time of patients with positive S100A4 expression was significantly shorter than the patients with negative S100A4 expression(χ2 =5.433,P =0.021).The joint detection showed that the disease -free survival time in CCL5(+)+S100A4(+)group was sig-nificantly lower than that of CCL5(+)or S100A4(+)group(χ2 =15.341,P <0.01)and CCL5 (-)+S100A4 (-)group(χ2 =15.341,P <0.01).Conclusion The expression of CCL5 and S100A4 in breast cancer can reflect the metastasis and staging of breast cancer,which can be used to judge the clinical pathological characteristics and prognosis of breast cancer.
6.Injecting Kanseran into Tumor Mass via Fibrobronchoscope in Combination with Chemotherapy for Treatment of Advanced Non-small-cell Lung Cancer

Objective:To observe the therapeutic effect and toxicity of local Kanseran injection in combination with chemotherapy in advanced NSCLC patients.Methods:A total of 18 cases was recruited in this study. After desensitization,Kanseran injection was injected into tumor mass through fibrobronchoscope,2 mg every time,once per week. Meanwhile,MVP regimen was administered.Results:Of the 18 patients,1 was completely relieved,7 cases were partially relieved. The response rate was 44.4%. After treatment,when examined by fibrobronchoscope the tumor masses of 15 patients became smaller. The main side effects were fever,alopecia and gastrointestinal reaction,which were mild and tolerable.Conclusion:Local Kanseran injection into tumor mass in conjunction with chemotherapy is effective and safe in the treatment of advanced NSCLC patients with bronchus obstruction.
7.Diagnosis of Bladder Cancer Using Fluorescence in Situ Hybridization Technique

Objective To evaluate the value of using fluorescence in situ hybridization (FISH)technique for the detection of chromosome aberration of urine exfoliated cells for the diagnosis of bladder tumor.MethodsFISH technique were used to detect the abnormalities of chromosome 3,7,17 and 9p16 site from 20 normal people, and to establish the threshold.The morning's first urinations were available from 75 patients with bladder cancer and 25 patients without urothelial tumor, then were detected using FISH technique and urine cytology respectively.The sample was considered positive if two or more probes results higher than the criteria,or one probe has two or more abnormal results.Results The sensitivity of single using were 73.3% (55/75),76.0% (57/75),62.7% (47/75) and 62.7% (47/75) for the 4 probes (3,7, 17 and 9p16)respectively.The sensitivity of combined detection was 85.3% (64/75) and specificity was 96.0% (24/25) The sensitivity and specificity of urine cytology examination was 9.3% (7/75) and 100% (25/25) .The sensitivity of FISH examination was significantly higher than that of urine cytology examination (85.3% vs 9.3% ,x2 = 57.00, P < 0.001) .Sensitivity of FISH examination was not correlated with cancer pathologic grading(low vs high : 84.2% vs 86.5%, x2 = 0.08, P > 0.05)and clinical stage (ta-tl : 82.9%, t2-t4 :87.5%, x2 = 0.32 ,P > 0.05) .ConclusionFISH technique is a non-invasive and effective method for the early diagnosis of bladder tumor and is more sensitive than urine cytology.Furthermore, FISH technique can be used to predict the tumor's biological behavivor and prognosis.
