2.Determination of Icariin in Yishengling Granules (Sucrose-free) by HPLC

Objective To establish a method for determination of Icariin in Yishengling granules (Sucrose-free) by HPLC. Methods Using a column packed with C18 and mixture of acetonitrile-water (30∶70) as the mobile phase. The flow rate was 1.0 mL/min and the temperature was 30 ℃. The detection wavelength was at 270 nm. Result The linear range of Icariin was 4.1~20.5 ?g (r=1). The mean recovery was 102.90% and RSD was 1.72%. Conclusion The method is simple, reliable and rapid, and can be applied to the quality control of Yishenling granules (Sucrose-free).
3.Analysis on six test results of sex hormone for 1 380 menstrual disorder women in Qingshan district of Wuhan City

Objective To investigation the serum sex hormone levels of 1 380 menstrual disorder women in Qingshan district of Wuhan city .Methods Applying Electrochemiluminescence immunoassay (ECLIA ) to detection the density of progesterone (PROG) ,testosterone(TEST) ,prolactin(PRL) ,estradiol(E2) ,luteinizing hormone(LH) and follicle‐stimulating hormone(FSH) in serum .Results 199 cases of PROG were abnormal ,which accounted for 14 .42% ;128 cases of TEST were abnormal ,which accoun‐ted for 9 .28% ;323 cases of PROL were abnormal ,which accounted for 23 .41% ;334 cases of E2 were abnormal ,which accounted for 24 .20% ;568 cases of LH were abnormal ,which accounted for 41 .16% ;553 cases of FSH were abnormal ,which accounted for 40 .07% .Conclusion In the menstrual disorder women in Qingshan district of Wuhan city ,LH and FSH anomaly is the highest proportion ,ovarian dysfunction may be the main reason for female menstrual disorder .
4.Mechanism of hsa-miRNA124-3p regulating the proliferation and migration of human lung cancer cell line NCI-H460

Objective · To study the regulation of hsa-miRNA124-3p on the proliferation and migration of human lung cancer NCI-H460 cells and its mechanism. Methods · Four pairs of lung cancer and para-carcinoma tissues were harvested in clinical and measured for hsa-miRNA124-3p and Krüppellike factor 4 (KLF4) levels. The theoretical binding site of hsa-miRNA124-3p in 3'-UTR of KLF4 was predicted by bioinformatics, and validated by luciferase report assay. NCI-H460 cells were transfected with pshRNA-Sponge-miRNA124 or pshRNA-KLF4, and 48 hours later, the proliferation of NCI-H460 cells after genetic intervention was assayed by the MTT method, and cell migration ability was observed by streak method. Results · For all four pairs of samples tested, hsa-miRNA124-3p was higher in the cancer tissues than in the adjacent tissue (P<0.01), and KLF4 protein was lower in the cancer tissues than in the adjacent tissue (P<0.01). The bioinformatic analysis showed there is a theoretical binding site (5'-UGCCUUAA-3') of hsa-miRNA124-3p in 3'-UTR of KLF4. Luciferase activity assay showed that hsa-miRNA124-3p could bind to the 3'-UTR region of KLF4 gene and negatively regulate the expression of protein. The proliferation of NC-H460 cells was suppressed by transfection with pshRNA-Sponge-miRNA12472 h after transfection (P<0.05 ). Compared with the control group, the proliferation activity of pshRNA-KLF4 transfection group was further enhanced (P<0.05) There was no significant difference in the proliferation of pshRNA-Sponge-miRNA124 and pshRNA-KLF4 cotransfection group and the control group (P>0.05). The data of cell migration assay showed that the changes of cell migration ability were the same as proliferation activity of the cells in groups 72 h after transfection. Conclusion · Hsa-miRNA124-3p increases the proliferation and migration in NCI-H460 cells via suppressing the expression of KLF4, and reducing the content of miRNA124-3p in NC-H460 cells can inhibit cell proliferation and migration via upregulating KLF4 expression.
5.Analysis of demands of African students for China-Africa malaria preven-tion training

Objective To understand the demands of African students on the China?Africa malaria prevention training pro?grams as well as explore further suggestions on the student selection and course content design. Methods A self?administered questionnaire survey was conducted,and all the students who attended in the malaria prevention training courses in 2014 and 2015 were included. The Chi?square test was conducted to analyse the correlations between professional backgrounds ,work stat?ues and training needs. Results A total of 161 individuals were sampled eventually. These participants were trained in either English(58.4%)or French(41.6%). Most of the participants were male(69.3%),the major of them were mainly clinical tech?nology specialty(40.0%),and most of them worked in malaria area within 10 years(56.4%). Moreover,48.2%of the partici?pants used more than 76%of total work time on malaria control,and more than 80%worked in national or provincial/municipal level. The working areas of these participants were focused on clinical field(41.4%)and official field(29.9%),and only a few of them were from research positions(11.9%). The most needed course content in malaria training was strategy and epidemiolo?gy knowledge for malaria prevention and control(65.5%),while clinical workers were most needed to be trained(39.2%). The participants who came from French speaking countries preferred strategy training(χ2=12.528,P<0.01),and those worked in the national level were aslo more likely to choose strategies training course(χ2=10.508,P<0.05). Conclusions Currently, the China?Africa malaria prevention training programs could basically satisfy African students’needs. However,more aimed courses should be designed according to their professional backgrounds,national situation,work experiences on malaria con?trol,and institutional levels.
6.Cellular expression profile of RhoA in rats with spinal cord injury.

RhoA, a small GTPase, is involved in a wide array of cellular functions in the central nervous system, such as cell motility, cytoskeleton rearrangement, transcriptional regulation, phagocytosis and cell growth. It is not known how spinal cord injury (SCI) affects the expression of RhoA in different nerve cells. In the present study, we investigated the changes of RhoA expression in remote areas of the injury at the 3rd, 7th and 30th day after SCI, which was established by T10 contusion method. Moreover, we examine its expression profile in neurons, astrocytes and microglia. RhoA was found to be weakly expressed in these nerve cells in normal spinal cord. Western blotting showed that, after SCI, the total RhoA expression was up-regulated, and the RhoA expression was increased and peaked at the 7th day. Double immunostaining revealed specific and temporal expression patterns of RhoA in different nerve cells. The expression of RhoA in neurons started to increase at day 3, peaked at day 7 and then decreased slightly at day 30. Expression of RhoA in astrocytes increased moderately after SCI and peaked at day 7. There was no obvious change in RhoA expression in microglia after SCI in remote areas. This study demonstrated that, after SCI, RhoA expression exhibited different patterns with different nerve cells of spinal cord. RhoA expression patterns also changed with time after SCI, and among different nerve cells in the injured spinal cord. These findings can help us better understand the roles of RhoA in SCI.
7.Advantages of population pharmacokinetics and its application in the field of traditional Chinese medicine.

The accurate medical treatment is based on the information of the genome, which is the best treatment for the patients. Population pharmacokinetic study can be formulated according to the individual differences of patients to the dose, in the accurate medical model which has a unique advantage. At present, there are many problems such as adverse drug reaction in Chinese traditional medicine, and it is necessary to introduce a group of medicine on the basis of precise medical treatment. However, due to the different characteristics of traditional Chinese medicine and chemical medicine, it is necessary to combine the population pharmacokinetics, genetics and statistical methods to establish a research method which is in line with the characteristics of Chinese medicine. The key scientific problem is to make clear the active components of Chinese medicine metabolism of the drug metabolic enzyme gene, and pay attention to the analysis of the polymorphism of the overall role of drug metabolism enzymes in the human body. Clear key scientific issues and break through the bottleneck, so as to achieve the precise medical treatment, to international.

8.Induced membrane technique for treatment of post-traumatic tibial osteomyelitis

Purpose To investigate the clinical outcome of induced membrane technique for treatment of post-traumatic tibial osteomyelitis.Methods A retrospective analysis was made on 17 cases of post-traumatic tibial osteomyelitis admitted from August 2011 to October 2012.There were 13 males and 4 females aged 19-67 years (mean,40.9 years).Mean length of bone defect was 6.7 cm (range,2.0-18.5 cm).Surgical treatments were performed including phase Ⅰ surgical debridement and antibiotics-loaded bone cement impregnation to induce a membrane and phase Ⅱ bone defect reconstruction by intramembranous bone grafting.Clinical effect of the treatment was evaluated.Results Bone healing was detected at the 27-month follow-up (range,24-32 months).Radiographic healing was obtained 4-6 months after operation.Two cases of recurrent infection were noted after phase Ⅰ surgery and had another debridement.External fixator proximal pin track infection occurred in one case and infection was controlled by hardware removal and dressing change.There was one case of metal rejection,two function disorders,one clubfoot,and one ankle stiffness.Conclusion Induced membrane technique is effective in the treatment of post-traumatic tibial osteomyelitis,especially in lone bone defect reconstruction,but limb functional complications are worth consideration during treatment.
9.Efficacy observation on acupuncture plus moving cupping with compound white mustard oil for lumbar myofascitis

Objective: To observe the clinical effect of acupuncture plus moving cupping with white mustard oil to treat lumbar myofascitis. Methods: A total of 90 patients with lumbar myofascitis who met the inclusion criteria were randomly divided into an observation group and a control group, 45 cases in each group. The observation group received acupuncture and moving cupping with white mustard oil; the control group received acupuncture plus warm needling treatment. The treatment was given twice a week for a total of 4 weeks. The visual analog scale (VAS) and Chinese version of Roland-Morris disability questionnaire (RMDQ) score of pain before the treatment, and respectively after 1-week and 4-week treatment were recorded for efficacy evaluation. Results: After 4 weeks of treatment, the difference in total effective rate between the two groups was not significantly (P>0.05). After 1 week of treatment, there was no significant difference in VAS score between the two groups (P>0.05). After 4 weeks of treatment, the VAS score of the observation group was lower than that of the control group, and there was a significant difference between the two groups (P<0.05). After 4 weeks of treatment, the Chinese version of RMDQ scores of both groups were significantly decreased, there were statistical differences in comparing with those before treatment (both P<0.05). After treatment for 1 week and 4 weeks, the Chinese version of RMDQ score in the observation group was lower than that in the control group, with significant differences between the two groups (all P<0.05). Conclusion: Acupuncture plus moving cupping and acupuncture plus warm needling both can relieve the pain of patients with lumbar myofascitis, improve the function of back muscles, while the curative effect of acupuncture plus moving cupping is better.
10.Effects of intelligent power-assisted functional electrical stimulation therapy on ankle joint function in post-stroke subjects

Objective To observe the effect of power-assisted functional electrical stimulation (PAFES)therapy on ankle joint function recovery in stroke patients.Methods Ninety hemiplegic stroke patients were randomly and evenly divided into a control group,a PAFES group,and a neuromuscular electrical stimulation (NMES)group.All groups received conventional rehabilitation training.PAFES group adopted PAFES treatment on affected lower extremities and NMES group was given the NMES therapy on the tibialis anterior of the affected lower limbs,in addition to conventional rehabilitation training.The active range of motion (AROM) of ankle dorsiflexion,FuglMeyer motor assessment (FMA),Barthel index (BI) and Ankle flexion and extension movement (AFEM) in 10 seconds were evaluated before the trial and after 4 weeks treatment.Results After treatment,there were significant differences in the AROM of ankle dorsiflexion,FMA,BI and AFEM (P < 0.05) compared with before treatment within each group.The improvement of AROM of ankle dorsiflexion in PAFES group (8.19 ± 3.39) ° and the values in NMES group (8.96 ± 3.68) ° were to a significantly greater extend than control group (3.88 ± 4.10) ° (P <0.05) ; the improvement of FMA and BI in PAFES group was also superior to those in NMES group and control group (P < 0.05).Conclusion The intelligent PAFES therapy could help improve AROM of ankle dorsiflexion,the motor function of the affected lower extremity and the ability of the activities of daily living in stroke patients.
