1.Down-regulating HIPK2 promotes cisplatin-induced apoptosis of human kidney tubular epithelial cells

Objective To explore the effect of HIPK2 on apoptosis of human kidney tubular epithelial cells (HKC) induced by cisplatin.Methods Apoptosis of HKC cells was induced by cisplatin and the expression of HIPK2 was detected by RT-qPCR and Western blot.Two HIPK2 siRNAs were designed according to gene sequence of HIPK2 and cell lines with HIPK2 knockdown were established through transfecting the HIPK2 siRNAs into HKC cells by liposome.The expression of HIPK2 mRNA and protein was detected by RT-qPCR and Western blot after induced by cisplatin.Then cell apoptosis was detected by Annexin V/PI after the HIPK2-knockdown cells were treated with cisplatin.Moreover,the expression of pro-apoptotic protein bax was detected by Western blot after HIPK2 was knockdown.Results The expression of HIPK2 mRNA and protein was down-regulated obviously on the process of HKC apoptosis which induced by dose-dependent cisplatin (P<0.05).The transfection of siRNA could significantly reduce the expression of HIPK2 mRNA and protein in HKC (P<0.05),which promotes the HKC cells apoptosis induced by cisplatin.Conclusions HIPK2 can suppress the HKC cells apoptosis induced by cisplatin.
2.Smoking Aggravates Large and Midio Arterial Stiffness in Patients with Essential Hypertensive

Objective To investigate the effect of smoking on large arterial stiffness in patients with essential hypertensive.Methods The aortic,carotid and brachial arterial dimension,intimal-medial thickness were determined by vascular ultrasound technique.Wall cross sectional area and indices of compliance and distensibility were calculated in essential hypertensive patients with smoking(n=75) or without smoking(n=70).Results Hypertensive patients with smoking have increased wall intimal-medial thickness and wall cross sectional area in aortic,carotid and brachial arteries associated with decreased compliance and distensibility.After adjusting for the confounding effects of other variables including Age,BMI,HR,BP,PP,FPG,LDL-C and TG,covariance analysis show smoking remained as predisposed factors for vascular remodeling.Conclusion Smoking aggravates large arterial stiffness in essential hypertensive patients.
3.Application of Virtual Reality Training System in Rehabilitation Medicine Education

Objective To apply the virtual reality training in clinical education of rehabilitation therapy. Methods 26 undergraduate students majored in rehabilitation therapy were randomly divided into 2 groups during clinical education of physiotherapy. The experimental group (n=14) received virtual reality training first, and then with routine teaching, while the control group (n=12) learnt with routine teaching only. Self-assessment and physiotherapy test were conducted after training. Results The experimental group reported more interested, initiative and positive in training than those of the control group. The experimental group achieved higher scores in practical tests than the control group (P<0.05), while no significant difference in the theoretical examination between the groups. Conclusion Application of virtual reality training may improve the teaching of physiotherapy.
4.Application of ultrasound in diagnosis of thyroid nodular disease

Objective To explore the diagnostic value of ultrasound in thyroid nodular disease.Methods Eighty-seven patients with thyroid nodule in hospital from 2010-2011 were taken ultrasound examination,including 23 cases with benign nodules and 64 cases with malignant nodules.Univariate analysis and mutivariate Logistic regression analysis were taken to test the ultrasonic index (internal echo,boundary,size,shape,microcalcifications,and aspect ratio) for discovery of the best diagnostic ultrasonic index to identify benign and malignant nodules.Results The ultrasound in the patients with malignant thyroid nodules showed the hypoecho,unclear border,irregular shape,microcalcifications,low blood supply,the aspect ratio > 1.The ultrasound in the patients with benign thyroid nodules showed high or equal echo,clear border,regular shape,high blood supply,and the aspect ratio≤ 1.The ultrasonic indices between two groups had statistically significant difference (P < 0.05).The shape and microcalcifications were the best ultrasonic diagnostic index to indentify the benign and malignant nodules with an EXP(B) 20.12 and 19.14,respectively.Conclusions The ultrasound is helpful in the diagnosis of thyroid nodular disease and identification of benign and malignant thyroid nodules.The ultrasonic images are the basic data for the clinical diagnosis of thyroid nodules.
6.Impacts of Kechuanning on Expression of Signal Transducer and Activator of Transcription 6 in CD4+, CD8+T Lymphocytes of Athma Rats Induced by Virus

Objective To observe the impacts of Kechuanning on expression of signal transducer and activator of transcription 6 (STAT 6) in CD4+, CD8+T lymphocytes of athma rats induced by virus, and analyze the mechanism of Kechuanning in treating asthma induced by RSV. Methods The PBMC was isolated from the peripheral blood, CD4+T, CD8+T lymphocyte was separated by immunomagnetic beads. The purity and activity of CD4+T and CD8+T lymphocytes were measured by using flow cytometry, trypan-blue dye exclusion test. The protein expression of STAT6 was detected by fluorescence-activated cell sorting. Results The descending progress of the trial were supported by the purity and activity of CD4+T, CD8+T lymphocyte. CD4+T lymphocyte in the asthma model group were increased (P<0.01), however the increase of CD8+T lymphocyte were not significant (P>0.05). The expression of the protein of STAT6 in the treated group with Kechuanning decoction were decreased than that in the model group (P<0.01). Compared with Dexamethasone and Salbutamol group, there were no significant differences (P>0.05). Conclusion Kechuanning decoction could decrease the protein of STAT6 in CD4+, CD8+T lymphocyte, thereby it could regulate the disorder of Th1/Th2, thus prevent and treat the asthmatic attack induced by virus.
7.Influence of morphine on synaptophysin expression and synapse structure in hippocampus of mice

Objective:To study the influence of morphine on the expression of synaptophysin(SYN)and synapse structure in mice hippocampus,so as to provide pathological evidence for studying the development and treatment of chronic morphine intoxication, addiction and abstinence symptoms of morphine.Methods:Twenty mice were evenly randomized into control group and experiment group.Mice in control group were injected with normal saline(0.1 ml daily)and those in experimental group were injected with morphine(0.1 ml,1 mg daily).Thirty days later the mice in 2 groups were killed and their brain tissues were harvested and made into slices,stained with immunohistochemical techniques(SP)and photographed under the light microscope.The images were analyzed with the image analytical system and the data were statistically analyzed.Results:In the control group,positive staining of SYN was found in the entorhinal area,subiculum,stratum plextiforme,polymorphic layer of gyrus dentatus,stratum oriens,and stratum radiatum of hippocampus;weak positive staining of SYN was noticed in the stratum lacunosum-moleculare of hippocampus;positive staining of SYN was also found the membrane of pyramidal cells and granule cells,with the mean gray scale value of the hippocampal structure being 132.84?8.67.Positively stained neurons was also found in the entorhinal area and the subiculum,with a intensity of(7.80?1.03)/ mm~2.In the experiment group,the suhiculum and polymorphic layers of gyrus dentatus were positively stained for SYN;the entorhinal area,stratum oriens,stratum radiatum and stratum lacunosum-moleculare of hippocampus were strongly positive of SYN;the membrane of pyramidal cells and granule ceils were also strongly positive of SYN,with the mean gray scale value of the hippocampal structure being 116.27?5.70.Strongly stained neurons were also found in the entorhinal area and the subiculum,with the intensity being(11.90?1.45)/mm~2.The number of SYN positive neurons and the intensity of SYN in the experimental group were higher than those in the control group(P
8.The therapeutic effects of intermittent hypoxia therapy in isolated systolic hypertension patients

Objective To study the therapeutic effects of intermittent hypoxia therapy(IHT)in isolated systolic hypertension(ISH)patients with elevated cerebral blood flow velocity(Vp),and to explore the mechanisms involved.Methods Seventy-six ISH patients with increasing Vp and normal pulsatility index(PI)of the middle ce- rebral artery(MCA)were randomly divided into a therapy group and a control group.IHT was administrated in the therapy group,and air in the control group.The Vp and PI of the MCA and blood pressure(BP)were observed be- fore and after treatment.Results Vp and systolic blood pressure(SBP)were significantly reduced after IHT(P<0.01)compared with the therapy group's scores betore treatment,but PI and diastolic blood pressure showed no sig- nificant difference.There was no significant change in BP,Vp or PI in the control group before or after treatment. Conclusion IHT has therapeutic effects on ISH by reducing Vp and moderating SBP.
9.Effects of multiple-trough sampling design and algorithm on the estimation of population and individual pharmacokinetic parameters.

The purpose of this study is to investigate the effects of multiple-trough sampling design and nonlinear mixed effect modeling (NONMEM) algorithm on the estimation of population and individual pharmacokinetic parameters. Oxcarbazepine and tacrolimus were used as one-compartment and two-compartment model drugs, respectively. Seven sampling designs were investigated using various number of trough concentrations per individual ranging from 1-4. Monte Carlo simulations were performed to produce state-steady trough concentrations. One-compartment model was used to fit simulated data from oxcarbazepine and tacrolimus. The accuracy and precision of the estimated parameters were evaluated using the median prediction error (PE), the median absolute PE and boxplot. The results indicated that trough concentrations could yield reliable estimates of apparent clearance (CL/F). For oxcarbazepine, as the number of trough concentrations per subject increased, the accuracy and precision of CL/F, between-subject variability (BSV) of CL/F and residual variability (RUV) tended to be improved. For tacrolimus, however, although no improvement were observed in the accuracy of CL/F and BSV of CL/F, the PE distribution ranges were significantly narrowed and the RUV estimates were less bias and imprecise. In terms of algorithm, Monte Carlo importance sampling (IMP) and IMP assisted by mode a posteriori estimation (IMPMAP) were consistently better than other methods. Additionally, the sampling design had no significant effects on the individual parameter estimates, which were only depended on the interaction between BSV and RUV in various algorithms. Decreased in BSV and RUV levels can improve the accuracy and precision of the estimation for both population and individual pharmacokinetic parameter estimates.
