China Oncology 2001;0(02):-
C erbB 2 is overexpressed in 20%~30% of human breast cancers, this oncoprotein plays an important role in predicting response to chemotherapy in breast cancer.Overexpression of c erbB 2 in breast cancer is associated with resistance or sensitivity to certain chemo therapeutic drugs, c erbB 2 can be expected to be a biological marker to direct chemotherapy in the treatment of breast cancer.

Objective To investigate the current statues of anticoagulation therapy for ischemic stroke patients with atrial fibrillation(AF) and compared the difference with guidelines. Methods Three hundred and fifty-eight patients with cerebral infarction and atrial fibrillation in Xinhua Hospital Affiliated to Shanghai Jiaotong University from Jan.2008 to Jun.2012 were selected as our subjects. The data related to the antithrombotic therapy were collected and analyzed. Results Among patients with ischemic stroke, the rate of preexisting AF was 70.9% (254/358),and warfarin taken rate was 4.3% (11/254) before stroke onset.Of warfarin taken patients,INR were less than 1.5.Of all out hospitalized patients,20.9% (68/326) were treated with warfarin therapy. There were the negative correlation between warfarin usage and HAS-BLED score,MRS score, antiplatelet drugs (OR =-1.974,-0.725,-4.170,P < 0.05 or P < 0.01).Among patients with warfarin usage in hospital,33.8% of subjects (23/68) still remain on warfarin even out of hospital. The medium dosage was 2.5 mg (1.25-3.75 mg),and INR was controlled at ranged of 1.5-3.1.Periods of warfarin therapeutic range (TTR) was (61.6 ± 21.2)%.Conclusion The anticoagulation treatment rate and INR were lower in patients cerebral infarction with atrial fibrillation.Warfar might no be universal use due to the bleeding concerns and un-onvenience of monitoring INR.

Objective To investigate the effect of nitric oxide(NO)on the prostatic proliferation/apoptosis in rat models. Methods Twenty adult male rats were randomly divided into 3 groups.Group 1 were normal controls (n=6);group 2 (n=7) and group 3 (n=7) were treated with subcutaneous injection of nitric oxide synthase (NOS) inhibitor-L-NG-Nitroarginine methyl ester (L-NAME) 50mg/kg and 100mg/kg, bid, respectively. The weight of the prostate was measured after 2 weeks and morphological changes were examined with light microscope.Detection of ki-67 and TUNEL in prostatic epithelial cells were undertaken so as to assess the proliferation and apoptosis index. The protein and mRNA expression of NOS were analyzed by means of immunohistochemistry and RT-PCR. Results The prostate weight of 3 groups was not significantly different.Compared with group 1, remarkable atrophy of prostatic epithelial cells, marked decrease in proliferation rate of epithelial and interstitial cells, and significant increase in apoptosis rate of epithelial cells were noted in group 2 and group 3 (P

China Biotechnology 2006;0(05):-
Eukaryotic expression plasmid pCI-neo-mdr1 which contains human multidrug resistance gene 1(mdr1),was constructed and transferred into human hepatocarcinoma HepG2 cells by use of liposome. G418 was used to screen the cells successfully with mdr1 and the selected cells was named HepG2/mdr1 Morphological and biological properties of HepG2/mdr1 cells were observed. The results show that the constructed HepG2/mdr1 cell line was high efficient and stationary in the expression of mdr1. The work was valuable and desirable for the establishment of multidrug resistant cell models, and for the study of MDR in human hepatoma. Furthermore, the work also provided a perfect model for the research of relationship between insulin resistance and MDR in hepatocarcinoma cells.

OBJECTIVE This study was to investigate the carrier and infection of Clostridium difficile in clinic feces specimen,to analyze clinic characteristics, and to improve isolation rate and to provide basis on efficient prevention.METHODS C.difficile toxin A&B kit and anaerobic culture was conducted in 20 cases with diarrhea.Colonies suspected to be C.difficile,on the basis of their macroscopic appearance and characteristic odor, oxygen tolerance experiment, were confirmed by their biochemical characteristics(API 20A,bioMerieux).RESULTS After C.difficile selective culture,8 suspected colonies from 20 feces specimen were conducted by feces smear and oxygen tolerance experiment.6 of 8 was G+ rod bacteria with positive oxygen tolerance experiment.4 stains of C.difficile were identified by API 20A,positive rate was 20%;toxin detect was positive in 1 specimen(5%).CONCLUSIONS Infection of C.difficile Is associated with the basic disease.Watery feces specimen was prone to culture positive.

Objective To explore the possible role of B7 family cell surface costimulatory molecules and their receptors in immunopathogenesis in children with asthma. Methods Forty children with acute exacerbation of asthma and 36 cases of age-matched healthy children were enrolled in the study.Interferon(IFN-?),IFN-? and lipopolysaccharide(LPS),phorbol 12-myristate 13-acetate and ionomycin or zero were used to stimulate peripheral blood mononuclear cells, flow cytometry was used to detect B7-1 and B7-2,B7H1,B7DC and B7H expression on CD14+ cells;CD4+ cells were separated by immune beads and stimulated by phorbol-12-myristate-13-acetate and ionomycin and fluorescence quantitative polymerase chain reaction was used to detect CD28,cytotoxic lymphocyte associated antigen-4(CTLA-4),programmed death-1(PD-1)and inducible costimulator(ICOS) mRNA expression. Results 1.The levels of B7-1 and B7-2 expression in children with asthma were higher than those of healthy control group [B7-1:(6.68?3.97)% vs(1.74?0.69)%;B7-2:(10.87?5.99)% vs(1.58?0.75)% Pa

OBJECTIVE:To discuss the mode and feasibility of informatization and automation construction in the inpatient pharmacy. METHODS:The practice of informatization and automation construction of inpatient pharmacy in our hospital was intro-duced,as well as the change of drug dispensing model. The influence of them on drug dispensing process was analyzed. RESULTS&CONCLUSIONS:2 automatic single dose tablets dispense packing machines,2 rapid dispensing machines and 2 automatic injec-tion chests are introduced in automatic inpatient pharmacy of our hospital. The informatization construction contain the establish-ment of inpatient pharmacy database, drug dispensing and recheck, the application of code, etc. Due to the implementation of auto-matic drug dispensing information system, drug dispensing mode have changed,i.g. drug dispensing by area instead of wards;in-jection and large volume transfusion are dispensed by wards;oral preparation and discharge medication are dispensed by prescrip-tion;dispensed drugs are distributed to wards with professional logistics. The implementation of informatization and automation of inpatient pharmacy in our hospital improve pharmaceutical administration and pharmaceutical care, and add automatic expiry date management and code tracking to guarantee the accuracy, timeliness and high efficiency of drug dispensing.

Objective To explore the correlation between the reliability and validity of Brunel balance assessment(BBA)with the activities of daily living(ADL).Methods The BBA,ADL and walking ability test were performed on 215 patients conforming to the inclusion standard by two professionals. The Pearson correlation analysis, factor analysis and descriptive statistical analysis were adopted. Results The BBA total score and Cronbach's α coefficient in 3 dimensions was 0.852~0.941(P<0.01),moreover the relative coefficient of each item with its dimension was higher that that of other dimensions;3 common factors were extracted by the factor analysis, the contribution rate was 83.916%;but BBA had the ceiling effect, the relative coefficient of BBA with ADL and walking ability was 0.284-0.709(P<0.05).Conclusion BBA has a good reliability and validity and can better reflect the functions of ADL and walk.
