1.Investigate the curative effect on treatment of 40 major aphthous ulcer patients by combined use povidone-iodine,H_2O_ and levamisole liniment

Objective To study the curative effect of major aphthous ulcer using povidone-iodine,H_2O_2 asso- ciated with levamisole liniment.Methods The RAU patients were divided into 2 groups randomly.The patients of curative group were instructed to use povidone-iodine,H_2O_2 and levamisole liniment;The control group were in- structed to take metronidazole,antibiotics,compound vitamin B and vitamin C.Then the patients were observed peri- odically.Results The success rate of curative group was 85 % and the control group was 55 %.The difference had statistical significance(P<0.01).Conclusion Treatment of major aphthous ulcer using povidone-iodine,H_2O_2 and levamisole liniment is effective.
2.Effect of rapamycin on proliferation, apoptosis and cell cycles of high glucose-cultured rat glomerular mesangial cell

Objective:To evaluate the effects of rapamycin on the proliferation,apoptosis and cell cycle of glomerular mesangial cells induced by high glucose,and to explore its significance in the prevention and treatment of diabetic nephropathy. Methods:The rat GMC HBZY-1 was divided into four groups:control group,high glucose group,the first group of high glucose plus rapamycin,the second group of high glucose plus rapamycin. CCK-8 assay was used to detect the proliferation of cells, flow cytometry was introduced to evaluate the apoptosis and cell cycle of HBZY-1,Real-time PCR was used to detect the mRNA of AngiotensinⅡ(ANGⅡ),transfor ming growth factor beta1 ( TGF-β1 ) and vascular endothelial growth factor ( VEGF ) . Results: The proliferation level of HBZY-1 induced by high glucose was significantly increased,and the level of apoptosis decreased,and the expression level of ANGⅡ,TGF-β1 and VEGF was increased. Rapamycin significantly inhibited,and there was a dose dependent,and down regulated the expression of ANGⅡ,TGF-β1,and VEGF. For the cell cycle,the S phase cells in the high glucose group were significantly higher than those in the normal group (P<0. 05),and the S phase cell proportion was decreased after rapamycin intervention (P<0. 05). Conclusion:Rapamycin can inhibit the proliferation of HBZY-1 in high glucose,promote its apoptosis and lead to G1/S arrest,and down regulate the expression of ANGⅡ,TGF-β1 and VEGF.
3.Studies on extraction and antioxidation activity of phycobiliproteins from Porphyra haitanensis

Objective To extract phycobiliproteins from Porphyra haitanensis,find out the optimum extraction conditions and investigate the antioxidation activity of phycobiliproteins in vitro and anti-oxygen free radical in vivo.Methods Porphyra haitanensis was extracted with deioned-water at 40 volumes for 6 days and tissue-triturated for 2 minutes.The crude phycobiliprotein(CPP) was purified by precipitating with 45% ammonium sulphate and dialyzing.The scavenging activities of the crude phycobiliprotein on?OH of the system of 1,10-phenanthroline-Fe(Ⅱ)/H2O2 and O-2? of pyrogallol autoxidation were studied.And the mice were treated by gastric incubation with crude phycobiliprotein,at doses of 100,200,300mg?kg-1?d-1.The activities of erythrocyte catalase(CAT) and glutathione peroxidase(GSH-Px),serum superoxide dismutase(SOD) and the content of erythrocyte MDA in mice were determined on the 25th day after treatment.Results CPP could scavenge free radicals,and also can increase the activities of erythrocyte CAT,GSH-Px,serum SOD and decrease the content of MDA significantly.Conclusion The crude phycobiliprotein from Porphyra haitanensis has antioxidation activity in different ways.
4.A platform setup for genetic diagnosis of Cryopyrin associated recurrent syndrome and a case report

Objective To setup a platform for genetic diagnosis of Cryopyrin associated periodic syndrome (CAPS) and to use it for clinical diagnosis.Methods The peripheral blood cells of the patient and the controls were collected for DNA extraction.Nine exons of CAPS associated gene NLRP3 were amplified using polymerase chain reaction (PCR) and subjected to sequencing.Blast was used to compare the sequencing results with the reference gene and to locate mutation.The clinical information of the patient was collected and the relevant literature was reviewed.Results We set up a platform for exon sequencing of NLRP3 gene.Using this platform,we identified a nonsynonymous mutation in a female patient (D303N,aspartic acid at locus 303 mutated to asparagine).Considering the clinical manifestations of the patient,chronic infantile neurologic cutaneous and articular syndrome (CINCA) was diagnosed.Conclusion The set up of the platform for NLRP3 genetic analysis will facilitate the clinical awareness and research on CAPS.
5.Investigation in psychological status and nursing care of patients with hand tenolysis

Objective To explore the psychological status of patients after hand tenolysis and the pertinent nursing countermeasures. Methods 47 patients after hand tenolyais were selected and investigated their psychological status and relevant influencing factors with serf-designed questionnaire and symptom checklist-90(SCL-90). Besides, corresponding psychological nursing were applied. The SCL-90 results of Chinese Norm were adopted to undergo comparison with them and t test was chosen. Results On the first day after operation, the score of the investigation objectives was significantly different compared with that of the Chinese norm. After pertinent mental nursing, their symptoms alleviated, and no significant difference was seen between them. Conclusions There are obvious psychological obstacles in patients after hand tenolysis. The pertinent mental nursing has important effect on rehabilitation training and functional rehabilitation after hand tenolysis.
6.Deep fungal infectious in patients with systemic lupus erythematosus:a clinical analysis

Objective To investigate the infection sites,the offending species,diagnosis and prognosis of deep fungal infections (DFI) in patients with systemic lupus erythcmatosus (SLE).Methods Fifty-one patients with fungal infections in 1466 SLE patients admitted to Peking Union Medical College (PUMC) Hospital from 2000 to 2006 were reviewed retrospectively.Results Candida albicans was ranked the first pathogen,followed by ncoformans and Aspergillus species.The infection sites were lungs,cerebral meninges and blood in the order of prevalence.The overall mortality was 20%(10/51).Aspergillasis carried the highest mortality which couht be as high as 80%.Hypoproteinemia,multiple focus of fungal infections,Aspergillasis and fungemia might he the independent risk factor for mortality.Conclusion Candida albicans is the most frequent species of fungal infections in SLE patients.Iungs are the most prevalent location of infection.Earlier diagnosis is important.Special attention should be paid to aspergillasis.
7.Influence of morphine on synaptophysin expression and synapse structure in hippocampus of mice

Objective:To study the influence of morphine on the expression of synaptophysin(SYN)and synapse structure in mice hippocampus,so as to provide pathological evidence for studying the development and treatment of chronic morphine intoxication, addiction and abstinence symptoms of morphine.Methods:Twenty mice were evenly randomized into control group and experiment group.Mice in control group were injected with normal saline(0.1 ml daily)and those in experimental group were injected with morphine(0.1 ml,1 mg daily).Thirty days later the mice in 2 groups were killed and their brain tissues were harvested and made into slices,stained with immunohistochemical techniques(SP)and photographed under the light microscope.The images were analyzed with the image analytical system and the data were statistically analyzed.Results:In the control group,positive staining of SYN was found in the entorhinal area,subiculum,stratum plextiforme,polymorphic layer of gyrus dentatus,stratum oriens,and stratum radiatum of hippocampus;weak positive staining of SYN was noticed in the stratum lacunosum-moleculare of hippocampus;positive staining of SYN was also found the membrane of pyramidal cells and granule cells,with the mean gray scale value of the hippocampal structure being 132.84?8.67.Positively stained neurons was also found in the entorhinal area and the subiculum,with a intensity of(7.80?1.03)/ mm~2.In the experiment group,the suhiculum and polymorphic layers of gyrus dentatus were positively stained for SYN;the entorhinal area,stratum oriens,stratum radiatum and stratum lacunosum-moleculare of hippocampus were strongly positive of SYN;the membrane of pyramidal cells and granule ceils were also strongly positive of SYN,with the mean gray scale value of the hippocampal structure being 116.27?5.70.Strongly stained neurons were also found in the entorhinal area and the subiculum,with the intensity being(11.90?1.45)/mm~2.The number of SYN positive neurons and the intensity of SYN in the experimental group were higher than those in the control group(P
8.Anatomical multiple hepatic segmentectomy by selective occlusion of hepatic inflow for the treatment of primary hepatic cancer

Anatomic hepatic resection not only enables enough tumor-free resection margin,but also guarantee the maximal remnant of normal liver tissue.A 61-year-old male patient with hepatic cancer was admitted to the Affiliated Hospital of Hainan Medical College in February 2012.Multiple space-occupying lesions were found in segment Ⅵ,Ⅶ and Ⅷ by computed tomography (CT).The results of CT volumetry analysis showed that the left hemihepatic volume was lesser than the minimal limit of survival,so anatomic hepatic segmentectomy of Ⅵ,Ⅶ and Ⅷ with preservation of segment Ⅴ was designed to guarantee the maximal remaining of normal liver tissue.Glisson's pedicle transection was used twice to divide the right hemihepatic Glisson's pedicle,segment Ⅵ and Ⅶ Glisson's pedicle,respectivley,then the resection line was determined,and anatomical hepatic segmentectomy of Ⅵ,Ⅶ and Ⅷ was completed.With the procedures adopted,the hepatic ischemia reperfusion injury and hemodynamic instability were maximally reduced during operation.
9.Mechanosignaling pathways in keloids

BACKGROUND:Keloid is a very chalenging problem in plastic surgery. Its pathogenesis is very complex, resulting from the combined action of many factors, such as various cytokines, signal transduction pathways, extracelular matrix,etc. At present, a critical role for mechanical force and mechanotransduction in the pathogenesis of keloids has been broadly concerned and becomes the focus of studying the pathogenesis of keloids.
OBJECTIVE:To summarize the progress of the mechanosignaling pathway involved in the pathogenesis of keloids in order to further understand the pathogenesis of keloids and provide new ideas for the prevention of keloids.
METHODS: The PubMed database and Elsevier database were retrieved for articles published from January 2000 to July 2014 by computer with key words of “keloid, molecular mechanism, mechanical stress, cutaneous scar, mechanobiology, mechanosignaling pathway” in English. A total of 23 articles were included which related to the molecular signal transduction mechanism and mechanosignaling pathway about keloids.
RESULTS AND CONCLUSION:The mechanosignaling transduction pathways, such as transforming growth factor-β/Smad, MAPK, integrin, Wnt/β-catenin, RhoA/ROCK and tumor necrosis factor-α/nuclear factor-κB signaling pathway, play an important role in the formation and development of keloids. A number of clinical trials have also shown the effectiveness of a part of mechanosignaling transduction pathway inhibitors in wound healing and reducing scar hyperplasia. The research about mechanosignaling transduction pathways involved in keloids has made some progress, but most stil remain in animal experimental stage. Secondly, various mechanosignaling transduction pathways about correlation and intersectionality stil need further studies to achieve a breakthrough in the prevention of keloids.
10.Effects of nitroglycerine of different concentrations on rat cardiac preservation for 8h

Objective To investigate the effects of Thomas solution in combination with nitroglycerine of different concentrations on rat cardiac preservation for 8 h. Methods Thirty Wistar rats were selected,and Langendorff heart perfusion models were established.Rats were randomly divided into Thomas solution group,Thomas solution+4?10-6 mol/L nitroglycerine group and Thomas solution+4?10-5 mol/L nitroglycerine group(n=10).After hypothermic preservation(4 ℃)for 8 h,the cardiac function of each group was determined.Tissues of left ventricular wall were obtained,and changes of myocardial ultrastructure were observed by electron microscopy. Results Compared with Thomas solution group,the cardiac function was significantly improved and the myocardial ultrastructure injury was less severe in Thomas solution+4?10-5 mol/L nitroglycerine group after hypothermic preservation(4 ℃)for 8 h,while there was no significant difference in myocardial ultrastructure injury and cardiac function between Thomas solution group and Thomas solution+4?10-6 mol/L nitroglycerine group. Conclusion Thomas solution in combination with 4?10-5 mol/L nitroglycerine can significantly improve the preservation effects for isolated hearts in rats.
