1.Research progress on peripheral blood circulating DNA detection for hepatocellular carcinoma

Peripheral blood DNA is a kind of extracellular free DNA that existing in plasma or serum.Many researchers have already found that circulating DNA level of cancer patients is significantly higher than that of healthy individuals. The alteration in peripheral blood DNA level and gene expression are closely associated with the occurrence, development and prognosis of cancer. Therefore, it is valuable to detect peripheral blood DNA expression for early diagnosis, therapeutic monitoring and prognosis. This article reviews recent advanced progress in research on peripheral blood DNA detection for hepatocellular carcinoma (HCC).
2.Food allergy and asthma

Food allergy and asthma may frequently coexist.Children with food allergy are more than 2 to 4 times as likely to have other atopic conditions such as asthma,eczema,or respiratory allergy compared to children without food allergies.One third of children with food allergy have asthma and 4%-8% of children with asthma have food allergies.Double-blind place-controlled food challenge test is the best way to investigate the relationships of asthma and food allergy.Apart from allergen avoidance in patients with documented IgE-mediated food allergies,there are no recommendations to avoid any foods in the treatment of asthma.For patients with asthma and food allergy together,information should be delievered to parents in order to recognize and avoid to affending foods properly.

6.Plasmid-mediated resistance to quinolones in Enterobacteriaceae

Although quinolone resistance results mostly from chromosomal mutations in Enterobacteriaceae,it may also be mediated by plasmid-encoded Qnr determinants.Qnr proteins protect DNA from quinolone action and compromise the effect of quinolones,such as nalidixic acid.Qnr proteins including QnrA,QnrB and QnrS,have been identified worldwide with a quite high prevalence among Asian isolates with a frequent association with clavulanic acid-inhibited expanded-spectrum b-lactamases and plasmid-mediated cephalosporinases.The QnrA genes are embedded in complex sul1-type integrons.A close relative and likely progenitor of the QnrA have been found in the water-borne species Shewanella algae.It may help to determine the location of in vivo transfer of the QnrA genes.Further analyses of the role of quinolones,if any,in enhancing this gene transfer may prevent the spreading of the drug resistance and possibly lead to the finding of a novel mechanism of antibiotic resistance.
9.AmpC Enzyme,ESBLs Production and Antibiotic Resistance in Gram-negative Bacilli Isolated in Intensive Care Unit

OBJECTIVE To investigate the production of AmpC enzyme and ESBLs in Gram-negative bacilli and their drug-resistance to provide basis for reasonable use of antibiotics in clinical practice.METHODS All bacteria were identified by VITEK-32 system and disk test.RESULTS The main five strains of all 184 Gram-negative bacilli were Pseudomonas aeruginosa(24.4%),Acinetobacter baumannii(19.3%),Enterobacter cloacae(14.8%),Escherichia coli(10.2%) and Klebsiella pneumoniae(9.7%).The AmpC enzyme-producing rate was 53.8% and 14.7% in E.cloacae and A.baumannii the ESBLs-producing rate was 35.3%(12/34) and 15.4%(4/26) in A.baumannii and E.cloacae.P.aeruginosa was sensitive to imipenem and the drug-resistance rate of K.pneumoniae was 0%.CONCLUSIONS ESBLs and AmpC enzymes are important mechanisms of the drug-resistance of Gram-negative bacilli.
10.Preliminary study on pharmacodynamic evaluation method of Houpo formula particles.

To discuss the feasibility of the pharmacodynamic evaluation method for traditional Chinese medicine (TCM) formula particles, with traditional decoction for reference and the intervention of Magnoliae Officinalis Cortex in rats with ulcerative colitis (UC). First of all, the similarity of traditional Magnoliae Officinalis Cortex decoction and formula particles of different manufacturers was defined by using the IR fingerprint. The UC rat model was established and given Houpo formula particles of different doses and manufacturers, with the decoction for reference, in order to observe disease activity index (DAI), colon mucosa damage index (CMDI), pathologic changes, nitric oxide (NO), endothdin (ET), substance P, vasoactive intestinal peptide (VIP). Their intervention effects on UC rats were compared to study the difference between Sanjiu and Tianjiang Houpo formula particles, in order to demonstrate the feasibility of the pharmacodynamic evaluation method for Houpo formula particles. According to the results, Houpo formula particles showed similar pharmacodynamic actions with the traditional decoction. The pharmacodynamic comparison of Houpo formula particles of different manufacturers showed no statistical significance. The experiment showed that on the basis of the TCM compounds, a prescription dismantlement study was conducted to define target points of various drugs. The traditional decoction was selected for reference in the comparison of corresponding formula particles for their pharmacodynamic equivalence. This method could avoid controversies about single or combined boiling of formula particles, and give objective comments on the pharmacodynamic effect of the formula particles. The method is proved to be feasible.
