1.Gottron papules and interstitial lung disease in patients with dermatomyositis

Objective To assess the relationship between Gottron papules and interstitial lung disease (ILD) in patients with dermatomyositis. Methods Clinical data were retrospectively collected from 83 patients with dermatomyositis (DM), who were classified into two groups based on the presence of Gottron papules. Results The prevalence of Gottron papules and ILD was 59.0% and 42.2%, respectively in these patients. A significant difference was observed in the incidence of ILD between patients with and without Gottron papules (24.5% vs 67.6%, P<0.01). In the patients with ILD, the average value of diffusing capacity of the lung for carbon monoxide (DLCO) in those without Gottron papules was significantly lower than that in those with Gottron papules (60.9±13.5 vs 72.3±12.7, t=2.427, P<0.05). No significant difference was observed between the patients with Gottron papules and those without in muscle strength (4.0±0.9 vs 3.8±1.2), the level of serum creatine phosphokinase (1047.9±1402.7 IU/L vs 1359.7±1752.4 IU/L), lactic acid dehydrogenase (397.2±226.5 IU/L vs 402.4±197.0 IU/L), aspartate aminotransferase (97.6±98.0 IU/L vs 120.1±82.3 IU/L),C reactive protein (11.9±8.3 mg/L vs 15.5±9.8 mg/L), erythrocyte sedimentation rate (26.5±16.4 mm/l h vs 33.3±25.3 mm/l h), the incidence of muscle tendemess (63.3% vs 70.6%) or malignant melanoma (21.4% vs 6.5%). Conclusions Patients presenting Gottron papules tend to show a decreased incidence and severity of ILD. No significant association was observed between the presence of Gottron papules and the degree of muscle changes or dermatomyositis activity, and the presence of Gottron papules seems to have no obvious relationship to the development of malignant melanoma.
2.Comparison of the diagnostic value of routine electrocardiogram and dynamic electrocardiogram in the diagnosis of arrhythmia and heart rate variability in children

Objective To compare the application value of routine electrocardiogram and dynamic electrocardiogram in the diagnosis of arrhythmia and heart rate variability in children.Methods In our hospital,pediatric arrhythmia patients were given routine electrocardiogram and dynamic electrocardiogram.The relationship between arrhythmia and heart rate variability in children was analyzed.Results Supraventricular tachycardia (13.33%),ventricular tachycardia(5.56%),premature ventricular contraction (21.11%),premature atrial contraction (14.44%),block (12.22%) in the dynamic electrocardiogram were significantly better than the conventional electrocardiogram,and the differences were statistically significant x2=11.93,4.56,9.22,10.74,11.55,all P<0.05).There were significant differences in the heart rate variability between ventricular arrhythmias and non-ventricular arrhythmias in children[(78.85±0.88),(4.68±2.43),(24.78±2.56),(21.66±0.94), t=10.88,11.79,9.84,9.92,all P<0.05].Conclusion Compared with routine electrocardiogram, dynamic electrocardiogram has better diagnostic value for pediatric arrhythmia of ventricular tachycardia.The detection rate of ventricular tachycardia,ventricular tachycardia,premature ventricular contraction,atrial premature contraction,conduction block and other arrhythmias are obviously superior.And dynamic ECG measurement of heart rate variability is helpful to assess the severity of the disease.
3.The discussion of major points of nursing work and its implementation strategy in health manage-ment department

According to the discipline construction, Nursing and Health Management Department of Nanfang Hospital of Southern Medical University conducted exploration in its nursing work from the points of health management center nursing care, nursing staff management, physical examination process optimiza-tion, service concept and etiquette and other aspects, and at the same time conducted satisfaction survey. Survey and statistical results showed that the customer satisfaction reached more than 90%. It indicated that great achievements were made in the health management nursing work, and a complete set of health man-agement nursing model was formed which adapted to the change of medical model and the development of social economy.
4.Renal cell tumor and renal cyst(Report 64 cases)

Purpose:To clarify the association between renal cysts and renal cell carcinoma (RCC), we analyzed the clinical characteristics of patients with renal cell carcinoma and renal cyst. Methods:From May 1996 to January 2004, a total of 198 patients were hospitalized for renal cell carcinoma in our department, and 64 patients had both renal cysts and renal cell carcinoma (RCC). The clinical characteristics of the 64 patients were evaluated, and compared to 106 renal cell carcinoma (RCC)patients without renal cysts from the 198 patients in the same period. Results:Renal cysts were identified by preoperative ultrasonography, computerized tomography and magnetic resonance imaging in 37%(22/59)、24%(15/61) and 35%(6/17) respectively. Histopathological examination revealed renal cyst in 15 patients (23%). The sizes of renal cyst were 0.5 to 12 cm. The pathological examinations showed clear cell carcinoma in 61, chromophilic cell carcinoma in 2 and a combined type of clear cell carcinoma and chromophilic cell carcinoma in 1. 5.Progress on malignant transformation of hepatic stem cells

Compelling evidences have shown that various types of cancer,including hepatocellular carcinoma,are originated from stem cells.The occurrenee,chemoresistance,metastasis and recurrence of hepatocellular carcinoma are associated with a small population of cells called cancer ster cells.Although the origin of hepatocellular carcinoma and hepatic cancer stem cells remain controversial after a decade of intensive investigation,increasing scholars tend to believe that hepatocellular carcinoma and hepatic cancer stem cells can derive from malignant transformation of hepatic stem cells being exposed to various kinds of carcinogens.In this review,we mainly focus on the recent progress on malignant transformation of hepatic stem cells.
6.Construction of the prokaryotic expression system for the prtC gene of Porphyromonas gingivalis and analysis of the relationship between thelocal level of PrtC protein and the periodontal damage

Porphyromonas gingivalis is a specific causative agent of human chronic periodontitis. This anaerobe produce collagenase PrtC encoded by prtC. To constructed the prokaryotic expression system for the prtC gene from P.gingivalis so as to be used to explore antigenicity and immunoreactivity of prtC gene product as well as the relationship between the local level of PrtC and the periodontitis damage, the entire length of prtC genes fragment from the ATCC-33277 and 47A-1 strains of P.gingivalis was amplified by PCR. After T-A cloning and sequencing, the prokaryotic expression system for prtC was constructed by using pET32a plasmid and E.coli BL21DE3 strain. The expression of the target recombinant PrtC protein was induce with different concentrations of IPTG. Western blot assay was used to detect the antigenicity and immunoreactivity of PrtC protein, and ELISA assay was used to detect the PrtC level in the subgingival plaque samples of patients with chronic periodontitis. The experimental results showed that the nucleotide sequences of the prtC genes from ATCC-33277 and 47A-1 strains of P.gingivalis were entirely identical, and the sequence similarities of nucleotides and amino acids were 98.46% and 99.07% respectively. Under the induction of different concentration of IPTG, the output of recombinant expressed product PrtC may reach up to 50% of the total bacterial proteins. It was also proved that the recombinant PrtC could bind with antibody against the whole cell of P.gingivalis, and could induce the production of specific antibodies in rabbit. In 91.39% of the subgingival plaque samples, the PrtC could be detectable, in which the level of positive rates of detection was higher in severe cases of chronic periodontitis than that in the mild cases. So far, a prokaryotic expression system for the prtC gene from P.gingivalis with high expression efficiency was successfully constructed in the present study, and the expressed product PrtC possesses well antigenicity and immunoreactivity, suggesting the possibility to be used as the candidate antigen for developing the serological kit and P.gingivalis vaccine.
7.Influence of intratracheal instillation of curosurf on right ventricular function in neonatal respiratory distress syndrome

Objective To investigate the effect of intratracheal instillation of curosurf on neonatal respiratory distress syndrome ( NRDS ) in children with right ventricular function.Methods 52 patients with NRDS were retrospectively selected and divided into two groups according to different treatment.The patients in the conventional group were treated with nasal airway ventilation.Based on the conventional group, the curosurf group was taken with curosurf.26 cases were in each group.The blood gas index (PaCO2, PaO2, pH), inflammatory reaction (TNF-α, IL-10), SF of the two groups were compared, the complications and curative effect of the two groups before and after treatment were taken for statistics.Results There was no significant difference in pH value between the two groups at each time point.The PaO2 expression levels in the curosurf group at different time points after treatment were significantly lower than the conventional group (P<0.05).The levels of TNF-αand IL-10 in the curosurf group were more stable than those in the control group at different time points after treatment (P<0.05), and the degree of SF increasing at different time points were higher (P<0.05).The total effective rate 80.77% of the curosurf group was significantly higher than that of the conventional group 61.54%(P<0.05).The total complication rate 19.24% had no significant differences with the conventional group 23.08%.Conclusion Intratracheal instillation of CsA in the treatment of NRDS has the advantages of simple operation, little side effect, rapid recovery of blood gas index and inflammatory factors, so it is a feasible method for clinical treatment of NRDS.
8.The role of protein kinase CK2 in signaling transduction and tumourigenesis

Protein kinase CK2 is a ubiquitously expressed protein serine/threonine kinase present in all eukaryotes. It can intervene transversally into a multitude of signal pathways.It plays a key role in regulation of cell growth and apoptosis in a hierarchical and vertical manner. It was demonstrated that modest deregulation in the CK2 expression impacts a potent oncogenic potential to the cells.Moreover,inhibition of CK2 with specific chemical inhibitors or CK2-targeting antisense RNA may result in depression of tumor. Thus, studying the regulation mechanism of CK2 in tumorigenesis and developing new anti-cancer drugs by employing specific strategies against CK2 are of significant clinical value.
9.Progress in Vaccine Development of Hepatitis C Virus

Hepatitis C virus (HCV) accounts for the majority of cases of transfusion acquired hepatitis and may cause chronic hepatitis, cirrhosis and hepatocellular carcinoma. Currently, there is no vaccine against HCV and treatment is expensive and not always effective.The adaptive host immune response in viral clearance of HCV infection was described and the recent progress in vaccine development of HCV, focusing on the fields of DNA vaccine candidates, recombinant viral vectored vaccine candidates and combined (prime-boost) vaccine candidates were summarized. Some challenges and limitations of developing a HCV vaccine were also analysed. In summary, a promising approach of developing an experimental HCV vaccine to induce extremely potent and broad T-cell responses based on prime-boost strategy was presented.
10. Expression of protein kinase CK2 subunits in non-small-cell lung cancer tissues and its significance

Objective: Casein kinase II is also called protein kinase CK2. This study was to explore the mRNA expression and enzyme activity of CK2 alpha, alpha' and beta subunits in the tissues of non-small cell lung cancer (NSCLC), and investigate their correlations with clinicopathologic features of NSCLC. Methods: Immunohistochemical method (SP) was used to examine the expression of CK2 alpha, alpha' and beta subunits in paraffin-embedded speciments of 7 benign lesions, 35 primary lung carcinomas and matched pericancerous tissues. The mRNA expressions of CK2 subunits were evaluated by reverse transcription-polymerase chain reaction (RT-PCR) in 4 benign lesions, 10 primary lung carcinomas and matched peritumor tissues and the activity of CK2 was measured by using isotope incorporation assay. Results: The positive rates of CK2 alpha and beta subunits (62.9% and 57.1%) were significantly higher in NSCLC tissues than those in matched adjacent noncancerous tissues (11.4% and 25.7%, P <0.05) and benign lesions tissues (28.6% and 42.9%, P >0.05). The positive rate of CK2 alpha' subunit (65.7%) was higher in NSCLC tissues than that in matched adjacent noncancerous tissues and benign lesions tissues (42.9% and 42.9%, P >0.05). The expression levels of CK2 subunits had no correlation with gender, age, and pathologic stage (TNM) of the patients (P >0.05). The expression level of CK2 alpha' subunit was not related with cell differentiation and histological classification; the expression level of CK2 alpha subunit was associated with cell differentiation (r = 0.388, P <0.05) but not related with histological classification. The positive rate of CK2 alpha subunit was significantly higher in poorly differentiated group than that in moderately differentiated and well-differentiated groups (P <0.05). The positive rate of CK2 beta subunit was significantly higher in squamous cell carcinoma than that in adenocarcinoma (P <0.05) but it was not related with the degree of cell differentiation. The expression level of CK2 beta subunit positively correlated with that of CK2 alpha or alpha' subunit in NSCLC (r = 0.439, P <0.05; r = 0.45, P <0.05). The mRNA expression levels of the three CK2 subunits were higher in lung tumors compared with the corresponding pericancerous lung tissues and non-tumor benigh lesions tissues. The average activity of CK2 in the lung carcinomas was 5.4-fold higher than that in matched adjacent noncancerous tissues and 3.6-fold higher than that in benign lesion tissues. Conclusion: Protein kinase CK2 subunits are highly expressed in NSCLC and their overexpression have correlation with the tumorigenesis and development of NSCLC. As a biological marker, CK2 alpha or beta subunit may be better than CK2 alpha′.
