Objective:To evaluate the biological functions and underlying mechanisms of transcription factor PAX5 in promoting cell proliferation and anti-apoptosis in multiple myeloma cells.Methods:a PAX5 knockdown cell line was established by stable transfection of vector-based PAX5-siRNA into multiple myeloma IM9 cells.The expressing level of PAX5,p53,XBP-1 and c-Myc was examined by either or both Western blot and RT-PCR for the targets.Cell proliferation and apoptosis were assayed by MTT and flow cytometry,respectively.Results:PAX5 was expressed selectively in IM9 cells,but neither in another common used multiple myeloma KAS6 cells nor in the prostate cancer cells DU145 and PC3.Knockdown PAX5 led to a significant up-regulation of p53 and XBP1,but decreased c-Myc expressions in IM9 cells that were correlated with the increased sensitivity of drug-induced apoptosis and decreased proliferation rates when compared to the control cells.Conclusion:PAX5 is specifically expressed in IM9 cells,which promotes cell proliferation and anti drug-induced apoptosis.In addition to inhibiting the expression of p53 and XBP-1,PAX5 is found to induce expression of oncogene c-Myc in IM9 cells,and this finding indicates an undiscovered signal pathway that may contributes to the malignancy of Multiple Myeloma cells.

Objective To investigate the effect of the serums containing Xuefuzhuyu capsules and Xuesetong capsules on the proliferation of vascular smooth muscle cells (VSMC) induced by 1ysophosphatidie acid (LPA). Methods Rat VSMCs were obtainded by tissue-piece inoculation and divided into seven groups:blank serum group, Xuefuzhuyu (Xuesetong) capsules-containing serum group, LPA+ Xuefuzhuyu (Xuesetong, captopril) containing serum group and LPA+blank serum group. Effects of different groups on the proliferation of VSMCs was evaluated by MTT colorimetry. Analysis of cell cycle and DNA content were performed by flow cytometry. Results The optical density (OD) and the S phase fraction (SPF) of LPA+blank serum group and Xuefuzhuyu capsules-containing serum group were more increased than the blank serum group (P 0.05, P 0.05). Conclusion Xuefuzhuyu capsules and LPA promoted the proliferation of VSMCs in vitro. Xuefuzhuyu and Xuesetong capsules can inhibit the proliferation induced by LPA, and effect of Xuesetong capsules was greater. Part of the mechanism may be associated with the inhibition of DNA synthesis and preventing the transition from G0/G1 to S phase of VSMCs.

Objective The aim of study is to explore the TORCH infection status on the pre-pregnancy women and analyze the relationship with pregnancy outcome in Hongkou District,Shanghai,and provide the reference for the eugenic and superi-or nurture.Methods Enzyme linked immunosorbent assay (ELISA)was used to detect the specificity antibodies to Toxo-plasma (TOX),Rubelavirus (RUV),Cytomegalovirus (CMV)IgG and IgM in sera of the 3 500 pre-pregnant women in Hongkou District,Shanghai.Chi-square test was used for comparative analysis of the different rates.Results The positive rate of TOX-IgG,TOX-IgM,RV-IgG,CMV-IgG and CMV-IgM in pregnancy women from Hongkou district were 1.37%, 0.34%,89.7%,96.2% and 0.60% respectively.The total positive rate of TOX and (or)CMV IgM was 0.94%.According to age group,the results showed that TORCH infections with three pathogens associated with the pre-pregnancy women’s age,and the difference was statistically significant (χ2=102.9,P<0.01).The abnormal pregnancy rate of TORCH infec-tion-positive pregnancy women (25.0%,3/12)was higher than that of TORCH-negative pregnancy women (6.62%,27/408),and the difference was statistically significant (χ2 =4.43,P=0.04).Conclusion The infection opportunity of TORCH in pregnant women in Shanghai Hongkou district was obviously existent and the TORCH infection status could effect on pregnancy outcome.TORCH testing has important practical significance as a routine examination of the pre-preg-nancy healthy service.

Objective In order to improve the diagnosis accuracy , 18F-FDG PET/CT results of 169 cases of lung lesions were analyzed. Methods The data of 169 pathological diagnosed patients were collected. SPSS 18.0 was used for the data analysis. Results In 169 cases, 122 cases were proved malignant by patholog-ical diagnosis, in which 110 cases were correctly diagnosed by PET/CT. 47 cases were proved benign by patho-logical diagnosis, in which 15 cases were correctly diagnosed by PET/CT. The sensitivity was 90.2%, specificity was 31.9%, and the accuracy was 74.0%. Conclusion In order to improve the accuracy rate of PET/CT exam-ination and reduce the existence of false positive cases and false positive cases , we should estimate SUV (max) objectively and diagnose according to the size of the lesion , clinical history and laboratory results.

Based on new material and technic, the design of general structure, stretcher fixture, vibration isolation, carriage baffle fixture, ergonomical parameters selection for a new kind of stretcher suspension named added equipment for transporting the wounded on the truck is carried out. The results of mechanical performance experiment, reliability experiment, road adaptability test, road comfort test, application experiment by troops indicate that the added equipment is safe, reliable, convenient for use and fairly comfortable to persons lying on it. Thus a new kind of equipment fit for the army to transport the wounded on the truck under the field condition is developed.

Objective To investigate the methylation status of promoter CpG islands of the estrogen receptors (ERs) in leukemia cell lines,including 3 promoters of ER? (ER?-A,-B and -C) and one promoter of ER?,and explore the possible relationship between the mRNA expressions of the promoters and the CpG methylation. Methods With RT-PCR and methylation specific PCR (MSP),the mRNA expressions of above promoters and their status of methylation in leukemia cell lines HL-60,K562,Jurkat,THP-1,U937 and MOLT-410,and health controls with or without treatment of 5-AzA-2′-Deoxycitidine (5-Aza-DC). Results Promoters of ER were all methylated or semi-methylated in the 6 strains of leukemia cells. Only the ER?-A was inactivated in these leukemia cell lines and was unmethylated in all 10 health controls,and the expression was recovered after the treatment of 5′-Aza-DC. Conclusion The CpG islands of ER promoter are hypermethylated in 6 strains of leukemia cells,and this inactivate the expression of ER?-A,which can be activated by the treatment of 5′-Aza-DC.

AIM To investigate whether pretreatment with prostaglandin E 1 (PGE 1) might protect myocardium against peroxidative injury in early phase and in delayed phase. METHODS Rats were pretreated with PGE 1 (25 or 150 mg?kg -1 ). Isoproterenol (ISOP)(80 mg?kg -1 ) were injected peritoneally (ip) to induce acute myocardial infarction 20 minutes (early phase) or 24 hours (delayed phase) after pretreatment. Hearts were removed punctually 24 hours after ip ISOP and were assayed for content of malonaldehyde (MDA), activity of glutathione peroxidase (GSH Px) and superoxide dismutase (SOD). RESULTS ISOP caused myocardial injury with increased MDA content and SOD activity, and decreased GSH Px activity. Objective To investigate the effects of Thomas solution in combination with nitroglycerine of different concentrations on rat cardiac preservation for 8 h. Methods Thirty Wistar rats were selected,and Langendorff heart perfusion models were established.Rats were randomly divided into Thomas solution group,Thomas solution+4?10-6 mol/L nitroglycerine group and Thomas solution+4?10-5 mol/L nitroglycerine group(n=10).After hypothermic preservation(4 ℃)for 8 h,the cardiac function of each group was determined.Tissues of left ventricular wall were obtained,and changes of myocardial ultrastructure were observed by electron microscopy. Results Compared with Thomas solution group,the cardiac function was significantly improved and the myocardial ultrastructure injury was less severe in Thomas solution+4?10-5 mol/L nitroglycerine group after hypothermic preservation(4 ℃)for 8 h,while there was no significant difference in myocardial ultrastructure injury and cardiac function between Thomas solution group and Thomas solution+4?10-6 mol/L nitroglycerine group. Conclusion Thomas solution in combination with 4?10-5 mol/L nitroglycerine can significantly improve the preservation effects for isolated hearts in rats.

Objective To explore the relationship between single nucleotide polymorphisms (rs12953-717) of SMAD7 and susceptibility of non-small cell lung cancer (NSCLC) in Chinese Han population.Methods A single nucleotide polymorphisms (rs12953717) from SMAD7 was detected via Sequenom system in 528 NSCLC cases and 762 healthy controls.Data was statistically analyzed by unconditional Logistic regression method.Results rs12953717 had significant differences between non-small cell lung cancer patients and the controls.Compared with CC/TT (CC combined with TT) genotype,the adjusted odds ratio for the CT genotype was 4.107 (95％ CI:3.206 ～ 5.260,P =0.000 1).Smokers had a 2.004 odds ratio (95 ％ CI =1.583 ～ 2.537,P =0.000 1) of NSCLC compared with the controls.There was a 10.074-fold increased risk of NSCLC among the subjects with CT genotype and smokers.Conclusion The polymorphism of rs12953717 may have relation with risk of NSCLC.Heterozygote (CT) is a susceptibility genotype of NSCLC.Smoking is one of the risk factors of NSCLC.Smoking and CT genotype have synergistic effects on NSCLC susceptibility.

Background Recently,there were many studies on corneal innervations during mammalian development.However,there were fewer studies on discussing corneal innervations before and after mouse eye openings.Objective The present study was to investigate the change in the regulation of corneal nerve fiber length and density before and after mouse eye openings to offer a basis for clinical research in human.Methods Thirty SPF C57BL/6 mice were divided into postnatal 1 day(P1 d),P7 d,P13 d(1 day before eye opening),P14 d(eye halfopened),P17 d(1 day after eye opening)and P23 d(7 day after eye opening)groups,with 5 mice and 10 eyes for each group.Entire corneal stretches were prepared and immunostaining with an anti-neuron-specific β-Ⅲ tubulin antibody was performed to label the corneal nerve fibers.Confocal microscopic pictures from the corneal dorsal-nasal region (DN),dorsal-temporal(DT),ventral-nasal region(VN)and ventral-temporal(VT)were taken using Delta Vision Core.From these pictures,the mouse corneal area,total length and density of nerve fibers in the 4 regions were calculated.The use of the animals complied with Statement of ARVO.Results Corneal areas of P1 d,P7d,P13 d,P14 d,P17 d and P23 d mice were(0.404±0.007),(1.362±0.154),(1.573±0.080),(1.603±0.046),(1.847±0.052),(2.445±0.798)mm2,respectively ; the total lengths of nerve fibers were(3.718±1.044),(19.065±3.350),(23.687±0.907),(27.309±2.477),(31.989±3.976),(41.214±1.573)mm,respectively ; the densities of nerve fibers were(9.592±1.138),(14.506±1.908),(15.088±1.241),(16.772±1.897),(16.821±2.102),(17.660±1.216)mm/mm2,respectively,all showing significant increases with age(F =22.906,P =0.000 ; F =0.424,P =0.000 ; F =2.375,P=0.000).A positive correlation of the increasing corneal areas and increasing lengths of nerve fibers was found(r=0.983,P＜0.01).Nerve fiber densities in the four corneal regions significantly increased with age(DN region:F =0.159,P =0.000 ; DT region:F =2.1 72,P =0.001 ; VN region:F =1.998,P =0.000 ; VT region:F=2.352,P=0.000).From P13 d to P14 d,the corneal nerve fiber densities in the DN region decreased by 6.0％ without significant difference(t =0.589,P =0.572); and the corneal nerve fiber densities in the DT region,VN region and VT region decreased by 4.6％,5.5％ and 0.1％,respectively,without significant difference from P14 d to P17 d(t=0.549,P=0.596;t=0.701,P=0.501 ;t=-0.100,P=0.919).Conclusions The development of nerve fibers in the whole cornea or the four corneal regions is influenced by eye opening in mouse to various extents.From P13 d to P14 d,the corneal nerve fiber densities in the DN region decreased by 6.0％ without significant difference.From P14 d to P17 d,the corneal nerve fiber densities in the DT region,VN region and VT region decrease by 4.6％,5.5％ and 0.1％,respectively,without significant difference.Afterwards,the growth of nerve fibers increased in pace and the growth rate is recovered.
