1.Advances in Study on Pathogenesis of Irritable Bowel Syndrome

Irritable bowel syndrome(IBS)is a commonly seen functional gastrointestinal disorder. Its main clinical manifestations were abdominal pain,abdominal distention and altered bowel habits. Currently,the pathogenesis of IBS has not been clarified. Studies showed that IBS was caused by many factors,including life style,gene polymorphism,food hypersensitivity,psychological factors,brain-gut axis abnormality and intestinal flora disorder. This article reviewed the advances in study on pathogenesis of IBS.
2.The influencing factors and reference value of sympathetic skin response in healthy adults

Objective To investigate the influencing factors and reference value of sympathetic skin response (SSR) in healthy adults. Methods SSR was studied in 40 normal subjects,with the waveform and onset latency analyzed. Five of them were treated with different intensities of electrical stimulus to observe the subsequent onset latency. Results ① SSR was present in all the hands and it was also true in all the feet except for one case. The waveforms were biphasic or triphasic in the hands and biphasic or triphasic or monophasic in the feet. ② No significant difference was seen between the left and right sides in amplitude and onset latency. ③ Age,height,weight and gender had no significant influences on onset latency of SSR. ④ The normal value on onset latency in the hands was within 1 664 ms and 2 234 ms in the feet,respectively. ⑤ The electrical stimulus intensity had no obvious effect on onset latency. Conclusion Onset latency is an important parameter in evaluating abnormality while amplitude is only a reference index.
3.Hospital phenomenon during SARS and the construction of hospital credit ethic

The fluctuation of medical staff thoughts and hospital rapid action during SARS epidemic, we name it "hospital phenomenon during SARS", has its specific characteristics. We analyzed this phenomenon from the following points: the conflict between panic response and the duty of the profession, the contradiction between the need to control infection and the available conditions, the special following phenomenon post - diagnosis of SARS and the hospital routine activity and SARS prevention and cure. We conclude that at least three fields should be improved, that is to strength education of duty morals, to normalize the medical procedure, and to build the dual system of hospital' s routine run and rapid action, in order to constructing hospital credit ethic.
4.MOOC-based flipped classroom construction for information literacy education course in colleges and universities

It is inevitable for colleges and universities to reform their information literacy education course with the coming of education informationization time.The construction elements of MOOC-based flipped classroom for information literacy education course in colleges and universities were studied in aspects of teaching design, classroom activities, hardware, teachers literacy based on the analysis of its necessity and feasibility, and strategies were put forward for colleges and universities to construct their multi-mainstay plus multi-element flipped classroom for information literacy education course.

6.Oxymatrine suppresses p-STAT1/PIAS1 signaling in LPS-induced human mesangial cells proliferation

Objective To observe the effect of oxymatrine(OM) on the expressions of p-STAT1 and PIAS1 signaling molecules at protein and mRNA levels in the proliferation of the human mesangial cells(HMC) induced by lipopolysaccharide(LPS) and explore the relationship between them.Methods HMCs were primarily cultured from a 4-month-old aborted human fetus(with informed consent and approved by the Ethics Committee of Lanzhou University),and then divided into 3 groups,that is,control group,LPS group(10 ng/ml) and OM group(LPS 10 ng/ml and OM 320 mg/L).After cultured for 12,24 and 48 h respectively,HMC proliferation were analyzed by methyl thiazolyl tetrazolium(MTT) assay and type Ⅳ collagen in the supernatants were detected by ELISA.At the same time points,the cells lysates were collected for the mRNA and protein expressions of p-STAT1 and PIAS1 by real-time quantitative RT-PCR and Western blot analysis.Results The cell proliferation of LPS group was faster and the type Ⅳ collagen protein was increased more than the control group(P

Objective:To improve teaching quality of laboratory teaching in medical cell biology on foreign students.Methods:To improve teaching language,teaching material,teaching methods and so on.Results:We got significant improvement on laboratory teaching of foreign students in medical cell biology.Conclusion:In order to improve teaching quality of laboratory teaching in medical cell biology on foreign students,we should improve on many different ways and accumulate experiences for future teaching.
8.Effects of Matrine on STAT1, STAT3, CTGF, PDGF expression in fetal glomerular mesangial cells under lipopolysaccharide

Objectives To determine the effects of Matrine (Ma) on signal transducers and activators of transcription (STAT) 1, 3, connective tissue growth factor (CTGF) and platelet -derived growth factor (PDGF) in glomerular mesangial cells (MC) induced by lipopolysaccharide (LPS), and to explore the protective mechanisms of Ma. Methods Ma were added to cultured glomerular mesangial cells which were exposed to LPS for 12, 24 and 48 hours. Real time polymerase chain reaction were used to determine STAT1, 3, CTGF, PDGF mRNA, The protein expression of STATI, 3 and p-STAT1, 3 were observed by Western blot. Results Compared with the control group, MC proliferation of the LPS group (10μg/ml) significantly increased, which were suppressed in the Ma (320 μg/ml) group (P < 0.01) at 12, 24, 48 hours. The expression of STAT1, 3, CTGF, PDGF mRNA and the levels of p-STAT1 , p-STAT3 protein was significantly increased in MC under LPS medium at 12, 24, 48 hours, which were obviously lower in Ma group than those of the LPS group, especially at 24, 48 hours. Conclusions The protective mechanisms of Matrine is considered to down-regulate the expression of STAT1, 3, CTGF and PDGF.
9.The effect of sciatic nerve injection of dexmedetomidine on the nociceptive behavior and spinal c-fos expression in formalin mice

Objective To investigate the effect of sciatic nerve injection of dexmedetomidine on the nociceptive behavior and spi-nal c-fos expression in formalin mice .Methods Adult male Kunming mice ,20-25 g ,were divided into 4 groups randomly :mice in group A and D were given an injection of NS (10 μL) and 0 .1 μg dexmedetomidine in 10 μL adjacent to the right sciatic nerve , sepretely .Mice in group B received intacutaneous injection of 10μL formalin(5% ) in the right hindpaws .Mice in group C were giv-en an injection of 0 .1 μg dexmedetomidine in 10 μL adjacent to the right sciatic nerve 15 min before formalin injection in the right hindpaws .Paw licking/biting time was counted every 5 min for an hour after hindpaw injections ,calculated pain score .1 hour after behavior tests ,the L4-5 of spinal cord were harvested to immunofluorescence for c-fos expression in the dorsal corn of spinal cord . Results Mice in group A、C and D showed no obvious foot licking/biting behaviors and a low level of c-fos expression in the dorsal corn of spinal cord .Compared with group A ,mice in group B displayed obvious two phases foot licking/biting behaviors(P<0 .05 , both in Phase Ⅰ and Phase Ⅱ) and a significant increasing expression of c-fos(P<0 .05) .Compared with group B ,dexmedetomi-dine injected to the adjacent of right sciatic nerve decreased the pain score and spinal c-fos expression in goup C(P<0 .05) .Conclu-sion Injection of dexmedetomidine to the right sciatic nerve alleviated intraplantar injected formalin-induced acute inflammatory pain ,and reduced the expression of spinal c-fos protein .

Objective To study the effect of oxymatrine on p-STAT3 and PIAS3 signaling molecule and it's mRNA expression in the proliferation of the human mesangial cells (HMCs) induced by lipopolysaccharide(LPS) and to explore their relationship. Methods HMCs were divided into three groups: control group, LPS group and oxymatrine group. HMC proliferation was detected by methyl thiazolyl tetrazolium (MTT) assay. Type Ⅳ collagen in the supernatants of the cultured HMCs was detected by ELISA at 12, 24, 48 hours respectively. At the same time, the protein and mRNA expressions of p-STAT3 and PIAS3 were measured by Western blot and real-time quantitative RT-PCR. Results The cell proliferation, the mRNA and protein expression of type Ⅳ collagen, p-STAT3 in LPS group were increased compared with the control group (P<0.01), but they were decreased in oxymatrine group (P < 0.01). The expressions of protein and mRNA of PIAS3 in LPS group were decreased significantly compared with control group (P<0.01), but they were increased in oxymatrine group (P<0.01). Conclusion Oxymatrine can down-regulate the expression of p-STAT3 and up-regulate the expression of PIAS3, which plays an important role in the process of LPS-induced HMCs proliferation.
