1.The effect of early cognitive rehabilitation on cognitive, physical function and quality of life in ICU survivors

Objective To explore the clinical effect of early cognitive rehabilitation on cognitive,physical function and quality of life in the patients transferred from intensive care unit (ICU).Methods A total of 120 cases of patients who were transferred from ICU to general wards was randomly divided into the control group and the observation group with 60 cases in each group.The control group was given symptomatic treatment according to their original disease,without cognitive intervention.On the basis of the control group,the observation group was treated with cognitive rehabilitation training,2 times / week,2 h/ times,12 weeks of treatment.The cognitive function,physical function and quality of life of two groups were compared with the memory and executive screening scale (MES),the daily activity scale (ADL),and the concise Health Survey (SF-36) before and after treatment.Results Compared to before treatment,after treatment,MES,SF-36 (in addition to bodily pain) and ADL scale project score in the observation group was significantly higher,SF-36 bodily pain scores decreased significantly,all the differences were statistically significant (P < 0.05);Compared to the control group,after treatment,the MES,SF-36 (in addition to bodily pain) and ADL scale scores in the observation group of were significantly increased,the bodily pain score of SF-36 in the observation group compared with the control group decreased significantly,all the differences were statistically significant (P < 0.05).Conclusions Early cognitive rehabilitation can significantly improve the cognitive,physical function and quality of life in the patients transferred from ICU,and it is worthy of clinical reference.
2.Comparison of soft tissue changes in angle class Ⅰ southern Chinese females with ELSID treated by orthodontic extraction

Objective To find out the changes of the soft tissue profile in Guangdong women with Angle's I teeth uncovered by lips after extraction of 4 first bicuspid teeth and to evaluate its significance. Methods Fifteen cases of adult Guangdong women with Angle's I teeth uncovered by lips were enrolled in this retrospective study. The X-ray film of lateral cephalometry was used to analyze the soft tissue profile in pre- and post- corrective tooth extraction. The paired t-test was statistically applied for comparison between the changes of lip tissue in pre- and post- correction. Results The changes occurred obviously in the angles of nose vs. lip and upper vs. lower lips, angle Z, and the length, thickness, esthetical plane distance and convex distance of upper and lower lips, but not in the angles of face and facial convex, basic angle of upper vs. lower lips and chin thickness. Conclusilons The facial dislocation and figure of these women have been positively improved after extraction of 4 first bicuspid teeth in spite of the larger thickness of lips than before correction, the just partly shortened distance between lips and the upper and front teeth still exposed and uncovered by lips to some extent. Further improvement depends on functional exercise of lip muscles.
3.Development of early human fetal testes after xenografting into mice

Objective To investigate the developmental feasibility of early human fetal testes (<3 months) using xenografting technique and to acquire an accessible donor derivation that is essential for studying human germ cell development. Methods Nine testes from 10-13 weeks aborted fetus were grafted under the back skin of 6 castrated nude mice. Grafts were collected at different time point according to the growth of the donor tissues and the health condition of the recipients. Morphological and histological analyses were performed for the observation of the development of grafted immature testicular tissues. Results The mass of grafts was increased from about 5-7mg to 84.1mg (the biggest). Six of 9 testes were to be in developing. Histological observations showed a significant expansion of seminiferous tubules from (44.26±3.14)μm to (77.69±7.47)μm. Cells dispersedly distributed in seminiferous cords at the time of grafting migrated towards the basal part of seminiferous epithelium. Some germ cells with spermatogonium-like characteristics located on the basement membrane. Sertoli cells were in stages from immature into matured with abundant cytoplasm which were orderly arranged around spermatogonia forming a niche-like structure. Conclusion Testes from early aborted human fetus grafted under the back skin of castrated nude mice showed further development and therefore could be used as an easier accessible donor tissues for the investigation of human spermatogenetic mechanism.
4.Investigation of tick bites in outpatients with fever from health care facilities in Tengchong County

Objective To study the prevalence of tick bites and relative factors of outpatients with fever from health care fa?cilities in Tengchong County. Methods From July to August,2014,the outpatients with fever in five health care facilities namely Tengchong Center for Disease Control and Prevention,Hehua Health Center,Jietou Health Center,Qushi Health Cen?ter and Xinhua Health Center were investigated by questionnaires. The factors related to tick bites were explored with the univari?ate analysis and multiple regression models. Results Totally 884 effect questionnaires were acquired. Among the 884 partici?pants,85(9.6%)had experienced tick bites. The frequency of tick bites was associated with locations,gender,age group,own?ership of cattle,working at foreign country,firewood cutting and lumbering in the forest(all P<0.05). The multiple regression revealed that the male and people raising cattle,working at foreign country,engaging in firewood cutting and lumbering in the forest had a more risk for tick bites. Conclusions Tick bites are common in the residents of Tengchong County. The risk of be?ing bitten varies in different populations. The local health departments should promote health education in the high?risk popula?tion to reduce the risk of infecting tick?borne diseases.
5.Conversion of human umbilical cord blood-derived cells into hepatocyte-like cells in a culture system mimicking hepatic injury

Objective:To explore the effect of the microenvironment induced by damaged mouse hepatic cells on the conversion of human umbilical cord blood-derived cells into hepatocyte-like cells. Methods: A hepatic injury-like microenvironment was mimicked using carbon tetrachloride damaged mouse hepatic cells, where mononuclear cells (MNC) from human umbilical cord blood were cultured in a compartment separated by trans-well membrane. Histochemical staining, reversed transcription-polymerase chain reaction (RT-PCR) and gene sequencing were performed for the information on the conversion of human umbilical cord blood MNC. Results: A number of PAS positive stained cells in MNC co-cultured with damaged mouse hepatic cells were observed after 72 h. Cells expressing mature hepatocyte markers, human albumin (hALB) and human GATA-4 (hGATA-4) mRNA were detected by RT-PCR, which was further confirmed with sequencing. Relevant control groups, MNC co-cultured with normal mouse hepatic cells and MNC cultured alone remained negative. Conclusion: The culture system using damaged mouse hepatic cells as stimulator could be a potential in vitro system for the conversion of human umbilical cord blood-derived cells into hepatocyte-like cells.
6.Effects of cytosine deaminase and HSV-tk genes in combination on gallbladder cancer cells mediated by retroviral vector

Objective To investigate cell-killing and bystander effects on gallbladder cancer (GBC-SD) cells transferred with double suicide genes (CD and HSV-tk). Methods CD and HSV-tk genes were transfected into PA317 cells using lipofectamine. GBC-SD cells were infected with viral supernatant. GBC-SD/CD+tk cells were treated with 5-FC and/or GCV. GBC-SD/CD+tk and GBC-SD were inoculated subcutaneously into the nude mice respectively and treated with GCV and 5-FC for 21 days. Results The killing effect of 5-FC and GCV in combination on GBC-SD/CD+tk cells is more effective than using 5-FC or GCV alone. The local bystander effect is significant while the distant bystander effect was not obvious. Conclusion The cell-killing efficacy of chemotherapy on GBC-SD transfected with double suicide gene was significantly increased, and the bystander effect was obvious.
7.Effects of pHGF on Renal Cellular DNA Synthesis after Partial Hepatectomy in Rats

The effects of pro-hepatocyte growth factor (pHGF) on the changes of renal cellular proliferative cycle of partial hepatectomized rats were observed by flow cytometry (FCM). S phase fraction (SPF) of control rats (group A) accounted for 7.58 % and increased gradually within 6 h, following a peak at 12th or 36th h after operation, but in pHGF-treated rats (group B) the peak appeared at 24th h after operation. Proliferation index (PI) of group A was 13.2%before partial hepatectomy, increased within 6 h and reached a peak at 12th or 36th h after operation, and in group B the peak appeared at 48th h after operation. There were significant differences in SPF and PI between two groups (P<0.01). These findings suggest that pHGF may nonspecifically promote the DNA synthesis of renal cells.
8.Comment on Prescription of Narcotic Analgesics in Patients with Cancer Pain

Objective To investigate current status of narcotic analgesic use in patients with cancer pain in our hospital, in order to promote the rationalization of prescribing narcotic drugs and the rational use of analgesics. Methods A total of 400 narcotic prescriptions ( 100 of outpatients and 300 of inpatients ) in Tongji hospital were randomly selected from March to May in 2014.Basic index and consumption of anesthetic drugs, money consumption distribution on different tumor types, and unreasonable prescription were analyzed. Results The overall prevalence of irrational prescriptions was 11. 00%, with 13.00% from inpatient and 10. 33% from outpatient, respectively. Reasons for irrational prescriptions mainly included vague instructions on dosage and administration, incomplete clinical diagnosis, lack of pharmacist check and inappropriate dosage. In the basic indexes, proportions of reasonable prescription and drugs within the national formulary were higher in the inpatient than in the outpatient service.The average money consumption of outpatient prescription was significantly higher than that of inpatient prescription.Most types of diagnosis were lung cancer in outpatient and inpatient departments, and total cost of lung cancer drugs was the highest.Morphine hydrochloride of outpatient had the highest average consumption, followed by codeine phosphate of the outpatient and the inpatient, and oxycodone hydrochloride controlled-release tablets cost more in inpatient. Conclusion The narcotic analgesic used in our hospital is basically rational. Most doctors prescribe according to the guidelines, but standardized training of prescription for cancer patients with pain medication needs to be strengthened, and supervision of narcotic analgesic medication should be enhanced.
9.Effect of Ginsenoside-rg1 on Rat's Cardiomyocytes With its Mechanism of Signal Pathwayin vitro

Objective: To investigate the effect of ginsenoside-rg1 (G-Rg1) on rat’s cardiomyocytes H9c2 with its mechanism of signal pathwayin vitro.
Methods: H9c2 cells were cultured and treated in different conditions by following groups:①Blank control group,②Hypoxia alone group, the cells were treated for (2, 6, 12, 24, 48) hr respectively,③G-Rg1 group, the cells were treated by G-Rg1 at (5, 10, 50) μmol/L respectively,④YC-1 group, which is the speciifc inhibitor of hypoxia inducible factor-1α (HIF-1α),⑤YC-1 + G-Rg1 group,⑥Wortmannin group, which is the speciifc inhibitor for protein kinase B (Akt) phosphorylation and⑦Wortmannin + G-Rg1 group. Each experiment was conducted with 5 replicates. The effects of G-Rg1, hypoxia and YC-1 on cell activity and injury were studied; intracellular mRNA expressions of HIF-1α, glucose transporter-1 (GLUT-1) and heme oxygenase-1 (HO-1) were examined by RT-PCR; protein expressions of HIF-1α, GLUT-1, HO-1, activating transcription factor-6 (ATF-6), CCAAT/enhancer binding protein homologous protein (CHOP) and Akt with its signal pathway factors were measured by Western blot analysis.
Results: The time of hypoxia was negatively related to cell activity (r=-0.8580,P<0.05) and positively related to LDH overlfow rate (r=0.9201,P<0.05). G-Rg1 (10μmol/L) group showed increased cell activity than Hypoxia alone (24 hr) group (87.8% vs 62.6 %,P<0.05), while decreased LDH overlfow (25.0% vs 74.8%,P<0.05), and up-regulated mRNA expressions of HIF-1α, GLUT-1 and HO-1, P<0.05. YC-1+ G-Rg1 group had decreased cell activity than G-Rg1 group (68.0% vs 87.8%,P<0.05), while increased LDH overlfow (56.4% vs 25.0%,P<0.05). Meanwhile, YC-1 clashed the effect of G-Rg1 on protein expressions of HIF-1α, GLUT-1, HO-1, ATF-6 and CHOP,P<0.05; wortmannin clashed the effect of G-Rg1 on protein expressions of HIF-1α, CHOP,P<0.05 and suppressed the two phosphorylation sites for Akt activation,P<0.05.
Conclusion: G-Rg1 may protect rat’s H9c2 cellsin vitro by activating expressions of HIF-1α with its downstream factors and inhibiting endoplasmic reticulum stress, which might be related to the effect of G-Rg1 on Akt activation.
