1.Study on the postmortem stabiliity of fibronectin for the diagnosis of myocardial infarction

Experimental acute myocardial ischemia model of dog was established, and the postmortem stability of fibronectin for the diagnosis of mycardial infarction was studied with immunohistochemistry and image analysis technique. The results showed that the positive reaction areas of fibronectin in ischemic myocardial tissues decreased along with the prolongation of postmortem interval, but positive reaction of fibronectin could still be found in ischemic myocardia kept for 4 weeks postmortem. No positive reaction for fibronectin could be found in normal myocardia when kept for different times. So fibronectin is a quite stable marker for the postmortem diagnosis of myocardial infarction, and is of practical value in the forensic practice.
2.Immunohistochemical study of C5 complement in experimental early myocardial ischemia

Experimental acute myocardial ischemia model of rat was established, and the changes of C5 complement in the ischemic myocardia were studied with immunohistochemistry and image analysis technique. The results showed that the positive reaction of C5 could be observed in ischemic myocardia at 15 min after ischemia, and the positive reaction area increased along with the prolongation of the ischemic period. It is concluded that the positive reaction of C5 in cardiomyocytes is a quite sensitive marker of early myocardial ischaemia.The immunohistochemical detection of C5 in cardiomyocytes will be a meaningful tool for the postmortem diagnosis of early myocardial ischaemia.
3.Study on the fibronectin for the diagnosis of myocardial infarction

In order to explore the value of fibronectin (Fn) in the postmortem diagnosis of myocardial infarction.The changes of Fn staining in normal, infarcted and other non infarcted myocardial injuries resulted from myocarditis, mechanical asphyxia, electrocution, hemorrhagic shock, cardiac contusion and organophosphate poisoning were studied with immunohistochemistry and image analysis system. The results showed that positive Fn staining could only be observed in groups of myocardial infarction and myocarditis, but could not be found in groups of mechanical asphyxia, electrocution, hemorrhagic shock, cardiac contusion, organophosphate poisoning. It is indicated that positive reaction of Fn could be affected only by myocarditis, so it is quite specific for the diagnosis of myocardial infarction.
4.Construction of eukaryotic expression plasmid pEGFP-N3-M-IL-2(88 Arg,125 Ala) and its effects on Glioma cell line U87

Objective:To construct a recombinant eukaryotic expressive vector of pEGFP-N3-M-IL-2(88 Arg,125 Ala),and to study the expression of this gene in the Glioma cell line U 87,and to detect its antitumor activities of the fusion protein M-IL-2(88 Arg,125 Ala).Methods:The target fusion gene M-IL-2 (88 Arg,125 Ala) was amplified by PCR from pPICZαA/M-IL-2 (88 Arg,125 Ala) and cloned into pEGFP-N3 vector after digestion to construct recombinant eukaryotic expressive vector pEGFP -N3-M-IL-2( 88 Arg,125 Ala).And then recombinant plasmid pEGFP-N3-M-IL-2(88Arg,125Ala) was transfected into Glioma cell U87 by LipofectamineTM2000 immediately after it was confirmed by restrictive enzyme analysis and sequencing .RT-PCR and Western blot were used to confirm expression of the fusion gene in the U87.Prohibitory effect of recombinant M-IL-2(88Arg,125Ala) protein on U87 was assessed by CCK-8 assay.Results:Restrictive analysis and sequence analysis revealed that M-IL-2(88Arg,125Ala) fusion gene was cloned into the vector pEGFP-N3 suc-cessfully,fusion gene M-IL-2(88Arg,125Ala) could express in U87 cells and could inhibit the growth of U87 cells.Conclusion:The eu-karyotic expression plasmid pEGFP-N3-M-IL-2( 88 Arg,125 Ala) was constructed and expressed in U 87 cells successfully ,the fusion protein could inhibit the growth of U87 cells.We laid a foundation for further research of gene M-IL-2(88 Arg,125 Ala).
5.Effect of modified Sanjia-Fumai decoction on the endocrine and immune function of patients with climacteric syndrome

Objective To observe the effect of modified SanJia-Fumai decoction with hormone replacement therapy on the endocrine and immune function of patients with climacteric syndrome. Methods A total of 84 patients with climacteric syndrome were divided into two groups according to the random number table, 42 cases in each group. The control group was treated with hormone replacement therapy. While the observation group was treated with modified Sanjia-Fumai decoction on the basis of the control grop. Both groups were treated for 4 weeks. The efficacy by scoring the changes of the patients clinical symptoms and signs was evaluated, and the serum hormones (LH, FSH, E2) levels were detected by radioimmunoassay, and the immune function index (CD3+, CD4+, CD8+) levels by immunofluorescence method, and the IL-2 by ELLSA. Results After treatment, the total efficiency of observation group was 92.8% (39/42), while the control group was 73.8% (31/42), and the difference was statistically significant (Z=-2.736, P=0.003). After treatment, the scores of hot flashes, menstrual disorders, insomnia and dreaminess, hot sweating, restlessness and dry mouth constipation in the observation group were significantly lower than those in the control group (t were 7.745, 10.364, 11.201, 16.765, 18.442, 18.625 respectively, P<0.001); the Serum LH and FSH levels in observation group were significantly lower than those in control group (t were 4.837, 5.016 respectively, P<0.01); the E2 level was significantly higher than that of control group (t=7.309, P<0.01). The levels of CD3+, CD4+, CD4+/CD8+, IL-2 in the observation group were significantly higher than those in the control group (t were 5.139, 6.083, 6.158, 5.932 respectively, P<0.01); The level of CD8+ was significantly lower than that of the control group (t=5.994, P<0.01). Conclusions The modified SanJia-Fumai decoction with hormone replacement therapy can regulate the endocrine function, improve the body's immunity and improve clinical symptoms of climacteric syndrome patients.
6.Research progress on pathogenesis and molecular targeted therapy of radiation-induced lung injury

Radiation-induced lung injury is a common complication of radiotherapy for thoracic tumor, usually involves the radiation-induced pneumonitis at early stage and radiation-induced pulmonary fibrosis at late stage, which seriously affects the treatment and prognosis of patients. At present the common treatments for radiation-induced pulmonary fibrosis include anti-inflammatory therapy, glucocorticoid therapy, antioxidant therapy and so on. Recentlywith the further research of radiation-induced pulmonary fibrosis, more and more attention has attracted in molecular targeted therapy. Molecular targeted inhibitors is a new class of drugs for the treatment of radiation-induced lung injury, mainly targeting a variety of cytokines, signaling pathways, tyrosine kinase receptors and other targets. This article systematically reviews the pathogenesis and molecular targeted therapy of radiation-induced lung injury.
7. Risk factors for large hematoma complication caused by percutaneous renal biopsy which guided by ultrasound and related nursing care

Objective:
To analyze the risk factors for large renal hematoma caused by percutaneous renal biopsy (PRB) in order to provide evidence for early clinical prevention and Effective nursing.
Methods:
The data of 707 patients who underwent PRB in nephrology department in Hangzhou Hospital of Traditional Chinese Medicine from January 2016 to January 2017 were retrospectively identified. Demographic and clinical data were collected, including general status (gender, age, body mass index, histological diagnosis, associated diseases), laboratory indexes and related examination during PRB (serum creatinine, estimated glomerular filtration rate, creatinineclearance rate, serumuricacid, serumalbumin, hemoglobin, platelet count, prothrombin time, activated partial thromboplastin time, kidney size), blood pressure(history of hypertension, systolic blood pressure, diastolic blood pressure and mean arterial blood pressure before PRB). Univariable logistic regression analysis, linear diagnosis, factor analysis, multivariable logistic regression analysis and receiver operating characteristic curve (ROC curve) were used to assess risk factors.
Results:
Over the period, 707 native kidney biopsies were performed. Hematoma occurred in 609 biopsies (86.1%), including 558 minorhematomacases (78.9%), 51 largehematoma cases (7.2%), no severe complications were observed. Univariable logistic regression analysis of risk factors in 51 patients with large hematoma after PRB found that there were significant differences in renal tubulointerstitial fibrosis, crescents > 25%, serum creatinine, history of hypertension, systolic blood pressure, diastolic blood pressure and mean arterial pressure before PRB (

Malnutrition in liver cirrhosis is associated with ascites, hepatic encephalopathy, infection, and other complications even death. To date, the hazard and disease burden of malnutrition in cirrhosis patients have been severely underestimated. This review summarized the most recent advancement in the field and discussed the techniques and methodologies in detection and evaluation of malnutrition in cirrhosis patients, nutritional support therapy, and future research directions and clinical care of the patients.
