Objective To find out the change of bone density in premenopausal women with systemic scleroderma (SSc) and its association with gluecorticosteroid. Methods Using a clinical bone densitometer manufactured by British McCue CuBA Cobroadband ultrasound attenuation (BUA) and velocity of sound (VOS) were measured at the left heel bone in 58 premenopausal patients with systemic scleroderma.Among them,35 patients had taken gluecorticosteroid, and the other 23 had not.Levels of serum estradiol (E 2) in 19 out of 58 patients with systemic scleroderma were determined and compared with healthy controls. Results Levels of broadband ultrasound attenuation and velocity of sound measured in permenopausal patients with systemic scleroderma who had taken gluecorticosteroid were significantly lower than those in ones who had not taken gluecorticoseroid ( P 0 05).Conclusion The bone mineral density and the level of BUA and VOS in premenopausal patients with systemic scleroderma, who had taken gluecorticosteroid, are obviously decreased, but there is no significant difference between patients without taking gluecorticosteroid and healthy controls. It indicates that systemic scleroderma is no dangerous factor of osteoporosis. And bone density in premenopausal patients with systemic scleroderma is related with the patients who have taken gluecorticosteroid for a long time.

Cultivation of laboratory physicians in accordance with the needs of discipline should be a priority of education on laboratory medicine in China. Unfortunately, the professional career of Chinese laboratory physicians is presently entrapped in a dilemma owing to unfavorable settings, in-cluding the late start of laboratory physician training in this country, unmatched installation between higher school faculty and curriculum, faulty programs of in-hospital training, disparity in management policies, low social esteem, uncertainty in personal intention of employment, and poor knowledge of position awareness. This paper puts forward a lot of measures such as the development trend of the university curriculum, which is in compliance with the subdivision and refinement of clinical medicine, the establishment of a medical degree for a professional laboratory physician, constantly improving the standardization of laboratory physician residency training and continuing education system for the in-spection of doctors , setting up physicians inspection posts and giving a clear mandate to examine physicians inside the hospital, while outside of the hospital continually improving awareness and accep-tance of the laboratory physicians, giving more educational resources to laboratory physicians, so as to improve the overall level of the laboratory medicine.

Objective To explore the effect of ionizing radiation and cisplatin on the expression of Transducer of erbB2,1 ( TOB1 ) in human lung adenocareinoma SPCA-1 and LTEP-α-2 cell lines. Methods SPCA-1 and/or LTEP-α-2 cells were respectively irradiated with 2,4,6,8,and 10 Gy X-rays generated by a linear accelerator (with the source skin distance of 100 cm and dose rate of 200 cGy/min).The cell molecular samples were subtracted at 6,12,18,and 24 h after 6 Gy irradiation. For X-rays and cisplatin combination treatment, cells were divided into radiation group (6 Gy X-rays),cisplatin group (20 mol/L cisplatin),and combination group (6 Gy X-rays + 20 mol/L cisplatin).SPCA-1 and LTEP-α-2 cells without any treatment were used as control group. The relative levels of TOB1 mRNA and protein expression were detected by rigorously controlled semi-quantitative RT-PCR and Western blot analysis with quantitation of the mRNA/protein bands by densitometry. Results The expression level of TOB1 was significantly increased in both mRNA and protein level after radiation exposure (t =8.25-24.48,P <0.05).For time-dependent induction of TOB1,the expression was significantly increased 6 h after X-rays exposure (t =14.23-15.82,P < 0.05 ).More significant increase of TOB1 expression was identified after the combination treatment of X-rays and cisplatin, compared to that of the radiation and/or cisplatin treatment alone (t =11.21-13.67,P <0.05).Conclusions lonizing radiation and cisplatin could upregulate the expression of TOB1 in lung cancer cells in both mRNA and protein levels.TOB1 may be a molecular target for ionizing radiation and cisplatin treatment in lung cancer and it may have potential implication in evaluating the curative efficiency of chemo-and radio-therapy.
4. Echocardiographic evaluation of myocardial impact injury caused by eccentric aortic regurgitation
Chinese Journal of Medical Imaging Technology 2020;36(4):494-498
Objective: To explore the effect of transthoracic color Doppler echocardiography on evaluating myocardial impact injury caused by eccentric aortic regurgitation (AR). Methods: Transthoracic color Doppler echocardiographic imaging data were collected in 26 cases with local deformation of some part of the ventricular septum or left ventricular free wall rammed by moderate and above eccentric AR, and the morphological characteristics of ventricular wall impact injury were analyzed. Results: Myocardial impact injury caused by eccentric AR located at the basal-mid segment of ventricular septum or left ventricular wall (26/26, 100%). Both inferoseptal and inferior wall were impacted in 4 cases (4/26, 15.38%), only inferior wall were impacted in 5 cases (5/26,19.23%), both inferior and inferolateral wall in 13 cases (13/26, 50.00%), inferolateral wall in 2 cases (2/26, 7.69%), while inferoseptal, inferior and inferolateral wall all were impacted in 2 cases (2/26, 7.69%).No anterolateral, anterior nor anteroseptal wall was impacted. Local dilatation was observed at the impacted wall in all cases (26/26, 100%). Severe hypokinesia was detected in 6 cases(6/26, 23.08%), mild hypokinesia in 10 cases(10/26, 38.46%),while normal motion was observed in the rest 10 cases(10/26, 38.46%). In addition to local wall dilatation and hypokinesia, myocardial fibrosis could be observed from LGE images in one case. Local perfusion and metabolism decreased on radionuclide perfusion images in severe cases, but not in mild cases. Conclusion: Moderate and above eccentric AR can cause damage to some part of the wall impacted directly by the eccentric aortic regurgitation, and the location and extent of damage can be evaluated using color Doppler echocardiography.

Objective To investigate the effect of EGCG on the radiosensitivity of human nasopharyngeal carcinoma CNE-1 cells. Methods CNE-1 cells were divided into four groups:control,EGCG treatment,UVC or X-ray exposure, and EGCG combined with UVC or X-rays.After treatment with different concentrations of EGCG for 24,48 and 72 h and UVC or X-rays,cell growth was determined with MTT assay, cell survival was measured with clonogenic assay, cell cycle was deteeted with flow cytometry, cell apoptosis was detected by the annexin V-FITC cell apoptosis kit,and protein expression was assayed by Westem blot. Results EGCG inhibited cell growth in a dose-and time-dependent manner(r =0.817and 0.364).Compared with UVC or X-ray irradiation alone, the radiosensitivity of CNE-1 cells was enhanced by 2 h pre-treatment of 50 μmol/L EGCG,which disrupted S phase arrest caused by UVC( t =18.68,P < 0.05 ) and increased the population of S and G2/M arrest caused by X-rays ( t =7.11 and 6.99,P <0.05 ).UVC could cause a significant increase of sub-G1 population( t =6.67,P < 0.05 ) and Annexin V-FITC assay indicated apoptosis was further elevated by EGCG ( t =10.28,P < 0.05 ).However,no significant induction of apoptosis was observed in the cells either irradiated with X-rays alone or combinationly treated with EGCG and X-rays.The combination treatment of EGCG and UVC significantly increased the expression of Bax and Caspase-3 proteins, but failed to affect Bcl-2 protein expression. Conclusions EGCG enhances the growth inhibition of CNE-1 cells caused by UVC or X-rays,which is relevant to apoptosis induction or cell cycle arrest.

Protein & Cell 2012;3(1):44-50
As a large family of hydrolases, GTPases are widespread in cells and play the very important biological function of hydrolyzing GTP into GDP and inorganic phosphate through binding with it. GTPases are involved in cell cycle regulation, protein synthesis, and protein transportation. Chaperones can facilitate the folding or refolding of nascent peptides and denatured proteins to their native states. However, chaperones do not occur in the native structures in which they can perform their normal biological functions. In the current study, the chaperone activity of the conserved GTPases of Escherichia coli is tested by the chemical denaturation and chaperone-assisted renaturation of citrate synthase and α-glucosidase. The effects of ribosomes and nucleotides on the chaperone activity are also examined. Our data indicate that these conserved GTPases have chaperone properties, and may be ancestral protein folding factors that have appeared before dedicated chaperones.
