1.Reversal of Multidrug Resistance Gene mdr1 of Drug-Resistant Human Hepatocellular Carcinoma Cells with Antisense Oligodeoxynucleotide in Vivo

Objective To investigate the reversal of the multidrug resistant gene mdr1 in vivo by antisense oligodeoxynucleotide (ASODN) on the basis of study in vitro. Methods The cultured drug-resistant human hepatocellular carcinoma cells were injected under the skin of axilla to establish the tumor model of nude mice. mdr1 ASODN accompanied by Lipofectamine were injected locally and ADM was injected intraperitoneally. Control 1 and control 2 were locally injected by Lipofectamine and normal saline separately, and ADM was also injected intraperitoneally. Results As time went on the tumor size increased and from the 5th day on alterations were marked, tumor size in different time phase showed marked difference to the prior time phase with significant difference (P 0.05). The results suggested that SODN and Lipofectamine showed no marked effect on tumor growth of nude mice and ASODN had marked inhibition effect on tumor growth.Conclusion mdr1 ASODN can also reverse multidrug resistance of drug-resistant human hepatocellular carcinoma cells in vivo. After the treatment the tumor’s growth in nude mice will slow down in a range of time.
2.Amlodipine Improves Endothelial Progenitor Cell Mobilization and Neo-vascularization in Experimental Diabetic Rats After Myocardial Infarction

Objective: To observe the effect of amlodipine on bone marrow endothelial progenitor cell (EPC) mobilization, neo-vascularization and cardiac function in diabetic rats after myocardial infarction (MI) with the possible mechanisms.
Methods: A total of 60 male SD rats were divided into 2 groups. Normal group, n=20. Diabetic group, n=40, the rats were fed with high fat diet (HFD) for 4 weeks and then received streptozotocin followed by left anterior descending coronary artery ligation to establish MI model, those rats were further divided into 2 sub-groups:Control group, the rats received sodium carboxymethylcellulose 1 ml/day with HFD and Treatment group, the rats received amlodipine 2 mg/kg/day with HFD, n=20 in each sub-group, all animals were treated for 4 weeks. The EPC level in peripheral blood CD45-/low+/CD133+/KDR+ at before and 1, 3, 5, 7, 14, 28 days after operation were examined by lfow cytometry, plasma vascular endothelial growth factor (VEGF) level was measured by ELISA, capillary density in MI area was determined by CD31 staining, EPC related protein expressions were detected by western blot analysis and the cardiac function was evaluated by echocardiography.
Results: EPC in CD45-/low+/CD133+/KDR+in Treatment group at 7 days after operation was increased than Control group at 5 days after operation (112 ± 30/106) vs (55 ± 10/106), plasma VEGF in Treatment group was higher than Control group (5.63 ± 1.33) ng/L vs (3.68 ± 0.98) ng/L; Treatment group presented increased expressions of protein kinase B, endothelial nitric oxide synthase (eNOS) and matrix metallopeptidase-9, increased capillary density in MI area, higher LVEF and left ventricular fractional shorting, all P<0.05-0.01.
Conclusion: Amlodipine improves EPC mobilization, neo-vascularization and cardiac function in diabetic-MI rats, it may be related to VEGF/eNOS cascade activation.
3.Nursing care of 12 patients with large area exposed bowel after abdomen opened

Objective To investigate the nursing care of patients with large area exposed bowel after abdomen opened, and the observation of complications. Methods Clinic data, treatment, difficulties of nursing, and clinical manifestations of 12 patients with large area exposed bowel after abdomen opened in the ICU ward of Beijing Hospital during the period January 2013 to October 2015 were reviewed. Comprehensive nursing interventions including intraperitoneal irrigation with active drainage, double tube maintenance of abdomen, protection of exposed bowel, attachment of intestinal fistula, heat preservation and observation on complications were used. Meanwhile, according to whether the intestinal fistula combined, different open wound nursing patterns and enteral nutrition support patterns were decided. Results Abdomen cavity of 12 patients were finally closed by surgery and all patients were cured and discharged. Conclusions Condition of patients with large area exposed bowel after abdomen opened is complex. If severe abdominal infection and multiple intestinal fistula are combined, the nursing would be more difficult. For such patients, we should apply holistic nursing care combined with individualized nursing mode.
4.Investigation of the current status of central venous pressure measurement operation

Objective To explore the investigation of central venous pressure ( CVP ) measurement operation in some hospitals throughout the country, in order to provide the evidence for the construction of CVP standardized measurement. Methods A convenience sample of 236, who was students of Chinese Nursing Association critical care specialist nurse training class in 2014, was investigated the various factors influencing the CVP measurement by self designed questionnaire according to literature. Results After central venous catheter insertion, 48. 7% of students′department checked the apex position of CVP as routine. A total of 55 students (23. 3%) believed it should remove PEEP before CVP measurement, 87 students (36. 9%) reckoned it was not available, while 94 of them ( 39. 8%) considered whether removed PEEP according to patients′condition. Most of students′ department utilized double channel catheter or multi-lumen catheter, while 131 students′department (55. 5%) chose the main catheter to measure CVP as regulation, department of 8 students (3. 4%) choosing lateral channel, and department of 97 students ( 41. 1%) having no clear regulation. Conclusions In some hospitals the CVP measurement has no united standard, even if there were factors related research had been confirmed. The results of researches had not been widely accepted in clinical. In order to improve the accuracy of clinical CVP measurement, CVP measurement needs to construct standardized operating norms.
5.Effects of DVT prevention and control project in inpatients of Vascular Surgery Department

Objective:To explore the effects of deep venous thrombosis (DVT) prevention and control project on preventing DVT in inpatients of Vascular Surgery Department.Methods:In June 2018, the DVT prevention and control team formed in Vascular Surgery Department of Beijing Hospital investigated the cognition of medical staff in Vascular Surgery Department on thrombosis by electronic questionnaire and built the thrombosis prevention and control management system. The cognition of medical staff on thrombosis and incidence of thrombosis in inpatients was compared before (January 2018 to June 2018) and after (July 2018 to December 2018) implementing the project.Results:Before and after implementing the thrombosis prevention and control project, the utilization rate of Caprini assessment scale and was 79.27% (65/82) and 100.00% (82/82) respectively with a statistical difference ( P<0.001) ; the accuracy rate of DVT prevention was 90.24% (74/82) and 98.78% (81/82) also with a statistical difference ( P=0.016) ; the incidence of DVT of inpatients was 4.88% (4/82) and 0 (0/82) with a statistical difference ( P=0.043) ; the implement rate of thrombosis education was 76.67% (23/30) and 96.67% (29/30) with a statistical difference ( P=0.023) ; the compliance of DVT prevention of patients after discharge was 91.02% (71/78) and 98.68% (75/76) with a statistical difference ( P=0.032) ; the incidence of lower limb swelling was 1.28% (1/78) and 0 (0/76) with no statistical difference ( P=0.322) . Conclusions:Implementation of the DVT prevention and control project in Vascular Surgery Department can effectively improve cognition of medical staff and patients on DVT, and can reduce the incidence of DVT of inpatients in Vascular Surgery Department.
6.Best evidence summary of application timing and management strategies for chronic wound oxygen therapy

Objective To summarize the most robust evidence on application timing and management strategies for chronic wound oxygen therapy and to provide an evidence-based basis for standardizing clinical practice.Methods A systematic literature search of BMJ Best Practice,UpToDate,NICE,SIGN,GIN,NZGG,NGC,RNAO,WOCN,WUWHS,APWCA,IWII,WHS,WI,AWMA,WCET,ESVS,AAWC,Medlive,JBI,Cochrane Library,PubMed,Embase,Web of Science,CNKI,Wanfang and VIP was conducted to collect the literature including clinical decision resources,guidelines,expert consensuses,evidence summaries,systematic reviews,and meta-analyses related to the application timing and management strategies for chronic wound oxygen therapy.The search period spanned from October 13,2018,to October 13,2023.There were 2 researchers who assessed the quality of the included literature,extracted relevant data,and synthesized the evidence.Results In total,we included 28 pieces of literature,consisting of 6 guidelines,4 expert consensus papers,3 evidence summaries,and 15 systematic reviews.From these sources,we distilled 18 pieces of evidence including 7 aspects of indications,contraindications,application timing,assessment,management strategies,effectiveness,and adverse reactions of hyperbaric oxygen therapy;12 pieces of evidence from 6 aspects of indications,application time,assessment,management strategies,effectiveness,and adverse reactions of local oxygen therapy.Conclusion Oxygen therapy can be used as an adjunct therapy for chronic wound management.Clinical practitioners should fully evaluate the scope and timing of the application of oxygen therapy,consider the feasibility,suitability,clinical signific ance,and effectiveness of evidence based on the specific clinical situations,and apply evidence in combination with patient preferences to promote chronic wound healing.
7.A New Micro-traumatic Laparoscopic Surgery Robot System.

At present, there still exist some limitations in the laparoscopic surgery robot represented by da Vinci surgical robot, such as the lack of force feedback function. Doctor can not feel the force feedback while operating. In this paper, a new minimally invasive laparoscopic surgery robot system is designed. Based on the master side surgeon's console, stereo vision subsystem and the slave side surgical cart, the multi-dimensional instrument force feedback technology and force feedback based safety protection strategy are introduced. The design realizes the force sensing function of full state operation. Besides, a number of different live pig experiments are carried out. The amount of bleeding in these experiments is relatively small compared with the data of the same kind of surgical robots, which effectively validates the force feedback and surgical safety protection strategies of the new robot system.

8.Exploring the mechanism of action of sour jujube nut compound formula for depression based on network pharmacology and experimental validation

Objective In this study,we aimed to use network pharmacology techniques to predict the key targets of a prescription of Ziziphi spinosae semen formula(ZSSF)compound for depression,and to verify its mechanism of action using a zebrafish model of rifampicin-induced depression.Methods The drug targets of ZSSF were retrieved from the TCMSP database,and the target names were corrected using the UniProt database.Depression-related targets were identified using the GeneCards,OMIM,and NCBI databases.Protein-protein interaction information for the shared targets was predicted using the STRING database.The collected data were then analyzed using the Metascape database to determine GO and KEGG pathway enrichment,and the result were visualized using microbiotics.Behavioral experiments and reverse-transcription quantitative PCR experiments were conducted to verify the therapeutic effects of ZSSF on a zebrafish depression model induced by risperdal.Results 188 targets were screened to find the interactions between depression and ZSSF.The protein-protein interaction result showed that ZSSF primarily targeted TNF-α,IL-2,IL-6,IL-1β,and IL-10 to produce its antidepressant effect.KEGG pathway enrichment analysis revealed that ZSSF exerted its effects on depression through various signaling pathways,including the TNF,PI3K-Akt,and cGMP-PKG signaling pathways.The result of the animal experiments showed that the treatment groups given high,medium,and low doses of ZSSF exhibited significant improvements in movement distance under acoustic and light stimulation compared with the model group(P<0.05).The speed of movement of the treatment groups was also significantly faster(P<0.01).Additionally,the mRNA expression levels of TNF-α,IL-2,IL-6,IL-1β,and IL-10 were up-regulated in the brain tissues of zebrafish in the high-,medium-,and low-dosage groups of ZSSF compared those in the model group(P<0.001).Conclusions ZSSF exerts its antidepressant effect through multiple components and targets,and its antidepressant effects may be associated with its inhibition of inflammatory factors.
9.cPKCγ Deficiency Exacerbates Autophagy Impairment and Hyperphosphorylated Tau Buildup through the AMPK/mTOR Pathway in Mice with Type 1 Diabetes Mellitus.

Type 1 diabetes mellitus (T1DM)-induced cognitive dysfunction is common, but its underlying mechanisms are still poorly understood. In this study, we found that knockout of conventional protein kinase C (cPKC)γ significantly increased the phosphorylation of Tau at Ser214 and neurofibrillary tangles, but did not affect the activities of GSK-3β and PP2A in the hippocampal neurons of T1DM mice. cPKCγ deficiency significantly decreased the level of autophagy in the hippocampal neurons of T1DM mice. Activation of autophagy greatly alleviated the cognitive impairment induced by cPKCγ deficiency in T1DM mice. Moreover, cPKCγ deficiency reduced the AMPK phosphorylation levels and increased the phosphorylation levels of mTOR in vivo and in vitro. The high glucose-induced Tau phosphorylation at Ser214 was further increased by the autophagy inhibitor and was significantly decreased by an mTOR inhibitor. In conclusion, these results indicated that cPKCγ promotes autophagy through the AMPK/mTOR signaling pathway, thus reducing the level of phosphorylated Tau at Ser214 and neurofibrillary tangles.
