Objective:To study the effects of panax notoginsenosides on the proliferation and oxidation indices of cisplatin-induced nephroxicity in HK-2 cells. Methods:HK-2 cells were cultured in vitro till the number was up to 1 × 106/ml. The cells were inoculated in 96-well culture plate and randomly divided into six groups:normal saline ( NS) group, the model group, the positive control group and the high dose group , medium dose group and low dose group of panax notoginsenosides ( PNS) . The nephroxicity model was dupli-cated with the addition of cisplatin (the final concentration was 6. 25μg·L-1). The model group, positive control group and the three panax notoginsenosides groups was treated with saline solutions, amifostine, panax notoginsenosides at the dose of 100,50 and 25 mg· L-1 , respectively. The cell viability was detected with an MTT method, the content of MDA and the activity of SOD, GSH-PX and LDH were measured and the cell structure was observed. DCFH-DA was used as the fluorescence probe to detect the level of ROS by a fluorescence microplate reader. Results:Compared with those in the model group, the cell viability and the activity of SOD and GSH-PX in the three PNS groups and the positive control group significantly increased (P<0. 05);the content of MDA, the level of ROS and the activity of LDH significantly decreased (P<0. 05); the cell structure was significantly improved. Conclusion: PNS can pro-mote the proliferation of HK-2 cells in vitro, and improve the biochemical parameters and enzyme levels. The results suggest that PNS has a protective effect on HK-2 cell, and the protective mechanisms may be related with its antioxidant effect.

OBJECTIVE:To study the spectrum-effect relationship of the antipyretic effect of Radix Bupleuri injection. METH-ODS:HPLC method was used to establish the fingerprint of Radix Bupleuri injection. The antipyretic effect of 10 batches of Radix Bupleuri injection on fever rats induced by dried yeast were determined respectively. The fingerprint peaks were screened, along with the temperature value of different time points(0,10,15,30,40,55,70 min). The principal components were extracted by principal component method and the indirect relationship between the fingerprint and antipyretic effect was analyzed, depending on the principal component correlation coefficient matrix. RESULTS:There were 39 common peaks in fingerprint(similarity>0.85), No. 8,12,14,19,26,31,34,35 and 39 common peaks with large peak area were included in the study. Four principal compo-nents were extracted by principal component analysis(87% of total variant). First principal component showed that the active com-ponents of the 12th peak may be related to the antipyretic effect of 6 to 13 hours. The second principal component showed that the active components of the 26th peak may be related to the antipyretic effect of 0.5 to 5 hours. The third principal component showed that the similar effect of the active components could be caused by 34th,35th and 39th peaks. The fourth principal component sug-gested that there were some similarities between the 14th and the 31st peaks. CONCLUSIONS:Radix Bupleuri injection have obvi-ous improvement for fever rats. There is certain corresponding relation between HPLC fingerprint and antipyretic effect of Radix Bu-pleuri injection .

7.Syndrome Differentiation and Treatment of Rheumatoid Arthritis with Anemia Based on Traditional Chinese Medicine Theory of "Deficiency Caused by Toxic Heat"
Chinese Journal of Experimental Traditional Medical Formulae 2022;28(20):218-225
Anemia is one of the most common complications of rheumatoid arthritis (RA). It is an important factor affecting the quality of life of patients and disease prognosis in addition to joint dysfunction. The pathogenesis of RA with anemia in western medicine is complex and diverse, and it is difficult to give consideration to medication. Traditional Chinese medicine (TCM) has certain therapeutic advantages because it pays attention to overall regulation and syndrome differentiation. In recent years, with the in-depth exploration of the pathogenesis of RA in modern medicine and the continuous improvement of the idea of diagnosis and treatment combined with disease and syndrome in TCM, the connotation of TCM pathogenesis of RA has gradually broken through the traditional theoretical framework of "arthralgia syndrome", and TCM etiology research of toxin in RA and its complications has received increasing attention. By tracing and sorting out the theory of "toxin caused arthralgia", combing with the research results of the pathological mechanism of modern medicine, this thesis expounds the importance of toxins in the pathogenic factors of RA in TCM. On this basis, it is further proposed that "deficiency caused by toxic heat" is the core pathogenesis of RA with anemia. Deficiency of healthy Qi, the six exogenous factors, the seven emotional factors, and improper diet can lead to the formation of toxic heat, and damage to the joint structure and viscera, resulting in blood deficiency. To treat RA with anemia, the syndrome differentiation by stages is conducted based on the principle of detoxification and dredging collaterals, and the mechanism of self-made prescription Ersi decoction on RA complicated with anemia is explored based on previous research, to enrich the theoretical basis for the treatment of RA from toxic heat and provide new ideas for TCM differentiation and treatment of RA with anemia and other complications.

Objective:To evaluate the expression of Vimentin and its relationship with clinical pathological data and prognosis in patients with metastatic renal cell carcinoma (mRCC).Methods:A total of 269 mRCC patients, from June 2007 to June 2017, treated with tyrosine kinase inhibitors (TKI) in Zhongshan Hospital, Fudan University were included in the study. Vimentin was assessed by immunohistochemistry on continuous paraffin embedded slides. To analyzed the relationship between Vimentin expression and clinicopathological data and prognosis. Measurement data were expressed as mean±standard deviation ( Mean± SD). The relationship between Vimentin expression and clinicopathological data of mRCC patients was determined by Chi-square test. Survival analysis used Kaplan-Meier survival curve, Log-rank test to judge the relationship between Vimentin expression and patient′s overall survival, and COX regression analysis was used for univariate and multivariate analysis. Results:Vimentin was expressed in cytoplasm of tumor cells. Vimentin expression in mRCC patients′ tissues was positively correlated with tumor Fuhrman grade ( P=0.002), and negatively correlated with prognostic overall survival ( P=0.001). Multivariate regression analysis showed that Vimentin expression was an independent risk factor for overall survival in patients with mRCC ( P=0.006). Conclusions:Vimentin expression can be used as a prognostic indicator of mRCC. It is expected that Vimentin can provide prediction for the prognosis of mRCC patients treated with TKI.
