1.Clinical application of Danhong injection combined with low-tnoiecular-weight heparins in treatment of unstable angina pectoris

Objective To observe the efficacy of Danhong injection combined with low-molecularweight heparins in treating unstable angina pectoris.Methods One hundred and three cases of unstable ansina pectoris were enrolled and divided into group A and B,Group A were given Danhong injection and lowmolecular-weight hepafins,Group B were given routine treatment.Results The overall response rate of group A was 90.7% and it was 74.0% for group B.The difference was significant (x2 =4.90,P<0.05).Conclusion Treatment with Danhong combined with low-molecular-weight heparins produced a good outcome in the patients with unstable angina pectoris.
2.Establishment of mus skin photo-damage model by 8-MOP plus UVA irradiation.

To establish a simple and reliable animal model of skin photo-damage, 20 mice were treated with 8-MOP and exposed to UVA (UVA 320-400 nm) for 24 h. After irradiation, the structure of the epidermis and dermis, collagen fibers, elastic fibers were observed by using HE staining and Weigert technique and compared with the normal controls. The acanthosis and epidermis proliferation with accompanying hyperkeratosis and parakeratosis were observed. Inflammatory infiltration was noted in the dermis. The elastic fibers became coarse, irregularly arranged and clustered, with their number increased. The collagen fibers showed obvious degeneration and some amorphous materials could also be observed. The blood vessels were irregularly dilated and vascular walls were thickened, with infiltration of inflammatory cells. It is concluded that murine photodamage model can be quickly, conveniently and reliably established by means of 8-MOP/UVA.

3.Construction of the eukaryotic expression vector containing the human perforin gene and Its expression in COS-7 cells

Objective: To clone human perforin(pfn) full-length DNA,construct the eukaryotic expression vector and observe the expression of the pfn gene in the transfected COS-7 cells.Methods: Full-length DNA of pfn was obtained by PCR from rPCR2.1/pfn,inserted into the pMD-18T vector and subcloned to the pcDNA3.1(+) vector to construct the recombinant eukaryotic expression vector pcDNA3.1(+)/pfn.The recombinant plasmid was transfected into COS-7 cells and the expression of the pfn gene in the transfected cells was detected by RT-PCR.Results: The pfn full-length DNA was successfully cloned and inserted into the pcDNA3.1(+) vector.The expression of pfn mRNA in the transfected COS-7 cells was confirmed by RT-PCR. Conclusion: The full-length human pfn gene can be expressed in transfected COS-7 cells.
4.Differences in gene expression profiles of hypoxic human embryonic retinal microvascular endothelial cells induced by cobalt chloride

Objective To study the differentially expressed genes and analyze its functional pathways of retinopathy of prematurity(ROP),in order to discover the pathogenesis and provide the theoretical basis for the prevention and treatment of ROP.Methods Fetal eyeballs of induced labor were obtained and retinal microvascular endothelial cells were isolated and cultured.The endothelial cells were divided into 7 groups according to the medium of cobalt chloride (CoC12) concentration (0 μ mol/L,100 μ mol/L,150 pμmol/L,200 μmol/L,250 pμmol/L,300 μmol/L,350 μmol/L),and 150 μmol/L CoCl2 was finally used to induce ROP model in vitro.Retinal microvascular endothelial cells were verified by adopting Ⅷ factor and CD31 antibody fluorescence staining.RNA purification,gene chip hybridization and signi-ficant analysis of microarrays were performed to screen differentially expressed genes.Genes functional pathways were studied by using gene ontology analysis software.Results (1) The proliferation activity of vascular endothelial cells decreased when CoCl2 ≥ 150 μmol/L(F =21,P < 0.05).(2) In 150 μmol/L CoC]2 group,blue nucleus and green cytoplasm were visible in the second and the third generation vascular endothelial cells stained by factor Ⅷ antibody,and red fluorescence could also be observed in the cytoplasm by means of CD31 monoclonal antibody staining.However,only blue nucleus was detected in the group without CoCl2.(3)There were 326 genes differently expressed in retinal micro vascular endothelial cells induced by CoCl2 in vitro,among whom,198 genes were up-regulated and 128 genes were down-regulated.Up-regulated expression genes were 1.5 times more than those of the down-regulated genes.(4)Ten biological pathways including cell hypoxia,angiogenesis suppression and iron ion transport etc.may play important roles in ROP pathogenesis.Conclusions Hypoxic retinal microvascular endothelial cells induced by CoCl2 can successfully be used to induce cell model of ROP in vitro.Differentially expressed genes may play an important role in ROP development.Functional pathway such as hypoxic cells,inhibition of angiogenesis,iron ion transport may be associated with ROP pathogenesis.
5.Expression and clinical significance of platelet-derived growth factor A in placenta of pre-eclampsia

Objective To investigate the expression and clinical significance of platelet-derived growth factor A (PDGF-A) on placenta tissue from pre-eclampsia.Methods The expression of PDGF-A in the placenta of 38 pre-eclampsia patients and 22 normal pregnant women at third trimester was detected by immunohistochemistry method.Results (1)PDGF-A was mainly expressed in the cytomembrane and cytoplasm of cytotrophoblasts and the endothelial cell of capillary in placenta.(2) The rates of PDGF-A expression of cytotrophoblasts were 63% (24/38) in pre-eclampsia group and 32% (7/22) in normal pregnancy group,which exhibited significant difference (P <0.05).(3) The rates of PDGF-A expression of endothelial cell were 68% (26/38) in pre-eclampsia group and 27% (6/22) in normal pregnancy group,which also showed significant difference (P <0.01).(4)The rates of PDGF-A expression of cytotrophoblasts were 39% (7/18) in mild pre-eclampsia patients and 85% (17/20) in severe pre-eclampsia,which reached statistical difference (P < 0.01).Conclusion The increasing expression of PDGF-A in cytotrophoblast and endothelial cell in placenta might confer the occurrence and progression of preeclampsia.
6.Clinical Effects of Levofloxacin Eyedrops in the Treatment of Acute Bacterial Conjunctivitis and Keratitis

OBJECTIVE: To observe the therapeutic effects of 0.3% levofloxacin eyedrops on the acute bacterial conjunctivitis and keratitis.METHODS: The patients were randomly divided into treated group and control group.They were given 0.3% levofloxacin and 0.3% ofloxacin eyedrops respectively.The effects were compared between two groups.RESULTS: From April 1998 to December 1998, 132 cases were enrolled in the study: treated group 67 cases, control group 65 cases.The positive rates of bacterial culture were over 80% in both groups.It showed that there was no significant difference between two eyedrops in the curative and effective rates, but the mean cure day of keratitis in levofloxacin group was significantly shorter than that in ofloxacin group.CONCLUSION: Levofloxacin eyedrops is effective and safe in treating bacterial conjunctivitis and keratitis, and the therapeutic course is short and no obvious ARDs occur.

Heat shock protein 90(Hsp90)is a highly conserved protein which have been proved to play an important role in the development and progression of malignant transformation .As one of small molecule inhibi-tors that has been detected to have potent antitumor activity in a wide range of malignancies ,NVP-AUY922 is a pyrazole scaffold drug with many advantages such as low toxicity and stable structure .As a result of this,NVP-AUY922 is extensively considered as a new promising kind of anti -tumor drug .This review intends to update the reader on advances made over the past four years in the clinical development of NVP -AUY922 in advanced cancers.
8.Reconstruction of personalized three-dimensional entity model of hip bone based on reverse engineering and rapid prototyping technology

BACKGROUND:Hip’s anatomy structure is complex, and the traditional two-dimensional imaging, such as X-ray film or CT scan, is susceptible to image overlap and other soft tissue. As a result, there are some limitations in the diagnosis of hip disease. The digital model is built based on reverse engineering and rapid prototyping. It is comprehensive, intuitive and accurate of the stereo configuration of the hip and the spatial relations of other parts’ anatomical structures. This study has a broad application prospect. OBJECTIVE:To reconstruct the personalized three-dimensional entity model of the hip bone with computer aided technology. METHODS: The human hips were scanned with a portable three-dimensional laser scanner, and then the point cloud data of the hips were gotten. The point cloud was processed and the three-dimensional model reconstruction was carried out by using the engineering software Geomagic and the computer aided software CimatronE. Three-dimensional print of hip was completed and the model was gotten, which was consistent with the geometry of hip bone, through the rapid prototyping machine. RESULTS AND CONCLUSION:A model with good geometric similarity and biomechanical characteristics was built by digital modeling method based on reverse engineering and rapid prototyping. This way of digital modeling provided the basis for digital manufacturing of artificial hip, virtual assembly, stress analysis and surgical simulation.
9.Analysis of MR findings of misdiagnosed cases with pituitary macroadenoma

Objective To investigate MR findings and analyze the misdiagnosed cases of pituitary macroadenoma.Methods MR features of 1 90 patients of pituitary macroadenoma confirmed by operation and pathology were reviewed.Results 1 6 cases were mis-diagnosed as craniopharyngioma,chordoma,or meningioma.Among the sixteen cases,eight cases with cyst degeneration and hemor-rhage were misdiagnosed as craniopharyngioma;five cases with clival and sphenoid sinus destruction were misdiagnosed as chordo-mas;three cases with suprasellar and anterior cranial fossa extension were misdiagnosed as meningiomas.Conclusion Craniophar-yngioma,chordoma and meningioma should be considered in the differential diagnosis of atypical pituitary macroadenoma.The com-prehensive analysis should be based on a variety of signs.
10.The expression of toll-like receptor 2 and 4 mRNA in local tissues of model of oropharyngeal candidiasis in mice.

To investigate the expression of Toll-like receptor (TLR) 2 and 4 mRNA in local tissues of model of oropharyngeal candidiasis in mice and to explore the potential role of TLR2 and TLR4 in earlier period of immune response, a murine model of oropharyngeal candidiasis inoculated by cotton wool balls saturated with Candida albicans was established. Mice were sacrificed at the indicated time points and the oropharyngeal tissues were excised. The expression of TLR2 and TLR4 mRNA was detected by RT-PCR. The results showed that low level of TLR2/4 mRNA could be detected in oropharyngeal tissues, but they were markedly up-regulated 6 h after inoculation, peaking after 12-24 h. Tissue TLR4 mRNA was gradually down-regulated 24-48 h, while TLR2 mRNA levels remained high up to the 72nd h. These data suggested that oropharyngeal infection of Candida albicans could result in up-regulation of TLR2/4 mRNA expression in local tissues, which might play important roles in earlier period of immune response.
