1.Regulation role of resveratrol in microRNAs expression in cancers
Journal of International Oncology 2013;40(8):587-590
As a kind of polyphenolic phytoalexin, resveratrol exerts protective roles in anti-tumor,antiinflammatory and promoting metabolism.Resveratrol's anti-tumor mechanisms may be involved in upregulating tumor suppressor microRNAs and downregulating oncogenic microRNAs,and affect its relative pathways and target genes in different cancers such as colon cancers and breast cancers. This will provide a new theoretical basis for the further clarification of resveratrol-associated tumor suppressing mechanisms and its application to the clinical.

Objective:To investigate the diagnostic values of detections of human papillomavirus (HPV) DNA combined with peripheral blood cyclin A mRNA and cyclin-dependent kinase 2 (CDK2) mRNA for cervical squamous cell carcinoma. Methods:Eighty patients with cervical squamous cell carcinoma treated in Jiangyin Hospital of Traditional Chinese Medicine from January 2018 to October 2021 were selected as the research objects. Eighty patients with benign cervical lesions such as cervicitis treated in the same period were selected as the control. The levels of HPV-DNA in paraffin-embedded tissues of cervical squamous cell carcinoma were detected by gene chip, and the mRNA expression levels of cyclin A and CDK2 in peripheral blood monocytes were detected by reverse transcription polymerase chain reaction. The related factors of cervical squamous cell carcinoma were analyzed by multivariate logistic regression. Taking the results of pathological biopsy as the gold standard, the diagnostic efficacy of HPV-DNA, peripheral blood cyclin A mRNA and CDK2 mRNA single and combined detection for cervical squamous cell carcinoma were determined by receiver operating characteristic (ROC) curve. Results:The positive rate of HPV-DNA in patients with cervical squamous cell carcinoma was higher than that in the control group [75.00% (60/80) vs. 13.75% (11/80), P < 0.05]; the relative expressions of cyclin A mRNA and CDK2 mRNA in patients with cervical squamous cell carcinoma were 0.26±0.08 and 1.49±0.07, respectively, which were higher than those in the control group (0.11±0.03 and 1.14±0.06), and the differences were statistically significant (both P < 0.05). Multivariate logistic regression analysis showed that the number of induced abortions > 1 ( OR = 3.093, 95% CI 1.386-6.899, P = 0.021), the age of first birth ≤18 years old ( OR = 3.684, 95% CI 1.651-8.219, P = 0.013), the positive HPV-DNA ( OR = 4.125, 95% CI 1.849-9.202, P = 0.001), the increased relative expression of cyclin A mRNA in peripheral blood ( OR = 3.800, 95% CI 1.703-8.478, P = 0.006) and the increased relative expression of CDK2 mRNA in peripheral blood ( OR = 4.821, 95% CI 2.161-10.756, P = 0.008) were risk factors for the occurrence of cervical squamous cell carcinoma. ROC curve analysis showed that the area under the curve (AUC) of HPV-DNA, peripheral blood cyclin A mRNA and CDK2 mRNA in single and combined diagnosis of cervical squamous cell carcinoma were 0.769 (95% CI 0.700-0.838), 0.756 (95% CI 0.688-0.823), 0.755 (95% CI 0.689-0.820) and 0.827 (95% CI 0.766-0.888), respectively. Conclusions:HPV-DNA and the levels of cyclin A mRNA and CDK2 mRNA in peripheral blood can be used to assist in the diagnosis of cervical squamous cell carcinoma, and the combination of the three has high diagnostic efficiency.

Objective To investigate effects of laryngeal mask ventilation combined with continuous nerve block analgesia versus the endotracheal intubation under general anesthesia on postoperative lung infection in elderly patients undergoing orthopedic surgery,so as to provide a theoretical basis for a reasonable anesthesia method. Methods A total of 180 elderly patients undergoing lower extremity orthopedic surgery at our hospital from January 2016 to December 2016 were enrolled and randomly divided into two groups. The control group (n =90) were treated with endotracheal intubation under general anesthesia and intravenous analgesia after the operation, and the observation group (n =90)received the ultrasound-guided continuous nerve block analgesia in spontaneous respiration with laryngeal mask ventilation. The anesthetic dosage, awakening time, visual analogue scale(VAS)scores, and incidence of lung infections at 7 days after surgery were compared between the two groups.Pathogenic strains causing lung infections were isolated and identified. Results The anesthetic dosage was lower in the observation group than in the control group(P < 0.05).The awaking time was shorter in the observation group than in the control group[(22.4±4.4) min vs.(34.1±8.5)min,P <0.05].The VAS scores under postoperative exercise were lower in the observation group than in the control group(P <0.05).The adverse reaction rate after surgery was lower in the observation group than in control group(3.3% or 3/90 cases vs.41.1% or 37/90 cases,P <0.05).The incidence of lung infections at 7 days after surgery was lower in the observation group than in the control group(3.3% or 3/90 cases vs.11.1% or 10/90 cases,P<0.05).Ten pathogenic strains were isolated from control group,of which 9 strains were gram-negative bacteria, accounting for 90.0%.Three pathogenic strains were isolated from the observation group,of which 2 strains were gram-negative bacteria, accounting for 66.7%.Conclusions Laryngeal mask ventilation combined with continuous nerve block analgesia can reduce the anesthetic dosage, shorten the awaking time, provide a better analgesic effect and decrease the incidence of lung infections in elderly patients undergoing lower extremity orthopedics. The main pathogenic bacteria are gram-negative bacteria.

Ginsenoside Rg1 (Rg1), the major effective component of ginseng, has been shown to have multiple bioactivities, but low oral bioavailability. The aim of this study was to develop a simple, sensitive and rapid high performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) method, which could be used to validate and quantify the concentrations of Rg1 and its metabolites in Sprague-Dawley rat bile, urine, and feces after oral administration (25 mg/kg). Calibration curves offered satisfactory linearity (>0.995) within the determined ranges. Both intra-day and inter-day variances were less than 15%, and the accuracy was within 80-120%. The excretion recoveries of Rg1, ginsenoside Rh1 (Rh1), and protopanaxatriol (Ppt) in bile, urine, and feces combined were all greater than 70%. The fecal excretion recoveries of Rg1, Rh1, and Ppt were 40.11%, 22.19%, and 22.88%, respectively, whereas 6.88% of Rg1 and 0.09% of Rh1 were excreted in bile. Urinary excretion accounted for only 0.04% of Rg1. In conclusion, the observed excretion profiles for Rg1 and its metabolites after oral administration are helpful for understanding the poor oral bioavailability of Rg1 and will aid further investigations of Rg1 as a pharmacologically active component.
