Objective To investigate the effect of 5-hydroxytryptamine 1A (5-HT1A) receptor agonists to improve micturition function in rats with diabetes mellitus (DM).Methods Fourteen female SD rats with the weight of 250 to 275 g were used.Seven rats were in the DM model group with intraperitoneal injection streptozotocin (STZ,65 mg/kg).Rats in the control group and DM group were anesthetized with urethane (1.3 g/kg) 8 weeks later.A polyethylene (PE)-50 catheter were placed in the left jugular vein for intravenous drug administration.A PE-90 catheter was inserted into the bladder, with the other end connected to a syringe pump for continuous infusion of saline and a pressure transducer for intravesical pressure monitor.Dose-response curves for 8-OH-DPAT were followed by WAY-100635 test. The capacity, residual volume, micturition volume, and EUS-EMG were measured. Results Compared to normal control,DM rats had a higher bladder capacity, residual volume, and a lower voiding efficiency. With increasing dose of 8-OH-DPAT (0.003-1.000 mg/kg,i.v.),the micturition volume increased from (2.15±0.49) ml to (2.85±0.21) ml,the residual volume decreased from (3.40±0.74)ml to (1.82±0.48) ml and voiding efficiency changed from (39.0±9.3)% to (61.6±6.9)%.Control rats showed little change in cystometic variable.During the micturition, there was a dose-dependent increased phasic EUS activity correlated with the improved voiding efficiency.WAY-100635 (0.300 mg/kg,i.v.) reversed the 8-OH-DPAT-induced changes. Conclusions Both the bladder voiding efficiency and the periodic EUS activity decrease in DM rats.5-HT1A receptor agonist could promote periodic EUS activity and improve voiding efficiency.

Huntington disease (HD) rat model was produced by injection of ibotenic acid (IA) into the head of right caudate putamen (CP). One month post lesion, fetal (El5-17) striatal cell suspension was implanted into the lateral ventricle ipsilateral to the lesion, and the rats were devided into four groups; group I normal control rats (n=8), group II model control rats (n=10), group III simple transplanted with fetal striatal cell suspension, (n=10, ST group), group IV grafted with striatal cell suspension containing laminin (n=6, LST group). Three and six months after grafting, active avoidence test was carried, the results showed that there were significant differences bewteen grafted groups and model control group at three and six months, and no significant differences between LST group and normal group either in three or six months whereas between ST group and normal group no significant difference only can be found at six months. Overnight locomotor activity was measured in each group at six months post grafting, the results indicated that the locomotor behaviour of model control group was hyperactive whereas the overnight hyperactivity was compensated completely in the grafted groups. After the behavioural test, the rat brain was investigated morphologically. The head of the lesioned CP was atrophied and the graft was located at the dorsal part of the atrophied CP and projected into the lateral ventricle. The volume of the graft area of LST group was larger than that of ST group. ChAT, GABA and Leu-ENK positive neurons were found in the graft area of the two grafted groups and their shape and size were similar to those of nomal CP. The processes of AChE positive neurons in the graft of LST group were more and longer than that in the graft of ST group. The results indicated that the fetal striatal neurons implanted into the lateral ventricle of HD model rats not only can survive and grow well but also ameliorate the behavioural deficits of the IA lesioned rats and the laminin may supports the neuronal suvival and growth in vivo.

A 47-year-old man presented with recurrent oral ulcerations for more than 1 year and generalized vesicles for 9 months.Physical examination revealed multiple oral ulcers, patchy labial erosions with yellow or dark brown crusts, maceration and blanching of the angle of mouth resulting in difficulty in mouth opening.Cinnamomeous crusts were seen on the nasal limen, palpebral margin and coronary sulcus of penis, with pale red macules after decrustation.Diffuse erythematous patches and bullae were present on the trunk and extremities. The walls of bullae tightly clung to the skin, and some bullae were ruptured, leaving an erythematous and moist surface and giving an erythema multiforme-like appearance; some bullae were turbid and covered with adherent drugcontaining white crusts or black crusts.Nikolsky's sign was negative.Shrinking, edematous dark-red patches were seen in periungual regions of all the fingers and toes.Histologic biopsy and direct immunofluorescence examination of bullous lesions confirmed a diagnosis of paraneoplastic pemphigus (PNP).Histopathological and immunohistochemical findings from the mediastinal tumor were consistent with Castleman's disease (hyalinevascular type) with proliferation of follicular dendritic cells.Laboratory examination revealed a decrease in serum complement C3 and the presence of antinuclear,anti-nucleosome and anti-dsDNA antibodies. The final diagnosis included Castleman's disease,PNP and systemic lupus erythematosus. After 2 months of treatment with low-dose prednisone and azathioprine, skin lesions completely regressed. Then, the dose of prednisone and azathioprine was tapered.Six months later, the patient himself suddenly withdrew prednisone and azathioprine, and began to take traditional Chinese medicine; thereafter, the lesions developed into toxic epidermal necrolysis (TEN),and the patient died finally. This case demonstrates that immunosuppressive therapy should be maintained for a long period of time in patients with Castleman's disease after tumor removal, otherwise, skin lesions may recur or get worse.
