Objective To observe the change of ?-smooth muscle actin (?-SMA) expression in carbon tetrachloride (CCl4)-induced liver fibrosis in rats after treatment with Ganle keli. Methods Liver fibrosis rat model was induced by CCl4 twice a week for 12 weeks. Ganle keli was used daily intragastric administration from the 7th week. Contents of NO, NOS and iNOS were determined by absorption spectrometry. Liver samples were collected after experiments and stained by immunohistochemistry of ?-SMA. Moreover ?-SMAmRNA expression in liver tissue was detected by RT-PCR technology. Results Ganle keli could significantly reduce NO, NOS and iNOS contents, effectively prevent the expression of ?-SMA, and significantly reduce ?-SMA mRNA in liver fibrosis rats. Conclusion Ganle keli can effectively inhibit the expression of ?-SMA in CCl4-induced fibrotic liver in rats.

The learning theory of constructivism, formed by the development of cognitive psychology, shaped well thecognitive structure of medical students. It paid attention to the curriculum configuration and integration, and also thoughtmuch of the situated instruction. The theory embodied the main ideology of regarding medical students as the instruc-tional central and the principle of independent study. It offered the theoretical basis and a new view for the setting ofclinical integrate curriculum.

Objective To optimize the extraction and inclusion process of mixed volatile oil of Rhizoma acori graminei, Fructus alpiniae oxyphyllae and Rhizoma chuanxiong in Huangpu Tongqiao Capsules. Methods With steam distillation method, extraction amount as indicator, the orthogonal experiment was used to optimze the extraction process of volatile oil from three factors such as water amount, soak time and extraction time. With saturated aqueous solution method, co-inclusion compound yield and volatile oil rate as indicators, the orthogonal experiment was used to optimize the inclusion process from three factors such as the proportion of volatile oil and β-CD, inclusion time and inclusion temperature. Results The optimum extraction was:adding eight times amount of water and soak for 2 hours to extract 7 hours. The optimum inclusion process was:the proportion of volatile oil andβ-CD was 1∶10, the inclusion time was 3 hours, the inclusion temperature was 40 ℃. Conclusion Optimal extraction process of oil has higher rate, and the volatile oil inclusion rate of inclusion process was also higher, the process is stable.

Objective To establish the quality standard of Qingshen Granules. Methods Thin-layer chrmatography was used to identify Rheum officinale Baill., Hedyotis Diffusa Wild. and Coptis chinensis Franch.. The contents of rhein, emodin and chrysophanol were determined by HPLC. The HPLC consisted of Kromasil C18 (4.6 mm×250 mm, 5 μm), methanol-0.1% phosphonic acid (70∶30) as mobile phase, flow rate of 1.0 mL/min, detection wavelength at 245 nm, and the column temperature was 30 ℃. Results The results of TLC showed that relevant spots were clear without interference against the negative sample. The calibration curves for rhein, emodin and chrysophanol were found to be linear within the range of 0.003 3-0.42 μg (r =0.999 9), 0.008 0-0.51 μg (r=1.000 0), 0.009 9-0.32 μg (r=1.000 0), respectively. AIM To observe the effects of Wuwei Wentong Chubi Capsules (Cinnamomi Ramulus,Poria,Epimedii Folium, etc.) on autophagy proteins of Beclin-1 and LC3-Ⅱ in adjuvant-induced arthritis rats and to explore the possible mechanism of action.METHODS Sixty SD rats were randomized into six groups:normal group, model group,Wuwei Wentong Chubi Capsules (0.8,1.6,3.2 g/kg) groups and tripterygium glycosides (TPT,40 mg/kg) group. In addition to the normal group, adjuvant arthritis (AA) was induced with Freund's complete adjuvant. From the 2th day after injection of FCA,Wuwei Wentong Chubi Capsules with different doses and TPT were given by gavage once a day for 12 days.At the end of the experiment,ankle-joint samples were taken to examine the degree of AA by HE.Beclin-1 and LC3-Ⅱ mRNA were determined by real-time fluorescent quantitative PCR.Meanwhile, the protein expression levels of Beclin-1 and LC3-Ⅱ were determined by immunofluorescence histochemical staining and Western blot.RESULTS As compared with the model group,Wuwei Wentong Chubi Capsules (1.60,3.20 g/kg) not only significantly reduced histopathological injuries, but also effectively up-regulated mRNA and protein expressions of Beclin-1 and LC3-Ⅱ.CONCLUSION Wuwei Wentong Chubi Capsules has a therapeutic action on AA in rats, which might be partly associated with promoting autophagy, decreasing excessive proliferation of synovial cells, leading to the reduction of damage to articular cartilage.

Objective To explore the pattern of spontaneous neural activity in different frequency bands in Parkinson's disease(PD).Methods The blood oxygen level-dependent signals in 39 PD patients and 37 normal controls were acquired and the amplitude of low-frequency fluctuation ( ALFF) in five differ-ent frequecy bands ( slow-2:0.198-0.25 Hz;slow-3:0.073-0.198 Hz;slow-4:0.027-0.073 Hz;slow-5:0.01-0.027 Hz;slow-6:0-0.01Hz) was cacaulated.A repeated-measures analysis of variance and Post hoc t-test were performed to detect differences of spontaneous activity between two groups in different frequency bands. A correlation analysis was examined between Unified Parkinson’ s Disease Rating Scale ( UPDRS) motor score and ALFF in patients with PD in each frequency band to discover the relationship between disease se-verity and spontaneous neural activity. Results Compared with the normal controls, patients with PD had decreased ALFF in the putamen(-24,-3,-9) (-30,3,0) ( -21,9,6) in the slow-3,slow-4 and slow-5, and had decreased ALFF in the left precentral gyrus (-21,-24,75) (-21,-24,72) (-27,-21,72) (-15,-15,72) and right supplementary motor area (21,-24,75) (6,-3,72) (12,-6,60) (15,-24,72) in all frequency bands except for the slow-5.Increased ALFF in the cerebellum was found in all frequency bands. The ALFF in the putamen was negatively correlated with the UPDRS motor score in the slow-3,slow-5 and slow-6.Conclusion This study shows that the abnormal activities in the striatum are significant in the slow-3,slow-4and slow-5.Most frequency bands except slow-5 are helpful in detecting neural changes in mo-tor cortex. The increased activity in the cerebellum is the common findings in all frequency bands.Multiple frequency analysis may provide insight into the pathophysiological mechanisms of PD.
