1.A FLUOROMETRIC METHOD FOR THE ASSAY OF LENS ALDOSE REDUCTASE AND ITS APPLICATION IN DIABETIC RATS
Chinese Journal of Endocrinology and Metabolism 1986;0(03):-
Rat lens aldose reductase (AR) activity was assayed in this study with a fiuorometric method. Rat lenses were homogenized in Na, K-phosphate buffer and centrifuged at 19127 ? g, 4℃ for 15 min. The supernatant was reserved as enzyme fraction. The incubation mixture contained 135 mmol KHaPO4-Na2HPO4 buffer (pH 7.0), 100 mmol Li2SO4, 6 ?mol NADPH, 1.0 mmol DL-glyceraldehyde or 10 mmol glucose and 20/ul enzyme fraction. AR activity in diabetic rat lent have not been shown to be significantly higher than that of the con rols. This method can be used properly to screen AR inhibitors in vitro.

We described a simple method for determining glycated lipoproteins (glc Lps) in serum by agarose gel film electrophoresis, and color development with nitroblue tetrazolium. The resulting blue bands on the film were measured densitometrically at 545 nm to quantify ?-, pre-?-, and ?-glc Lps. The concentration of gfc ?-Lp in serum from diabetics was 2,2-fold higher than that in normal individuals. Diabetic patients with vascular complications had higher concentrations of glc ?-Lp than did patients without vascular complication. The concentration of glc ?-Lp (glc LDL) in serum may be a useful diagnostic indicator of diabetic vascular complications and their severity.

OBJECTIVE:To provide reference for clinical prevention and treatment of ceftizoxime-induced hemolytic anemia by investigating the situation of ceftizoxime-induced hemolytic anemia. METHODS:Using Chinese and English“ceftizoxime”“he-molysis”and other words as key words, related literatures during 1986-2015 were retrieved from CNKI,Wanfang database and PubMed database. Those literatures were analyzed statistically in respects of general information, clinical drug use and manifesta-tion,occurrence time and outcome, etc. The mechanisms of hemolytic anemia were analyzed to put forward prevention and treat-ment measures. RESULTS & CONCLUSIONS:A total of 11 literatures were retrieved, including 4 foreign literatures and 7 domes-tic ones. There were 15 cases in total, including 5 foreign cases and 10 domestic cases. According to the patient’s age, there were 7 cases of infants,4 cases of the elderly,3 cases of middle-aged people and 1 case of youth. The hemolytic anemia often occurred on the 2-7 day(8 cases). Most of the patients were improved after treatment, but there were 3 patients who died. The mechanism of hemolytic anemia mainly included the production of immune complex and non-immunologic protein adsorption. It is suggested to investigate allergic reaction history, pay attention to sensitization test on skin, select suitable drug dosage and avoid drug combina-tion. The patient’s medications should be monitored closely. Once hemolytic anemia occurs,it should be immediately stop using the drug and take appropriate measures as hormone treatment, comprehensive treatment, blood transfusion treatment,to avoid en-dangering the life safety of patients.

Objective To investigate the effect of nutrition intervention on psychological stress combining with sleep deprivation in rats. Methods Seventy-five male SD rats were randomly divided into five groups:control group, composite factor group, composite factor control group, nutrition group Ⅰ and nutrition group Ⅱ. The rat model of 14d psychological stress combining with 48h sleep deprivation was established by Communication Box and improved small platform methods. Rats in the 2 nutrition groups were gavaged with corresponding nutriment everyday; those in other groups were treated with gavage of physiological saline. Morris water maze place navigation test was employed to train rats and measure the ability of rats' spatial working memory. The antioxygenic potential of rats' blood serum and some nutrients were measured with corresponding kits. Results Compared with the composite factor group, the escape latency of all quadrants shortened significantly (P

Objective:To study the coronal bone structure matching of distal radius in normal population and some patients with postoperative distal radius fracture, and to explore the clinical significance of coronal bone structure reduction of distal radius fracture. Methods:CT scans of 80 asymptomatic wrists were performed. Mimics 20.0 and 3-Matic research software were used to measure the matching data of coronal bone structure of the distal radius. Total of 44 patients with distal radius fractures treated with open reduction and volar plate fixation were collected. According to the data coronal bone structure of the distal radius, the matching group was in the normal range, and the mismatching group was less than the normal range. X-ray films were used to evaluate fracture healing, humeral height, ulnar angle and palm tilt angle at 3 months and 12 months after operation. The clinical indexes of wrist pain, wrist function, grip strength and activity were recorded in 2 groups. The DASH score was used for evaluation, and statistical comparisons was made between the two groups of related indicators. Results:The coronal bone structure matching value of the distal radius in 80 normal adults was 45.0%±16.2%. All the 44 patients with distal radius fracture were followed up for an average of 16 months. The postoperative wound healing was good, and the bone healing standard was reached 3 months after the operation. 3 months after surgery, radius height, ulnar deviation angle and palmar inclination angle of the mismatched group were all smaller than those of the matched group, but the differences had no statistical significance. The pronation angle in the mismatched group (68.82°±11.62°) was lower than that in the matched group (76.91°±9.14°), and the difference was statistically significant ( t=2.567, P=0.014). The DASH score in the mismatched group (15.53±2.36) was higher than that in the matched group (13.62±2.52), and the difference was statistically significant ( t=2.591, P=0.013). 12 months after surgery, the VAS score of the matched group (2.08±2.95) was less than that of the mismatched group (2.95±1.24), and the difference was statistically significant ( t=2.348, P=0.024). There was no significant difference in wrist range of motion, grip strength and DASH score between the two groups. Conclusion:The coronal bone structure matching of distal radius is about 45.0% in normal population. Early wrist dysfunction, limited pronation, and wrist pain may occur when the postoperative matching degree of the distal radius fracture is not within the normal range.
