1.Comparison of the behavior characteristics in SHR, WKY and SD rat models of attention deficit/hyperactivity disorder

Objective To compare the differences in behavior characteristics among SHR, WKY and SD rat models of attention deficit/hyperactivity disorder (ADHD), and explore an ideal control model of SHR rats.Methods Using open field test to analyze the rat movement distance, speed, wearing numbers and the number of grooming to evaluate the spontaneous movement in SHR, WKY and SD rats.Using the Morris water maze to test the learning and memory ability among the three rat groups.Results The result of open field test showed that the SHR rats had significantly increased (P< 0.01) total amount of exercise, average speed and wearing numbers than WKY and SD rats.Compared with the WKY rats, SD rats had a significantly higher movement distance (P< 0.01), slightly higher movement speed and wearing number (P< 0.05).In the Morris water maze hidden platform period test, the SD rats had a significantly longer latency than the SHR rats (P< 0.05).SD rats showed longer latency distance on the first, third and fourth days of training, as compared with the SHR rats (P< 0.05 or P< 0.01).Compared with the WKY group, SD rats showed a shorter latency distance in each training time (P< 0.05 or P< 0.01).In the probe trial period, the SD rats showed shorter time and distance ratio to the target quadrant than SHR rats (P< 0.05), while significantly longer than the WKY rats (P< 0.05 or P< 0.01).Conclusions There are significant behavioral differences between SHR and WKY rats, showing certain disadvantages in comparison of the two types of rats.To add SD rats as a control group for SHR rats can improve the comparability of behavior characteristics of SHR rats, and to get more objective evaluation of the behavior characteristics of SHR rats.
2.Biliometric Analysis on TCM Therapy for ADHD in Recent Ten Years

Objective To discuss the research status of TCM therapy for ADHD in China in recent ten years; To provide references for clinical workers. Methods Clinical research papers about TCM therapy for ADHD in CNKI, VIP, and CBM from 1st January 2005 to 1st April 2015 were retrieved. Metrology analysis was conducted from the aspects of publication time, diagnostic criteria, contrasted medicine, and types and features of TCM therapy. Results By eliminating duplication literature, 670 papers were screened out. After the screening of exclusion criteria, 148 papers were included. DSM-Ⅳ and CCMD-3 were the most commonly used diagnostic criteria in the 148 papers; internal therapy was the main therapy in TCM therapy; decoction was the main dosage form; liver-kidney yin deficiency syndrome and heart-liver fire syndrome were the main patterns of syndrome; Acori Tatarinowii Rhizoma, Rehmanniae Radix Praeparata and other 19 Chinese medicinal herbs were the main Chinese mateia medica; Xiaoer Zhili Syrup was the most commonly used Chinese patent medicine; acupuncture and moxibustion and auricular point therapy were the main external therapy, and external combined with internal therapy had the best efficacy. Conclusion TCM therapy for ADHD is with high effectiveness. External combined with internal therapy has the best efficacy.
3.An optimized extraction protocol of prefrontal cortical and striatal synaptosomes from SHR rat

Objective To introduce an improved extraction method of prefrontal cortical and striatal synaptosomes from SHR rat. Methods Synaptosomes were prepared from SHR rat brain tissue by Percoll density gradient centrifugation.Transmission electron microscopy was used to assess the morphology and structural integrity of the synaptosomes.Results The obtained synaptosomes showed oval structures surrounded by an intact membrane.Presynaptic components contained one or more mitochondria and a large number of synaptic vesicles.The synaptic clefts were clearly visible, and prominent part of the characteristic compact structure was clear, complete and with higher electron-density. The synaptosome presynaptic membrane, synaptic cleft, and postsynaptic membrane were well preserved, and the synaptosomes were densely distributed, showing typical morphological characteristics of synaptosomes.Conclusions The results of our study improved the traditional preparation method and provide a less time-consuming, highly productive protocol for preparation of structurally typical and intact synaptosomes, suitable for further research on neuroscience and neurological diseases.

5.Effect of salt-inducible kinase 2 on checkpoint in response to γ-ray irradiation

Objective To investigate the effect of salt-induced kinase 2 (SIK2) in the G2/M checkpoint in response to ionizing radiation and the possible mechanism.Methods HeLa cells were irradiated with 60Co γ-rays.The cell model of knockdown SIK2 expression was constrcuted by transfecting HeLa cells with a pSicoR-based lentivirus vector of expressing SIK2 shRNA by lipofectamin 2000.Western blot and flow cytometry were performed to measure the changes of SIK2 protein level and cell cycle distribution.The phosphorylated histone protein H3 on Ser 10 was used as a molecular marker of mitotic cells for detecting the function of G2/M checkpoint.Results The expression level of SIK2 protein increased in HeLa cells after 60Co γ-ray irradiation.A cell model of knockdown SIK2 expression was successfully generated by transfecting the specific shRNA against SIK2.Depression of SIK2 significantly increased the cellular sensitivity at 1,2,4,6 Gy post-irradiation (t =-3.445,-2.581,-3.251,-2.553,P <0.05),and led cells to release earlier from the G2/M boundary arrest compared to control cells at 5,6 h post-irradiation (t =4.341,6.500,P < 0.05).Western blot analysis indicated that the irradiation-induced phosphorylated CHK2/T68 in SIK2 knock-down cells was earlier than that in control cells.Conclusions salt-induced kinase 2 (SIK2) participates in the regulation of G2/M checkpoint induced by ionizing radiation and affects cellular radiosensitivity.
6.Event-related potentials of non-mediated obsessive-compulsive disorder patients in an emotional Stroop task

Objective To study the difference of attentional bias towards general negative words between non-mediated obsessive-compulsive disorder (OCD) patients and healthy controls.Methods Event-related potentials (ERPs) were collected from 26 non-mediated OCD patients and 24 normal controls while they performed an emotional Stroop task.Results Compared to healthy controls,OCD patients showed significantly longer reaction time (OCD:(467 ± 14.88) ms,Controls:(409.65 ± 15.49) ms,P< 0.01),and had significantly larger P2 and P3 amplitude to all three types of words (P2 amplitude:OCD:(10.41±0.49) μV,Controls:(8.89±0.48) μV,P<0.05;P3 amplitude:(10.04±0.68) μV,Controls:(7.68±0.67) μV,P<0.05).In addition,the P2 peak latency of the frontal area in OCD patients was significantly longer than that in healthy controls(OCD:(190.09±4.71) ms,Controls:(179.60±4.72) ms,P<0.05).Conclusion Current findings suggest that OCD patients are characterized by a general attentional inhibition deficit but not an attentional bias to the negative words.
7.Effect of rehabilitation skills training program on suicide and relapse prevention of patients with depression

Objective This study aims to investigate the effect of rehabilitation skills training on suicide and relapse prevention of patients with depression.Methods Eighty patients were randomly divided into two groups.One group accepted depression rehabilitation skills training and the other group accepted general health education for 4 weeks.Both groups were followed up by 12 months,and the number of relapse and suicide and the score of Health-related quality of life made by Word Health Organization (WHOQOL-BREF) were recorded.Results The rate of relapse (10.0% vs.42.5%) and hospitalization (5.0% vs.20.0%) were lower in skills training group than in control group (P<0.05).Rate of seeking help of suicide was higher in skills training group than in control group (25.5% vs.7.5%) (P<0.05).The suicide mortality was insignificantly different between two groups (0.0% vs.2.5%) (P>0.05).The scores of WHOQOL-BREF were significantly higher in skills training group than in control group in follow-up (P<0.05).Conclusions Rehabilitation skills training program can not only reduce the rate of relapse and suicide but also improve the quality of life of patients with depression.

Malignant tumors continue to pose a significant threat to human life and safety and their development is primarily due to the activation of proto-oncogenes and the inactivation of suppressor genes.Among these,the activation of proto-oncogenes possesses greater potential to drive the malignant transformation of cells.Targeting oncogenes involved in the malignant transformation of tumor cells has provided a novel approach for the development of current antitumor drugs.Several preclinical and clinical studies have revealed that the development pathway of B cells,and the malignant transformation of mature B cells into tumors have been regulated by oncogenes and their metabolites.Therefore,summarizing the key oncogenes involved in the process of malignant transformation of mature B cells and elucidating the mechanisms of action in tumor development hold significant importance for the clinical treatment of malignant tumors.
9. Application evaluation of failure mode and effect analysis in optimization of vascular recanalization process

Objective:
To investigate the clinical application and effect evaluation of failure mode and effect analysis (FMEA) in the optimization of vascular recanalization in patients with ST-segment elevation myocardial infarction (STEMI).
Methods:
A total of 389 STEMI patients admitted to the emergency department of the Fifth Central Hospital in Tianjin from January 2014 to January 2015 were served as the control group, and 398 STEMI patients admitted to the chest pain center of the Fifth Central Hospital in Tianjin from January 2016 to October 2017 were served as the experimental group. In the control group, routine emergency treatment was used. At the same time, the intervention room was 24-hour prepared for emergency vascular recanalization. The experimental group used FMEA. Through the usage of FMEA, the main factors those caused the delay in revascularization treatment were determined, and the revascularization process was optimized for these influencing factors, thereby shortening the "criminal" blood vessel opening time of patients. The door-to-balloon dilatation time (D-to-B time), troponin testing time, placement time of the catheterization room, initiation of the catheterization room to balloon dilatation time, and preoperative and 1 week postoperative N-terminal pro-brain natriuretic peptide (NT-proBNP) levels, heart function parameters [left ventricular ejection fraction (LVEF), left ventricular short axis shortening rate (FS), left ventricular end-systolic diameter (LVESD), and left ventricular end-diastolic diameter (LVEDD)] within 1 week, 3 months and 6 months after intervention, and the incidence of main cardiovascular adverse events within 1 month after intervention, hospital mortality, the length of hospital stay, and readmission within 1 year in the patients of two groups were recorded.
Results:
D-to-B time (minutes: 70.6±3.6 vs. 79.4±8.7), troponin testing time (minutes: 17.1±2.3 vs. 65.2±6.5), placement time of the catheterization room (minutes: 28.9±9.8 vs. 52.3±12.2) and activation of the catheterization room to balloon expansion time (minutes: 47.3±9.3 vs. 65.1±7.2) in the experimental group were significantly shorter than those in the control group (all

Objective To provide evidence for the long-term development of biobank in China,we retrieved literature on biobank for analyzing the development status and trend of biobank in the world and China.Methods Using the topic of "Biobank" to search articles from 2008 to 2017 in the core collection of Web of ScienceTM,we conducted statistical analysis covering the annual published quantity,the corresponding national/regional distribution,h-index,and the authors of these articles.Results A total number of 2012 articles on biobank published from 2008 to 2017 were retrieved from the core collection of Web of ScienceTM,in which the annual published quantity on biobank in the world and China was basically similar,and was on the upward trend;among them,the top three countries/regions with the published quantity were United States,England and China;the top three countries/regions with the h-index were United States,England and the Netherlands,and China ranked fifth;the cooperation degree and co-authorship rate of the authors in China were higher than that in the world.Conclusions The annual growth trend of published quantity on biobank in China is basically consistent with that in the world.Chinese biobank plays an important role in the world,and the Chinese researchers on biobank attach more importance to cooperation.However,the scientific research achievements with significant academic influence need to be further improved.Therefore,in order to build a standardized and high-quality biobank,Chinese biobank still needs innovation and exploration continuously.
