1.The effects of low intensity pulsed ultrasound on the osteogenetic differentiation of BMSCs on titanium with different surface topography

Objective:To observe the effects of low intensity pulsed ultrasound(LIPUS) on the osteogenic differentiation of rat bone marrow mesenchymal stem cells(BMSCs) on titanium surface.Methods:BMSCs from Wistar rat bone marrow were respectively cultured on the flat titanium surface and the large grain blast acid etched(SLA) titanium surface,and induced by mineralization medium.Then,the cells were interfered by LIPUS and a control condition.Alkaline phosphatase(ALP) were quantitative determinated after 3 and 7 d mineralization induction respectively,ALP staining were observed after 14 d induction.Alizarin red staining were observed after 21 d mineralization induction.Osteogenic related protein and gene expressions were detected after mineralization induction.Results:ALP in culture medium of LIPUS group was higher than that of the control group after 3 d and 7 d mineralization induction(P<0.05).LIPUS group showed stronger ALP staining and alizarin staining,and more mineralized nodules than control group.The expression of osteogenic related proteins,including Runx2,BMP2,OPN in LIPUS group increased.Osteogenic related genes expression,including ALP,Runx2,BMP2,OPN,OCN and Col-1 of the LIPUS group increased.Conclusion:The osteogenic differentiation of BMSCs on the fiat titanium surface or SLA titanium surface can be promoted by LIPUS.
2.Study on Anti-RSV Activity of Natural Curcuminoids

Objective:To study the anti-human respiratory syncytial virus ( RSV) activity of natural curcuminoids in Curcuma lon-ga in vitro. Methods:The anti-RSV activity of ethyl acetate part of 95% ethanol extracts of Curcuma longa and natural curcuminoids in Curcuma longa in vitro were detected by cytopathic inhibition method. Results:Bisdemethoxycurcumin exhibited anti-RSV activity with IC50 of 12. 5 μg·ml-1 and SI of 1. Conclusion:Natural curcuminoids in Curcuma longa exhibit anti-RSV activity.
3.Expression of gene encoding mature protease of Schistosoma japonicum asparaginyl endopeptidase (Sj32) and diagnostic application of recombinant protein

Objective To express the gene encoding mature protease of Schistosoma japonicum asparaginyl endopeptidase (Sj32) and evaluate the potential of the recombinant protein rSj32 in diagnosis of domestic animal schistosomiasis. Methods The DNA fragment encoding mature protease of Schistosoma japonicum asparaginyl endopeptidase was cloned with PCR from pET-28(a)/Sj32, and a recombinant plasmid was previously constructed in the laboratory, which contained the ORF of the gene encoding the pro-enzyme Sj32. The amplified DNA fragment was subcloned into pET-28a( + ) and the recombinant plasmid was transformed into E. coli BL21 (DE3) to express the mature protease of Sj32. Then the recombinant antigen (rSj32) was used in ELISA assay to diagnose schistosomiasis of mice, rabbits and water buffalo artificially infected. The detection effects of soluble Schistosoma japonicum egg antigen (SEA) , rSj32 and the recombinant 23 KDa membrane protein were compared. Results The recombinant antigen rSj32 with a molecular weight 41 KDa was successfully produced in E. coli BL21 ( DE3) and was purified with His Column with a yield of 25 mg/L E. coli culture. By using rSj32 as coating antigen in ELISA assay to detect the specific antibody in artificially infected mice, rabbits and buffalo, the sensitivities were 88.9% , 85.0% and 71.8% , respectively, the specificities were 100% , 96.7% and 96. 9% , respectively. There were no significant differences among the detection results of rSj32, SEA and rSj23. Conclusion rSj32 is a promising antigen for serological diagnosis of domestic animal schistosomiasis.
4.Bibliometric analysis of Biobank based on Web of ScienceTM

Objective To provide evidence for the long-term development of biobank in China,we retrieved literature on biobank for analyzing the development status and trend of Biobank in the world and China.Methods Using the topic of "Biobank" to search articles from 2008 to 2017 in the core collection of Web of ScienceTM,we conducted statistical analysis covering the annual published quantity,the corresponding national/regional distribution,h-index,and the authors of these articles.Results A total number of 2012 articles on Biobank published from 2008 to 2017 were retrieved from the core collection of Web of ScienceTM,in which the annual published quantity on Biobank in the world and China was basically similar,and was on the upward trend;among them,the top three countries/regions with the published quantity were United States,England and China;the top three countries/regions with the h-index were United States,England and the Netherlands,and China ranked fifth;the cooperation degree and co-authorship rate of the authors in China were higher than that in the world.Conclusions The annual growth trend of published quantity on Biobank in China is basically consistent with that in the world.Chinese Biobank plays an important role in the world,and the Chinese researchers on biobank attach more importance to cooperation.However,the scientific research achievements with significant academic influence need to be further improved.Therefore,in order to build a standardized and high-quality biobank,Chinese biobank still needs innovation and exploration continuously.

6.Buyang Huanwu decoction promotes angiogenesis and improves hemorheological parameters after cervical spinal cord injury

Objective:
To explore the effects of Buyang Huanwu decoction (BYHWD) on vascular neogenesis and hemorheological parameters following cervical spinal cord injury (SCI).
Methods:
An acute cervical SCI model was established using 84 female Sprague–Dawley rats. Functional recovery of the rats was evaluated using the forelimb locomotor scale score, forelimb grip strength test, and Basso-Beattie-Bresnahan score. The animals were subsequently euthanized at days 7 and 28 postoperatively. The gross morphology, neuronal survival, and myelin sheath in the injured area were evaluated using hematoxylin and eosin (HE), Nissl, and luxol fast blue (LFB) staining, respectively. Immunofluorescence staining was used to observe CD31 expression 7 days post-injury. Furthermore, the expression of CD31, neuronal nuclear protein (NeuN), and myelin basic protein (MBP) were evaluated 28 days post-injury. Additionally, vascular endothelial growth factor A (VEGFA) and VEGF receptor-2 (VEGFR-2) expression was evaluated using western blotting. Whole-blood viscosity, plasma viscosity, and red blood cell aggregation were measured using a hemorheometer.
Results:
From postoperative days 3–28, motor function in the BYHWD group began to recover considerably compared to the SCI group. BYHWD effectively restored spinal cord histopathology. In addition, the number of NeuN-positive cells, and fluorescence intensity of CD31at 7 and 28 days and MBP significantly increased in the BYHWD group compared with the SCI group (all P < .05). Moreover, this decoction significantly upregulated the expression of VEGFA and VEGFR-2 (all P < .05). BYHWD improved the hemorheology results (i.e., except erythrocyte aggregation index in the low-dose group), revealing statistically significant differences compared with the SCI group (all P < .05).
Conclusion
BYHWD effectively promoted angiogenesis, improved hemorheological parameters, and protected neurons and myelin sheaths, ultimately promoting the recovery of neurological function after cervical SCI in rats. These findings suggest that BYHWD promotes vascular neogenesis through the VEGFA/VEGFR-2 pathway.
7.Effects of intensive self-care intervention in patients undergoing day self-retaining laryngoscope vocal polyp resection

Objective To explore the effects of intensive self-care intervention on self-care agency, reactive mode, psychology and voice functional recovery in patients undergoing self-retaining laryngoscope CO2 laser vocal polyp resection after leaving hospital. Methods From January 2016 to June 2017, 75 patients with selective self-retaining laryngoscope CO2laser vocal polyp resection were selected. From January to September 2016, a total of 37 patients with conventional nursing were selected as control group. From October 2016 to June 2017, a total of 38 patients accepting intensive self-care intervention were chosen as observation group. The self-care agency, reactive mode and voice functional recovery of patients in two groups were compared at different times. Results The score of self-care agency of two groups all increased one and three months after leaving hospital compared with that on admission (P< 0.05). The score of self-care agency of observation group one and three months after leaving hospital were (125.32±6.75) and (122.46±8.24) significantly higher than those [(117.69±12.47) and (115.38±10.25)]of control group (P< 0.05). The score of positive coping style of observation group one and three months after leaving hospital were (23.87±1.76) and (24.00±1.83) significantly higher than those [(20.13±1.20) and (20.08±1.35)]of control group (P< 0.05). The fine rate of voice functional recovery in observation group one month after leaving hospital was 78.95% higher than that (51.35%) in control group with a significant difference (P<0.05). However, there was no statistically significant difference in the fine rate of voice functional recovery between two groups after 3 months leaving hospital (P>0.05). Conclusions Intensive self-care intervention can improve the self-care agency, positive coping style and promote voice functional recovery in patients undergoing self-retaining laryngoscope CO2laser vocal polyp resection.
8.Effect of doctor-nurse integration mode health education on the compliance of patients with breast cancer treated with Fulvestrant injection

Objective To explore the effect of doctor-nurse integration mode health education on the compliance of patients with breast cancer treated with Fulvestrant injection. Methods A total of 89 patients with breast cancer who received Fulvestrant injection in Breast Center of the Affiliated Hospital of Qingdao University were selected from October 2015 to June 2016 by convenient sampling method and they were divided into observation group (n=46) and control group (n=43) using randomly number table method. Patients in the control group received routine outpatient clinic medication treatment, while patients in the observation group received extra doctor-nurse integration mode health education during the treatment circle. Endocrine therapy compliance questionnaire for breast cancer patients was applied to assess treatment compliance of patients in two groups. Results After the implementation of doctor-nurse integration mode health education, scores of treatment compliance and each dimension of observation group were significantly higher than those of the control group (P<0.05). Conclusions For patients with breast cancer treated with endocrine therapy for a long time, the implementation of doctor-nurse integration mode health education could improve the compliance of patients, ensure the completion of the treatment cycle and promote the rehabilitation of patients.
9.Investigation on antimicrobial usage in 451 patients with complicated intra-abdominal infection

Objective To investigate the microbiological epidemiology and clinical use of antibiotics in patients complicated intra-abdominal infection (cIAI),therefore to optimize antibiotic use and to promote antimicrobial stewardship.Methods A total of 451 patients with cIAI from a Chinese tertiary hospital between January 2015 and December 2016 were retrospectively reviewed.The infection severity,timing of microbiological specimen sampling,culture results,initial antibiotic selection and later anti-infective regimen adjustment were analyzed.Results Three hundred and sixteen (70.1%,316/451) patients undergone microbiological investigation at infection sites within 3 days and 133 (42.1%) patients had a positive culture,of which 64.5% were Enterobacteriaceae.Three hundred seventy-four patients (82.9%) initially received broad-spectrum antibiotics against gram-negative bacilli.Sixty-five patients (14.4%) initially received combined antibiotic therapy,of which 30.8% were deemed as overuse.Among 308 patients who initially received broad-spectrum antibiotic therapy,268 patients (87.0%) clinically improved in five days,while de-escalation was only conducted in 72 cases (26.9%).On average,patients were treated with (2.29±1.30) antibiotics for a duration of (10.6±6.5) days,and 42.4% received combined antibiotic therapy during hospitalization.Conclusions The major microbiological pathogens in cIAI patients in our hospital were Enterobacteriaceae.However,there are phenomena such as excessive usage with broad-spectrum antibiotics,insufficient awareness of de-escalation,and long course of anti-infective therapy,which needs to be further improved.

DDX5 helicase (DEAD box helicases 5), also known as P68, is an important member of an ATP dependent RNA helicase.Studies have shown that DDX5 is abnormally expressed in a variety of cancers, targeting a variety of tumor related signal pathways, regulating upstream and downstream factors to affect the occurrence, invasion and migration of tumor cells. This article describes the biological role of DDX5 in malignant tumors and provides prospects for targeted treatment of tumors.
