1.Determination of Syringin and Syringaresinol in Ciwujia Injection by RP-HPLC

Objective To establish the determination method of syringin and syringaresinol in Ciwujia Injection with RP-HPLC. Methods C18 ODS was used as a stationary phase. The mobile phase consists of water and acetonitrile. The gradient condition was 0→20 min, A:90%→50%. The flow rate was 1.0 mL/min. The detective wavelength was set at 220 nm. Results The linear range of syringin was 0.03~0.16 ?g (r =0.999 7, n=5) and the linear range of Syringaresinol was 0.02~0.13 ?g (r =0.999 3, n=5). The average recovery of syringin was 99.34% and RSD=0.58%. The average recovery of Syringaresinol was 99.22% and RSD=0.71%. Conclusions The method was simple, convenient and accurate, and can be used for content determination of syringin and syringaresinol in Giwujia injection.
2.Analysis of the volatile oil in fructus lycii(Lycium barbarum L.) by GC-MS and HELP method

Objective Gas chromatography-mass spectrometer (GC-MS) technology combined with Heuristic evolving latent projections (HELP) method were used to qualitatively analyze the volatile oil of fructus lycii (Lycium barbarumL.)Methods With the best temperature-programmed chromatographic condition, overlapping peaks among the total ion chromatogram were separated. Then an automatic mass spectral deconvolution and identification system (AMDIS) was used to identify volatile components comprehensively. Finally, the pure chromatographic peaks were matched with NIST 05a mass spectra library for giving the precise qualitative result. Results Totally, 29 compounds in fructus lycii (Lycium barbarum L.) were identified accurately.Conclusions Compared with the traditional qualitative analysis method, the new established method which GC-MS technology combined with HELP could get tremendous advantages. It not merely enhanced the accuracy of identification but also solved the bottleneck of handling the inseverable compounds.
3.Effects of aerosolized prostaglandin E1 before one lung ventilation on lung protection in patients undergo-ing surgery for oesophageal cancer

Objective To observe the effects of aerosolized prostaglandin E1 (PGE1 )via right lung before one-lung ventilation (OLV ) on shunt rate (Qs/Qt ) and oxygenation in patients undergoing surgery for oesophageal cancer.Methods Sixty patients scheduled for elective trans-left-thoracic esophagectomy for esophageal cancer were randomly and single-blindly located into two groups.Patients in each group received different therapy before OLV,namely inhaling PGE1 0.2μg/kg via right lung in group P and inhaling normal saline in group C.The PaO 2 and hemodynamic indicators of two groups were recorded at these points:before OLV(T1 ),OLV 10 min (T2 ),OLV 1 5 min (T3 ),OLV 30 min (T4 ),OLV 60 min (T5 ),OLV 120 min (T6 ),.Results PaO 2 in both groups were declined straightly since OLV and fell to the lowest point at T4 in group C.PaO 2 in group P at T2-T4 were significantly higher than that in group C (P <0.05),and the lowest point of which was recorded at T5 .Qs/Qt in group P was significantly lower than that in group C at T2-T4 (P <0.05).There were no significant differences in hemodynamics indicators between the two groups. Conclusion Inhalation of 0.2 μg/kg PGE1 before OLV via one lung can reduce pulmonary shunt and improve PaO 2 in thoracic surgery patients.
4.Role of the dynamic contrast-enhanced MRI in assessing the response to neoadjuvant chemotherapy of breast cancer

Background and purpose:Neoadjuvant chemotherapy to breast cancer has become a mature treat-ment method. The purpose of this study was to evaluate the dynamic contrast-enhanced MRI in assessing the response to neoadjuvant chemotherapy (NAC).Methods:Twenty-four female patients with breast carcinoma (24 were all inva-sive ductal carcinoma) underwent dynamic contrast-enhanced MRI (DCE-MRI) before, after the second and ifnal cycle of NAC. For each patient and each MRI examination, the maximum diameter of tumor, volume transfer constant (Ktrans), exchange rate constant (Kep), and extravascular extracellular volume fraction (Ve) were compared. According to the method of response evaluation criteria in solid tumor (RECIST), the results of neoadjuvant chemotherapy were divided into responder and non-responder.Results:All 24 patients were unilateral and single breast cancer; there were 17 cases of responders and 7 cases of non-responders according to RECIST criteria. For 17 cases of responders, both the average values ofKtrans andKepwere signiifcantly descended after neoadjuvant chemotherapy (allP<0.05).Conclusion:The quantitative parameters ofKtrans andKep can evaluate objectively and veridically the response to neoadjuvant chemother-apy for breast cancer in dynamic contrast-enhanced MRI.

6.Arterial Plaques Identification Based on Intravascular Ultrasound Elasticity Imaging.

Intravascular ultrasound (IVUS) is widely used in coronary artery examination. Ultrasonic elastography combined with IVUS is very conspicuous in identifying plaque component and in detecting plaque vulnerability degree. In this study, a simulation model of the blood vessel based on finite element analysis (FEA) was established. The vessel walls generally have radial changes caused by different intravascular pressure. The signals at lower pressures were used as the pre-deformation data and the signals at higher pressure were used as the post-deformation data. Displacement distribution was constructed using the time-domain cross-correlation method, and then strain images. By comparison of elastograms under different pressures, we obtained the optimal pressure step. Furthermore, on the basis of the obtained optimize pressure step, the simulation results showed that this method could effectively distinguish characteristics between different component plaques, and could guide the later experiments and clinical applications.

7.Clinical efficacy of argon- helium knife cryotherapy followed by chemotherapy for advanced non- small cell lung cancer

Objective To explore the short- term efficacy of argon- helium knife cryotherapy followed by chemotherapy in treating advanced non- small cell lung cancer (NSCLC) and to investigate its effect on the long- term survival. Methods During the period from March 2005 to March 2008, a total of 61 patients withⅢb or Ⅳ stage NSCLC received argon- helium knife cryotherapy followed by chemotherapy (study group), and other 52 patients with Ⅲb or Ⅳ stage NSCLC were treated with chemotherapy only (control group). The clinical data were retrospectively analyzed. The pain was evaluated by numeric rating scale (NRS) and the quality of life (QOL) was assessed by functional assessment of cancer therapy- general (FACT- G) scale. The clinical effect was evaluated according to RECIST criteria for solid tumor, and the patient’s survival time was recorded. Results (1) Twenty- six patients had local pain before the treatment, and the pain was relieved in different degrees after cryotherapy. The QOL, including all respects of FACT - G, was significantly improved after cryotherapy in all 61 patients. (2) The remission rate of the study group and the control group was 34.4% and 15.4% respectively, the difference between the two groups was statistically significant (P <0.05). (3) The median survival time of the study group and the control group was 12.9 months and 9.5 months respectively, and the one- year survival rate of the study group and the control group was 53.6% and 35.4% respectively. The differences between the two groups were statistically significant (P < 0.01). Conclusion Argon- helium knife cryotherapy is a safe and effective local treatment for advanced NSCLC, which can quickly reduce the tumor load, relieve the pain and improve patient’s quality of life. Cryotherapy with subsequent chemotherapy is superior to simple chemotherapy in improving the patient’s survival rate.
9.Study on the protective effect of lycium barbarum polysaccharide on light aging resistance in rats

Objective To study the mechanism of the protective effect of lycium barbarum polysaccharide on light aging resistance rats by using the metabolic profile and metabolic target analysis technique.Methods Ultraviolet irradiation induced Wistar rats were induced to produce skin photo aging model and 24 rats were randomly divided into three groups, including control goup, model group, Lycium barbarum polysaccharide group (LBP). After modeling for 24 hours, LBP group was conducted with Lycium barbarum polysaccharide solution of 10 mg/kg. Control group and model group were given same volume of stroke-physiological saline solution for 14 days. The biochemical indexes such as rat serum antioxidant activity of related substances and MDA were measured in model group and drug group; the urine metabolomics study was also investigated for the mechanism of lycium barbarum polysaccharide against light aging band.Results Compared with the model group, the LBP group rats total superoxide dismutase activity (301.51 ± 42.56 U/mgvs.93.41 ± 56.31 U/mg), hydroxyproline (8.91 ± 5.78μg/mgvs.4.74 ± 1.54μg/mg) content significantly increased (P<0.05), but the malondialdehyde (8.54 ± 6.41 nmol/mgvs.21.31 ± 6.58 nmol/mg) decreased without any statistics difference (P<0.05). The urine metabonomics results showed that LBP could regulate the skin photoaging of multiple metabolic pathways and key metabolic enzymes in the process, such as peanut four acid, tyrosine, taurine, citric acid and hippuric acid, L-cysteine, inositol, threonine etc.Conclusions In the process of skin photoaging in rats, multiple metabolic pathways in vivo were disordered, and Lycium barbarum polysaccharide could play a protective role by regulating the key metabolic enzymes in the network.
10.Expression of bone morphology proteins in mice with collagen-induced arthritis

Objective To establish collagen‐induced arthritis (CIA) model used of DBA/1J mouse ,a preliminary study of the expression levels of BM Ps in mononuclear cells of peripheral blood and the joints tissue of CIA mice .Methods We induced DBA/1J mice and developed arthritis pathology by using of bovine type Ⅱ ;then ,we detected mRNA and protein expression levels of BMPs by using quantitative PCR and immunohistochemical staining .Results We successfully established CIA mouse model .The expres‐sion of BMP4 and BMP9 mRNA was significantly down‐regulated in peripheral blood mononuclear cells and in the pathogenesis of joint tissues of CIA mice (P<0 .05) .It showed that BMP9 protein significantly decreased in joint tissues of CIA mice by immuno‐fluorescence technique (P=0 .002) .Conclusion At the genetic level ,the expression of BMP4 and BMP9 could be significantly down‐regulated in the CIA mouse .At the protein level ,BMP9 could be significantly down‐regulated in the CIA mouse .
