1.Effect of modified Yulin Decoction on reproductive endocrine related indexes and ovarian function in patients with premature ovarian failure

Objective To analyze the effect of modified Yulin Decoction on hormones and ovarian function in patients with premature ovarian failure (POF).Methods 142 cases of patients with POF and kidney-yang deficiency syndrome were selected and randomLy divided into the control group (n=64) and the observation group (n=78).Levels of serum hormones and ovarian function,the traditional Chinese medicine (TCM) symptom score in the two groups were observed before and after treatment.Results Before treatment, there were no significant differences between the two groups in the scores and levels of hormones.After treatment, levels of serum FSH, LH and E2 were significantly decreased in the two groups, and the changes were more obvious in the observation group than the control group (P<0.05).After treatment, TCM symptom scores of two groups were significantly decreased, and the observation group were significantly lower than those in the control group (P<0.05).Conclusion Modified Yulin Decoction is significantly effective in the treatment of POF of spleen and kidney-yang deficiency syndrome.It can significantly improve the level of estrogen, adjust endocrine and relieve clinical symptoms.
2. Effect of insulin in combination with selenium on blood glucose and GLUT4 expression in skeletal muscle of streptozotocin-induced diabetic rats

Objective: To evaluate the effect of low-dose insulin [1 U/(kg·d)] in combination with selenium [180 g/(kg·d)] on general physiological parameters and glucose transporter (GLUT4) level in skeletal muscle of streptozotocin (STZ)-induced diabetic rats. Methods: Diabetic rats were treated with insulin, selenium, and insulin and selenium in combination for four weeks. The level of blood glucose was determined using One Touch SureStep Blood Glucose meter and the level of GLUT4 in skeletal muscle was examined by immunoblotting and immunohistochemistry. Results: Our data showed that insulin in combination with selenium could significantly lower blood glucose level and restore the disturbance in GLUT4 level in skeletal muscle. Treatment with insulin was only partially effective in restoring diabetic alterations. Conclusion: It can be concluded that there is a synergistic action between insulin and selenium, and that treatment of diabetic rats with combined doses of insulin and selenium is effective in the normalization of blood glucose level and correction of altered GLUT4 distribution in skeletal muscle of diabetic rats.

4. The pharmacokinetics of irinotecan in rats with immunological liver injury

OBJECTIVE: To investigate the pharmacokinetics of irinotecan (CPT-11) in SD rats with immunological liver injury. METHODS: The animal model of immunological liver injury was established by intraperitoneal injection of lipopolysaccharide (LPS) plus Bacille-Calmette-Guerin (BCG) in rats. Rats were randomly divided into two groups; normal control group and immunological liver injury model group. The two groups were injected with CPT-11 (20 mg · kg-1) via tail vein and plasma concentrations of CPT-11 and its metabolite SN-38 were determined by HPLC. Pharmacokinetical parameters were calculated. RESULTS: Compared with the normal control group, AUC0-24h, AUC0-∞ and ρmax of CPT-11 in immunological liver injury rats were increased significantly (P < 0.05), but AUC0-24h, AUC0-∞ and ρmax of SN-38 were decreased significantly (P < 0.05). CONCLUSION: Under the state of immunological liver injury, the hydrolysis of CPT-11 is inhibited significantly. The underlying mechanism may be related to the down-regulation of the expression of carboxylesterase 2 by LPS. Copyright 2012 by the Chinese Pharmaceutical Association.
5.Optimization of hydrolysis process of linarin using response surface methodology and research about ARI activity of glycosylation-acacetin.

OBJECTIVETo optimize the hydrolysis process of linarin by response surface methodology, and to use the model of aldose reductase to study the acacetin's activity of aldose reductase inhibitory.
METHODThe model of acacetin enzyme in vitro was established by the determination of fluorescence absorption of NADPH, the inhibition rate of acacetin aldose reductase was calculated, and then the IC50 of hydrolysis was obtained. The hydrolysis process of linarin hydrolysis condition was optimized by using response surface method.
RESULTThe results indicated that the IC50 of acacetin (2.74 mg x L(-1)) was less than the IC50 of linarin (3.53 mg x L(-1)). Hydrolyzation time of 7.4 h, sulphuric acid concentration of 0.54 mol x L(-1) and the ratio of material to liquid of 3 : 1 were the optimum conditions.
CONCLUSIONHydrolyzate acacetin has preferable inhibitory activity of aldose reductase. The optimized hydrolysis condition of linarin is convenient to use with good predictability.

6.Developments of neutrophil function and the relationship between neutrophils dysfunction and periodontitis.

Polymorphonuclear neutrophil leukocyte (PMN) is an important member of the human immune cells. Recentyears, the recognition of the PMN function and the relationship between PMN and periodontitis have been updated. Besidesthe pathogens killing and phagocytosis, PMN also play an important role in immunoregulation and proresolving. The maintaining of PMN homeostasis is an intricate process and the precondition of defense function, which involves activation, adhesion, recruitment, apoptosis and efferocytosis. The regulatory mechanism of PMN homeostasis called neutrophil rheostat, it works through several cytokines and cells. Any factors that break the homeostasis will result in the damage of host immunity,and may relate to the occurrence of periodontitis. Moreover, PMN dysfunction, because of host factors or microorganism factors, is closely related to periodontitis, especially those associated with systemic diseases and gene defect.

9.Effect of osteogenic growth peptide on proliferation and differentiation of bone marrow stromal cells

BACKGROUND: Osteogenic growth peptide (OGP) is a newly discovered growth factor to promote osteogenesis, the effect on proliferation and differentiation of bone marrow stromal cells (MSCs) arouses more and more attention.OBJECTIVE: To elucidate the effect of OGP on proliferation and differentiation of MSCs in vitro.DESIGN, TIME AND SETTING: A randomized COntrol experiment was performed at the Key Laboratory for Oral Biomedical Engineering by the State Ministry of Education from February to June 2006.MATERIALS: A 6-month-old male Sprague-Dawley rat was used for the extraction of MSCs; and OGP was provided by Sigma Company, USA.METHODS: MSCs were cultured individually in a-MEM medium containing various COncentrations of OGP (10-11 mol/L,10-10 mol/L, 10-9 mol/L, 10-8 mol/L and 10-7 mol/L) and fetal bovine serum. The cells in the control group were cultured with α-MEM medium containing FBS alone.MAIN OUTCOME MEASURES: Identification of MSCs; cell proliferation using MTT method; alkaline phosphatase (ALP) activity using enzyme-linked immunosorbent assay; core binding factorα1 (Cbfa1) mRNA expression using RT-PCR.RESULTS: The addition of OGP resulted in an increase of cell proliferation (10-9mol/L OGP, P < 0.05), ALP activity (10-8 mol/LOGP, P < 0.05) and Cbfal mRNA expression (P < 0.05).CONCLUSION: OGP stimulates proliferation and sequential differentiation of MSCs via inducing Cbfal mRNA expression.

Objective To select the optimum condition for the determination of formaldehyde in the car and to understand the formaldehyde pollution situation,dynamic change law and the influence factors.Methods The national standard method,acetyl acetone spectrophotometer,was used to determine formaldehyde.The investigated cars were the ones which left the factory for one month,three months,six months and one year.The sampling time,rate of flow and coloration time were determined.The influence of temperature and ventilation on the formaldehyde content in the car was analyzed.Results The formaldehyde content in the car was influenced by the temperature,ventilation,date of production and the type of the car.The optimum conditions for the determination of formaldehyde in the car were as follows,10 min of sampling time,0.3 L/min of sampling flow rate and 3 min of coloration time.Conclusion Ventilation is an effective measure to reduce the formaldehyde content in the car.Higher temperature is good for the release of formaldehyde.The formaldehyde content in the car will reach the standard limit in one year after left factory.
