1.Effects of etomidate preconditioning on expression of HL-60 cell apoptosis-related proteins

Objective To investigate the effects of etomidate preconditioning on the expression of procaspase-3, caspase-9 p35 and caspase-8 p20 in HL-60 cells. Methods HL-60 cells were purchased from Shanghai life science institute and cultured in RPMI-1640 culture medium at 37℃ in 5% CO2 incubator. The cells were randomy divided into 3 groups ( n = 3 each): control group (group C), etomidate group (group E) and etomidate preconditioning group (group EP). In group E, the cells were exposed to 500 μmol/L etomidate and incubated for 24 h. In group EP, the cells were exposed to 1μmol/L etomidate for 1 h and was allowed to recover for4 h after etomidate washout, then etomidate 500μmol/L was added and the cells were incubated for 24 h. The expression of procaspase-3, caspase-9 p35 and caspase-8 p20 was determined using Western blot. Results The procaspase-3 expression was significantly down-regulated, while the expression of caspase-9 p35 and caspase-8 p20 was up-regulated ingroup E and EP as compared with group C ( P < 0.05). The procaspase-3 expression was up-regulated,while the expression of caspase-9 p35 and caspase-8 p20 was down-regulated in group EP as compared with group E ( P < 0.05). Conclusion Etomidate preconditiong can inhibit etomidate-induced down-regulation of procaspase3 expression and up-regulation of caspase-9 p35 and caspase-8 p20 expression, resulting in suppression of HL-60 cell apoptosis induced by etomidate.
2.Preparation and in vitro Drug Release of Betahistine Dihydrochloride Sustained-release Matrix Tablets

Objective: To prepare betahistine dihydrochloride sustained-release matrix tablets. Methods: The tablets were pre-pared with water soluble HPMC matrix, and the release behaviors were investigated by single factor study. The formula and preparation procedures were optimized by orthogonal design. Results:The optimal technology was as follows:using 60% HPMC K15M as the ma-trix material, calcium hydrogen phosphate as the filler, 10% PVP in 90% alcohol as the bonding agent;wet granulation compression technique was used to prepare the tablets with the tablet weight of 500mg. The in vitro drug release fits a Higuchi equation and the drug can be sustained-released within 12 h. Conclusion:The preparation technology is simple and the tablets have sustainol release behav-ior.

4.Clinical application and prospect of suspension array technology

Suspension array technology,a bead-based multiplex assay,allows a fast and automatic detection of multi-analytes and batch samples in limited sample volume,and it is great potential clinical application in the determination of autoimmune antibodies,muhitumor markers,the phenotype of Human Leukocyte Antigen and the genes of infectious pathogens.However,currently,there are various limitations which hinder the clinical application of this technology broadly.The globally accepted panel of multi analytes,performance criteria,and quality control programs should be established before we get benefit from this high throughput,sensitivity and repeatability platform,which will help to provide scientific and accurate laboratory data for the clinical diagnosis,treatment and prevention.
5.Clinical and genetic analysis of a Chinese family with Walker-Warburg syndrome

Objective To analyze the clinical,molecular and genetic features of a Chinese family with WalkerWarburg syndrome(WWS).Methods The clinical data of the proband and his family members were collected.Genomic DNAs from the patient and his parents were extracted with standard procedures from the peripheral blood leukocytes.Polymerase chain reaction and DNA direct sequencing were employed to analyze all of the 20 exons of the POMT1 gene to determine the mutation,and the relationship between genotype and phenotype was analyzed.Results The proband presented with delayed psychomotor development,muscle hypotonia and early joint contractures,his serum creatine kinase was elevated moderately and the brain magnetic resonance imaging (MRI) displayed brain structural malformations,cerebellar cyst,bilateral dilatation of the lateral ventricle,cerebellum and brainstem dysplasia.Further genetic testing detected a compound heterozygous mutation of c.313C > T,p.Arg105Cys inherited from his father,a frameshift mutation c.2208delG,p.Trp736X inherited from his mother,both of which were known as pathogenic mutations.Conclusions According to the study,the proband carried compound heterozygous mutation of POMT1 gene,and his parents were heterozygous carriers,which is consistent with autosomal recessive inheritance.The child is definitely diagnosed as WWS.Genetic counseling and prenatal diagnosis are available for this family.
6.Protein Folding Study Based on The HNP Model and The Relative Entropy Approach

Twenty kinds of amino acids are simplified into 3 types: hydrophobic amino acids (H), hydrophilic amino acids (P) and neutral amino acids (N). Each residue is reduced to a bead which locates in the position of the C?琢 atom. The off-lattice model is adopted and the relative entropy is used as a minimization function to predict the tertiary structure of a protein. A new contact intensity function is given to consist with protein design research based on the relative entropy. Testing on several real proteins from Protein Data Bank (PDB) shows the good results obtained with the model and method. The root mean square deviations (RMSD) of the predicted structures relative to the native structures range from 0.30 to 0.70 nm. A foundation for studying protein design using the HNP model and the relative entropy was made.
7. Diterpenoid alkaloids in aerial parts of Aconitum transsectum

Objective: To study the diterpenoid alkaloids in the aerial parts of Aconitum transsectum. Methods The compounds were isolated and purified by silica gel and Sephadex LH-20 chromatography from the aerial parts of A. transsectum, and the structures were identified by spectral analysis (1H-NMR, 13C-NMR, MS). Results: A total of 16 compounds were isolated from A. transsectum and characterized as longtoaconitine A (1), 14-acethyltalatisamine (2), 8-O-methylsachaconitine (3), talatisamine (4), vilmorrianine D (5), 14-acetylsachaconitine (6), forestine (7), chasmanine (8), crassicausine (9), homochasmanine (10), crassicautine (11), 8-deacetylyunaconitine (12), liljestrandisine (13), vilmorrianine B (14), yunaconitine (15), and isotalatizidine (16). Conclusion: Compounds 1-16 are isolated from aerial parts of A. transsectum for the first time, and compounds 1, 3, 9-11, 13-14, and 16 are isolated from A. transsectum for the first time.
8.Analysis of thigh pain after treatment of femoral trochanteric fractures by proximal femoral nail an-tirotation Ⅱ

Objective To analyze the causes for the thigh pain after treatment of femoral trochanteric fractures by proximal femoral nail antirotation Ⅱ( PFNA Ⅱ) . Methods Included in this ret-rospective study were 236 patients who had been treated by us for femoral trochanteric fracture from October 2011 to December 2015. They were 103 men and 133 women, aged from 42 to 86 years (average, 50. 3 years) . According to AO classification, 13 cases belonged to type 31-A1. 2, 32 to type 31-A1. 3, 35 to type 31-A2. 1, 27 to type 31-A2. 2, 33 to type 31-A2. 3, 38 to type 31-A3. 1, 39 to type 31-A3. 2 and 19 to type 31-A3. 3. All the fractures were single, fresh and closed and treated with PFNAⅡinternal fixation. Results This cohort was followed up for 8 to 26 months (average, 13. 2 months). Nonunion occurred in one case who had to accept artificial hip replacement. The remaining 235 cases obtained bony union after 22 to 39 weeks (average, 29. 3 weeks). By the Harris evaluation at final follow-ups, the affected hips scored from 81 to 93 points (average, 85. 1 points) . Post-operative thigh pain was reported in 19 cases (8. 05%) . The causes included varied anatomic morphology of the proximal femur in 6 cases, distal defects of the intramadullary nails in 4, insufficient stability of internal fixation or uneven biomecanical distribution in 3, unskillful operation in 2, and severe oesteoporosis in 4. Avascular necrosis of femoral head was not observed during follow-ups. Conclusions Postoperative thigh pain is worthy of serious atention from orthopaedists following PFNA Ⅱtreatment of femoral trochanteric fractures. PFNA Ⅱshould be modified according to the specific Chinese features of the proximal femur, especially in the respects of anterior arch and distal structure of the main nail and lateral declination as well.
9.Coating technique of pH-time lag colon-specific matrine delivery mini-pill

AIM:To study the coating technique of pH-time lap colon-specific matrine delivery mini-pill consisted of the time lag release coating (inner layer) and enteric coating (out layer). METHODS:To filter the coating composition based on the index of dissolution of matrine and oxymatrine in vitro and the appearance rating of miui-pill. RESULTS:4 The coating composition of inner layer was the alcoholic solution,consisted of 2% EC,0.4% DEP and 2% talc powder. Then the coating composition of out layer was the alcoholic solution consisted of 5% Eudragit S100,3% talc powder and 0.5% TEC. The dissolution tests in vitro indicated that matrine and oxymatrine were not dissolved in the simulated gastric juice in 2 h. The accumulative amount of matrine and oxymatrine were less than 15% in the simulated intestinal fluid in 4 h. The amount of matrine and oxymatrine were 80.7% and 83.5% in the simulated colon juice in 2 h. CONCLUSION:The mini-pill could achieve the goal of delivering in the specific colon.
10.Relationship between whole blood chemical dements and coal-burning borne endemic fluorosis

ObjectiveTo investigate the differences of whole blood chemical elements and urinary fluorine between patients with endemic fluomsis and patients without endemic fluorosis,and to find out the elements associated with endemic fluorosis and further lay a theoretical basis for clarify the pathogenesis of the disease.MethodsUsing case-control study,100 children aged 8 - 12 with dental fluorosis in Wushan and Fengjie counties of Chongqing from December 2010 to February 2011,and 30 adults with skeletal fluorosis were enrolled as case group; 100 children aged 8 - 12 without dental fluorosis and 30 adults without skeletal fluorosis were enrolled as internal control group; and 50 children without dental fluorosis and 30 healthy adults were selected as external control group in non-epidemic areas in Yubei district.Whole blood copper,zinc,calcium,magnesium,iron and urinary fluorine of all subjects were determined,and differences of these indexes were compared between groups.ResultsThe levels of copper,zinc,calcium,magnesium,iron and urinary fluorine of children in the case group were (30.08 ± 2.83),(74.04 ± 9.75)μmol/L,(1.65 ± 0.29),(1.37 ± 0.17),(6.79 ± 1.27)mmol/L,and (0.73 ±0.37)mg/L,respectively; the levels of these elements of children in internal control group were (28.65 ± 3.96),(72.83 ± 11.35)μmol/L,(1.62 ± 0.27),(1.36 ± 0.18),(6.73 ± 1.22)mmol/L,and (0.48 ± 0.21)mg/L,respectively; in external control group were (32.03 ± 2.99),(77.78 ± 10.85)μmol/L,(1.41 ± 0.11),(1.43 ± 0.13),(7.66 ±0.55)mmol/L,and (0.49 ± 0.26)mg/L,respectively(all P< 0.05),the comparison between any two groups indicated the levels of copper,zinc,magnesium,iron of the case group were lower than that of external control group,urinary fluorine was higher than that of internal and external control groups(all P < 0.05).The levels of copper,zinc,calcium,magnesium,iron and urinary fluorine of adult case were (26.93 ± 4.37),(95.89 ± 12.45)μmol/L,(1.50 ± 1.76),(1.56 ± 1.96),(8.15 ± 1.00)mmol/L,and (2.17 ± 0.99)mg/L; internal control group were (26.26 ±4.96),(94.86 ± 12.18)μmol/L,(1.57 ± 0.12),(1.46 ± 0.16),(7.64 ± 1.00)mmol/L,and (1.44 ± 1.22)mg/L;external control group were (26.20 ± 2.96),(96.52 ± 11.11)μmol/L,(1.48 ± 0.14),(1.45 ± 0.16),(7.81 ±0.91 )mmol/L,and (0.55 ± 0.21 )mg/L,respectively.The levels of magnesium,iron and urinary fluorine of case group were higher than that of internal control group,magnesium and urinary fluorine were higher than that of external control group(all P < 0.05).ConclusionsIn vivo anti-fluorine elements are deficient in the areas with endemic fluorosis.Other chemical elements,the environment and genetic factors may be related to the pathogenesisof the disease,which needs a further comprehensive analysis.
