1.Interaction of Polymorphisms of CYP2E1 and Environmental Risk Factors in Gastric Cancer

Objective To study the interaction of genetic polymorphisms of CYP2E1 and environmental hazard factors in gastric cancer. Methods 1∶1 case-control study was carried out in Nanjing area, using PCR-RFLP technology to determined the genetic polymorphisms of CYP2E1 and epidemiological data about environmental exposure of 121 cases and controls were collected. Interaction indexes (?) were calculated to determine the type of gene-environment interaction. Results After confounding adjusted, the result showed that interaction existed in genetic polymorphisms of CYP2E1 RsaⅠand the family history of digestive system disease, smoking, pickled food, irregular diet habit, interaction indexes (?) value were 1.855, 2.626, 1.736 and 1.714 respectively. A low exposure-gene effect interaction was found in CYP2E1 RsaⅠgenotype and the frequency of pickled food consumed, while a high exposure-gene effect interaction exist in CYP2E1 RsaⅠgenotype and irregular diet habit. Conclusion Genetic and environmental hazard factors co-effect the development of gastric cancer, genetic polymorphisms of CYP2E1 and environmental risk factors show an interaction in gastric cancer.
2.Efficacy of aripiprazole combined with modified electroconvulsive therapy in treatment of female refractory schizophrenia

Objective To investigate the efficacy and side effects of aripiprazole combined with modified electroconvulsive therapy (MECT) in the treatment of female refractory schizophrenia.Methods One hundred and five female refractory schizophrenia patients in the Second Affiliated Hospital of Xinxiang Medical University from July 2014 to May 2016 were selected and divided into control group and observation group.Fifty-two patients in the control group were treated with aripiprazole.In the observation group,53 patients were treated with aripiprazole combined with MECT.All the patients were treated for eight weeks.Clinical efficacies were assessed with the positive and negative syndrome syndrome scale (PANSS);Wechsler memory scale (WMS) were used to evaluate the effect of MECT on memory.Adverse drug reactions were evaluated by treatment emergent symptom scale(TESS) before and after treatment in both groups.Results There was no significant difference in PANSS scores between the two groups before treatment (P > 0.05).The positive symptom scores,the general mental pathology scores and the PANSS total scores in the two groups at the end of the second week were significantly lower than before treatment(P < 0.05);at the end of fourth and eighth week of treatment,the negative symptom scores,the positive symptom scores,the general psychopathological scores and the PANSS total scores of the two groups were significantly lower than those before treatment (P < 0.05 or P < 0.01).The positive symptoms,the general psychopathological scores and the PANSS total score in the observation group were significantly lower than those in the control group at the end of fourth week of treatment (P <0.05).At the end of the eighth week of treatment,the negative symptom scores,the positive symptom scores,the general psychopathological scores and the PANSS total scores in the observation group were significant lower than those in the control group (P < 0.05).At the end of the eighth week of treatment,the decreasing rates of PANSS score in the observation group and control group were (39.89-8.62) % and (31.97-± 8.17) %,and the total effective rates were 66.67% and 48.08%,respectively.The decreasing rates of PANSS score and total effective rates in the observation group were higher than those in the control group (P < 0.05).The WMS score of the observation group decreased more significantly at the end of the second week of treatment compared with that before treatment;at the end of the fourth week of treatment,the score of the picture was louer than that before treatment(P < O.05);but there was no significant difference in WMS score between that at the end of the eighth week and before treatment(P > 0.05).There was no significant difference between the two groups in the TESS score and adverse reaction rate (P > 0.05).Conclusion Compared with single application of aripiprazole,MECT combined with aripiprazole can significantly improve effects in treating female refractory schizophrenia and it can not increase the side effects.
3.Treatment of 34 cases of non small cell lung cancer in the elderly patients using vinorelbine and oxaliplatin

Purpose:To investigate the effects on non small cell lung cancer(NSCLC) and untoward reactions in elderly patients using vinorelbine(NVB) and oxaliplatin. Methods:NVB 25mg/m 2 ivgtt on day 1 and 8,oxalipatin 85mg/m 2 ivgtt on day 1 and 8, at 21 days per cycle. And every case should receive at least 2 cycles. Results:Among 34 patients,complete response(CR):2.9%,partial response(PR):35.3%,stable disease(SD):44.1%,progressive disease(PD):17.6%.Total response rate(RR) is 38.2%. The side effects were myelosuppression and peripheral sensory nueropathy. Conclusions:NVB combined with oxaliplatin treating NSCLC in elderly people has some effect,and the toxixity can be well tolerated.

Objective To determine the association of glutathione-stransferase M1(GSTM1), T1(GSTT1) genetic polymorphisms, smoking and alcohol consumption with the risk of gastric cancer. Methods 1:1 matched case-control study and molecular epidemiology technique were used to analysis the genetic polymorphisms of GSTM1, GSTT1 genotype among 60 cases of gastric cancer patients and sex-and age-matched controls, who were selected from Nanjing Zhongda Hospital and Jiangsu Province Cancer Hospital. Objective To study the insulin‐like growth factor binding protein (IGFBP)‐2 and IGFBP‐6 expression and clini‐cal significance in colorectal adenomas .Methods A collection of Chengde Medical College Hospital from July 2012 to March 2013 after surgical treatment of colorectal cancer confirmed by pathology (colorectal cancer ,CRC) tissue samples of 50 patients ,colorec‐tal adenomas (colorectal adenoma ,CRA) 50 cases ,20 cases of colorectal normal mucosa .Immunohistochemistry and RT‐PCR were used to detect the expression of IGFBP‐2 and IGFBP‐6 protein and mRNA ,combined with clinical and pathological data were statis‐tically analyzed .Results IGFBP‐2 protein positive expression and the amount of mRNA expression in the CRA group compared with normal colorectal mucosa had a rising trend;While compared with CRC group had a tendency to reduce ,and the differences are obvious statistical significance (P<0 .05) ,and IGFBP‐6 protein positive expression in the CRA group compared with normal color‐ectal mucosa had a lower trend;While compared with CRC group amount IGFBP‐6 mRNA expression in the CRA group compared with normal colorectal mucosa had a rising trend;While compared with CRC group had a tendency to reduce ,and the differences were obvious statistical significance (P<0 .05) .In the CRA group ,IGFBP‐2 ,IGFBP‐6 positive expression and the patient′s age , sex ,tumor and the number of parts were no significant statistical difference (P>0 .05) ,but with the degree of hyperplasia had sig‐nificant statistical difference(P<0 .05);In the CRA group ,IGFBP‐2 and IGFBP‐6 expression were negatively related to each other , and the difference was statistically significant (P<0 .01) .Conclusion Colorectal adenomas in normal colorectal mucosa of colorec‐tal cancer to the middle part of the transformation process ,and in its occurrence and development process ,insulin‐like growth factor family (IGFs) and IGFS‐R axis plays an important and irreplaceable role ,so IGFBP‐2 ,IGFBP‐6 may be used as diagnostic colorectal adenomas and early predictors of prognosis ,clinical studies on colorectal adenomas is important .
6.Cancer genomics-related data management and its application

Objective To provide reference for developing and perfecting the open resources of national-wide cancer genomics-related data by collecting, systematizing, organizing, sharing and applying the related data of the National Genome Atlas ( TCGA) Program. Methods The technical process, sharing and use of TCGA Program data were in-vestigated. Results TCGA established the whole linkage data management process by cooperating with multiple cen-ters, including tissue sample collection, processing, quality control, sequencing, characteristics analysis, data sharing and application of research achievements. The data were classified according to the related cancer, data types and their processing. The mutation, amplification and deficit of related cancer characteristic genes and the af-fected signaling pathways were studied according to the two sharing mechanisms underlying open access and con-trolled access to the collected data and individual data. Conclusion Studies on TCGA Program can provide experi-ences and reference in data management for the implementation of large scale TCGA Program.
7.Effect of pigment epithelium-derived factor on rat retinal Müller cells under high glucose conditions

Objective To investigate the effect of pigment epithelium-derived factor (PEDF) on rat retinal Müller cells under high glucose conditions. Methods Müller cells cultured in 25 mmoL/L glncose were incubated with 100 ng/ml PEDF or 10 ng/ml interleukin-1β(IL-1β) for 24 h. The expression of IL-1β or PEDF in Müller cells was measured by indirect immunofluorescence, Western blot or realtime RT-PCR. The survival of Müller cells was detected by MTT assay. Results Under high glucose conditions, expression of IL-1β or PEDF was decreased after treated with 100 ng/ml PEDF or 10 ng/ml IL-1β for 24 hours by the methods of immunocytochemistry, Western blot or realtime PCR (P < 0.05). Activity of Müller cells was increased significantly by PEDF (0.48±0.09 vs 0.64±0.17, P<0.05). Conclusion In mimic diabetic conditions, PEDF decreases expression of IL-1β in rat retinal Müller cells and enhances the cell activity. To some degree, PEDF may block the process of inflammation in diabetic retinopathy.
8.Cyclooxygenases, lipoxygenases, their targeted drugs and the prevention of Alzheimer's disease.

Many studies have shown that chronic inflammation occurs in the brain of patients with Alzheimer's disease (AD). It is well known that long-term administration of non-steroidal anti-inflammatory drugs (NSAIDs) can alleviate the cognitive decline of AD patient and elderly. Several inflammatory cytokines produced in the metabolism of arachidonic acid (AA) are closely related to inflammatory diseases. Lipoxygenases (LOXs) and cyclooxygenases (COXs) play a crucial role in the AA network, the products eicosanoids have an important impact on the progression of AD. Although there are many arguments and conflicting evidence, currently LOXs and COXs are still the hot topics in the research on AD pathogenesis and drug development. Here, we review the progress in research on COXs and LOXs, including their actions on CNS and their association with AD, and explore the feasibility of LOXs and COXs as targets for the drugs to prevent and/or treat AD.

9.Expression of THANK in human PBMC activated with different stimulators

Aim To analyze THANK gene expression in peripheral blood mononuclear cells(PBMC) stimulated with different stimulators and to clone whole length human THANK gene. Methods PBMC were conventionally isolated and cultured in RPMI1640 containing 10% FCS. After stimulated with LPS,TNF α ,IL 2,IFN γ ,PHA or PMA,the THANK gene expression in PBMCs was analyzed by RT PCR and THANK cDNA was cloned. Result RT PCR detection showed that THNAK gene expressed in PBMCs after stimulated with interferon γ for 3 days, whereas THANK'expression could not be detected after stimulated with LPS,TNF α ,IL 2,IFN γ ,PHA or PMA respectively. Then THANK gene was cloned by cloning PCR product and sequenced. Conclusion Human THANK gene is cloned successfully, thus providing the possibility for further research of THANK'function.
10.Stability of the salt iodization agent potassium iodate: a differential thermal analysis

Objective To investigate the thermal stability of solid potassium iodate and potassium iodate as additive in the sodium chloride,vitamin E,vitamin C and yellow prussiate.Methods HCR-2 type Differential Thermal Analyzer was used to carried out the differential thermal analysis of the potassium iodate and the potassium iodate in the sodium chloride,vitamin E,vitamin C and the yellow prussiate,and differential thermal curves were obtained and analyzed.Results The decomposition temperature of solid potassium iodate was 525℃ ; when mixed with sodium chloride,potassium iodate was stable below 300 ℃ ; vitamin C was unstable at 170-200 ℃ and underwent chemical changes; iodate and vitamin C underwent oxidation-reduction reaction at 145 to 160 ℃;potassium iodate with vitamin E at 300 ℃ was stable; yellow prussiate at 300 ℃ was stable; iodized salt was stable at cooking temperature below 300 ℃.Conclusions The potassium iodate has good stability below 525 ℃,however,potassium iodate iodized salt in the cooking process is easy to react with vitamin C in vegetables causing iodine losses,so iodized salt should be added just before the dish is done.
