1.The Ultrasonographic Diagnosis of the Complications in Mammoplasty by Different Operation Methods

Objective To evaluate the clinical value of highfrequency ultrasonography in diagnosis of the complications in mammoplasty.Methods One hundred and one cases of mammoplasty with complications were examined.They were divided into implanted group and injected group,and the prosthetic position,shape,internal echo and capsula were observed.Results Among the cases,91 breasts were capsular contraction and hardening,24 were asymmetry,38 were rupture,4 were infection and 1 was bleed.All cases were proved by operation.Conclusion High-frequency ultrasonography was easy and effective to diagnose the complications of mammoplasty.
2.Analysis in application of flexible management in nursing care of general surgery department

Objective To investigate the application effect of flexible management in nursing of general surgery department. Methods 21 nursing staff and 836 patients from January to October, 2007 were selected as research object, and were given flexible management. 21 nursing staff and 797 patients from January to December were selected as the control. The influence of flexible management on nurses and patients and satisfaction degree with nursing work before and after application of flexible management were compared. Results Working and learning enthusiasm greatly improved,educational background and professional titles of nurses increased after application of flexible management. The average hospitalization time was 28 days,the per capita cost was 12 131 yuan,satisfaction degree with nursing was 87.0% before application of flexible management, but the average hospital stay of patients decreased to 20 days, the per capita cost decreased to 8764 yuan and satisfaction degree with nursing increased to 97.0% after application of flexible management. Conclusions Application of flexible management in nursing care of general surgery department showed significant effect,it is suitable for more application in practice.
3.Role of histamine on the expression of tight junctions gene in airway epithelia

OBJECTIVE To investigate and elucidate the role of histamine on the regulation of tight junctions(TJs) gene expression of the airway epithelial cells. METHODS We cultured primary human bronchial epithelial cells from healthy and asthmatic individuals in the presence of histamine for 96 hours in air-liquid interface (ALI) culture system. The TJs expression on the level of mRNA and protein was determined by RT-qPCR and immunofluorescence (confocal microscopy), respectively. RESULTS A RT-qPCR analysis revealed that the mRNA expression of several TJs were up- or down-regulated by histamine. However, examination by confocal microscope revealed no detectable alterations in the morphology of the tight junctions after stimulating with histamine. CONCLUSION Our findings that histamine could regulate the mRNA expression of TJs of bronchial epithelial cells. That might contribute to the pathogenesis of chronic airway inflammatory diseases.
4.IL-13 inhibits differentiation of human nasal ciliated epithelial cells

[ABSTRACT]OBJECTIVETo explore the role and mechanism of IL-13 in regulating the differentiation of ciliated cells on cultured human primary nasal epithelial cells.METHODSHuman nasal epithelial cells were cultured in vitro and treated by IL-13, the differentiation of epithelial cells was observed by western blot detection ofβ-tubulinⅣ and ELISA detection of MUC5AC; the percentage of ciliated and goblet cells was quantified by cytospin followed by immunohistochemical staining; the expression level of central ciliary genes was detected by real-time PCR.RESULTSCompared with control group, IL-13 treatment for 14 day significantly decreased the percentage of ciliated cells and expression ofβ-tubulinⅣ, additionally increased the goblet cells and the secretion of MUC5AC; IL-13 treatment significantly reduced the expression of ciliary key transcription factors such as FOXJ1, RFX2 and RFX3, as well as basal body biogenesis genes like CCNO and CETN2.CONCLUSIONIL-13 inhibits the differentiation of human nasal ciliated epithelial cells, which may be by down-regulating the expression of ciliary key transcription factors and basal body biogenesis proteins.
5.Effects of Jinzhuyalong Ershiwuwei coral capsule on PLA2 in degenarated cervical intervertebral discs

Objective To study the anti-inflammation mechanism of Jinzhuyalong Ershiwuwei coral capsule on cervical spondylosis by investigating Jinzhuyalong Ershiwuwei coral capsule on PLA2 in degenerated cervical intervertebral discs.Methods 60SD rats (SPF) were divided into a control group (15),a medicine model control group (15),a medicine modle 1 group (15),and a medicine model 2 group (15) by random lottery; Medicine model 1,2 groups were given Jinzhuyalong Ershiwuwei coral capsule powder 0.33 × 5 g/kg 0.66×5 g/kg,and added 1 ml distilled water dilution lavage,l/d,90 d,The unbalance of power cervical spondylosis animal models were chosen and C4~5 neck intervertebral discs were taken after 90 days.Millipore colorimetry methods were used to measure PLA2 activity in C4~5 cervical intervertebral discs.Results The PLA2 activity of the cervical intervertebral disc (12.37 ± 4.21) IU/mg · min-1 in the model group was significantly higher than the control group(2.56± 1.19)IU/mg· min-1 (P<0.05).The cervical intervertebral disc PLA2 activity of medicine model group 1 (8.56± 2.13)IU/mg · min-1 and medicine model group 2 (5.58 ±2.39)IU/mg· min-1 was lower than the model control group(12.37±4.21)IU/mg· min-1,with significant differences (both P<0.01).Conclusion Jinzhuyalong Ershiwuwei coral capsule could down regulate PLA2 activity in degenerated cervical intervertebral discs,and also decrease the production of some inflammaroty mediators.
6.Experiment of treatment of bone defect with adenovirus-mediated expression of bone morphogenetic protein-2

Objective To explore the feasibility of mouse muscle-derived cells with adenovirus-mediated expression of BMP-2 in the treatment of mouse tibia bone defect via collagen sponge as scaffold. Methods Mouse adenovirus BMP-2 plasmids were prepared and identified by PCR.The adherent cells of muscle-derived stem cells cultured in vitro were passaged,cells of the third generation were selected,and recombinant Ad-BMP-2 plasmids were expressed and incubated in mouse myoblasts.The number of BMP-2 positive cells was measured using indirect immunofluorescence method,and alkaline phosphatase(ALP) method was adopted to determine the cell activity in each group.The mouse models of proximal tibial bone defect were established(n=80) and were divided into Ad-BMP-2 transfection group,Ad-LacZ transfection group,blank group and Ad-BMP-2 implantation group.The histological and radiological changes were observed 3 weeks after operation. Results It was revealed by indirect immunofluorescence that BMP-2 positive cells were frequently found in Ad-BMP-2 transfection group,while none were observed in Ad-LacZ transfection group and blank group.The ALP activity was higher in Ad-BMP-2 transfection group,while that was extremely low in Ad-LacZ transfection group and blank group.There was most typical imaging of repair and healing of bone defect in Ad-BMP-2 transfection group,followed by Ad-BMP-2 implantation group,while there was no typical imaging in Ad-LacZ transfection group and blank group. Conclusion The constructed Ad-BMP-2 plasmids can effectively transfect mouse muscle-derived cells,and the transfected cells can continuously express effective amount of BMP-2.The mouse muscle-derived cells with adenovirus-mediated expression of BMP-2 play a definite role in the treatment of bone defect,which provides an effective method and material for clinical application.
7.Content Determination of Rutin in Xueguankang Tea by Reversed Phase HPLC

Objective To establish a method for determination of rutin in Xueguankang Tea. Methods RP-HPLC method and C18 column were used for determination. Methanol-water-acetic acid (40∶60∶1) was the mobile phase with a flow rate of 1.0 mL/min. The detection wavelength was set at 257 nm. Results There was a good linear relationship of rutin within the range of 0.06~0.3 ?g with r=0.999 2. The average recovery of rutin was 98.48 % with RSD=0.87% (n=5). Conclusion This method is convenient,sensitive,accurate and reproducible,and can be used for the quality control of Xueguankang Tea.
8.Role of nitric oxide in ischemia/reperfusion injury and ischemic preconditioning

AIM: To clarify the role of nitric oxide(NO) in ischemic preconditioning(IP) and its effects on apoptosis. METHODS: Seventy-two male Wistar rats were divided into the following six groups:ischemia/reperfusion (IR) group,IP group,IR+L-arg group,IP+L-arg group,IR+L-NAME group and IP+L-NAME group,The following changes were measured:cardiac hemodynamic parameters,infarct size,PMNs counting myocardial MPO activity and TUNEL staining.RESULTS: ①L-arg significantly attenuated ischemia/reperfusion-induced heart injury,reduced PMNs infiltration and cardiomyocyte apoptosis.②L-NAME also significantly reduced infarct size,PMNs infiltration and cardiomyocyte apoptosis compared with IR group,however,L-NAME aggravated ischemia/reperfusions-induced cardiac functional injury.③L-arg or L-NAME did not significantly alter the protective effect of ischemic preconditioning. CONCLUSION: Increased production of endogenous NO before prolonged ischemic period can protect hearts and inhibit apoptosis.L-NAME can inhibit iNOS activity and ONOO- production in reperfusion period to protect heart.
9.Clinical Observation of Enhanced Immunosuppressive Therapy in the Treatment of Refractory Nephrotic Syndrome

OBJECTIVE:To observe the clinical efficacy and safety of enhanced immunosuppressive therapy in the treatment of refractory nephrotic syndrome (RNS). METHODS:Totally 76 RNS patients were selected from 2 hospitals during Jan. 2012-Mar. 2015,and then divided into control group and observation group according to random number table,with 38 cases in each group. Two groups were given Prednisone acetate tablet 50 mg,qd;decreasing to 15 mg,qd,after urine protein returned to normal. Based on it,control group was given Leflunomide tablets 50 mg,qd;decreasing to 20 mg,qd,3 days later. Based on con-trol group,observation group was additionally given Mycophenolate mofetil dispersible tablet 750 mg,bid;decreasing to 500 mg, qd,3 months later. Both groups were treated for 6 months. Clinical efficacies,follow-up recurrence rate as well as renal function in-dexes and inflammatory cell factors before and after treatment,and the occurrence of ADR were compared between 2 groups. RE-SULTS:The total response rate of observation group(92.11%)was significantly higher than control group(73.68%),and fol-low-up recurrence rate(5.26%)was significantly lower than control group(23.68%),with statistical significance(P<0.05). Be-fore treatment,there was no statistical significance in renal function indexes or inflammatory cell factors between 2 groups(P>0.05). After treatment,24 h urinary protein quantification,urinary IL-6 and IL-8 levels of 2 groups decreased significantly,while the content of serum protein increased significantly;the observation group was significantly better than the control group,with sta-tistical significance(P<0.05). There was no statistical significance in the content of serum creatinine between 2 groups before and after treatment(P>0.05). The incidence of ADR in the control group and the observation group was 34.21% and 44.74% respec-tively,without statistical significance between 2 groups(P>0.05). CONCLUSIONS:Enhanced immunosuppressive therapy in the treatment of RNS can improve renal function,reduce inflammatory reaction and long-term recurrence risk,and have good therapeu-tic efficacy and safety.
