2.Expression and Relationship of OPN and MMP-9 in Epithelial Ovarian Carcinoma

Objective To study the expression and relationship of osteopontin (OPN) and matrix metalloproteinase-9 (MMP-9) in epithe-lial ovarian carcinoma. Methods Immunohistochemical staining S-P methods was used to detect the expression of OPN and MMP-9 in the samples of 60 epithelial ovarian carcinoma,20 benign ovarian tumor,20 borderline ovarian tumor and 10 normal ovarian tissue. Results The expression level of OPN and MMP-9 in epithelial ovarian carcinoma(109.86±21.51) were significantly higher than those in borderline ovarian tumor(45.81±8.93),benign ovarian tumor(11.86±3.01) and normal ovarian tissue(5.32±2.74). The expression level of OPN and MMP-9 related to the tumor grading,tumor stage and lymph node metastasis,while had no relationship with histological grading. There was significant positive correlation between OPN and MMP-9 in epithelial ovarian carcinoma (r = 0.80,P < 0.05). Conclusion The increased expression of OPN and MMP-9 may play an important role in the process of carcinogenesis, progression and differentiation of epithelial ovari-an carcinoma. OPN and MMP-9 may be a useful targets for treatment of ovarian carcinoma.

4. Development and function analysis of SSR molecular markers in Magnolia officinalis transcriptome

Objective: SSR markers, primers and unigene function in the transcriptome of Magnolia officinalis were analyzed in this study to provide powerful tools for molecular marker-assisted breeding and resources protection in this plant. Methods: A total of 16 369 unigenes obtained by transcriptome sequencing were used to explore SSR. SSR primers were designed by Primer.0 3 and 45 pairs were randomly selected for PCR. The gene function analysis of SSR unigene were obtained by Blast. Results: A total of 8 635 SSR were identified, accounting for 52.75%. Among them, mononucleotide, dinucleotide and trinucleotide type were the dominant types, and the type which repeated for 10 times was the most. Additionally, the dominant repeat types were A/T, AG/CT and AAG/CTT, and the distribution frequencies were 47.16%, 31.74%, and 6.53%. Twenty-two pairs of 45 primers produced fragments with expected molecular size. Unigenes containing SSR were annotated to the process of energy and redox reaction, RNA transport, spliceosome and plant hormone signal transduction metabolic pathway. Conclusion: The SSR markers in the transcriptome of M.officinalis show rich type, strong specificity and high potential, which will benefit to the candidate gene mining and marker-assisted breeding.
5.Effects of Porphyromonas gingivalis infection on intercellular adhesion molecule-1 expression in rat vascular smooth muscle cells.

OBJECTIVETo observe the effects of Porphyromonas gingivalis (P. gingivalis) ATCC 33277 infection on expression of intercellular adhesion molecule-1 (ICAM-1) in rat vascular smooth muscle cells(VSMC).
METHODSAn infection model of rat VSMC invaded by P. gingivalis was established in vitro. The mRNA of ICAM-1 was measured through reverse transcription-polymerase chain reaction (RT-PCR).
RESULTSCompared with the control group, an apparent and statistically significant increase in expression of ICAM-1 mRNA was observed after 8, 16, and 24 h in P. gingivals-infected rat VSMC (P<0.05). The expression reached its peak at 16 h. Statistically significant differences were observed in the 8 h group and in the other two experimental groups (P<0.05).
CONCLUSIONInfection of P. gingivals in rat VSMC can cause increased expression of ICAM-1, which may have an important function in the progression of atherosclerosis.

6.Effects of background music on psychological state of patients and medical personnel in operation room

Objective To investigate the effectiveness of background music in operating room on psychology of patients and medical personnels. Methods A total of 127 patients who would receive selective operation and 65 surgical staff in our hospital were randomly divided into the intervention group and the control group. There were 63 patients and 30 medical personnel in the intervention group, 64 patients and 35 medical personnel in the control group. Routine care was given to all patients, and background music was broadcasted after patients and medical personnels coming into operating room in the intervention group. The blood pressure and heart rate of patients were contrasted, and the anxiety status was evaluated by Self-Rating Anxiety Scales (SAS), Visual Analogue Scale was used to appraise the horror, discomfort and pain between the control group and the intervention group. Results Diastolic pressure and systolic pressure was higher and heart rate was more rapid in the control group than in the intervention group. SAS score of patients and medical personnel was higher in the control group than in the intervention groups, degree of horror, pain and complaint in patients was more apparent in the control group than in the intervention group. Conclusions Background music could release negative emotion and was helpful for stabling the mood of medical personnel and preserving physical and mental health of patients.
7.Significance of cerebrospinal fluid matrix metalloproteinase-9, interleukin-6, and tumor necrosis factor-alpha protein in children with viral encephalitis

Objective To explore the significance of changes of matrix metalloproteinase-9 (MMP-9),interleukin-6 (IL-6),and tumor necrosis factor-alpha (TNF-oα) protein level in the cerebrospinal fluid (CSF) of children with viral encephalitis (VE).Methods The concentration of neuron-specific enolase (NSE),structural proteins 100B (S100B),and MMP-9,IL-6,TNF-α in the CSF of VE children were detected by an enzyme linked immunosorbent assay,and the correlations of them were analyzed.Results NSE,S100B,MMP-9,IL-6,TNF-α protein expression could be found significantly higher than those in the control group,and there were significant differences according to statistics expression trends(all P <0.05).The NSE protein expression was significantly positive related with S100B in the VE group (r =0.467,P =0.009),and the concentration was markedly negative related with the duration of viral encephalitis (r =-0.472,P =0.008).MMP-9,IL-6 protein expression were significantly positive related with NSE,S100B respectively (r =0.698,P =0.00 ; r =0.559,P =0.00 ; r =0.812,P =0.00 ; r =0.664,P =0.00).TNF-α protein expression was positive related with CSF S100B(r =0.363,P =0.049),but there was no correlation between TNF-α and NSE (r =0.245,P =0.193).Conclusions The neurons and the neuroglial cells are damaged in the viral encephalitis children.MMP-9,IL-6,TNF-α protein may participate in the pathological damage process of nerve cells in VE children in different degrees.
8.Inhibition of notoginsenoside R1 on SOCE in pulmonary arterial smooth muscle cells of pulmonary hypertension rats

Aim To evaluate the effects of notoginsen-oside R1 on store-operated calcium entry ( SOCE ) in pulmonary arterial smooth muscle cells ( PASMCs ) of chronic hypoxia ( CH)-and monocrotaline ( MCT)-in-duced pulmonary hypertension ( PH) rats. Methods Mn2+ quenching of Fura-2 and measurement of intra-cellular free calcium concentration ( [ Ca2+] i ) using fluo-3 were examined in PASMCs of CH-exposed and MCT-treated rats. Results ①CH-exposed and MCT-treated rats exhibited profound PH when examined 3 weeks after hypoxia exposure or MCT injection, respec-tively. ②In the presence of 3 μmol·L-1 nifedipine, 10 μmol · L-1 notoginsenoside R1 significantly re-duced cyclopiazonic acid ( CPA )-induced the percent reduction in Fura-2 fluorescence measured 500 sec af-ter application of Mn2+, the maximal rate of Mn2+quenching, the amplitude of the Ca2+ influx transient and the resting [ Ca2+] i in PASMCs of CH-exposed and MCT-treated rats. Conclusion Notoginsenoside R1 inhibits SOCE and reduces resting [ Ca2+] i in PASMCs of CH-and MCT-induced PH rats.
9.Screening of Antitumor and Antimicrobial Activities of the Endophytic Fungi Isolated from Cephalotaxus hainanensis Li

Seventy-two endophytic fungi isolated from Cephalotaxus hainanensis Li were screened for antitumor and antimicrobial activities. The results showed that 9 strains presented cytotoxic activities to at least one of the tested tumor cells, five strains exhibited strong antimicrobial activity against Staphylococcus aureus, one strain had inhibitory effect on Phytophthora parasitica. It was evidently that the endophytic fungi of C. hainanensis are potential resources to find valuable bioactive components.
10.Determination of Uric Acid Based on Active Copper-Containing Carbon Nanodots-modified Electrochemical Biosensor

The active copper-containing carbon nanodots were prepared by hydrothermal method, and then characterized by fluorescence spectroscopy and UV-visible absorption spectroscopy.Subsequently, a highly sensitive and selective electrochemical biosensor was fabricated on the basis of this synthesized carbon nanodots with electro-deposition technique.The electrode behavior was investigated by cyclic voltammetry, electrochemical impedance spectroscopy, and differential pulse voltammetry.Furthermore, the catalysis mechanism was studied.The experimental results indicated that the biosensor exhibited a strong electrocatalytic activity toward the oxidation of uric acid (UA).What′s more, the interference from ascorbic acid and dopamine was eliminated effectively.Under the optimum conditions, there were linear relationships between the anodic peak current and the concentration of UA (1.00-300.0 μmol/L), and the limit detection was 0.30 μmol/L (S/N=3).The prepared biosensor had advantages such as easy fabrication, strong anti-interference ability, high sensitivity, and wide detection range, and could be used for real sample detection.
