1.Changes of c-jun expression in temporalis muscles caused by hypergravity in Rhesus macaque

Objective:To observe the changes of c-jun expression in the monkey's temporalis muscles induced by stress reaction under +Gx loads.Methods:Nine male Rhesus macaques were randomly divided into four groups according to Gx loads and its lasting time:control group was exposed to +1 Gx/300 s overloads(n=2),and experimental groups 1(n=2),2(n=2)and 3(n=3)were exposed to +15 Gx/200 s,+21 Gx/165 s or +21 Gx/140 s respectively.Temporalis muscles tissue was fixed with 40 g/L buffered formaldehyde and embedded in paraffin.Histopathological changes were observed under light microscope.The expression of c-jun was detected by immunohistochemical PicTureTM method in the temporalis muscles.Results:Gross and histological analyses showed that no significant pathological changes in the temporalis muscles in different experimental groups.Immunohistochemical staining demonstrated that c-jun was over-expressed in the nuclei of the muscles in the experimental groups,while negative or slight positive staining was observed in the nuclei of muscles in control animals.There was no obvious difference of c-jun expression in different experimental groups.Conclusion:Overload gravity can induce enhanced expression of c-jun in temporalis muscles of the monkeys received +Gx loads.

Objective To study whether peripheral blood (PB)CD34+ cell absolute count can reliably predict peripheral blood progenitor cell (PBPC) autograft CD34+ cell contents. Methods We examined the PB CD34+ cell count and PBPC CD34+ cell count with ProCOUNT by flow cytometry. Objective To explore the role of type 1 plasminogen activator inhibitor (PAI-1 ) in mediating renal tubulointerstitial injury of patients with IgA nephropathy. Methods The mRNA and protein production of PAI-l in renal tubulointerstitium were defected using in stiu hybridization and immunohistochemistry. Detections of antigens of ?-smooth muscle actin(?-SMA) and proliferating cell nucleus antigen (PCNA) were also performed. To explore expressions of ? smooth muscle actin (? SMA), plasminogen activator inhibitor 1 (PAI 1), matrix metalloproteinase 9 (MMP 9)and tissue type inhibitor of metalloproteinase 1 (TIMP 1) in renal vessels of 38 patients with immunoglobulin A nephropathy (IgAN), immunohistochemistry method was used to detect abundances of the above proteins. The results showed that ? SMA staining was found mainly in vascular smooth muscle cells (SMCs); apparent PAI 1 staining was also shown by vascular SMCs; MMP 9 expressed considerably in vascular both SMCs and endothelial cells; TIMP 1 was located only in vascular endothelial cells. 5.Anatomical study and clinical application of minimally invasive internal fixation in the treatment of intra-articular fractures of calcaneum via sinus tarsi approach

Objective To investigate the anatomical features, operative method and efficacy of internal fixation in the treatment of iutra-articular fractures of caleaneum via the sinus tarsi approach. Methods The pathway, branches distribution and anastomosis of perforating descending branch of peroneal artery were observed on 18 adult cadaveric lower limbs. A sinus tarsi approach was designed. From July 2001 to January 2008, 71 intra-articular calcaneal fractures in 68 patients were treated with open reduction and internal fixation via sinus tarsi approach at lateral sides of calcaneus. According to the Sanders classification, there were 26 type Ⅱ fractures, 32 type Ⅲ fractures and 13 type Ⅳ fractures. Results All patients were followed up for a mean period of 39.3 months (13-85 months), and the fractures were completely healed. There was a significant difference in the length, width and height of the calcaneus, Bohler angle and Gissane angle before and after operation (P < 0.01). According to Maryland Foot Score, the operative effect was excellent in 33 feet, good in 29 feet, fair in 6 feet and poor in 3 feet. Conclusion Open reduction and internal fixation via sinus tarsi approach is an effective method for minimally invasive treatment of intraarticular fractures of the calcaneus, with the advantages of good clinical results and causing minimal damage to soft tissues.
6.Radiological and therapeutic study of posterior bicondylar tibial plateau fractures

ObjectiveTo investigate the radiological characteristics,morphological characteristics of fracture fragments and treatments of posterior bicondylar tibial plateau fractures.MethodsA total of 47 patients with posterior bicondylar tibial plateau fractures treated between March 2005 and February 2009 were enrolled in the study.Characteristics of fractures and morphologies of fracture fragments were measured precisely with CT-chip Starpacs system.A retrospective study was carried out on the therapeutic results of the 47 patients undergone lateral condylar plate and medial condylar lag screw fixation via anterolateral combined with anteromedial tibial plateau approaches.Results Posteromedial condylar fracture was split one with small displacement,but the fracture was unstable.In the meantime,the fracture fragments were inverted three prism in shape and remained quite intact.Posterolateral condylar fracture was compression one and was often associated with soft tissue injury of knee joints.According to the Rasmussen radiology score,the results were excellent in 24 patients,good in 18,fair in three and poor in two.According to the Rasmussen function score,the results were excellent in 13 patients,good in 27,fair in two and poor in five.ConclusionsThe morphous of posteromedial condylar fracture is similar with that of Hohl type E fracture.The posterolateral condylar fracture is compression one.Anterolateral combined with anteromedial approaches for posterior tibial plateau fractures allow stage Ⅰbone fusion with internal fixation,simultaneously repair recombined injuries and achieve excellent radiological and clinical results.
7.Treatment of open pelvic fracture concomitant with perineal laceration using external fixation combined with limited percutaneous internal fixation plus colostomy

Objective To introduce the methods and key points for the external fixation combinel with limited percutaneous internal fixation plus colostomy in the treatment of open pelvic fracture concomitant with perineal laceration.Methods Nine patients with open pelvic fractures concomitant with perineallaceration were treated by external fixator combined with limited percutaneous internal fixer plus colostomy. Data including injury details, management and outcomes were collected for comparison.Results All the patients survived and attained bony union except for two patients with local infection that was healed several days after repeated wound debridement and use of wide spectrum antibiotics. All the patients had good extremity function at the latest follow-up. Conclusion External fixation combined with limited percutaneous internal fixation plus colostomy is a reliable, safe and less invasive procedure for the treatment of open pelvic fractures concomitant with perineal open wound.
8.Coefficients between TEOAEs and DPOAEs in infants

bjective:To investigate the coefficients of TEOAEs and DPOAEs based on various pass/fail criteria. Method:Thirty-six Australia infants were tested (age range: 1~6 months) using both TEOAE and DPOAE. Cohen′s Kappa (K) were used to analyze different criteria.Result:There is a significant low agreement in the screening outcomes between TEOAEs and DPOAEs, and different criteria for TEOAEs.Conclusion:The findings from the study of coefficients among commonly used screening pass/fail criteria suggested that there is no simple relationship among them. The possible reasons for weak agreement were discussed.

10.Isolation of chondrocytes from rabbit rib cartilage with three-step enzymatic digestion and their biological characteristic in vitro

Objective To observe the effect of isolating and harvesting the chondrocytes from rabbits rib cartilage with the method of three-step enzymatic digestion, and the biological characteristic of the isolated chondrocytes during cultivation in vitro to evaluate their biological activity. Methods The method of three-step enzymatic digestion was designed that the rib cartilage was digested one by one with 1 g/L trypsin and 1 g/L EDTA, 1 g/L hyaluronidase and 2 g/L collagenaseⅠ in the culture medium to isolate chondrocytes. The harvesting and viability rate of the primary chondrocytes were detected. During the passage cultivation in vitro, the changes of the chondrocyte shape and growth were observed, and the changes of the collagen typeⅠ and Ⅱ and aggrecan in the extracellular matrix were detected. Results ① The extracellular matrix of rib cartilage was completely dissolved by the three-step enzymatic digestion, and the chondrocytes were completely isolated from the solid matrix. The number of the harvested chondrocytes from every gram of wet cartilage was (4 295.7)?10~(4) on average,and their viability rate was 97.2% on average. ②The primary and first passage chondrocytes had triangle or multi-angle shape, and became elliptic shape at the growing confluence with the positive immunohistochemical staining of collagen type Ⅱ and the strong heterochromia to toluidine blue. The content of sulfate glycosaminoglycans(GAG) in the extracellular matrix of the primary passage cells was (80.61?11.40) ?g/cm~(2). The chondrocytes after the third passage gradually became spindle shape with the negative staining of collagen typeⅡ and the weak heterochromia to toluidine blue. The content of sulfate GAG of the fourth passage cells was (44.74?10.18) ?g/cm~(2). Conclusion ① The method of three-step enzymatic digestion can make the extracellular matix of rib cartilage to be completely degraded, and has advantages of the high efficiency of harvesting primary chondrocytes with high cellular viability rate and simple manipulation. ②The primary and first passage chondrocytes have fine biological activity.
