1.Treatment of Carotid-Cavernous Fistula with Intravascular Embolization——An Analysis of 7 Cases

Objective:To investigate the therapeutic effect of intravascular balloon emolization of carotid-cavernous fistula.Materials and Methods:7 cases(5 of traumatic origin, and 2 spontaneous),undewent DSA of brain vessels showing the exact sites of fistulae.The key points of angiography in this procedure were described.Results:5 of the 7 cases complete patency of ICAs.I case resulted in occlusion of ICA,and another of type D was unsatisfactory. Conclusion:DSA of brain vessele could dipiot the site,size and type fistula.Most cases of simple carotid-cavernous fistula are of traumatic arigin and intravascular balloon embolization should be the first-choice of treatment
2.Balloon Catheter Dilatation and Internal Stent Placement in Treating Takayasu's Arteritis with Carotid Arterial Stenosis

Purpose:To evaluate the feasibility of balloon catheter dilatation and internal stent placement in the treatment of Takayasu's arterial stenosis.Materials and Methods:Three patients with stenosis of common carotid arteries caused by Takayasu' s disease were treated with PTA by balloon catheter and intravaseular stent placement,the lenghs of stenosis were all beyond 8cm.Balloon angioplasty was performed in one patient,the others were treated with Wallstent stents after PTA.Result:Immediately after treatment,angiography and ultrasound showed that the proportion of stenosis was zero and all appeared to achieve in good result.The artery treated with PTA was completely occluded after 1 year follow-up;the other two patients with stents placement were also examined by angiography and ultrasound at 4,5 months and 4 months respectively after the praeedure.For the patient with left carotid artery stenosis,the proximal part of the segment with the intravascular stent showed a restenosis;the other patient was normal.Conclusion:We considered that intravascular stent placement be might to the treatment for Takayacu' s arteritis with long segment stenosis of common carotid artery.
3.Construction of a recombinant Escherichia coli for high trehalose production.

Trehalose, a compatible solute, is widely used in food, cosmetics, pharmaceutical products and organ transplantation. Nowadays, trehalose is mostly produced by enzymatic synthesis with many secondary products and lowpurity. In this study, high amount of trehalose was produced by recombinant E. ccli fermentation. First, a bifunctional trehalose gene TPSP was amplified from genome of C. hutchinscoii. Second, an expression vector pTac-HisA containing TPSP was constructed and transformed into the host E. coli. Expression of this bifunctional enzyme-TPSP converted glucose to trehalose. The result suggested that TPSP from C. hutchinsonji has been successfully expressed in E. ccoi. High amount of extracellular trehalose generated from glucose by whole-cell catalysis and After optimization, the production of trehalose in shake flasks was improved to 1.2 g/L and the relative conversion rate reached 21%. The production in bioreactor reached 13.3 g/L and the relative conversion rate reached 48.6%. It is the first time to realize the functional expression of the bifunctional enzyme-TPSP of C. hutchinsonii in E. coli and achieved the conversion form glucose to trehalose. This study laid a foundation for industrial large-scale production of trehalose.

4.Effects of tacrolimus on cell proliferation of and secretion of stem cell factors by cultured human keratinocytes

Objective To study the effects oftacrolimus on the cell proliferation of and secretion of stem cell factors (SCF) by cultured human keratinocytes. Methods Human keratinocyte cell line HaCaT was cultured and treated with various concentrations (0, 10, 102, 103, 104 nmol/L) of tacrolimus. After 48 hours of treatment, cell proliferation was measured with MTT assay, the levels of stem cell factor in the culture supernatant of HaCaT cells were detected with ELISA. Results Inhibited proliferation was observed in HaCaT cells treated with tacrolimus of 103-104 nmol/L. The secretion of SCF by HaCaT cells was enhanced in the presence of 10-102 nmol/L of tacrolimus, was inhibited in the presence of tacrolimus of 104 nmol/L, and remained unchanged with 103 nmol/L. Conclusion Certain concentrations of tacrolimus could enhance the expression Of SCF in HaCaT cells, which may be associated with the efficacy of tacrolimus in the treatment of vitiligo.
5.Lower limb muscle mechanics during isokinetic pedaling

Objective To document and analyze thepeak force and work during isokinetic pedaling at different linear speeds and the flexion-extension peak force ratio.Methods Thirty healthy adults were tested using the Legpress system of the IsoMED 2000 apparatus.Peak force,work and the flexion-extension peak force ratio were recorded and analyzed during isokinetic pedalingat 5 and 50 cm/s.Results The peak force applied and work done by the lower limbs were both significantly lowerwhen pedaling at 50 cm/s than at 5 cm/s.There was no significant difference in peak force or work between the left and right legs at 50 cm/s.The peak force ratio at 50 cm/s was significantly higher than at 5 cm/s.Conclusion The mechanical characteristics of the leg muscles during isokinetic pedaling vary greatly.It is advisable to choose different linear speeds or training modes accordingly.

Objective To study the effects of endothelin-1 (ET-1) and adrenocorticotropic hormone (ACTH) on the proliferation of cultured normal human melanocytes. Methods Normal human melanocytes were obtained from the foreskins of healthy men, and cultured in minimum essential medium (MEM) with 10% bovine serum. The cultured melanocytes of the 3rd passage were treated with various concentrations of ET-1 and ACTH. Cell proliferation was assessed by the MTT method. Objective To explore the effects of cognitive rehabilitation on aphasia for the patients with severe traumatic brain injury.
Methods 40 patients with traumatic brain injury following aphasia were randomly divided into simple language group (n=20) and language-
cognition group (n=20). Both groups received conventional therapy and language therapy, and the language-cognition group received
cognitive rehabilitation additionally, 30 minutes 1 day for 1 month. Results The index of Chinese Rehabilitation Research Center Aphasia
Examination (CRRCAE) improved significantly after treatment in both groups (P<0.01). There were significant differences in all the indexes
of CRRCAE (P<0.05) between 2 groups after treatment except dictation (P>0.05). Auditory comprehension correlated with orientation,
visual perception, spatial perception, motor praxis, visuomotor organization and thinking operations. Conclusion Cognitive rehabilitation
can improve the language function of traumatic brain injury patients with aphasia.
8.High Performance Liquid Chromatogram-Mass Spectrometry for Helicobacter pylori Infection-associated Differentially Expressed Proteins in Urine with Relative Molecular Mass More than 10 kDa

Background:Helicobacter pylori(Hp)is an important pathogen for peptic ulcer and gastric cancer,and is reportedly associated with a variety of extragastrointestinal diseases. However,there is no body fluid detection technique for Hp infection in clinical practice. Aims:To identify Hp infection-associated differentially expressed proteins in urine with relative molecular mass more than 10 kDa and provide potential biomarkers for diagnosis of Hp infection through body fluid detection. Methods:Midstream urine was collected from volunteers in the morning,and 13 C-urea breath test was performed to determine Hp infection. Each of 15 Hp-negative and 15 Hp-positive urine samples were mixed respectively for protein extraction. Spectra data were acquired by high performance liquid chromatogram-mass spectrometry,and label-free technology was used for relative quantitative analysis. The other 26 urine samples(15 Hp-negative and 11 Hp-positive) were used for validation by full scan. IPA software was employed for bioinformatics analysis. Results:A total of 475 urinary proteins were detected by label-free quantitative analysis and 42 differentially expressed proteins were identified. Finally,11 significantly up-regulated differentially expressed proteins were confirmed by external scanning validation. Bioinformatics analysis revealed the molecular functions,biological pathways,and related diseases of these differentially expressed proteins. Conclusions:These 11 differentially expressed proteins more than 10 kDa identified in urine might be potential biomarkers for diagnosis of Hp infection and provide molecular evidence for the correlation of Hp infection with extragastrointestinal diseases.
9.Cytotoxic effect of Guillain-Barre syndrome-associated and diarrhea-associated Campylobacter jejuni

In order to assess the cytotoxin effect of Campylobacter jejuni (C.jejuni), the whole cell lysates from 9 strains of Guillan-Barre Syndrome (GBS)- associated C.jejuni and 4 strains of diarrhea-associated C. jejuni were co-cultivated with HeLa cells at the concentration of 0.001-5.00 μg/mL in vitro. The morphologic change of HeLa cells was observed under Olympus BX51 microscope after treatment with different concentration of bacteria lysate in the following 4 days. The morphological changes including swelling, irregularity and lysis of the affected cells over 50% was selected as the cut off for positive change and C.jejuni 81-176 and Helicobacter J99 strains were chosen as the positive and negative control. It was found that the minimum concentration to induce the positive changes in 10 strains(8 GBS associated and 2 strains from diarrhea patients)was 0.1μg/mL and 1 strain with the positive change at the minimum concentration of 1 μg/mL. There was only one GBS-associated strain causing no morphologic change on HeLa cells at the concentration of 5 μg/mL. It was evident that the cytotoxic effect of C.jejuni strains on HeLa cell was strain-specific, and there was no significant difference in the cytotoxic effect on HeLa cell between GBS-associated and diarrhea associated C.jejuni strains.
