1.Clinical analysis of 6 cases with sinonasal malignant melanoma.

OBJECTIVE:
To explore clinical characteristics of sinonasal malignant melanoma and curative effect of the combined modality therapy.
METHOD:
Clinical data of 6 cases with sinonasal malignant melanoma was retrospectively analyzed. All patients received surgery and postoperative radiotherapy. In addition, 3 cases received postoperative chemotherapy which scheme was CDBT and bioimmunotherapy consisted of INF-α and IL-2 after surgery, of which, 2 cases received one cycle of preoperative chemotherapy.
RESULT:
Six cases were followed up. The survival time ranged from 15 months to 98 months. The average survival time was 62.7 months. Analyzed by direct method, the 1-year, 3-year and 5-year survival rates were 100%, 83% and 67% respectively. Three cases which received the combined modality therapy, of whch, 2 cases received preoperative chemotherapy have survived by now.
CONCLUSION
The combined modality therapy should be adopted in case of sinonasal malignant melanoma with operation indication. For the patients who can not be operated recently Postbiopsy, it was beneficial to improve the efficacy of therapy that one cycle of preoperative chemotherapy and bioimmunotherapy should be implemented.

2.Calcitonin gene-related peptide suppresses isoprenaline-induced cardiomyocyte apoptosis through regulation of microRNA-1 and microRNA- 133 a expression

Objective To explore the inhibitory effect of calcitonin gene-related peptide (CGRP) on cardiomyocyte apoptosis and the underlying mechanism.Methods In cultured rat cardiomyocytes,apoptosis was induced by the incubation of isoprenaline ( 10ˉ5 mol/L) for 48 h.CGRP ( 10ˉ8 or 10ˉ7 mol/L) was administrated for 1 h before incubating isoprenaline to evaluate its effect on cardiomyocyte apoptosis.At the end of the drug treatment,the rate of apoptotic cells and intracellular reactive oxygen species (ROS) were determined,and RNA was extracted to examine the expression of microRNA-1 and microRNA-133a.Results Isoprenaline significantly increased the rate of apoptotic cells and intracellular ROS production concomitantly with the increased microRNA-1 expression and the decreased microRNA-133a expression,which were inhibited by pretreatment with CGRP.The effects of CGRP were reversed by CGRP receptor antagonist.Conclusion CGRP can inhibit the isoprenaline-induced cardiomyocyte apoptosis.The beneficial effect of CGRP is related to regulating microRNA-1 and microRNA-133a expression through the prevention of isoprenaline-induced ROS production.
3.Anti-inflammatory Effect and Underlying Mechanism of Ethanol Extracts of Ageratum Conyzoides. L. in Guangxi

Objective:To explore the anti-inflammatory effects and the underlying mechanisms of ethanol extract of Ageratum cony-zoides. L. from Guangxi. Methods:The auricle edema model was induced by dimethylbenzene in the mice and the paw edema model was induced by carrageenan respectively in the mice and rats to study the anti-inflammatory effects of ethanol extract of Ageratum cony-zoides. L. from Guangxi. The content of malondiadehyde (MDA) and proateglandin E2 (PGE2), and the activity of superoxide dis-mutase( SOD) in the mouse edema paw was measured. The contents of tumour necrosisfactor-α ( TNF-α) , interleukin-1β ( IL-1β) and interleukin-6(IL-6) in the rat serum were detected as well. Results:Compared with the model control group, the ethanol extracts of Ageratum conyzoides. L. from Guangxi could remarkably inhibit auricle edema in the mice and paw edema in the mice and rats( P<0. 05 or P<0. 01), the inhibition ratio for high, medium and low dosage group(6. 0, 3. 0, 1. 5 g·kg-1)was 29. 24%,16. 42% and 11. 21% in the auricle edema mice and 28. 66%,18. 79% and 13. 13% in the paw edema mice , respectively. It could remarkably re-duce MDA and PGE2 content and enhance the activity of SOD in the mouse inflammatory tissue(P<0. 05 or P<0. 01). In the paw e-dema rats, the inhibition ratio for high, medium and low dosage group(4. 5,2. 3, 1. 2 g·kg-1)was 43. 69%, 36. 01% and 23. 29%at the 3rd h, respectively , and it also could remarkably reduce serum TNF-α, IL-1βand IL-6 content(P<0. 05 or P<0. 01). Con-clusion:The ethanol extracts of Ageratum conyzoides. L. from Guangxi show significantly anti-inflammatory effects, and the mecha-nisms may be related to the ability of scavenging oxygen free radicals and reducing the release of inflammatory cytokines and proinflam-matory cytokines.
4.Radiation combination with DDP plus tegafur treated for advanced esophageal cancer

Objective To study on the curative effect of DDP and Tegafur plus radiation for advanced cancer of esophagus. Methods 148 Patients with advanced cancer of esophagus were randomized into two groups(each 74), one group were accepted radical radiotherapy followed by DDP and Tegafur (chemoradiotherapy group),the other group were accepted radical radiotherapy only (radiotherapy group), radiotherapy in both groups was similar (60 ~ 65 Gy/6~6.5 W with routine fractionation). Results The effect was 81.1 % in radiotherapy group and 90.5 % in chemoradiotherapy group, the difference in effect was no statistically significant (?2=2.719, P =0.09), The 1, 3 and 5-year survived rates were 43.2 %, 16.2 %, 9.5 % respectively in radiotherapy group and 71.6 %, 33.8 %, 21.6 % respectively in chemoradiotherapy group. 5.Losartan attenuates vascular remodeling of the aorta in spontaneously hypertensive rats and the underlying mechanism

Objective To determine the effect of losartan on vascular remodeling and the underlying mechanism in spontaneously hypertensive rats(SHR). Methods SHR of 12 weeks old were given losartan orally [0,15,30 mg/(kg·d),n=12]. The tail arterial pressure was measured every week.Eight weeks later, the pathological changes and p22phox expression in the thoracic aorta, the activity of catalase (CAT), the contents of H2O2 and AngⅡ in the plasma were evaluated. Results Blood pressure was increased in the SHR accompanied by the thickened wall and increased p22phox expression in the thoracic aorta. The plasma levels of H2O2 and AngⅡwere elevated while the CAT level was decreased in the SHR. Administration of losartan reversed the thickened wall and increased the CAT activity concomitantly with the decreased plasma levels of H2O2 and p22phox expression in the SHR. The plasma level of AngⅡincreased after the losartan treatment. Conclusion Oxidative stress induces the vascular remodeling of the aorta in the SHR. Losartan can reverse the vascular remodeling through down-regulating p22phox expression and inhibiting the oxidative stress.
6.Effect of calcitonin gene-related peptide on isoprenaline-induced cardiac fibroblast proliferation and collagen expression.

OBJECTIVE:
To explore the inhibitory effect of calcitonin gene-related peptide (CGRP) on cardiac fibroblast proliferation and collagen synthesis induced by isoprenaline and the underlying mechanism.
METHODS:
The primary cultured cardiac fibroblasts were incubated with isoprenaline (10(-5) mol/L) for 48 h after pretreatment with CGRP (10(-8) or 10(-7) mol/L) for 1 h. Cell activity was detected by MTT. The mRNA expression of collagen (types I and III) and connective tissue growth factor (CTGF) was determined by RT-PCR, and the levels of intracellular ROS were determined by DCFH-DA fluorescent probe.
RESULTS:
Isoprenaline significantly promoted fibroblast proliferation and up-regulated collagen (types I and III) and CTGF mRNA expression concomitantly with an increase in ROS production, which were attenuated by CGRP. The effect of CGRP on cardiac fibroblasts was inhibited by CGRP8-37, a selective antagonist of CGRP receptor.
CONCLUSION
CGRP is able to protect cardiac fibroblasts against isoprenaline-induced proliferation and collagen expression, which might be related to the down-regulation of CTGF expression through inhibition of ROS production.

7.MicroRNA-1 in Cardiac Diseases and Cancers.

MicroRNAs (miRs) are endogenous approximately22-nt non-coding RNAs that participate in the regulation of gene expression at post-transcriptional level. MiR-1 is one of the muscle-specific miRs, aberrant expression of miR-1 plays important roles in many physiological and pathological processes. In this review, we focus on the recent studies about miR-1 in cardiac diseases and cancers. The findings indicate that miR-1 may be a novel, important biomarker, and a potential therapeutic target in cardiac diseases and cancers.

8.Ring suture technique for renorrhaphy during partial nephrectomy in the management of renal hilar cancer.

OBJECTIVETo evaluate the safety and efficacy of "ring suture technique" during partial nephrectomy in the management of renal cell cancer located in renal hilar.
METHODSThe data of 31 patients with renal cell cancer located in renal hilar who underwent partial nephrectomy from January 2009 to January 2014 in Peking University First Hospital were collected. Of the 31 patients, 21 were male, 10 were female ranging from 48 to 75 years (average age of (58±13) years). "Ring suture technique" was adopted in all the operation which were performed following the same steps: a ureteral catheter was placed into ipsilateral ureter pre-operatively; renal artery was clamped first and after 30 s renal vein was also clamped; tumor was resected from kidney; the edge of renal parenchyma was sutured continuously; methylthioninium Chloride solution was injected into pelvis retrograde through the ureteral catheter, and the collecting system was repaired if needed; remove renal vein clamp and then the renal artery, repair any injury of the vascular when necessary. The renal ischemic time, blood loss and postoperative complications were analyzed retrospectively.
RESULTSRetroperitoneal laparoscopic surgery and laparotomy were performed in 14 patients and 17 patients each. Average maximum diameter of tumor was (3.3±1.2) cm, and median R.E.N.A.L.nephrometry score was 8 (7-11). Average ischemic time was (24±8) minutes (18.4-37.5 minutes). Median blood loss during operation was 130 ml (90-350 ml), average blood loss was (125±45) ml, and there was no uncontrollable massive bleeding. Radical nephrectomy was adopted in 1 case due to tumor embolus in branch of renal vein. All patients had good recovery after surgery without complications such as hemorrhage, urine leakage or renal atrophy during the first 3 months after surgery.
CONCLUSIONRing suture technique is a safe and effective method in the management of renal hilar cancer during partial nephrectomy.
