1.A study of anti-gastric carcinoma activity of tumor infiltrating lymphocytes stimulated by dendritic cells

Objective To investigate the killing activity of tumor infiltrating lymphocytes (TIL) stimulated by dendritic cells (DC) on gastric carcinoma cells in vitro. Since DC is the strongest antigen presenting cell (APC) which has been known, it can present antigens to T lymphocytes, including TIL, and can induce cytotoxic T lymphocyte (CTL) responded in vivo and vitro. Methods DC were isolated from the peripheral blood of patients with gastric carcinoma and stimulated by granulocyte/macrophage colony stimulating factor (GM CSF), interleukin 4 (IL 4) and tumor antigen. Then TIL were stimulated by DC and their killing activity on autogenous gastric carcinoma cells and SGC 7901 cells in vitro were observed. Results TIL stimulated by DC had very high killing activity on autogenous gastric carcinoma cells and the killing rate was (89.39?3.05)%, which was much higher than those of TIL not stimulated by DC and T lymphocytes stimulated by DC as well as T lymphocytes not stimulated by DC on autogenous gastric carcinoma cells. The killing rates of the last three ones were (54.37?1.50)%, (53.92?1.46)% and (3.55? 0.25) % respectively. However, their killing activity on SGC 7901 cells was lower. Conclusions The results indicate that DC from patients with gastric carcinoma can induce efficient and specific anti gastric carcinoma immune responses.
2.Diagnostic Value of Low-field MR Imaging in Lumbar Disc Degeneration with End-Plate Osteochondritis

Objective To investigate the diagnostic value of low-field MR imaging in lumbar disc degeneration with end-plate osteochondritis. Methods Imaging data of 504 patients with lumbar disc degeneration diagnosed by MRI were retrospectively analyzed. Among these cases, the total of 330 end-plate osteochondritis in 148 patients was found. According to MR signal findings of end-plate and it's adjacent vertebra, the lesions were classified as 4 types. Results 330 end-plates were involved in 148 patients,including type I in 68(20.6%),type II in 53(16.1%),type III in 174(52.7%) and type IV in 35(10.6%).Among all 148 patients,the end-plates at one lumbar disc,two disci and over three disci involved in 98(66.2%),42(28.4%) and 8(5.4%),respectively.The lesions were usually found in the lower lumbar vertebra.Conclusion MR imaging could accurately show the end-plate osteochondritis and degenerative lumbar disc.
3.Underlying mechanisms and potential of adipose-derived stem cells in the treatment of bronchial asthma

BACKGROUND:Currently, inhaled glucocorticoid is still the classic treatment for asthma. Adult stem cell transplantation has made significant progress in a variety of diseases, and it also provides new insights into the treatment of asthma.OBJECTIVE:To summarize the recent advances in the treatment of asthma with adipose-derived stem cells and related adult stem cells, and to discuss the therapeutic safety of adipose-derived stem cells and possible research directions in asthma therapy.METHODS:Relevant articles published from 2001 to 2016 were searched in PubMed, CBM, CNKI, and Wanfang databases. The keywords were (adipose-derived stem cells[All Fields]) OR (adipose stem cells[All Fields]) OR (adipose-derived mesenchymal stem cells[All Fields]) AND (asthma[All Fields]) in English and Chinese, respectively.RESULTS AND CONCLUSION:A total of 125 literatures were initially searched, and finally 54 representative papers were selected. Adipose-derived stem cells may reduce airway inflammation, airway hyperresponsiveness, ease collagen deposition and scar tissue formation, promote neovascularization, and reconstruct damaged airways in the mouse asthma model through immune regulation. It is necessary to understand its treatment mechanism of action deeply and comprehensively and carry out genomic analysis before introduction of adipose-derived stem cells as a conventional clinical treatment. In summary, adipose-derived stem cells may be a therapeutic potential for the treatment of airway allergic inflammatory diseases such as asthma.
4.Determination of Total Chloride in Oral Rehydration Salt Powder (Ⅲ) by Ion Chromatography

Objective:To establish a method for the determination of total chloride in oral rehydration salts powder (Ⅲ) by ion chromatography (IC). Methods:The analysis was performed on an IonPac AS 14A (250 mm × 4 mm) column with an IonPac AG 14A (50 mm × 4 mm) guard column and 9mmol·L-1 Na2 CO3 solution as the mobile phase. The flow rate was 1. 0 ml·min-1 with isocratic elution and the sample size was 25 μl. The suppressor was AERS 500(4mm) with 45mA suppression current, and an ECD was used for the detection. The quantity of chloride ion ( Cl-1 ) was calculated by the peak area with an external standard method. Re-sults:The linear range of Cl-1 was 0. 05-20. 00 mg·L-1(r=0. 9999);the recovery was 98. 89%(RSD=0. 56%, n=6). Conclu-sion:The results demonstrated that the method has the advantages of high sensitivity, simple sample pretreatment, promising accuracy and good reliability, which is suitable for the determination of total chloride in oral rehydration salt powder (Ⅲ) .

Diffuse large B-cell lymphoma (DLBCL) is the most common type of non-Hodgkin lymphoma with highly heterogeneous clinical course. How to recognize patients with high risk is coming into the focus of the 57th American Society of Hematology (ASH) annual meeting. The International Prognostic Index (IPI) score as a classic prognostic system is challenged by variety of new prognosis systems. The tumor microenvironment plays an important role in progression of lymphoma, and its prognostic value draw much more attention. The value of PET-CT for prognosis of DLBCL has also been affirmed in this meeting.
6.Application of dual-color interphase fluorescence in situ hybridization in sex-mismatched hematopoietic stem cell transplantation

Objective To evaluate the significance of dual-color interphase fluorescence in situ hybridization (FISH) in the engraftment estimation and minimal residual disease (MRD) monitoring after sex-mismatched allogeneic hematopoietic stem cell transplantation (allo-HSCT).Method The fluorescence signal of samples from 35 cases in different periods after sex-mismatched allo-HSCT were detected by interphase FISH using chromosome enumeration probes (CEP) X and Y. Results All of 35 patients had been determined to obtain engraftment after allo-HSCT. When the disease relapsed,FISH showed that the percentage of donor chromosomes was decreased and when the disease got remission,the percentage of donor chromosomes increased. When conventional cytogenetics showed 100 % XX or 100 % XY,FISH showed different percentage of host chromosomes.Conclusions The test of dual-color interphase FISH is reliably sensitive and simple for engraftment evaluation and MRD monitoring post HSCT. It is a good complement method to cell morphology and traditional karyotype analysis.
7.Prophylaxis of cytomegalovirus infection with ganciclovir in renal transplant recipients: a prospective randomized controlled study

Objective To investigate the prophylactic treatment of cytomegalovirus (CMV) infection in renal transplant patients.Methods Fifty-five patients were divided into two groups. Group A (n=27) was administered with ganciclovir (5 mg/kg every day) intravenously for 30 days. Group B(n=28) was the control group. According to donor (D) or recipient (R) CMV antibody categories,each group had D+/R-,D-/R+,D+/R+ three subgroups. All patients were followed up prospectively for 6 months with measurements of CMV antibody,CMV-DNA antigen,acute rejection for monitoring activity of CMV infections.Results The rate of CMV infection and disease was similar in both groups. 8.A study on cancer vaccines made up of cells treated by TNF-? gene modifying combined with freezing

Objective:To investigate the possibility of producing vaccine with higher effect of anti-hepatoma by means of freezing the tumour cells which have been modified by TNF-? genes before. Methods: H22 cells which had been modified by TNF-? genes were treated by means of freezing and 60Co radiation or of 60Co radiation, respectively. Then, they were put to use to produce two kinds of vaccines. Effects of these two kinds of vaccines on anti- plant-hepatoma in mice were compared. Results: Effects of increasing the inhibiting rates, the levels of lectin-induced IL-2 and TNF from splenocytes, and the splentic NK activity, were all obviously seen stronger in vaccine made up of cells treated by the method combined with freezing, than that in vaccine made up of cells by no freezing. Conclusion: Method of freezing, after procedure of TNF-a gene modifying, could raise up the immunogenicity of the tumour cells, thereby enhance the anti-plant-hepatoma effect in mice induced by the vaccine.
9.An in vitro study of the effects of different enamel conditioners on the shear bond strength and the adhesive remnant index

Objective:To evaluate the effects of different enamel conditioners in combination with an adhesive on the shear bond strength and the adhesive remnant index in vitro.Methods:One hundred and eight permanent healthy premolars from orthodontic patients and fresh saliva from a healthy young volunteer were collected.The teeth were cleaned,dried and etched by phosphoric acid,polyacrylic acid or Transbond plus SEP respectively(36 teeth for each agent).Then the teeth were bonded to brachet by RMGICs with the presence of water,saliva on the tooth surface or without any of them respectively(12 teeth in each group).After bonding,all samples were tested on a testing machine of Instron.Shear bond strength and adhesive remnant index were recorded.Results:Highest mean shear bond strength was observed in the group of teeth etched by Traspond plus SEP and bonded by RMGICs with the presence of saliva.Higher mean shear bond strength was obtained with the presence of saliva or water on the tooth surface than without any of them.Conclusion:Transbond plus SEP conditioning followed by RMGICs bonding may produce high shear bond strength between moistured tooth surface and brachet.
10.Treatment of the floating knee with interlocking nails (a report of 6 cases)

Objective To evaluate the clinical effects of the interlocking intramedullary nails in treatment of the floating knee. Methods Internal fixation with the interlocking nail in unilateral femur and tibia was performed on 6 patients with the floating knee, and the treatments of complications were analyzed and summarized. Results After operation the 6 patients were followed up and their X ray films were reviewed after 3, 6, and 12 months respectively. Fractures in unilateral femur and tibia healed at once in 4 patients, the union of one femur fracture delayed in one case and the union of tibia fracture delayed in one case. The knee joint could flex to 80~90 degrees in 5 of the 6 patients, and the other one could flex to 60 degrees. No shortening of the limb occurred in all the patients who healed after 6~18 months. Conclusion As the “floating knee”is a kind of high energy injury, often complicated with injury of important organs, the complication should be treated while the internal fixation be conducted as soon as possible. The interlocking intramedullary nail is a good device with which the function of the limb can recover fast with the postoperative dysfunction of the knee and ankle joints greatly reduced.
