1.Determination of Diosbulbin B in Dioscorea bulbifera L. from Different Regions

Objective To establish a HPLC method for determining and comparing the contents of diosbulbin B in
Dioscorea bulbifera L. from different regions. Methods The medicine powders were extracted twice by water, then the water extracts were combined and detected by HPLC at 35℃,with Kromasil-C18 column (4. 6 mm×250 mm,5μm) and a mobile phase of acetonitrile-water-glacial-acetic acid (34660. 1). The flow rate was 1. 0 mL·min-1 and the detection wavelength was 210 nm. Results The linear range of diosbulbin B was 26. 0-260. 0 μg·mL-1(r=0. 999 9). The average content of diosbulbin B in Dioscorea bulbifera L. from different areas was that of Hebei>Guangxi>Jiangsu>Sichuan>Zhejiang. Conclusion The method is simple,quick,accurate and suitable for the determination of diosbulbin B in Dioscorea bulbifera L. from different regions.
2.AFLP Analysis on genetic diversity for germplasm resources of Fritillaria thunbergii cultivated in Zhejiang Province

Objective To study the genetic diversity of Fritillaria thunbergii,a traditional Chinese herb in Zhejiang Province in China.Methods The genetic diversity of six representational populations of F.thunbergii including 32 individuals was investigated by amplified fragment length polymorphism(AFLP) maker technique.Results The genetic diversity was revealed as follow: the Nei′s genetic diversity index(He) 0.169 0?0.175 7,Shannon′s information index(I) 0.269 8?0.245 3,percentage of polymorphic loci(PPB) was 76.85% at the species level;Ht 0.169 0?0.030 9,and Hs 0.150 8?0.024 0,I 0.233 3?0.261 9, PPB was 50.38% at population level.The genetic differentiation index(Gst) was 0.107 6,Nm 4.147 0.The result of dendrogram of six populations indicated that Dongyang and Yongkang populations shared the minimum genetic distance(0.015 0),they were classified into a group,and Xiangshan and Jinyun populations shared the maximum genetic distance(0.032 4).Conclusion The genetic diversity of F.thunbergii cultivated in Zhejiang Province is very rich,which could ensure the long-term survival of F.thunbergii.But the genetic diversity of F.thunbergii is relatively higher in population levels while lower at the species levels and the degree of genetic differentiation occured among the populations is not significant.The germplasm resources are relatively stable among these six populations.These populations could be used to breed the fine strains of F.thunbergii as the bases.
3.Genetic structure analysis of cultivated Scrophularia ningpoensis in Zhejiang Province

Objective To observe the genetic structure of cultivated Scrophularia ningpoensis in Zhejiang Province.Methods The genetic structures of six typical S.ningpoensis populations were analyzed by fluorescence AFLP marker.Results Bands(12 552) were generated by seven pairs of AFLP primer combinations,of which 8 808 were polymorphic,and the polymorphic rate was 70.17%.The variety ranges of PPB among different populations were 41.67%—55.56%,and 47.30% in average.I was between 0.190 8—0.238 3,and 0.221 8 in average.Ne was between 1.201 4—1.280 6,and 1.236 9 in average.Gst was 0.127 1,Nm was 3.432 4.UPGMA Cluster analysis showed that the six populations can be divided into two clusters,as that of Tiantai,Jinyun,and Jingning were one sub-cluster,and Dongyang,Pan′an,and Xianju were another one sub-cluster.Conclusion There is a relative high genetic diversity level in cultured S.ningpoensis of Zhejiang Province.Genetic differentiation exists among populations,but it exists in population mostly.There is a relative high genetic intercommunion among populations.The genetic distance is not related to the geographic environment.
4.Effect of portal vein blocking on permeability of the intestinal mucosa in pigs

Objective To study the effect of portal vein blocking on the permeability of the intestinal mucosa in pigs. Methods Healthy Rongchang pigs were divided into 3 groups: ① sham operation group(SO), ② portal vein clamping for 45 min group (PVC-45'), ③ portal vein clamping for 60 min group (PVC-60'). Urine lactulose/mannitol(L/M) ratio was measured after portal vein blocking. Results The L/M ratio was increased significantly (P<0.05) in PVC-45' and 60' groups than in SO group, with that of PVC-60' higher than that of PVC-45' group, but not significantly. Conclusion The increase of intestinal mucosal permeability after portal vein blocking is an early and important index for the damage of the intestinal mucosa barrier.

Objective: To study the cellular immunoregulatory effect of Huangqi through intravenous injection in patients with chronic hepatitis B. Methods: In treated groups, light and mild chronic hepatitis B (15 patients in each group) were treated with Huangqi and Ganlixin. In control groups, 10 patients in each group with light and mild chronic hepatitis B were treated with Ganlixin. Normal control group included 10 healthy volunteers. 6.Treatment progress of diffuse large B-cell lymphoma: reports from the 57th American Society of Hematology annual meeting

Diffuse large B-cell lymphoma (DLBCL) is one of the most common non-Hodgkin lymphomaaccounting for 30 %-40 %. The most common first-line therapy for DLBCL is rituximab in combination withchemotherapy. About two thirds of patients treated by the first-line therapy achieve a complete remission (CR)and are cured finally, but nearly one third of patients can not reach CR after frontline treatment appearingrefractory or relapse early, especially for the high risk patients or cases with MYC alterations, the regimenimproving the long-term survival is not much. In the 57th American Society of Hematology (ASH) annualmeeting, a plenty of treatment as focus on these patients brought in encouraging results, which makes itpossible to further improve the CR rate. The progresses on DLBCL of relapse and refractory, high risk andspecial types were summarized in this paper based on the reports in the 57th ASH annual meeting.
7.Effects of Apr-1 gene on cell cycle regulation of cholangiocarcinoma QBC939 cell lines

Objective To investigate the role and the mechanism of Apr-1 gene on cholangiocarcinoma QBC939 cell lines proliferation and cell cycle regulation. Methods Apr-1 gene was transfected into QBC939 cells by using liposomes to establish a QBC939 cell model ( QBC939-Apr-1 ) stably expressing Apr-1 gene. Apr-1 mRNA expression and the changes in cell cycle and cell growth of QBC939 cells were analyzed by RT-PCR, flow cytometry ( FCM ) and growth curve before and after transfection. The regulatory effect of Apr-1 gene on the expression of cell cycle-related genes was investigated in QBC939 cells before and after Apr-1 transfection using cell cycle gene microarrays. Results Significant suppression of cell growth was observed with the cell model stably expressing Apr-1 gene. Apr-1 over-expression caused cell arrest from 9% to 13% (P <0. 01 ) increase in G2 population. Cell cycle gene microarrays demonstrated that the expression of Skp2 、UBE1 was up-regulated, while the expression of MRE11A 、CKS2 、CDK8 、CDC45 was down-regulated by more than 3 folds. Conclusions Apr-1 gene suppresses QBC939 cell proliferation in vitro, QBC939 cells presented with differences in the expression of cell cycle-related genes after Apr-1 gene transfection.
8.The chromosomal aberration detected by comparative genomic hybridization in lung cancer

Objective To understand the molecular aberration at whole genomic level,CGH (comparative genomic hybridization) was used to investigate genetic abnormality in lung cancer.Methods Comparative genomic hybridization was performed in 17 cases to detect the global genomic aberration in cancer tissue cells.Results All of 17 cases detected by CGH showed chromosomal aberrations.The average numbers of chromosomal gains and losses in each case were 7.0 and 4.8 in NSCLC and 8.4 and 9.6 in SCLC,respectively.The frequency of gains and losses on chromosome had no significant differences between NSCLC and SCLC.The frequencies of gains on chromosomal arms 3q24 -28 and 11q13(58.3% and 58.3% ) in NSCLC were significantly higher than that in SCLC(0% and 0% ) ( P <0.05 and <0.05,respectively).Conclusions The cytogenetic aberration generally existed in lung cancer cells.Several regions ( more than one) of chromosomal aberration were involved in the carcinogenesis of NSCLC and SCLC.The regions and frequencies of chromosomal aberration in NSCLC were somewhat different from that in SCLC,which might result in the different biological behavior of the two types of lung cancer.The chromosomal aberration might be served as a marker to differentiate the two types of lung cancer.
9.Chemical components in Hemsleya chensnsis (III).

OBJECTIVETo study the chemical constituents of Hemsleya chensnsis.
METHODVarious chromatographic techniques were used to separate and purify the constituents. The spectral data (MS, NMR) were obtained for structure elucidation.
RESULTEight known triterpenoid saponins were isolated from the root of H. chensnsis. Their structures were elucidated as dihydrocucurbitacin B (1), 25-O-acetyl-dihydrocucurbitacin F (2), cucurbitacin F (3), 3-O-(6'-ethyl ester) -beta-D-glucuropyranosyl oleanolic acid-28-O-alpha-L-arabinopyranoside (4), oleanolic acid-3-(6'-methyl ester) -beta-D-glucuropyranosyl (1-3) -alpha-L-arabinopyranoside (5), oleanolic acid-28-O-beta-D-glucopyranosyl-(1-6) -beta-D-glucopyranoside (6), 3-O-(6'-methyl ester) -beta-D-glucuropyranosyl oleanolic acid-28-O-beta-D-glucopyranosyl-(1-6) -beta-D-glucopyranoside (7), 3-O-(6'-methyl ester) -beta-D-glucuropyranosyl (1-2) -beta-D-glucopyranoside-oleanolic acid-28-O-beta-D-glucopyranoside (8).
CONCLUSIONAll compounds except for 2 were isolated from H. chensnsis for the first time.

10.Effect and mechanism of curcumol on angiogenesis activity of zebrafishes.

OBJECTIVETo conduct a preliminary study on the effect of curcumol in promoting angiogenesis activity and its mechanism in zebrafishes, in order to provide basis for clinical prescription.
METHODZebrafishes biological model was established to, observe curcumol's effect on embryo blood vessel growth, blood vessel regeneration of adult fishes after tail-cutting and tissue regeneration of fish fries after tail-cutting. The relative fluorescence quantitative PCR method was adopted to determine the gene expression of vascular endothelial growth factor (VEGFA) and receptor VEGFR2 of fish fries after tail-cutting.
RESULTCurcumol contributed to angiogenesis of intersegmental blood vessels in zebrafishes embryos and speed up regeneration of blood vessels in adult fishes after tail-cutting. Furthermore, curcumol can increase the gene expression of VEGFA and VEGFR2 in fish fries.
CONCLUSIONCurcumol can promote angiogenesis in zebrafishes, and enhance the gene expression of VEGFA and VEGFR2 in fish fries after tail-cutting and speed up the regeneration of their tails.
