1.Application of New Zealand rabbit femoral puncture anti-leakage performance verification biodegradable mesh-like microporous balloon

Objective Biodegradable is validated by the New Zealand rabbit femoral puncture mesh porous seepage prevention performance of the balloon.Methods The experiment was divided into the experimental group and control group, by piercing the New Zealand rabbit left leg near the femur, establish channel introduced after bone drill, fracture model is established, degradable mesh implant microporous balloon and/or bone cement.Observe and record the operation process of each New Zealand rabbit blood oxygen saturation change and cases of bone cement occur breakup and shortness of breath and groupings.Results Surgery under general anesthesia, bone cement injection process smoothly, balloon group of intraoperative breathing smooth, blood oxygen saturation has no obvious change; CPC group of blood oxygen saturation in the cement perfusion and perfusion after 81.63 +/-32.02, 32.02 +/-32.26, respectively, compared with perfusion 96.67 +/-1.71 in front of the difference is statistically significant, of which 3 cases died in 5 cases, shortness of breath, direct infusion of bone cement is more likely to lead to pulmonary embolism.Conclusions Further confirmed that the degradable mesh porous balloon seepage prevention of CPC and the breakup of the effect is obvious, comply with the design requirements.
2.Experimental study on the protective effects of ischemic preconditioning on brain ischemia-reperfusion injury in mice

Objective To develop a forebrain ischemic preconditioning model in C57BL/6 mice and determine the protective effects of ischemic preconditioning on brain ischemia-reperfusion injury. Methods Forty-eight 8-10 week old C57BL/6 mice weighing 19-23 g were randomly divided into four groups of 12 animals, A: sham-operation group; B: ischemic preconditioning group; C: ischemia-reperfusion group (I/ R); D: ischemic preconditioning + I/R group. The animals were anesthetized with halothane. Bilateral common carotid artery (BCCA) was exposed and occluded for 6min (ischemic preconditioning) or 18min followed by 3h reperfusion (I/R). In group D BCCA was firstly occluded for 6 min, then released and 48h later occluded again for 18min followed by 3h reperfusion. 6 animals in each group were sacrificed 72h after reperfusion and brain was immediately removed for detection of DNA fragmentation of neurons in hippocampus by TUNEL. Another 6 animals were sacrificed on the 7th day after I/R and neuronal damage was identified by microtubule-associated protein-2 immunochemistry and quantified by cresyl violet staining. Results I/R resulted in severe damage to hippocampus in C57BL/6 mice. The ischemic preconditioning caused neither noticeable hippocampal neuronal damage nor DNA fragmentation but significantly reduced hippocampal neuronal damage and DNA fragmentation caused by I/R. Conclusion Our results indicate that hippocampal neuronal injury induced by I/R can be greatly reduced by ischemic preconditioning in C57BL/6 mice. Many kinds of gene-altered C57BL/6 mice are available, this preconditioning model may provide an useful method for investigating the molecular mechanisms of ischemic preconditioning.
3.Correlations between intracranial and extracranial artery stenosis and ambulatory arterial stiffness index

Objective To investigate the correlations between ambulatory arterial stiffness index and intracranial/extracranial arterial stenosis.Methods One hundred and twenty-eight cases of ischemic cerebrovascular disease were collected in our hospital from January 2010 to March 2012.Joint diagnosis of cranial computer tomography(TCD) and magnetic resonance angiography (MRA) and,or CT angiography (CTA) were used to detect the degree and number of intracranial arteries,and in accordance with the lesions level,patients were divided into stenosis group,the mild stenosis group,the moderate stenosis group and severe stenosis group.24 h ambulatory blood pressure was monitored and ambulatory arterial stiffness index (AASI) was calculated and statistically analyzed.Results (1) Age,sex,hypertension proportion of diabetes,body mass index(BMI) of different Intracranial arterial stenosis in four groups did not have significant differences (P >0.05),but in AASI the without stenosis group is 0.48 ± 0.15 ; the mild stenosis group 0.62 ± 0.16,the moderate stenosis group 0.61 ± 0.17,severe stenosis group 0.64 ± 0.15,and there was significant difference (F =3.955,P =0.001).(2) Age,sex,hypertension proportion of diabetes,BMI of different extracranial arterial stenosis in four groups did not have significant differences (P > 0.05),but in AASI the without stenosis group was 0.48 ± 0.01 ; the mild stenosis group 0.57 ± 0.11,the moderate stenosis 0.59 ± 0.12,and severe group 0.60 ±0.15,and there was significant difference (F =3.643,P =0.002).In comparison between any two group:light,moderate and severe stenosis AASI were significantly higher than those without stenosis,and there was significant difference (P < 0.05).And there was significant different in AASI among different intracranial and extracranial arterial lesions (F =7.395,P < 0.001).Compared to 0 branch pathological changes,1 branch,2 branch,3 branch and above,there was was significant difference(P < 0.05).Conclusion Based on a 24-hour ambulatory blood pressure monitoring indicators,AASI was mainly reflecting the impact of atherosclerosis on blood pressure,associated with intracranial and extracranial artery stenosis.AASI would play a major role in clinical diagnosis and treatment of ischemic cerebrovascular and forecast.

Research on effects of environmental endocrine disrupting chemicals (EDCs) is an important field in environmental science.The research progress on mechanism of EDCs in environment was present in this paper.Not only the action mechanism of EDCs mediated via receptor was expatiated,but also the interference influencing the synthesis and metabolism of endogenous hormone and the effects of EDCs on neural and immune system were discussed in this paper.This paper will conduce to the further studies on the mechanism of EDCs and the development of screening method for new EDCs.
5.A PRELIMINARY APPROACH TO BIOLOGICAL FUNCTION OF SOYBEAN SELENOPROTEIN

Objective: To investigate the biological function of soybean selenoprotein by the study of anti-oxidation,immunological and anticarcinogenic efficacy. Method: The soybean selenoprotein at different doses of Se were fed intra-gastrically in Kunming mice. A low Se feedstuff was used as basic diet. Result: (1) when dose is below 202.5 ?g/kg bw Se, the number of white cell and red cell was increased remarkably, and the activity of GSH-Px and SOD in liver and serum was increased at dose dependent manner, but LPO in serum and liver was decreased noticeably. At level of 607.5?g/kg, all the indices were reversed; (2) A dose-dependent cancer protective effect was expressed in range of 22.5-202.5?g/kg Se of soybean selemoprotein in the diet. Feeding soybean selenoprotein can postpone the death of mice with carcinoma . Total tumour yield was consistently reduced by 78% with 202.5?g/kg supplementation; (3) Any abnormal response was never noticed during all trial by supplementing 202.5?g/kg selenoprotein. Conclusion: The increased effect of anti-oxidation and immunological modulation might be the mechanism of tumour suppression by soybean selenoprotein, and Kunming mice tolerated the soybean selenoprotein very well without any side effects.
6.Relationship between vascular endothelial growth factor and microvessel density in placenta and placenta increta

Objective To investigate the relationship between the expression level of vascular endothelial growth factor (VEGF) and microvessel density (MVD) in placenta tissue and placenta increta.Methods Thirty singleton pregnant women who received antenatal care and underwent cesarean section in Fujian Maternity and Child Health Hospital between November 2013 and August 2014,were enrolled in this study.They were divided into placenta previa group,placenta increta group and control group,with ten patients in each group.Placenta tissue was collected from each patient.Expressions of VEGF and its mRNA were detected by immunohistochemistry,Western blot,and real-time polymerase chain reaction.MVD in placenta tissue was measured by immunohistochemistry.Rank sum test,t test,Kruskal Wallis test,one-way ANOVA and Spearman correlation test were used for statistical analysis.Results (1) Gestational age at admission and delivery in placenta previa and placenta increta groups was lower than in control group (all P<0.05).Compared with control group,the placenta previa and placenta increta groups had more blood loss,and longer operating duration and hospital stay (all P<0.05).(2) The expression levels of VEGF and its mRNA in placenta increta and placenta previa groups were higher than in control group (VEGF:0.691±0.032,0.695 ± 0.027 and 0.518±0.025,respectively,F=373.401;VEGF mRNA:1.667±0.661,1.832±0.678 and 0.767±0.269,respectively,F=27.399;both P<0.05).But there was no significant difference between placenta previa and increta groups.(3) There was no significant difference of VEGF expression in increta location,border site and normal site in placenta increta group,but its mRNA was decreasing (2.519± 0.116,1.482 ± 0.232 and 1.000± 0.000,respectively,F=240.827,P<0.05).(4) Expression level of VEGF at the attachment of umbilical cord,upper and lower margin of placenta in placenta previa group was higher than in control group (0.702 ± 0.026 vs 0.528± 0.020,t=12.302;0.698 ± 0.026 vs 0.519±0.035,t=12.715;and 0.685±0.029 vs 0.509±0.010,respectively,t=17.891;all P<0.05).Expression of VEGF mRNA in placenta previa group was higher than in control group (2.080± 0.539 vs 1.024±0.272,t=8.093;1.587±0.757 vs 0.546±0.083,t=2.401;1.828±0.704 vs 0.731 ±0.157,t=4.259;all P<0.05).(5) MVD in placenta increta group and placenta previa group was higher than in control group (171.2± 14.7,155.7± 14.6 vs 147.8±12.3,respectively,F=7.277,P<0.05).(6) Expression level of VEGF in placenta increta group and control group was positively associated with MVD (r=0.825,P<0.05).Conclusions There may be some common mechanisms in the occurrence of placenta previa and placenta increta.Overexpression of VEGF in placenta and abnormal formation of villous vessels may be important factors in the pathogenesis of placenta increta.
7.Relationship between Umbilical Artery Blood Flow, Levels of sEng and NO and Perinatal Outcome in Patients with Preeclampsia

Objective: To explore the association of fetal umbilical artery (UA) Doppler blood flow parameters with umbilical vein (UV) serum sEng,nitric oxide (NO) level and the perinatal outcome in patients with preeclampsia(PE). Methods: Color ultrasonography was used to detect the resistive index (RI), pulse index (PI), the peak systolic velocity (Vs), diastolic velocity (Vd) and the peak-systolic/diastolic ratio(S/D) of UA in 39 PE patients(20 cases of mild preeclampsia, 19 cases of severe preeclampsia), 20 cases of normal full-term pregnant women and 20 gestational hypertension(GH) women. The UV serum level of sEng was analyzed using enzyme-linked immunosorbent assay (ELISA). The level of NO was determined using nitric acid recovery, and to analysis of perinatal outcome. Results: Compared with normal pregnancy and GH, levels of S/D, RI, PI and sEng were significantly higher in the mild and severe preeclampsia group(P < 0.05), and levels of Vs, Vd and NO were significantly lower(P < 0.05). There was a negative correlation between the values of PI, RI and S/D with the level of UV serum NO in the mild and severe preeclampsia group(P < 0.05). There was a positive correlation between the values of PI, RI and S/D with the level of UV serum sEng in the mild and severe PE group (P < 0.05). The incidence of perinatal disease was significantly higher in PE (P < 0.05). Compared with normal perinatal infants, the abnormal perinatal infants had obviously higher levels of S/D, RI, PI and sEng (P < 0.05), and lower levels of Vs, Vd and NO(P < 0.05) in PE. Conclusion: The umbilical artery blood flow parameters can reflect the disease condition and fetal status, which has an important clinical value.

Objective To observe the mitochondrion and nucleus ultrastructural damage and caspase-3 expression in hippocampal CA_3 neurons during kainic acid(KA) induced status epilepticus(SE) in rats.Methods SE was induced for 2 h with KA in adult male Wistar rats.3,12 and 24 h later the rats were killed and the hippocampal CA_3 subareas were taken out to make brain sections.The neuronal damage on the whole with light microscope and the ultrastructure of mitochondrion and nucleus with electron microscope.Caspase-3 expression of the same area was examined with immunohistochemical staining.Results 24 h after SE,by the light microscope examination,the KA group showed that the neurons put scattered disorder and nucleus shrink firmly.3 h after SE,electron microscope examination showed swelling cristae and ruptured membrane of mitochondria.The change of nucleus were significant margination of chromatin 24 h after SE.Compared to normal control group,the caspase-3 expression increased 12 h after SE,the average number of positive cell and the gray scale were obviously higher(all(P

10.Peripheral cholangiocarcinoma associated with hepatolithiasis

Objective To understand the clinical features of peripheral cholangiocarcinoma (PC) developed against the background of hepatolithiasis. Methods Between 1995 and 2002, a total of 11 PC patients with definite history of hepatolithiasis were treated surgically in our hospital. The diagnosis, treatment and prognosis of this clinical entity were retrospectively analyzed. Results Peripheral cholangiocarcinoma occurred in 5 1% of patients with hepatolithiasis in this group and the tumor was located in the left lobe in 7 (64 7%) cases. All patients underwent laparotomy, with diagnosis established preoperatively in 3 (27 2%) cases and intraoperative diagnosis was missed in 2 (18 1%) cases. Radical resections were performed in only 3 (27 2%) cases. The overall operative morbidity and mortality were 72 7% and 9 1% respectively . Infection was the main postoperative complication. The postoperative 1 and 2 year survival rate was 33 3% and 11 1% respectively. Conclusions Cholangiocarcinoma may develope in patients with a history of hepatolithiasis. The prognosis of PC is still poor because of the difficulty to achieve early diagnosis and radical resection.
