1.Effects of Industrial Wastewater Containing Chromium on the Mutagenicity of Ground Water and Drinking Water

Objective To study the effects of industrial wastewater containing chromium from Anyang Bicycle Works (ABW) on the mutagenicity of ground water and drinking water. Methods The ground water samples were collected from wells located on the southwest of the pollution source of ABW, near the gate of ABW(A) and at a distance of 60 m from the east of ABW(B) respectively, and also from the following sampling points closely located along the lower reach of sewage ditch: cigarette work(C), iron alloy works(D), match works(E), chemical and chemical fiber works(F). The raw water and finished water samples were collected from water plant. The contents of chromium(Cr6+) in water samples were measured and the mutagenicity of the water samples were tested by Vicia faba root tips cell micronucleus test. Results The contents of Cr6+ in ground water samples showed a decreaseing trend (0.358 5-0.010 0 mg/L) with the increasing distances from the sampling points to the pollution source of ABW, the order of which was the following: A, B, C, D, E, F. The contents of Cr6+ in ground water samples collected from the gate of ABW and the location at a distance of 60 m from the east of ABW exceeded the related standard 6.17 and 2.08 times respectively.The contents of Cr6+ in raw water and finished water samples were 0.0123 and 0.0095mg/L respectively.All of the water samples except that from the chemical and chemical fiber works showed higher micronucleus rates in different degree compared with that of negative control. Objective To observe the influence of standardized deep well water on mortality and morbidity of esophageal cancer (EC) in Linzhou. In order to test the hypothesis of "nitrogenous compound metabolised cycling" in etiology of EC. Methods Based on the investigation data collected in 1999 among which some data were from Linzhou Institute of Prevention and Treatment of EC. Three kinds of water were included, standardized deep well water(SDWW) from the well with the depth exceeding 100 meters ,finished time exceeding 6 years; unstandardized deep well water(UDWW) and mountain spring water(MSW). The mortality and morbidity of EC in the the related population. Results There were 281 458 villagers of 165 villages to drink SDWW, 183 289 villagers of 129 villages to drink UDWW and 45 274 villagers of 50 villages to drink MSW. 3.Detection of Mutations in the Wilms' Tumor Gene (WT1) in Leukemias

Objective To analyze mutations in the Wilms tumor gene (WT1) in Ieukemogenesis.Methods WT1 gene in peripheral blood was determined by PRC-SSCP technique,in 32 cases of acute leukemia,included 9 cases of acute granulocytic leukemia,8 cases of chronic granulocytic leukemia(mean age-33 years) and 16 specimens of normal subjects were also detected.Results Mutations in the WT1 gene in 3 of 32 leukemias were found .WT1 mutations were found in 11% of cases of acute lymphoblastic leukemia and in 13% of cases of acute myeloid leukemia,in which they were associtated with a poor response to chemotherapy.Conclusions The mutations in Wilms tumor gene WT1 are associated with leukemogenesis and its therapy,which WT1 transcripts may prove a significant tumor marker, as a MRD monitor in evaluating remission status and early relapse,and may be useful in prognosis of acute leukemia.
4.Clinical Observation on Combined Tuina and Medicated Bath for Early Intervention of Neonatal Brain Injuries

Objective: To observe the clinical effect of early intervention of combined tuina with medicated bath for neonatal brain injuries.Methods: Brain-benefiting tuina manipulations,medicated bath and music plus exercise therapies were employed in 60 infants with medium or severe brain injuries.The Gesell measurement methods were adopted prior to and 3 months after treatment to evaluate the development quotient(DQ).In addition,the long-term follow-up was made after one and a half years.Result: The scores of DQ prior to treatment,3 month after treatment and 1.5 years after treatment were(34.98±28.94),(66.17±14.91)and(75.40±14.69)respectively,showing a marked improvement after 3months and 1.5 years of treatment(P=0.000).Conclusion: Early intervention using tuina manipulations could enhance the brain development,repair the nerve injury,improve the motion and cognitive ability and prevent the sequela of the nervous system.
5.Effects of breathing exercise on preoperative pulmonary function in adolescent scoliosis patients

BACKGROUND: Acute respiratory insufficiency is common complication of surgical treatment of adolescent scoliosis patients. The preventive effects of breathing exercise before surgery is still uncertain.OBJECTIVE: To explore the feasibility and efficacy of breathing exercises for scoliosis patients before operation.DESIGN: Nonrandomized cross sectional study.SETTING, PARTICIPANTS and METHODS: Totally 35 hospitalized kids with scoliosis and pulmonary dysfunction aged from 9 to 15 years old were selected from Shanghai Changhai Hospital. A set of breathing exercise including which included pursed lip breathing, abdominal breathing, diaphragmatic breathing and blowing balloon was performed to these patients one week before surgery. The changes of pulmonary function were recorded and compared before and after exercises.MAIN OUTCOME MEASURES: The change of vital capacity, lung capacity, forced ventilation capacity(FVC), maximal ventilation volume(MVV) before and after breath exercises.RESULTS: All sorts of indes are improved comparing with those before exercises. The vital capacity, lung capacity, FVC and MVV were greatly improved from(1.77 ±0. 36), (2.67 ±0. 84), (2.07 ±0.46), (52.5 ± 14.0) L before exercises to(2.65 ±0. 29), (3.56 ±0.79), (2.41 ±0.41), (70. 1 ± 17.0) L respectively( t = 3.63 - 11.28, P < 0.01).CONCLUSION: Preoperative breathing exercises can improve the pulmonary functions of kids with scoliosis and improve their tolerance to surgery and prevent the postoperative respiratory insufficiency.
6.Effect of Motor Imagery on Patients Accepted Radiotherapy after Operation for Cerebral Glioma

Objective To observe the effect of motor imagery therapy on patients accepted radiotherapy after operation for cerebral glioma. Methods 34 patients were randomly divided into control group (n=17) and observation group (n=17). Both groups accepted routine physiotherapy during radiotherapy, 45~60 min/d, 5 times a week for 4 weeks, while the observation group underwent motor imagery additionally, 15 min/d for 4 weeks. They were evaluated with Fugl-Meyer Assessment (FMA), Manual Muscle Test (MMT) and modified Barthel Index (MBI) before and after treatment., and with Karnofsky Performance Scale (KPS) 1 year later. Results There was no significantly difference in the scores of FMA, MMT and MBI between 2 groups before treatment (P>0.05). There was no significantly difference in the score of FMA in both groups before and after treatment (P>0.05). The score of MMT improved in the observation group after treatment (P<0.01). The scores of MBI improved in both groups after treatment (P<0.05). And there was no significantly difference in the D-value score of FMA, MMT and MBI between 2 groups (P>0.05). After 1 year follow-up, KPS showed that the number of survival patients and their quality of life were better in the observation group than in the control group (P<0.05). Conclusion Motor imagery therapy can improve the muscle force, activities of daily living and long-term quality of life in patients during radiotherapy after neurosurgery for cerebral glioma.
7.Construction, expression, and bio-activity assay of an anti-IL-1βscfv and TNFR1 fusion protein

Objective To express the anti-IL-1βscfv and soluble TNF receptor 1 (sTNFR1),and analyze their bio-activities.Methods sTNFR1 was obtained by RT-PCR from the total RNA of HeLa cells,and fused with IL-1βscfv by the hinge fragment of IgG molecule.The fusion gene IL-1scfv:TNFR1 was cloned into the expression vector pET27b(+).The fusion protein was expressed and purified from inclusion bodies.Results The ELISA analysis showed that the fusion protein could bind hIL-1β and hTNF-α respectively in a dose-dependent manner,indicating that scfv and sTNFR in the fusion protein can form the correct spatial configuration.The dolt-blot analysis showed that the fusion protein could concurrently bind with hIL-1β and hTNF-α,indicating that the combination of the two parts of the fusion protein does not influence each other for binding to their target molecules.The bioactivity assay showed that the fusion protein could inhibit both the cytotoxicity of hTNF-α on L929 cells and hIL-1β-induced proliferation of L929 cells,indicating that the fusion protein has the ability to neutralize hTNF-α and hIL-1β.Conclusion A bispecific fusion protein IL-1scfv:TNFR1 was successfully constructed.The fusion protein has the ability to inhibit the biological activity of hTNF-α and hIL-1β,and provides a drug candidate for the treatment of rheumatoid arthritis.
8.Expressions of CD70 mRNA and protein and methylation status of CD70 gene promoter in T cells from patients with systemic lupus erythematosus

Objective To detect the expressions of CD70 mRNA and protein and to determine the methylation status of CD70 gene promoter in T cells from patients with systemic lupus erythematosus (SLE).Methods Peripheral blood CD4+ and CD8+ T cells were isolated from 15 patients with active SLE,15 patients with inactive SLE and 15 healthy control subjects.Real-time quantitative reverse transcription-PCR was carried out to quantify the mRNA expression of CD70,flow cytometry to determine the frequency of CD4+CD70+ and CD8+ CD70+ T cells,and bisulfite sequencing to evaluate the methylation status of CD70 gene promoter in CD4+ and CD8+ T cells.Differences in these parameters among these groups were analyzed by one-factor analysis of variance and SNK-q test.Results Compared with the healthy controls,the patients with active SLE and inactive SLE showed a significant increase in CD70 mRNA expression in CD4+ T cells (0.82 ± 0.12 and 0.73 ± 0.11 vs.0.45 ±0.09,F =53.017,P < 0.01) and in the frequency of CD70+CD4+ T cells (80.30% ± 11.04% and 66.80% ± 3.98% vs.12.48% ± 3.45%,F =311.517,P < 0.01).Also,the expression of CD70 mRNA in CD4+ T cells and the frequency of CD70+CD4+ T cells were significantly higher in patients with active SLE than in patients with inactive SLE (both P < 0.05).There was a positive correlation between the frequency of peripheral CD70+CD4+ T cells and disease activity in SLE in these patients (r =0.792,P < 0.01).The average methylation index of the region between-600 bp and-300 bp of CD70 gene promoter in CD4+ T cells was 0.32 ± 0.05 and 0.36 ± 0.05 respectively in the patients with active and inactive SLE,significantly lower than that in the healthy controls (0.62 ± 0.05,F =152.64,P < 0.01),and the patients with active SLE showed a significantly lower methylation index than those with inactive SLE (P < 0.05).Conclusions The CD70 gene promoter in CD4+ T cells is significantly hypomethylated in patients with SLE,which may directly lead to the overexpression of CD70.
9.Establishment and effect of nursing information technology platform

Objective To explore nurses' bonus assessment and distribution methods based on the information technology platform and to improve the efficiency of nursing management. Methods We build a hospital informa-tion platform in order to collect nursing work efficiency data by using existing computer information network. We distributed the bonus through the information platform according to evaluate nursing quality and performance effi-ciency. Results After the information flatform used for 2 years,the satisfaction of nurses with different professional titles and different experience showed no significant difference (P>0.05). The time of department management person-nel collecting and distributing data change from [103.75 (180.00,215.00)] min per month to [42.50 (60.00,90.00)] min per month.The difference showed statistical significance(P<0.05). Conclusion The technology platform can im-prove the satisfaction and the nursing management efficiency.
10.Treatment of Helicobacter pylori-infected gastritis in BALB/c mice by HP1188-IgY

AIM:To evaluate the effects of treatment with HP 1188-immunoglobulin yolk ( HP1188-IgY) on Helicobacter pylori ( H.pylori)-infected gastritis in BALB/c mice.METHODS:BALB/c mice were used to establish an animal model of H.pylori-infected gastritis, and the mice were divided into 8 groups (10 mice per group).Oral antibiotics were used in group 1, 1 mg HP1188-IgY in group 2, 1 mg HP1188-IgY plus 30%sucralfate in group 3, 5 mg HP1188-IgY in group 4, 5 mg HP1188-IgY plus 30%sucralfate in group 5, PBS in group 6, and 30% sucralfate in group 7 with the treatment once per day for 10 d;and 2.5 mg HP1188-IgY was injected hypodermically twice with a 48-h interval in group 8.Another 10 mice were used as normal control in group 9.The planting of bacteria in the stomach was assayed by bacteri-al culture, rapid urease test, PCR and pathological sectioning .RESULTS:Intragastric administration with 1 mg HP1188-IgY plus 30%sucralfate per day effectively cured the injury of gastric mucosa caused by H.pylori infection, and the effect has no significant difference compared with antibiotics (P>0.05).CONCLUSION:We establish a BALB/c mouse mod-el infected with H.pylori successfully.Sucralfate (30%) is an ideal protectant for HP1188-IgY, which might decrease H. pylori infection in the stomach of BALB/c mice by oral inoculation .
