1.Progresses on relationship of conotruncal heart defects and retinoic acid signaling pathway

Congenital conotruncal heart defects (CHD),including tetralogy of Fallot (TOF),double outlet of right ventricle(DORV),transposition of the great arteries(TGA),persistent truncus arteriosus (PTA),is a variety of common and serious congenital cardiovascular diseases which are threatenting to the health of neonates and infants.Previous studies have demostrated that TBX1 contricuted to some of the CHD.Recent studies have also confirmed that retinoic acid signaling pathway plays a curcial role of development of CHD.Further studies have also found that TGF-β2 differential expression is closely related with phenotypic polymorphism of conotruncal heart defects in retinoic acid receptor mutant mice,of which the mechanism is not yet clear.This paper provides an overview of the relationship between TBX1-RA signaling pathway and congenital conotruncal heart defects and TGF-β2-RA and CHD during the development of CHD.
2.Relationship of conotruncal anomalies and chromosome 22q11.2 deletion

Congenital cardiovascular anomalies are present in approximately 80% of children with 22q11.2 deletion syndrome.Three genes in chromosome 22q11.2 ( TBX1,CRKL,and ERK2 ) have been identified whose haploinsufficiency causes anomalies of 22q11.2 deletion.The most common diseases are conotruncal anomalies,which include tetralogy of Fallot ( TOF),pulmonary atresia with ventricular septum defect (PA-VSD),truncus arteriosus,and interrupted aortic arch.In major phenotypes,a high prevalence of the deletion is noted in patients with TOF with pulmonary atresia,TOF associated with pulmonary atresia and major aortopumonary collateral arteries,persistent truncus arteriosus,and type B interruption of aortic arch.In minor phenotypes,right aortic arch,aberrant subclavian artery,and major aortopulmonary collateral arteries are frequently associated with cardiovascular anomalies associated with 22q11.2 deletion.In conclusion,conotruncal anomaly associated with aortic arch and branch anomalies should increase the suspicion of 22q11.2 deletion.
3.Mechanism of FTY720 on cardiovascular complications of diabetes mellitus

Objective To study the mechanism of FTY720 on type Ι diabetic rats. Methods The typeⅠdiabetes rat model was established by feeding male SD rats with high energy urea and injecting into the abdominal cavity with low dose cephalosporins (STZ, 30 mg/kg). In the following, the treated rats were divided into two groups: model group and FTY720 group. Another group of untreated rats was assigned as normal control group. FTY720 group was given 1 mg/kg FTY720, the normal control group and model group was given the equal amount of water. Results The cardiac function of FTY720 group was recovered markeyly. FTY720 activated the expression of vascular endothelial cells S1P1 , diabetes and reduced the expessions of S1P3 and PKCβ Ⅱand it restored the migration ability of diabetes cardiovascular endothelial cell , as well as the abnormally elevated cardiovascular endothelial cells induced by high sugar permeability. Conclusion The S1P1 and S1P3 cut is an important reaction pathway to the complications of diabetes and cardiovascular dysfunction. FTY720 may reduce the damage to core blood vessels caused by diabetes , and pathological angiogenesis which functionally depends on the PKCβⅡ signaling pathways. Therefore, FTY720 may provide a potential therapy for cardiovascular function impaired by diabetes.
4.Vacuum sealing drainage combined with external fixator in the treatment of post-traumatic osteomyelitis

BACKGROUND:Posttraumatic osteomyelitis is bone infection after surgery of open fractures or other joint, which leads to more secretions in antrum or bone exposed wounds, long treatment cycle, delayed healing of fracture, even disunion. The key link in the treatment is complete debridement, adequate drainage, and the use of external fixators with low interference and far away from lesions. <br> OBJECTIVE:To observe the clinical efficacy of external fixator combined vacuum sealing drainage (VSD) in the treatment of post-traumatic osteomyelitis, and compared with conventional catheter drainage. <br> METHODS:Since June 2010 to June 2013, 21 patients of post-traumatic chronic osteomyelitis were included in this study and divided into VSD group (n=11) and conventional catheter drainage group (n=10). Al patients in VSD groups underwent debridement and VSD was used to fil dead space, then the wounds were sutured. Cases appeared nonhealing fractures underwent external fixation, 10 cases using external fixators and 1 case retained original external fixator. Al patients achieved the closed wounds at the second phase, 9 cases were directly sutured, and 2 cases received skin flap transplantation. Among them, 1 case received autologous bone graft at the third phase because of bone defects. In the conventional catheter drainage group, al patients underwent debridement and conventional catheter drainage to rinse the wounds after surgery, 3 cases failing to close the wounds changed to receive ordinary dressing and skin flap transplantation at the second phase. Cases appeared nonhealing fractures underwent external fixation. Local swel ing, pain and fever were observed after treatment. The time of total hospitalization stay and fracture healing was recorded. <br> RESULTS AND CONCLUSION:The VSD group quickly control ed preoperative symptoms such as swel ing and pain and fever than the conventional catheter group (P<0.05). The length of hospital stay showed no significant difference between the two groups (P>0.05). Al patients were obtained through clinic service and fol owed up for 24-36 months. The healing time in the VSD group was shorter than that in conventional catheter group (P<0.05). Until the end of fol ow-up, no patients in the VSD group appeared infection recurrence, and two cases in the conventional catheter group had recurrent infection. The results suggest that VSD combined with external fixator can repair post-traumatic chronic osteomyelitis due to rapid control of infection, easy care, shortened treatment time, and reduced rate of postoperative recurrence. In addition, external fixator can be used as the final fixation for post-traumatic chronic osteomyelitis.
5.The influence of disposing the shoulder by the dental pneumatic ultrasonic hand-piece on the edge sealing prop-erty of the zirconium dioxide crown

15 extracted premolars were selected and randomly divided into 3 groups, with 5 in each group. MANI TF-13 bur、TR13-EF bur and the dental pneumatic ultrasonic hand-piece were respectively used to dispose the shoulder. 15 Zirconium dioxide full crowns were made. The shoulder marginal microleakage was observed by staining method. Dental pneumatic ultrasonic hand-piece showed the best anti-mic-roleakage effect in shoulder refinement.
6.Case-control study on the serum sex steroids levels in prepuberal children with autism

Objective To investigate the changes of the serum sex steroids levels in prepuberal children with autism,and analyze the possible clinical implications.Methods The serum sex steroids,including folliclestimulating hormone(FSH),luteotropic hormone(LH),testosterone(T),estradiol(E2),progesterone(P),dehydroepiandrosterone sulfate(DHEA-S) and androstenedione were measured in 44 autistic children(35 boys and 9girls,47.5 ± 18.9 months) and 44 normal children(35 boys and 9 girls,45.0 ± 18.7 months),using chemiluminescence immunoassay.Results ①The progesterone levels in autism group were significantly lower than control group((< 0.48,2.08) nmoL/L vs (< 0.48,6.95) nmol/L,Z =-3.564,P < 0.01),but no statistical differences were observed in other sex steroids levels(P> 0.05).②The progesterone levels in < 48 months autism group showed no significant differences from ≥48 months autism group (Z =0.150,P > 0.05).There were no statistical differences in progesterone level between the boys and girls of the autism group(Z=1.972,P>0.05).Conclusion The progesterone levels are lower in autistic children,and they are not associated with the children's age or gender.This study indicates that the progesterone levels are closely associated with autism.
7.Three-dimensional finite element analysis on the support and protection of spine protector for thoracic-lumbar vertebra

BACKGROUND:Human spine protector can protect human thoracic-lumbar vertebra segments against injury, and the design and development of a novel dynamic protector needs the verification of various experimental means.
OBJECTIVE:Using the three-dimensional finite element method, we evaluate the effect of spine protector and the biomechanical reaction of thoracic-lumbar vertebra under the axial loading.
METHODThe thoracic-lumbar vertebra were cut from the whole spine three-dimensional finite element model. Then the thoracic-lumbar vertebra models carrying spine protector were taken as experimental group, while the models without the protector served as control group. Al the specimens were evaluated, constrained, loaded and figured out by its properties. The results of equivalent stress and strain distribution were obtained from the data. RESULTS AND CONCLUSION:In both groups, the stress was distributed at axial and posterior column of L 2 when the load was applied in axial direction. According to the data obtained from the experiment, both the experimental group and the control group had achieved the maximum stress at 16 ms, 3.919 Mpa and 5.727 Mpa, respectively. The statistical analysis result showed that the stress varied significantly at T 12 and L 2 in two groups (both P<0.05). However, the stress distribution at T 11 and L 1 showed no significant difference between the two groups (both P>0.05). Experimental findings indicate that, spine protector can significantly reduce the vertical stress of the thoracic-lumbar vertebra when fal ing on the ground, and share the vertical load, which is protective to thoracic-lumbar vertebra.
8.Sex differences in cognitive and behavioral characteristics in children with autism spectrum disorder indifferent age

Objective To explore sex differences in cognitive and behavioral characteristics in children with autism spectrum disorder (ASD) in different age.Methods The study involved 621 children(503 boys,118 girls) diagnosed with ASD from June 2013 to June 2015.Using raw scores to examine cognitive and behavioral characteristics in children with ASD based on the Psycho-Educational Profile-3rd Edition (PEP-3).The subjects were divided into two groups according age:<4Y group and ≥ 4Y group.The sex differences of cognitive and behavioral characteristics were analyzed in different age groups.Results No sex differences were found in characteristic motor behaviors (CMB) and characteristic verbal behaviors (CVB) raw scores in <4Y group(P>0.05),while girls with ASD had lower raw scores on CMB and CVB raw scores in >4Y group((21.56±7.33) vs (24.09±4.80),(9.79±7.05) vs (13.27±5.75) ; P<0.05)).Girls with ASD had lower raw scores on cognitive verbal/preverbal (CVP),CMB and CVB in ~ 4Y,~5Y,and ~6Y group (P<0.05).However,there was no sex difference on CVP,CMB and CVB in ~ 3Y and ~ 7Y group (P>0.05).Conclusions Characteristic verbal and motor behaviors may not be obvious in girls with ASD in early childhood,which may increase the difficulties of early clinical identification.The development of cognitive ability in girls with ASD is far behind that in boys in the crucial 3-5 years old,as well as social reciprocity and characteristic behaviors.It suggests that the behavior characteristics are closely related to cognitive development.
9.Upregulation of miR-27b Facilitates Apoptosis of TNF-α-Stimulated Fibroblast-Like Synoviocytes

PURPOSE: The aim of this study was to explore the function of microRNA-27b (miR-27b) in fibroblast-like synoviocytes (FLSs) stimulated by tumor necrosis factor α (TNF-α). MATERIALS AND METHODS: mRNA expression of miR-27b in FLS cells (MH7A) treated with or without TNF-α was determined by q-PCR. MiR-27b mimics was transfected into MH7A cells to upregulate miR-27b expression. MTT assay and flow cytometry analysis were performed to investigate the effect of miR-27b on MH7A cell viability and apoptosis. The targets of miR-27b were predicted by TargetScan. The direct regulation of miR-27b on IL-1β expression was verified by luciferase assay. The protein expression levels of apoptosis-related proteins, IL-1β, and NF-κB signaling-related proteins were detected by Western blot. RESULTS: We discovered that miR-27b expression was decreased in MH7A cells stimulated by TNF-α. Upregulation of miR-27b by miR-27b mimics significantly inhibited the proliferation and promoted the apoptosis of TNF-α-stimulated MH7A cells. Consistently, upregulation of miR-27 decreased the level of Bcl-2 and increased Bax and caspase-3 expression in MH7A cells stimulated by TNF-α. Luciferase assay revealed that IL-1β was indeed a target of miR-27b. By quantitative real-time PCR and Western blot, we found that the expression of IL-1β is negatively regulated by miR-27b. Moreover, the NF-κB signaling pathway was significantly inhibited by miR-27b. CONCLUSION: Taken together, our results illustrated that enhanced miR-27b expression results in the suppression of proliferation and the promotion of apoptosis in FLSs stimulated by TNF-α, partially by regulating IL-1β expression and NF-κB signaling.

10.A comparison of dual- and triple-freeze protocols for liver cryoablation in a Tibet pig

Objective This study compares a dual-freeze protocol with a triple freeze protocol for hepatic cryoablation in the Tibetan pig model.Method Cryoablation with a dual-(10-5-10-5 min)and triple-freeze (5-5-5-5-10-5 min) protocol for the normal livers of 9 Tibet pigs was performed under exposed operation.Temperature changes of cryoprobes and diameter changes of iceballs were measured during the ablation,and seven days later the pathological changes of cryozones were reviewed and the surface and depth cryolesions were measured.Results Compared with cryoablation with two freeze-thaw cycles,there was a greater iceball diameter for cryoablation by three freeze-thaw cycles.Also,seven days after cryosurgery,there were similar surface and deep cryolesions in dual-and triple-freeze protocols.Pathologically,the triple freezing protocol was associated with a longer zone of complete necrosis.Conclusions With the same freezing time (20 min),the triple-freeze protocol may become a more powerful liver-ablation method in cryosurgical application.
