Lower urinary tract symptoms due to benign prostatic hyperplasia is of great prevalence among ageing males,and it can markedly affect patients' quality of life.Personalized drug therapy according to genetic characteristics is the ideal choice for this problem.Here we summarized the latest research findings on the associations of gene polymorphisms with drug efficacy and adverse reaction in the treatment of benign prostatic hyperplasia/urinary tract symptoms.

3.The Predictive and Monitoring Value of VEGF in Ovarian Hyperstimulation Syndrome

Objective To explore the value of vascular endothelial growth factor(VEGF) in predicting and mornitoring ovarian hyper-stimulation syndrome(OHSS). Methods 11 cases of OHSS patients and 16 cases of high-responder patients were enrolled in this study, respectively from IVF-ET or ICSI programme. Serum VEGF level before giving gonadotrophin, on the days of hCG administration, OPU and ET, and at the 7th and 14th days after ET as well as the VEGF level of follicular fluid (FF) on the day of OPU were measured. The serum VEGF of hospitalized women with severe OHSS were also measured during the whole therapy. Results Compared with high-responder patients, serum VEGF level on mid luteal and late luteal phases significantly increased in OHSS patients, and FF VEGF level obviously increased as well. Serum VEGF level gradually decreased along with clinical and biological improvement of OHSS. Conclusion FF VEGF level could predict the occurrence of OHSS, and serum VEGF level could monitor the progress of OHSS.
4.Effects of 4 sorts of growth factors on the cell growth of androgen independent prostate cancer cell line PC-3M

Objective To evaluate the effects of TGF ? 1, EGF, bFGF, KGF on the cell growth of androgen independent prostate cancer cell line PC 3M in vitro. Methods We cultured androgen independent prostate cancer cell line PC 3M in vitro and added TGF ? 1, EGF, bFGF, KGF to the medium at different concentrations of 1, 5,10 and 50 ng/ml. Brdu incorporation method was used to determine the status of cell proliferation. Results The absorbance ( A ) value of the control group PC 3M was 0 759. Seventy two hours after adding the growth factors separately, the inhibition was enhanced with the increasing of concentrations. At the dosage of 50 ng/ml, A value decreased to 0 400(TGF ? 1),0 455(EGF),0 532(bFGF),and 0 491(KGF),respectively, showing significant difference compared to the controls. When the mixture of 1 ng/ml EGF with 0 5 ng/ml or 5 ng/ml TGF ? 1 was added into the medium, A values were 0 522 and 0 299, respectively. Conclusions All these four growth factors inhibited the proliferation of PC 3M cell line in a dose dependent manner. TGF ? 1 and EGF have no interaction to the cell proliferation.
5.The regulatory effect of transforming growth factor-?_1 on the proliferation and apoptosis of human prostatic epithelial cells

Objective To investigate the regulatory effect of transforming growth factor-? 1 (TGF-? 1)on the proliferation and apoptosis of human prostatic epithelial cells in vitro. Methods The primary culture was prepared by collagenase dissociation of minced prostatic tissue obtained from 8 BPH patients (53 to 70 of age) on open prostatectomy and from 5 organ donors (23 to 36).Cells were cultured in serum free prostate epithelial cell growth media and identified by immunocytochemical studies.The cells were passaged and cultured in media free of bovine pituitary extract and EGF and TGF-? 1 and EGF in different doses were then added either alone or in combination according to experimental requirement.The rate of cell proliferation was assessed by counting the cell number and by 5-bromo-2'-deoxyuridine (ELISA).Apoptosis induced by TGF-? 1 was examined by in situ transferase-mediated dUTP nick and labeling. Results The cultured cells had the typical morphological features of epithelium and stained positive for PSA and Pan-Keratin.The cells from normal prostates of young donors showed more mitogenic activity than those from BPH prostates.EGF enhanced cell proliferation whereas TGF-? 1 inhibited cell proliferation which could be reversed by increasing concentration of EGF.TGF-? 1 induced apoptosis of human prostatic epithelial cells.The effect of TGF-? 1 and EGF was concentration dependent. Conclusions TGF-? 1 is a direct physiological regulator of cell proliferation and cell apoptosis of human prostatic epithelial cells.
6.The regulation effect of transforming growth factor-?_1, epithelial growth factor on the proliferation of human prostatic epithelial cell in vitro

Objective To investigate the effect of TGF ? 1 and EGF on the proliferation of cultured human prostatic epithelial cells. Methods The primary culture was prepared by collagenase digestion of minced prostatic tissue obtained from prostatectomies of 8 benign prostatic hyperplasia(BPH) patients (aged 53 to 70) and 5 organ donors (aged 23 to 36). Cells were cultured in serum free prostatic epithelial cell growth media and identified by immunocytochemical method. The cells were passaged and cultured in media without bovine pituitary extract (BPE) and then, EGF and TGF ? 1 with different concentration were added into the culture either alone or in combination according to the experimental protocol. The cell proliferation was assessed by counting the cell number (BrdU) and ELISA. Results Cultured cells showed typical morphological features of epithelium and stained positive for PSA and Pan Keratin. After 5 7 passages cells went senescence. BPE and EGF enhanced whereas TGF ? 1 inhibited the cell proliferation and the inhibition was reversed by increasing the concentrations of EGF. The effect of both TGF ? 1 and EGF displayed concentration dependent manner. Conclusions Cell proliferation of the cultured human prostatic epithelial cells could be modulated by TGF ? 1 and EGF. These results indicated that TGF ? 1 and EGF, as the negative and positive growth factors might play an important role in the pathogenesis of BPH.
7.The affection of recombinant adenovirus vector expressing wild type p53 to prostate cancer cell line PC3M

Objective To investigate the availability and mechanism of recombinant adenovirus vector expressing wild type p53 in gene therapy of prostate cancer. Methods Prostate cancer cell line PC3M was cultivated and recombinant adenovirus vector expressing wild type p53 was added to the media. Then the levels of proliferation and apoptosis were detected and changes of bax were exa mined . Results AdCMVp53 is effectively transfected to the cells. The transfecting ratio is 95%. In the cells transfected with AdCMVp53, the level of proliferation decreased. Tested by BrdU, the absorbance rate decreased from 1.454 in the control group to 0.496 in the experimental group( P
8.Causes and management of readmission after prostatectomy for BPH (report of 106 cases)

Objective To explore the causes of readmission after prostatectomy for BPH and to discuss the treatment approaches. Methods A total of 106 cases who were eligible for the study were reviewed.47 cases had residual and recurrent adenoma;of these cases,42 underwent TURP and 5 open prostatectomy again.32 cases developed urethral stricture;of these cases 27 underwent transurethral cold knife incision plus electric resection of scar and 3 urethral orifice reconstruction and 2 suprapubic cystostomy.11 cases who had experienced bleeding after TURP underwent transurethral bladder washing and coagulating.7 cases had neuropathic bladder dysfunction;of these cases 2 underwent transurethral bladder neck circle muscle resection,3 intermittent self-catheterization and 2 suprapubic cystostomy.6 cases had prostatic cancer;of these cases 1 underwent radical prostatectomy,3 palliative TURP plus orchiectomy and 2 orchiectomy.2 cases had incontinence after TURP;of the 2 cases 1 used external collecting device and 1 was transfered to other hospital for surgery.1 case had abdominal incision hernia and then underwent hernia repair. Results Among 106 cases, 97 cases underwent operations and of them 84 (86.6%) cases achieved successful results.The follow-up time was 6 months to 15 years with a mean of 5 years and 8 months.9 cases with urethral stricture required regular urethral dilation after surgery.4 cases who had had unsuccessful surgery underwent suprapubic cystostomy.Other 9 cases did not undergo operations due to poor health and other reasons. Conclusions Correct preoperative diagnosis, rational surgical type and adequate intraoperative and postoperative management are key to preventing readmission. Transurethral surgery is most favorable treatment choice for readmitted patients after prostatectomy for BPH.
9.Construction and expression of cell-penetrating peptide CCL fusion protein expression vector

Objective To evaluate the construction of expression vector for fusion protein of cell-penetrating pep-tide CCL (PEP-CCL).Methods CCL6-PEP-6XHis was inserted into plasmid pABP,pABP-CCL6-PEP plasmid was extracted and then transfected into HEK293 cells,CCL6-PEP-6XHis was expressed and purified by chromatog-raphy and detected with Western Blot.Results PEP-CCL express vector was successfully constructed and purified. PCR product of CCL6-PEP-6XHis Tag was ligated with T vector,recombinant was transferred into the host cells, then host cells were cultured,plasmid was extracted and sequenced,the sequence was identical to targeted gene. CCL6-PEP-6XHis was successfully inserted into the eukaryotic expression vector pABP,plasmid was extracted and digested,electrophoresis results revealed that a fragment with 430bp was digested by Hind Ⅲ+XbaⅠ,which was identical to the expected value.Western Blot revealed that CCL6-PEP fusion protein could be recognized by His monoclonal antibody.Conclusion PEP-CCL express vector can be constructed and expressed in eukaryotic cells.
10.THE PSYCHOLOGICAL DIFFERENCES IN ABDOMINAL AORTIC ANEURYSM PATIENTS BEFORE AND AFTER OPERATION

To investigate the psychological difference in abdominal aortic aneurysm patients before and after operation and its influence on life quality, 24 patients who can understand and complete the questionnaire were enrolled in the study, the inquiry was performed after admission and discharge with the standard SCL 90 table. The results showed that the general conditions, depression, anxiety, compulsive symptoms and fear of the patients had marked differences before and after operation. It is concluded that operation of abdominal aortic aneurysm can not only prevent the rupture of aneurysm but also relieve the patients′ psychological pressuer and improve their life quality.
