1.Effects of microbe-derived antioxidant on sleep and antioxidant ability in the rat

Objective To investigate the effect of different doses of microbe?derived antioxidant on sleep and antioxidant ability in mice. Methods Sixty male Kunming mice with similar body weight were randomly divided into 4 groups. The control group received normal saline, and the experimental groups received microbe?derived antioxidant in a dose of 0?5 g/kg bw, 1?0 g/kg bw or 1?5 g/kg bw once per day, respectively. The experiment period was 30 days. At the end of experiment, the mice of each group were intraperitoneally injected sodium pentobarbital to induce sleep. The mice fall sleep was judged by righting reflex. After the test of sleep, blood was taken for detection of serum antioxidant ability. Results Compared with the low dose and high dose groups, the middle dose group showed a significantly prolonged sodium pentobarbital?induced sleeping time (P<0?05). Compared with the control group, low and high dose groups, the middle dose group had highly significantly increased GSH?Px activity ( P <0?01 ) and significantly increased content of SOD. Under these conditions, the middle dose group reduced both the contents of MDA and 8?ISO?PGF2α ( P<0?05 ) compared with the control group. Conclusions Our results suggest that microbe?derived antioxidant exerts effect on sleep and antioxidant ability in rats. Supplement of 1?0 g/kg bw/d shows the most significant effects.
2.Three kinds of engineered hepatic tissues constructed using rat bone marrow mesenchymal stem cells

BACKGROUND:Engineered hepatic tissue is considered a promising strategy for healing acute liver failure. But, there are series of hindrances in the construction of engineered hepatic tissues, including acquisition of vital hepatocytes, choice of scaffolds and culture system, and nutrition supply. OBJECTIVE:To construct three kinds of engineered hepatic tissues in hope to screen the optimal one for transplantation in acute liver failure. METHODS:After purification, amplification, bone marrow mesenchymal stem cells were induced to differentiate into hepatocyte-like cells which were co-cultured with acellular amniotic membrane, DACRON PATCH cardiovascular surgical patch, biological surgical patch, respectively to construct three kinds of engineered hepatic tissues. After 3 days of culture, morphological and functional detections were performed. RESULTS AND CONCLUSION:Rat bone marrow mesenchymal stem cells with higher purity were successful y harvested by using density gradient centrifugation and adherent methods, and then the cells were differentiated into hepatocyte-like cells. In the three kinds of engineered hepatic tissues, hepatocyte-like cells were found to be combined with the biological surgical patch to the maximum extent, and their combination exhibited stronger ability of urea synthesis and albumin secretion, which provides experimental basis for treatment of acute liver failure.

4.Application of perioperative enteral nutrition support in patients with gastrointestinal cancer

Objective To study the application method and effect of perioperative enteral nutrition support in patients with gastrointestinal cancer.Methods 82 patients with gastrointestinal tumor were randomly divided into two groups.41 cases in the control group were given intravenous nutrition.41 patients in the study group were given enteral nutrition support 7d before operation,outage on the day of surgery,postoperation consistently given enteral nutrition support for 7d.The preoperative nutritional status,immune index,postoperative complications,the length of time and hospitalization expenses were compared between the two groups.Results Preoperative 1d,7d,the ALB,TRF,TP, PAB levels in the study group had no statistically significant differences compared with admission (all P >0.05).7d after operation,the TRF,PAB of the study group were (1.71 ±0.17)g/L,(174.67 ±22.81)mg/L,which of the con-trol group were (1.46 ±1.46)g/L,(142.73 ±27.32)mg/L,the differences were statistically significant (t =2.662, 2.917,all P <0.05).3d,7d after operation,the CRP,IL -6 and TNF - levels in the two groups increased signifi-cantly,compared with preoperation the differences were statistically significant (P <0.05).3d,7d after operation,the team CRP levels of the study group were (121.5 ±15.6)mg/L,(55.17 ±7.62)mg/L respectively,IL -6 levels were (81.42 ±13.56)pg/L,(56.14 ±9.73)pg/L and TNF -levels were (67.25 ±17.23)pg/L,(63.23 ±11.80)pg/L;In the control group,3d,7d after operation,CRP levels were (161.3 ±20.7)mg/L,(99.26 ±20.74)mg/L,IL -6 levels were (103.62 ±21.54)pg/L,(77.43 ±18.26)pg/L,the TNF -levels were (79.01 ±16.9)pg/L,(68.25 ± 12.48)pg/L,the differences between the two groups were statistically significant (P <0.05).The incidence rate of complications of the study group was 2.44%,which of the control group was 12.20% (χ2 =2.717,P <0.05 ). Conclusion Gastrointestinal tumor patients with malnutrition,perioperative enteral nutrition support can promote the synthesis of protein metabolism,relieve excessive postoperative systemic inflammatory reaction,reduce the risk of surgery.
5.The expression and significance of IL-23/IL-17 axis in esophageal tissues of rats with reflux esophagitis

Objective To detect the expressions of interleukin-17(IL-17) and interleukin-23(IL-23) in esophageal tissues of rats with reflux esophagitis(RE) and explore the role of IL-23/IL-17 axis in the pathogenesis of RE.Methods Thirty SD rats were randomly divided into 3 groups,namely,control group,sham-operated group and model group.RE was induced by adopting partial pyloric ligation plus cardiomyotomy.Then rats in each group were fasted for 24 h but had free access to water.They were fed 24 h after operation.Four weeks later,rats were killed and pathological changes of esophagus mucous membrane were observed by HE staining in each group.The expressions of IL-23p19 and IL-17 mRNA in esophageal tissues were examined by real-time fluorescent quantitative reverse transcriptase polymerase chain reaction(RT-FQ-PCR).IL-17 protein content in esophageal tissues was measured by ELISA.Results After four weeks,compared with control group,model group had significant pathological changes of esophagus mucous membrane.Expressions of IL-23p19 and IL-17 mRNA in the esophageal tissues of model group were significantly higher than those of the other two groups;the content of IL-17 protein was significant higher in model group than in the other two groups.There were no significant differences between control group and sham-operated group.Conclusion IL-23/IL-17 axis is an important cellular factor involved in the pathogenesis of RE and may be involved in the chronic inflammation of RE.
6.Analysis of Volatile Oils and Ethanol-Extracts from Different Parts of Radix Fici Hirtae

To investigate the quality of different parts of Radix Fici Hirtae. The content and components of volatile oils and ethanol-extracts from different parts of Radix Fici Hirtae were analyzed by the combination of gas chromatography and mass spectrometry (GC-MS). The content of volatile oils in the cortex was higher than that in the xylem and the chemical components either in the cortex or in the xylem are similar. The main components in volatile oils are hexadecanoic acid, oleic acid+linoleic acid, linoleic acid amide, palmitic acid amide,stearic acid amide and dibutyl phthalate. The main components in ethanol-extracts are psoralen, hexadecanoic acid, hexadecanoic acid ethyl ester, bergapten, oleic acid and linoleic acid. The content of psoralen in the cortex was higher than that in the xylem.[Conclusion]The chemical components in different parts of Radix Fici Hirtae are similar but the contents of volatile components and psoralen are higher in the cortex than those in the xylem.
7.Factors influencing pneumococcal vaccination uptake among elderly people in Guangzhou

Objective To explore the factors influencing pneumococcal vaccination uptake among the el-derly people in Guangzhou. Methods A survey by questionnaire was performed among 827 subjects aged 60 years or above and living in Guangzhou for five consecutive years. Chi-square test and multiple logistic regression analysis were applied to identify factors influencing pneumococcal vaccination uptake among the elderly. Results The positive factors for vaccination uptake among the elderly people included age of over 70 years (OR=1.677, 95%CI: 1.156 ~ 2.434), mental workers (OR = 1.837, 95%CI: 1.214 ~ 2.779), education background of over-three-year-course training (OR=1.769, 95%CI:1.039~3.012), and history of chronic diseases (OR=1.659, 95%CI:1.096~2.512) were positively associated with pneumococcal vaccination uptake. Monthly disposable income was not an influencing factor (OR=1.420, 95%CI: 0.895 ~ 2.251). Conclusion Strengthened publicity of pneumo-coccal vaccination among the elderly people and flexible measures tailored to the needs of different groups are rec-ommended in order to improve pneumococcal vaccination uptake among the elderly people.
8.Improvement of immunoprecipitation/immunoblotting of endogenous protein by TrueBlot antibody

Objective:To improve the immunoblotting of immunoprecipitated proteins and decrease the interference of immunoprecipitation antibody in the interaction of endogenous proteins. Methods:Transient transfect cells with fusion protein expression vector containing the targeted S5b gene and the FLAG tag, the transfected cells or untransfected cells were harvested to study the exogenous or endogenous protein interaction. The total cell lysate was immunoprecipitated by specific antibody. Then the eluted immunocomplex was separated by SDS-PAGE, and the TrueBlot antibody or conventional antibody was used as the secondary antibody for immunoblotting detection of S5b and its partner (Rpt1 and Rpt2). Results:Clear immunoblotting bands for S5b, Rpt1 and Rpt2 were obtained. Conclusion:TrueBlot antibody prefers the immunoblot antibody to immunoprecipitation antibody, and decreases the interruption of immunoprecipitation antibody to display clear protein band.
9.Effects of natural walking on human plantar pressure and surface electromyography of lower extremities

Objective To study the effects of natural walking on human plantar pressure and surface electromyogrphy (sEMG) of lower limbs.Methods 28 healthy male volunteers were enrolled,and their plantar pressure and surface electromyogrphy related parameters were measured before and after 30 min natural walking.These parameters not only include plantar peak pressure and peak impulse,but also include time domain parameter (integral electromyogrphy,iEMG) and frequency domain parameter (mean power frequency,MPF) of tibial anterior muscle,gastrocnemius,femoral rectus,and biceps femoris muscle.The correlations between peak pressure and height as well as weight were analyzed.Results The feature parameters showed significant difference between left and right foot before and after 30 min natural walking (all P<0.05).The differences of peak pressure and peak impulse values between left and right limb were found in different plantar areas.In the metatarsal areas M3,M4 and M5,the pressure parameters of left side were larger than those of right side (all P<0.05).The values of iEMG and sEMG of left limb were larger than those of right side in tibialis anterior,gastrocnemius,rectus femoris and biceps femoris (all P< 0.05).The MPF values of sEMG of left limb were larger than those of right side in tibialis anterior and gastrocnemius (all P<0.05).After 30 min natural walking,the peak pressures in right toe2-5 areas were decreased (all P<0.05),the differences of iEMG of sEMG in all muscles were not significantly,and the MPF value of sEMG in left rectus femoris was decreased.Weak correlations were observed between peak pressure and height as well as weight.Conclusions The differences of left and right lower limbs were found in plantar pressure and sEMG values during natural walking.The results showed that 30 min natural walking did not cause significant stress-related fatigue in plantar areas.Therefore,the stability of gait can be maintained for a 30 min natural walking.However,that may lead to fatigue of the femoris rectus in the left limb.
10.Immunization coverage and safety of the 23-valent pneumococcal polysaccharide vaccine in Guang-zhou from 2010 to 2015

Objective To analyze the immunization coverage of 23-valent pneumococcal polysac-charide vaccine(PPV23)in a large population in Guangzhou and to evaluate its safety by analyzing the ad-verse events following immunization(AEFI)reported to the passive surveillance system. Methods Immu-nization data of PPV23 in Guangzhou from 2010 to 2015 were collected from Information Management System of Biological Products and the Information System of Immune Programming of Guangzhou. AEFI reported to the AEFI Information System during 2010 to 2015 was collected for safety evaluation. The collected data were analyzed by using descriptive methodology. Results A total of 621 059 doses of PPV23 were pre-scribed in Guangzhou from 2010 to 2015. Most of the recipients were children younger than 10 years old,ac-counting for 79. 44% . Only 9. 38% of the subjects received PPV23 were older than 60 years. A total of 243 AEFI cases were reported at a rate of 39. 13 cases per 100 000 doses,among which 199 cases(32. 04 / 105 ) showed minor vaccine reactions,25 cases(4. 03 / 105 )occurred adverse events,16 cases(2. 58 / 105 )de-veloped coupled diseases and 3 cases(0. 48 / 105 )were classified as psychogenic reactions. No rare adverse reactions were observed. Conclusion The majority of people immunized with PPV23s in Guangzhou were children,while the immunization coverage among the elderly was relatively low. PPV23 was safe for vaccina-tion as the reported AEFI cases were similar to that of other vaccines.
