2.Peroxisome proliferator-activated receptor-gamma coactivator-1 alpha and exercise-induced skeletal muscle adaptations

BACKGROUND:Peroxisome proliferator-activated receptor-gamma coactivator-1 alpha (PGC-1α) may play an important role in the exercise-induced skeletal muscle adaptation, which is involved in the regulation of a variety of exercise-induced biological reactions.
OBJECTIVE:To review the PGC-1αand endurance training induced skeletal muscle adaptations.
METHODS:The relevant articles about relationship between PGC-1αand endurance training-induced skeletal muscle adaptations were searched from PubMed database (1995-01/2010-10) by using the keywords of“PGC-1α, skeletal muscle, exercise, mitochondrial biogenesis, adaptations”, and the language was limited to English. Repetitive contents were deleted. The 59 col ected articles were searched. According to the criterion, 37 were classified and sorted.
RESULTS AND CONCLUSION:Endurance training can typical y increase the expression/activity of membrane transporters and mitochondrial metabolic enzymes as wel as increase capil arisation in the skeletal muscle, together enhancing the oxidative capacity of the muscle and the ability to oxidize both carbohydrates and fatty acids. Studies in PGC-1αknockout and overexpression mice have clearly demonstrated that PGC-1αplays an important role in maintaining the expression of mitochondrial metabolic and anti-oxidant enzymes in the skeletal muscle and does influence exercise-induced adaptations of mitochondrial proteins. However, PGC-1αis not exclusively required, and additional factors must be involved in the regulation of both basal expression and exercise-induced adaptations. Exercise-induced PGC-1αexpression and potential y increased PGC-1αactivity are likely the mechanisms contributing to skeletal muscle mitochondrial adaptations and concomitant health beneficial effects of regular physical activity.
3.Transcriptional Model of Nucleus Isolated from Lymphocytes of Rat-spleen in vitro

The nuclei of the lymphocytes of rat spleen were isolated with the modified Kohji Ueno method and then were purified. Four kinds of nucleoside triphosphate (ATP, GTP, CTP, UTP) as well as 3H-UTP and certain inorganic ions were added in to study the transcriptional function, DNA-dependent RNA polymerase activity, of the nuclei in vitro. The influence of the substrate concentration, temperature, pH value, the concentration of certain inorganic ions(Mg, Mn, NH4 + ), actinomycin D and ?-amanitin on the polymerase activity was evaluated.It was found that Mn and (NH4)2SO4 in high concentration(0.2M) could markedly enhance the transcriptional activity while actinomycin D and a-amanitin inhibited it. A-amanitin in low concentration (1?g/ml) could inhibit 80% of the transcriptional activity. If proper ion concentration was given, relatively high activity of RNA polymerase could be shown. The transcriptional activity was in linear relation with certain range of the nucleus concentration. Substrate requires the existence of the four kinds of nucleoside triphosphate: this means that the transcriptional model reflects the activity of the DNA-dependent RNA polymerase and is suitable for the study of transcriptional mechanism.
4.Research progress in stem/progenitor cells derived from human placenta

Placental tissues have many cell types with stem/progenitor cell potential and immunomodulatory properties. Moreover, placental tissue is readily available and easily procured without invasive procedures, and its use does not elicit ethical debate. Thus, placental tissue has been a key focus in studies of searching a new source of human stem/progenitor cells. Most of the research published in this area focuses on mesenchymal stromal cells isolated from various parts of the placenta or epithelial cells isolated from amniotic membrane. Considering the complexity of the placenta, an urgent need exists to define the region of origin and methods of isolation of cells derived from this tissue.
5.Construction of the eukaryotic expression vector of mouse 24p3 gene and its expression in NIH3T3 cell lines

Objective To construct the eukaryotic expression vector of mouse 24p3 gene and express it in NIH3T3 fibroblast cell line and verify its biological function of delivering iron into cells. Methods 24p3cDNA was cloned by RT-PCR and the eukaryotic expression vector of mouse 24p3 gene was constructed by gene recombination technique and the recombinant plasmid was verified by restriction enzyme digestion analysis and sequenceing, then transfected into NIH3T3 cells using DOTAP transfection reagent. Then the positive clones were selected and amplified. The expression of 24p3 gene in the culture supernatant of positive clones was analyzed by ELISA and Western blotting and its biological activity of delivering iron into cells was tested by atomic absorption spectrometer. Results The eukaryotic expression vector was constructed successfully. The NIH3T3 cells that stably expressed 24p3 were screened out. 24p3/SIP24 protein could express in the culture supernatant of positive clones and deliver iron into NIH3T3 cells. Conclusion The 24p3 gene was expressed successfully in the transfected NIH3T3 cells with good biological activity, which laid the foundation for further studying its biological function and mechanism involved in the iron metabolism.
6.The relationship among big five-personality, resilience and general health of new recruits

Objective To discuss the relationship among Big Five-Personality, Resilience and General Health of the new recruits in a certain troop. Methods 205 new recruits completed the questionnaires, which include NEO Five-Factor Inventory ( NEO-FFI) , Connor-Davidso Resilience Meter (Chinese version) , and General Health Questionnaire (GHQ-12). Results The neuroticism trait of personality positively correlated to General Health( r = 0.50, P<0.001) and significantly predicted mental disease (β =0.4, P<0.01). The self-strengthening dimension of resilience was negatively related to general health ( r = - 0. 51 , P<0. 01 ). Furthermore, self-strengthening played partial mediating role between neuroticism and general health ( Sobel Z =2. 92, P<0. 01). Conclusion The neuroticism trait of personality is a risk factor for new recruits'mental health, while self-strengthening is a protective factor for it.
7.Natural forgetters' episode memory and personality characteristics

Objective To explore the natural forgetters' episode memory and personality characteristics.Methods Situational Self-Awareness Scale(SSAS),Russell Loneliness Scale and Zimbardo Time Perspective Inventory(ZTPI)are used upon 127 natural forgetters and subjects of control group.in which 90 subjects participate tions,natural forgetters performance worse than subjects of control group(t=1.76 one-tail,P<0.05),but subjects getters score lower in the SSAS and the future subscale of ZTPI(t1=1.73,P<0.05 one-tail;t 2=2.70,P<0.05),and tend to endorse in the present fatalistic subscale of ZTPI(t=2.19,P<0.05),but don't feel more lonely.Conclusion Natural forgetters episodic memory is selectively micro-injured under the enactment conditions,which is different from the aging impairment mode of selective source memory,and they have some special personality characteristics.

Finite element analysis (FEA) is broadly used in biomechanics.Being widely used in clinical studies on cervical spine injury,cervical degeneration and stimulating a variety of cervical spine surgeries,cervical finite element model is becoming more and more accurate in recent years.This paper aims to review the development of cervical finite element model,to introduce the application progress of the modeling and analysis in cervical spine injury,cervical disc arthroplasty,cervical interbody fusion,cervical degeneration and cervical instability,and to prospect the foreground of cervical finite element model in future.
9.Mental Health Status and Its Affecting Factors among Residents of Weihai City

Objective: To well understand the mental health status and its affecting factors of the residents aged 15 and above of Weihai city. Methods:The sample was randomly chosen by using cluster-based sampling method and was interviewed by using both self-made questionnaire and Kessler 10 rating scale. SPSS was used to analyze the field data.Results:11652 individuals were interviewed, the score distribution in total was 13.3?5.6 and about 22% of them were likely to have a mild or medium or high mental disorder. The factors affecting mental health status were various, not only including general factors such as age, sex, but also other factors such as physical condition.Conclusion:The factors affecting mental health are multiple, about one in five residents are in poor mental status.
10.Effect of 1,25-dihydroxyvitamin D3 on renal expression of TGF-1/Smads in diabetic nephropathy rats

Objective To explore the effects of 1,25-dihydroxyvitamin D3 on renal expression of TGFβ1,Smad3 and Smad7.Methods 40 Sprague-Dawley mts were randomized into 4 groups:normal control rats (group A),diabetic nephropathy group (group B),small dose 1,25-dihydroxyvitamin D treatment group (0.5 μg · kg-1 · d-1,group C) and large dose 1,25-dihydroxyvitamin D treatment group (1 μg · kg-1 · d-1,group D),each group had 10 rats.After 12 weeks,the renal function,blood glucose,glycosylated hemoglobin and the urine trace albumin content of each rats were tested.Results The biochemical indicators in group B were higher than those in group A,(t =-16.566,P <0.05;t =-16.949,P <0.05;t =-11.844,P <0.05;t =-19.778,P <0.05;t =-14.013,P < 0.05).Compared with group B,the biochemical indicators and the expression of TGFβ1,Smad3 mRNA reduced in group C and group D,and the expression of Smad7 mRNA increased (F =37.892,P < 0.05;F =70.068,P < 0.05;F =21.95,P <0.05;F =77.619,P <0.05;F =37.670,P <0.05;F =1062.562,P <0.05;F =2463.789,P <0.05;F =81.745,P < 0.05).There were no significant differences between group C and group D (t =0.538,P>0.05;t =1.737,P>0.05;t =0.671,P>0.05;t =1.763,P >0.05;t =0.997,P >0.05;t =1.653,P >0.05;t=1.543,P>0.05;t =-1.313,P >0.05).Conclusion 1,25-dihydroxyvitamnin D3 has protective effect on diabetic nephropathy rats model,the mechanism may be associated with inhibiting the expression of TGF β1 and Smad3,increasing the expression of Smad7.
