1.Optimization of Ultrasonic Extraction for Polysaccharide from Rosa Laevigatae Michx in Guizhou by Central Composite Design-Response Surface Methodology

Objective To select the optimum ultrasonic extracting method of polysaccharides from Rosa Laevigatae Michx in Guizhou. Methods Independent variables were solid-liquid ratio, extraction time and extraction temperature, dependent variable was extraction rate ratio of polysaccharides from Rosa Laevigatae Michx.Linear or nonlinear mathematic models were used to estimate the relationship between independent and dependent variables. Response surface methodology was used to optimize the process of extraction.Prediction was carried out through comparing the observed and predicted value. Results The optimum extraction condition for the polysaccharides was as follows: extracting at 75 ℃ for 60 min and ratio of liquid to solid 30??1 ( mL??g ) . The average extraction yield of polysaccharides was 37. 54% under the optimum extraction condition. Conclusion This optimized extraction process is simple and reliable for the extraction of polysaccharides from Rosa Laevigatae Michx in Guizhou.
2.Adverse Reactions Induced by Lidocaine: Literature Analysis of 44 Cases

OBJECTIVE: To study the general pattern and characteristics of the ADR induced by lidocaine.METHODS: A total of 44 ADR cases caused by lidocaine reported in CHKD during 2002~ 2006 were collected and analyzed statistically. RESULTS: Lidocaine-induced ADR were mostly occurred in local anesthesia and block anesthesia,especially in the first time medication and within ten minutes.ADR could involve multiple organs and systems,and the clinical manifestations were complicated and diversified,which were mainly allergic reactions or even death in severe cases.CONCLUSION: Clinical physicians and pharmacists should be alert to the ADR induced by lidocaine and they should stick to the principle of rational drug use.
3.The effects of insulin like growth factorⅠ on the intestinal adaptation in growth hormone-treated rats

Objective To understand the effect of insulin-like growth factorⅠ (IGFⅠ) on the adaptation of the small intestine in growth hormone (GH)-treated rats. Method 20 SD rats were randomized into GH group and STD group. Parenterally fed, short bowel rat models were established. The morphological changes of the intestinal mucosal epithelia were examined, serum GH, IGFⅠ concentrations were determined by RIA method. The local intestinal IGFⅠ mRNA was evaluated by Northern blot method. 4.Generation and Regulation Technology of Stimulating Signals for Implantable Neural Stimulators.

The accuracy and robustness of stimulating signals are critical important for implantable neural stimulators since they stimulate neurons directly. The characteristics and applications of stimulating signals were depicted in the paper. The principles and features of five common generation and regulation technology of stimulating signals for implantable neural stimulators were introduced. The technical characteristics of them were analyzed. Finally, the development of the implantable neural simulators was prospected.

5.Effect of human papilloma virus16 E6 shRNA mediated by lentivirus on infection ratio and invasive ability of cervical cancer cell

Objective To investigate the influence of lentivirus mediated short hairpin RNA(shRNA)target to human papilloma virus(HPV)16 E6 on invasive ability of cervical cancer Caski cells.Methods Lentivirus was produced after shRNA target to human papilloma virus(HPV)16 E6 and to nonsense was cloned to lentivirus work vector.Infection ratio was assessed by assay of EGFP positive cells of Caski.Total mRNA of E6 was determined by RT-PCR after Caski cells were infected by lentivirus.The change of E6 protein expression was analyzed by Western blot.The invasive ability of Caski cells was assayed employing Transwell.Results The optimal MOI (Multiplicity of infection)of lentivirus to Caski was 2.5.Total mRNA and protein of E6 were decreased (by 70%and 63%)in interfering group compared with control group.The invasive ability of Caski cells also reduced after infected by lentivirus.Conclusion shRNA mediated by lentivirus can inhibit expression of HPV16 E6 and invasive ability of cervical cancer cells.
6.Study on bilingual teaching of functional experiment for medical undergraduates

7.The difference and relationship of gene expression profiles between gastric cancer and chronic atrophic gastritis

Background and purpose:The development of gastric cancer has a long course from chronic mucosal inflammation and involves a lot of genes.We investigated the different gene expression profiles with gene chip of high flux between gastric cancer and chronic atrophic gastritis so as to analyze their biologic significance,combined with endoscopic findings and surgical pathology.Methods:Paired pathological and non-pathological specimens from 23 patients with gastric cancer and 15 with chronic atrophic gastritis were enrolled for the study.Gene chip with 13 824 genes/ESTs was employed for detection of the expression profile,while endoscopic and pathologic findings were collected for evaluation.Data were analyzed by average-linkage hierarchical clustering,t test and Fisher calculation of exact probability.Results:Fifty-three specific genes were filtered from gastric cancer group,and 21 from chronic atrophic gastritis group.In the former group,STS,HAP1 and some other relatively specific genes were expressed in correlation to the Borrmann's classification,while in the latter group,BRD3 and BF508879 were demonstrated positively in correlation to intestinal metaplasia.However,there were also some specific genes,such as SLC26A3,related to precancerous condition,commonly expressed in both two groups.Conclusions:There was either homogenous or heterogeneous gene expression profile in gastric cancer as well as in chronic atrophic gastritis.Abnormal gene expressions were related to precancerous conditions or precancerous lesions and to biologic behavior of tumor.The difference and comparability of gene expression in the two groups studied shows the continuity and pertinence of the two diseases.
8.Utility of whole-body diffusion-weighted imaging in the detection of bone metastases in malignant tumors

Objective To investigate the application of whole-body diffusion-weighted imaging (WB-DWI),and to detect bone metastasis in malignant tumor patients.Methods A total of 118 malignant tumor patients prospectively underwent WB-DWI,standard MRI/CT in suspicious bone metastases parts within one week after inspection.Final diagnosis of the bone metastases in each patient was determined on the basis of results of all radiologic and follow-up examinations.The differential diagnosis value of apparent diffusion coefficient (ADC) in benign and malignant bone metastases was compared with the variance analysis.Results Area under ROC curve value of WB-DWI with standard MRI/CT (0.999) was larger than that of WB-DWI (0.944) and standard MRI/CT (0.983),and the specificity,accuracy and positive predictive value were also the highest(100.0 %,97.4 %,100.0 %).The ADC values of malignant bone metastases were significantly lower than those of benign lesions[(0.71±0.15)×10-3 mmTs vs (1.50±0.23)×10-3 mm2/s] with a statistically significant difference(F =261.587,P < 0.001).Conclusion WB-DWI is the beneficial supplements of standard MRI/CT.Which can be used for assessment and differential diagnosis of bone metastases in malignant tumor patients combined with ADC value.
9.Pathogens and Their Drug-resistance in Severe Pneumonia Induced by Different Underlying Diseases

OBJECTIVE To analyze the pathogens of severe pneumonia induced by different underlying diseases in(hospital) and to evaluate the difference of the pathogens to antimicrobial susceptibility test.METHODS The(bacteria) through sputum culture with VITEK-AMS,and the antimicrobial susceptibility against(bacteria) by K-B method were tested in hospital from 2002 to 2005.RESULTS There were 106 patients with 173 strains of isolated(pathogenic) bacteria,including Klebsiella pneumoniae(KPA) 20.9%,and Escherichia coli(ECO)(16.3%) from the aspiration severe pneumonia;Pseudomonas alcaligenes(PAL) 18.9%,P.aeruginosa(PAL)(16.2%) and Staphylococcus aureus(SAU) 16.2% from the obstructive severe pneumonia;KPN 22.6%,SAU(18.9%),and PAE 17.0% from the COPD complicated with severe pneumonia and PAE 25.0% and SAU 20% from the(hospital-)acquired pneumonia(HAP).CONCLUSIONS The constructed ratio of pathogens is different between(severe) pneumonia infected by different underlying diseases and community-acquired pneumonia without any(underlying) disease.The resistant pathogens are increasing significantly in cases with HAP.
10.Practice and Thought of Physiology Experimental Teaching for the Medical Foreign Students
