1.Association between calcium channel ?1 subunit gene and thyrotoxic periodic paralysis

Objective To explore the relationship between calcium channel ?1 subunit (Cav1.1) gene intron 26 -67 A/G polymorphism and thyrotoxic periodic paralysis(TPP). Methods Cav1.1 gene polymorphism at position -67 was determined by polymerase chain reaction-single-strand conformation polymorphism (PCR-SSCP) in 46 male patients with TPP, 68 male Graves’ disease (GD) patients without TPP and 72 healthy male controls. The difference of genotype and the variation of allele frequencies were analyzed by Chi-square test. Results (1) Frequencies distribution of AG+GG genotype in TPP, GD and control groups were 47.83%, 14.71% and 29.17% respectively, and those of allele G were 44.57%, 13.24% and 27.78% for the three groups respectively. Objective To assess the anesthesia efficacy of remifentanil-propofol or remifentanil-desflurance in patients undergoing video-assisted thoracoscopic surgery(VATS).Methods Forty ASA Ⅰ-Ⅱpatients. undergoing VATS were randomly divided into remifentanil-propofol group(group P,n=20)and remifentanil-desflurance group (group D,n=20).MAP and HR were monitered during the entire procedures. Conscious recovery, spontaneous breathing recovery, the endotracheal extubation time and OAAS score were recorded and compared between two groups. Results During the operation, MAP was decreased significantly in group D (P<0.05).There was no significant difference in conscious recovery, spontaneous breathing recovery, the endotracheal extubation time and OAAS score between two groups. Conclusions The anesthesia efficacy of remifentanil-propofol or remifentanil-desflurance in patients undergoing VATS were both with quick recovery, but the fronter has more stable hemodynamics.

4.Regulation of glutamate current by orexin A on pyramidal neurons in rat prefrontal cortex

Objective To investigate the modulatory effect of orexin A on glutamate receptor-mediated current in the freshly isolated pyramidal neurons from the rat prefrontal cortex (PFC). Methods Deep layer (Ⅴ-Ⅵ) prefrontal cortical pyramidal neurons from postnatal 10 to 14 day-old Wistar rats were acutely dissociated by a combination of mechanical and enzymatic method. Subsequently, the effect of orexin A on the current induced by glutamate was studied by the technique of whole cell patch clamp. Results Both orexin A and glutamate dose-dependently evoked the inward transmembrane current. The current was evoked by 1 mmol/L glutamate as a control group(100%). Objective To investigate the effect of extragenous adenosine(AD) by microinjection in the orexinergic area of rat lateral hypothalamus(LH) on the sleep-wake cycle.Methods A total of 14 male SD rats were given 1,10 or 20 nmol AD and artificial cerebrospinal fluid(aCSF) as own control through three different models of microinjections into the LH.Their electroencephalogram(EEG) and electromyogram(EMG) were recorded by a polygraph recording instrument to observe the changes of sleep-wake after microinjections.Results AD administration to LH at doses of 1,10 and 20 nmol decreased the total amount of wakefulness by 84%,62% and 60% fold during that first 3-hour recording period and concomitantly increased non rapid eye movement(NREM) and REM sleep,when compared with aCSF treatment.The sleep-inducing effect was maintained even in 3 hour after the 20 nmol AD microinjection.The maximal sleep-inducing effect was obtained at the 20 nmol dose of AD administration into the orexinergic LH(P

Objective To study the influences of polysacch arides(PS)from Radix Codonopsis on immune function and hematopoiesis in mice.Methods Several experiments have been condu cted for detecting delayed type hypersensitivity(DTH),hemolysin antibody concentration,endogenous spleen colony,3 H-TdRpermeate nuleated cells in bon e marrow.Results By oral administration,PS from Radix Codonopsis can promote the humoral immunity and PS at a small dosage en-hance the cellular immunity.PS from Radix Codonopsis can also elevate the hemoglobin cont ent of the hemolysis -induced blood -deficiency mice and increase the formation of endogenous spleen c olony,but has less effect on DNA synt hesis in the bone marrowof blood -deficiency mic e caused by 60 Co -?ray.Conclusion PS from Radix Codonopsis can enhance the im-mune function and improve the compen satory hematopoiesis of spleen and t heir mechanisms need to be further studied.
7.Effect and significance of Shenmai injection on value of vascular endothelial active factors of heart valve replacement patients.

OBJECTIVETo observe the effect of Shenmai injection on the value of vascular endothelial active factors nitric oxide (NO), endothelin-1 (ET-1) of patients undergoing heart valve rveplacement and cardiac pulmonary bypass (CPB).
METHODThirty patients of cardiac valve replacement undergoing open heart surgery under cardiopulmonary bypass (CPB) were randomized single-blind method divided into Shenmai injection group (SM) and control group (C) with 15 cases each. Shenmai injection group (SM) were injected Shenmai injection 0.6 mL x kg(-1) added to physiological saline 250 mL after anaesthesia before CPB, the control control group were injected only physiological saline 250 mL at the same time. Blood samples were taken before induction of anesthesia and at 0.5, 2, 6, 24 hours after terminating CPB. To calculate P(A-a) DO2 and respiratory indexs (RI) by blood gas analysis, nitric oxide (NO) and endothelin-1 (ET-1) ET-1 were measured. At the same time, the time of CPB and artery blockage were recorded.
RESULTThere was no statistical significance before operation between 2 groups to every blood index. After CPB, P(A-a) DO2 , RI and ET-1 was higher than pre-operation after CPB (P < 0.05). But P(A-a) DO2, RI and ET-I of Shenmai group were lower than control group evidently at every point after CPB (P < 0.05 or P < 0.01). The concentrations of NO were reduced obveiously after CPB (P < 0.05), but in shenmai group, the range of descent was lower than control group (P < 0.01 or P < 0.05).
CONCLUSIONThe concentrations of NO and ET-1 is connected with the lung injury after CPB. Through rise the level of NO and reduce the level of ET-1, Shenmai injection can alleviate the lung injury in some degree after CPB and improve pulmonary oxygenation function.

8.Association of BCAS3 rs11653176 gene polymorphism with the occurrence of gout

Objective To explore the relationship between the polymorphisms of the primary gout predisposing gene (BCAS3) rs11653176 locus and the incidence of gout in Han Chinese men in coastal areas of Shandong Province. Methods One hundred and fifty-two cases of patients with gout remission,68 cases of acute stage,252 patients with hyperuricemia, and 280 healthy subjects, total males, were enrolled. Genotyping the rs11653176 locus of BCAS3 gene by TaqMan probe technique. The expression level of BCAS3 gene mRNA in each PBMC was measured by RT-qPCR. The levels of tumor necrosis factor-α(TNF-α), interleukin-6(IL-6), and interleukin-18(IL-18)in serum were measured by ELISA. Results The change of allele frequency of rs11653176 locus in BCAS3 gene was associated with gout(P<0.01). BCAS3 mRNA in patients with gout was significantly higher than that of healthy people and patients with hyperuricemia(P<0.01). In gout patients, the expression level of BCAS3 gene containing C allele was higher than that of T allele(mRNA,P<0.05). The inflammatory factors in the acute phase of gout were significantly higher than those in phases of remission and hyperuricemia(P<0. 01). Conclusion Changes in the allele frequency of BCAS3 alleles rs11653176(high C, low T)may contribute to the expression of this gene,and lead to gout. And the onset of gout is closely related to the production of inflammatory factors.
9.Isomangiferin, a Novel Potent Vascular Endothelial Growth Factor Receptor 2 Kinase Inhibitor, Suppresses Breast Cancer Growth, Metastasis and Angiogenesis.

PURPOSE: Vascular endothelial growth factor (VEGF) signal transduction mainly depends on its binding to VEGF receptor 2 (VEGFR-2). VEGF downstream signaling proteins mediate several of its effects in cancer progression, including those on tumor growth, metastasis, and blood vessel formation. The activation of VEGFR-2 signaling is a hallmark of and is considered a therapeutic target for breast cancer. Here, we report a study of the regulation of the VEGFR-2 signaling pathway by a small molecule, isomangiferin. METHODS: A human breast cancer xenograft mouse model was used to investigate the efficacy of isomangiferin in vivo. The inhibitory effect of isomangiferin on breast cancer cells and the underlying mechanism were examined in vitro. RESULTS: Isomangiferin suppressed tumor growth in xenografts. In vitro, isomangiferin treatment inhibited cancer cell proliferation, migration, invasion, and adhesion. The effect of isomangiferin on breast cancer growth was well coordinated with its suppression of angiogenesis. A rat aortic ring assay revealed that isomangiferin significantly inhibited blood vessel formation during VEGF-induced microvessel sprouting. Furthermore, isomangiferin treatment inhibited VEGF-induced proliferation of human umbilical vein endothelial cells and the formation of capillary-like structures. Mechanistically, isomangiferin induced caspase-dependent apoptosis of breast cancer cells. Furthermore, VEGF-induced activation of the VEGFR-2 kinase pathway was down-regulated by isomangiferin. CONCLUSION: Our findings demonstrate that isomangiferin exerts anti-breast cancer effects via the functional inhibition of VEGFR-2. Pharmaceutically targeting VEGFR-2 by isomangiferin could be an effective therapeutic strategy for breast cancer.

10.Transgelin inhibits proliferation and invasion of human thyroid cancer cells by blocking the mitogen-activated protein kinase/extracellular signal-regulated kinase signaling pathway

Objective:To explore the role of transgelin(TAGLN) in the occurrence and development of papillary thyroid carcinoma (PTC) and its possible signal pathway.Methods:One hundred cases of PTC tissues and corresponding paracancerous normal thyroid tissues were collected. Realtime quantitative PCR (RT-qPCR), Western blotting, and immunohistochemistry were used to analyze the expression of TAGLN in PTC tissues and corresponding paracancerous normal thyroid tissues. PTC cells were transfected with plasmid and shRNA lentivirus vector respectively to up-regulate or down-regulate the expression of TAGLN in order to detect the effects of them on the proliferation, invasion, and migration by cell proliferation assay(cell counting kit-8, CCK-8)and cell invasion and migration assays (Transwell). The effects of TAGLN on mitogen-activated protein kinase (MAPK)/extracellular-signal regulating kinase (ERK) signal pathway was detected with Western blotting.Results:RT-qPCR showed that there was no difference in the expression of TAGLN mRNA between PTC and corresponding paracancerous normal thyroid tissues ( P>0.05); Western blotting demonstrated that the expression of TAGLN protein in PTC tissues was significantly lower than that in corresponding paracancerous normal thyroid tissues ( P<0.01). Immunohistochemical results revealed that the expression of TAGLN in PTC tissues was significantly lower than that in corresponding paracancerous normal thyroid tissues. Overexpression of TAGLN inhibited the proliferation, invasion, and migration of PTC cells ( P<0.01), but knockdown of TAGLN promoted the proliferation, invasion, and migration of PTC cells ( P<0.01). Overexpression of TAGLN decreased the expression of phosphorylated ERK ( P<0.05), whereas silencing TAGLN increased phosphorylated ERK level in PTC cells( P<0.01). Conclusion:The expression of TAGLN in PTC is significantly decreased. It is related to the occurrence and development of PTC, and its mechanism may be related to MAPK/ERK signal pathway.
