Objective Nowadays ,in the field of stem cell transplantation ,improving the bone mesenchymal stem cells′ability to be implanted into target organs is still a hot topic .Objective of this study is to observe the effects of Naomaiyihao ( NMYH) capsule, a compound traditional Chinese herbal medicine invented for regulating thesea of blood in brain, and survival rate of BMSCs trans-plantaion in cerebral ischemic tissue of rat . Methods 60 rats were randomly divied into 4 group:untreated group ( serum-free L-DMEM +isotonic saline ) , BMSCs transplantation group ( cell suspension+isotonic saline ) , NMYH group ( serum-free L-DMEM+NMYH) and Combination of NMYH and BMSCs group ( cell suspen-sion+NMYH ) .A rat of model of middle cerebral artery occlusion ( MCAO) was established .The third generation BMSCs were isolated and cultured from bone marrow by the holo-bone marrow adherence , the CM-Dil labeled BMSCs were transplanted into corpora striata with stereotactic technique , the rat′s brains were removed and sliced up after 7-,14-and 21-day′transplantation .Then the survival and distribution of cells were observed under the fluorescence microscope . The expression of CD34 in the brain tissue were tested by immunohistochemical method . Results The fluorescence absorbance of combination group was 1.61 ±0.25 on day 14, increased significantly (P<0.05).when compared with BMSCs group (1.23 ±0.17). The fluorescence absorbence of combination group was 0.36 ±0.09 on day 21, increased significantly (P<0.05) when compared with BMSCs group(0.23 ±0.09).Compared with the model group ,numbers of CD34 positive cells on day 7,14 and 21 significantly in-creased in all treated groups(P<0.01).Numbers of CD34 positive cells were significantly more increased(P<0.05) in combination group than those in BMSCs group . Conclusion Survival, distribution and migration trending to boundary regions of ischemia were observed under the fluorescence microscope after transplantation .Naomaoyihao can increase blood supply around the transplanted cell region, and decrease infarct volume .Therefore it could promote BMSCs to better survive in the brain tissue .

China Oncology 2000;0(06):-
Purpose:To investigate palitaxel (Tax) induct io n of apoptosis in human retinoblastoma Y79 cells and the role of Bcl-2 phosphor ylation in the mechanism. Methods:Antiproliferation effects of Tax on Y79 cells were dete rmined by 3H-thymidine incorporation. Apoptosis of the Tax-treated cells was determined by DNA fragmentation analysis. Bcl-2 phosphorylation was determ ined by Western blot analysis. Cell transfection was used in mutant Bcl-2 trans fecting in Y79 cells. Results:After 24 hours treatment with 1 ?mol/L Tax, 3H- thymidine incorporation decreased in Y79 cells and the effects were time and dos e dependent. DNA ladder appeared in DNA fragmantation analysis after 12-24 hour s treatment with Tax. Bcl-2 Phosphorylation was significantly increased in 12- 24 hours treatment with Tax. Bcl-2, a mutant of Bcl-2, blocked Tax activation process. Conclusions:Tax inhibits cell growth and induces apoptosis in h uman Y79 cells, and phosphorylation of Bcl-2 might be involved in this process.

Journal of Chinese Physician 2000;0(12):-
Objective To analyze the clinicopathological features and enhance the knowledge of Askin tumor in children. Methods The clinicopathological features of 4 cases of children with Askin tumor were described. Results Clinically, the tumor was often seen in elder child patients with painful thoracopulmonary masses.Histopathologically, the nest-like or lobular structures composed of small round-to-oval cells were surrounded by sparse fibrovascular stroma. Among tumor cells there were no or few resticular fibers, however in the surrounding of tumor masses the fibers were found. PAS staining in the tumor in a part of cases was positive. The positive rates of CD99 and Syn detected by immunohistochemistry were high. Neurosecretory granules can be observed by electron microscopy. Two patients received both chemotherapy and radiotherapy three patients died of metastatic tumor of lung and brain and chemotherapy complication in 24,25 and 5 months, respectively. Conclusions Askin tumor is a highly malignant neurosecretory tumor in thoracopulmonary region, which often be misdiagnosed as neuroblastoma, lymphoma, embryonal rhabdomyo sarcoma, tuberculosis and lung abscess, and requires a synthetical treatment plan consisting of pre-and postoperative chemotherapy, radiotherapy and surgical resection. The treatment methods of Askin tumor needs to be further studied.
