1.Relationship between the advanced glycation end products content and expressions of RAGE,ICAM-1 in vascular tissue of diabetic rats

In this study,the relationship between the advanced glycation end products(AGEs) and the expressions of receptor for AGEs(RAGE),intercellular cell adhesion molecule-1(ICAM-1) was investigated.The diabetic rat model was reconstructed and the fluorescence method,RT-PCR and in-situ hybridization techniques were used to detect AGEs content and the expressions of RAGE and ICAM-1 gene in the aorta and cardiac tissues.The results showed that AGEs content in aortic and cardiac tissues increased(P<0.01) in diabetic rats; The expressions of RAGE and ICAM-1 enhanced (P<0.05~0.01) and were positively correlated with the quantity of AGEs accumulation(P<0.01) in the aorta and cardiac tissue.These parameters change in the diabetic rats can be improved with aminoglumine(AG) treatment.Suggesting that AGEs might induce RAGE and ICAM-1 expression.It's postulated that AGEs binding to RAGE play an important role to result in diabetic endothelial cells dysfunction and lesion.

3.Inhibition of human hepatocellular carcinoma cell proliferation by galactose receptor-mediated c-myc antisense oligonucleotide

AIM: To evaluate the inhibitory effect of galactose (Gal)-polyethyleneimine (PEI)-c-myc antisense oligodeoxynucleotide (ASODN) complex on proliferation of human hepatocellular carcinoma cells. METHODS: Human hepatocellular carcinoma cell line Bel-7402 was treated with Gal-PEI-ASODN complex. Cell proliferation was tested by trypan blue dye at different time points and with various concentrations of ASODN treatment. Cell morphology was observed under inverted microscope, cell hypodiploid percentage was analyzed by flow cytometry and cell ultrastructure was observed through electron microscopy. RESULTS: Compared with ASODN group (20 ?mol/L) from (0 h) to 96 h, Gal-PEI-ASODN complex (with ASODN 0.75 ?mol/L) significantly suppressed Bel-7402 cells proliferation, the ASODN concentration within Gal-PEI-ASODN complex and time course acquired were significantly lower and shorter, respectively. Incubated with pure ASODN at different concentrations for 72 hours, cell proliferation was inhibited and IC_(50) was 20.9 ?mol/L; while mediated with galactose receptor for 48 hours, ASODN significantly inhibited cell proliferation and IC_(50) was only 0.294 ?mol/L, the inhibitory efficacy of ASODN enhanced 70.9 folds. While Bel-7402 cells were incubated with Gal-PEI-ASODN complex for 48 hours, cell hypodiploid percentage was much higher than ASODN groups and cell apoptosis was seen under electron microscopy. CONCLUSIONS: Galactose receptor mediated ASODN delivery may significantly increase proliferation inhibition efficacy on Bel-7402 cells. [
4.HPLC Fingerprint of Semen Cassiae

Objective To establish HPLC fingerprint for the identification of Semen Cassiae. MethodsChromatographic fingerprint of Semen Cassiae was investigated by RP-HPLC and the gradient elution mode was applied to chromatographic separation. Data were analysed by fingerprint similarity evaluation software to compare the similarity of the samples. ResultsThe HPLC fingerprint of Semen Cassiae was established preliminarily. ConclusionHPLC Fingerprint method is repeatable and can be used in quality control of Semen Cassiae.
5.Purification extraction of water-extraction solution of Ginkgo leaves with chitosan

AIM:In order to improve the clarity and quality of Ginkgo leaf water-extraction solution,and optimize the production process. METHODS:The experiment was based on the optical absoiption and scattering rate,flocculation rate and loss in flavanones were used as two main parameters to estimate the effect of influencing factors on flocculation.It could be proved by analyzing the particle size in water-extraction solution before and after flocculation. RESULTS:The best flocculation technological parameters of water-extraction solution of Ginkgo leaf with chitosan were as follows:at flocculant concentration of 1.071 g/L,flocculation temperature of 40(?C),pH of 4.9. CONCLUSION:The flocculation of water-extraction solution of Ginkgo leaf with chitosan is effective and selective,it has the advantage over ethanol precipitation method.
6.CT Differential Diagnosis of Thyroid Neoplasm

Objective To investigate the value of CT scans in differential diagnosis of thyroid neoplasm.Methods CT findings ofthyroid neoplasm in 29 cases including 8 cases thyroid adenoma,4 cases nodular goiter and 17 cases thyroid carcinoma identified by operation and pathology were analyzed retrospectively.Results The contour of thyroid adenoma was distinct,a completely enhanced ring in 5 cases and enhanced nodules in 3 cases were showed on contrast-enhanced scan.4 cases of nodular goiter showed homogenous cystic low density the lesions were localized and distinct in contour,the cystic wall could be seen.Thyroid carcinoma appeared unhomogenous density,the edges contour of lesion was not distinct and infiltration on adjacent tissue might be seen.Conclusion CT scan might play an important role in diagnosis and differential diagnosis of thyroid neoplasm.
7.Effects of two modes of ventilation on pediatric patients undergoing lung lobectomy

Objective To investigate the effect of two modes of ventilation on pediatric patients undergoing lung lobectomy.Methods Forty ASA Ⅰ~Ⅲ patients aged 2~10 years,undergoing lung lobectomy were randomly divided into VCV and PCV group(n=20).After general anesthesia,patients in VCV group were ventilated on volume-control mode maintaining 30min and then changed into pressure-control mode;patients in PCV group were ventilated on pressure-control mode maintaining 30min and then changed into volume-control mode.Ppeak,Pplat,Raw,MVI,MVE and EtCO2 were measured according to SSS monitor system.All the patients were treated with traumatic blood pressure measurement,and all the patients were analyzed during the period of pre-operation,30min and 60min after ventilation.Results Compared with VCV group,breathing mechanics MVI and MVE in the PCV group were increased(t=4.135,3.988,P<0.05)and PaO2 in the PCV group was significantly increased(t=3.183,P<0.05),as compared with those on volume-control mode.Conclusion PCV mode was benefit for preventing low oxygen in lung lobectomy surgery.
9.Application of Tracheal Intubation Guided by Fiberoptic Bronchoscopy in Children Snoring Disease Operation

Objective To explore the value of tracheal intubation guided by fiberoptic bronchoscopy in children snoring disease operation.Methods A total of 50 patients underwent tracheal intubation guided by fiberoptic bronchoscope or laryngoscope from August 2015 to June 2016.There were 25 cases of fiberoptic bronchoscopy assigned into group A, 25 cases of laryngoscopy into group B.The intubation time, success rate and adverse reactions of the two groups were compared and analyzed.Results All the intubation in group A succeed at the first time.The intubation time was (20.1±4.3) s and the position was right.There was no need for replacement during the operation.Three cases in group B succeed at the second time because the tonsil was too big to expose the pharynx.The intubation got out of the pharynx in 3 cases in group B and were adjusted to the right position.The intubation time was (50.7±6.9) s and the success rate at the first time was 76% (19/25).Groups A behaved better than group B in aspects of the success rate of signal intubation and intubation time (Fisher''s Exact Test, P=0.022;t=-18.725, P=0.000).There were no adverse reactions in group A, such as pharyngorrhagia, odontoseisis or loss of tooth, laryngospasm and so on.There were 3 cases of hemorrhage from throat after intubation, 3 cases of loss of tooth in different degrees and 1 case of laryngospasm at second intubation time in group B.There was no significant difference in adverse reactions between the two groups (P>0.05).ConclusionFiberoptic bronchoscopy can be used for children snoring disease with good safety as well as rare adverse reactions.
10.Prevention and treatment of peritoneal laceration in the laparoscopic totally extraperitoneal hernia repair during learning curve

Objective To explore the causes and managements of peritoneal laceration in the laparoscopic totally extraperitoneal (TEP) hernia repair during learning curve.Methods The retrospective cross-sectional study was conducted.The clinical data of 120 patients with inguinal hernia who underwent laparoscopic TEP hernia repair in the Third Affiliated Hospital of Anhui Medical University (98 patients) and Anhui Provincial Hospital (22 patients) during surgeons' learning curve between February 2012 and January 2017 were collected.Patients underwent laparoscopic TEP hernia repair,meshes were intraoperatively placed and then fixed by medical glue.Observation indicators:(1) intraoperative situations:surgical procedure,operation time,using of mesh,intraoperative peritoneal laceration;(2) postoperative situations:time to anal exsufflation,time for fluid diet intake,occurrence of complications,duration of hospital stay;(3) follow-up:number of patients receiving follow-up,follow-up time,recurrence of hernia during follow-up,pain in inguinal region,intestinal adhesion and obstruction induced abdominal pain,incisional infection.Follow-up using outpatient examination and telephone interview within 10 days postoperatively and using telephone interview at 10 days postoperatively was performed to detect the recurrence of inguinal hernia,pain in inguinal region,intestinal adhesion and obstruction induced abdominal pain and incisional infection up to May 2017.Measurement data with normal distribution were represented as (x)±s.Results (1) Intraoperative situations:of 120 patients,112 underwent laparoscopic TEP hernia repair,5 converted to laparoscopic transabdominal preperitoneal hernia repair and 3 converted to open surgery due to adhesion between hernial sac and surrounding tissues induced bleeding of separation.Total operation time of 120 patients was (71 ± 13) minutes,including (63± 7) minutes in 106 patients with unilateral hernia and (79 ± 11)minutes in 14 patients with bilateral hernia.All the patients used intraoperatively meshes of 10.0 cm×15.0 cm and 16.0 cm× 10.8 cm.Forty-eight patients had intraoperative peritoneal laceration,peritoneal laceration occurred for reconstruction of preperitoneal space in 10 patients,separation of anterolateral preperitoneal space in 11 patients and improperly operating equipment or hernial sac in 27 patients.Of 48 patients with peritoneal laceration,40 continued to finish operation through acupuncturing into the abdominal cavity for exsufflation and then received peritoneal suture and repair,including 5 with recurrence of indirect inguinal hernia (receiving tissue repair) undergoing peritoneal repair through opening hernial sac,and 8 intraoperatively converted to other or open surgery.(2) Postoperative situations:time to anal exsufflation and time for fluid diet intake in 120 patients were (18± 4) hours and (15±6) hours.Of 120 patients,14 had postoperative complications,scrotal emphysema of 6 patients disappeared in 24 hours anti inguinal and scrotal seroma of 8 patients disappeared after puncture treatment.All the patients were discharged from hospital in 2 days postoperatively.(3) Follow-up:112 of 120 patients were followed up for 3-65 months,with a median time of 31 months.During follow-up,there was no occurrence of recurrence of hernia,pain in inguinal region,intestinal adhesion and obstruction induced abdominal pain and incisional infection.Conclusion During surgeons' learning curve,identifying anatomy of the groin clearly,a right way to treat the hernia sac and broken peritoneum in the operation can ensure the smooth completion of the laparoscopic TEP hernia repair.
