2.Inhibition of human hepatocellular carcinoma cell proliferation by galactose receptor-mediated c-myc antisense oligonucleotide

AIM: To evaluate the inhibitory effect of galactose (Gal)-polyethyleneimine (PEI)-c-myc antisense oligodeoxynucleotide (ASODN) complex on proliferation of human hepatocellular carcinoma cells. METHODS: Human hepatocellular carcinoma cell line Bel-7402 was treated with Gal-PEI-ASODN complex. Cell proliferation was tested by trypan blue dye at different time points and with various concentrations of ASODN treatment. Cell morphology was observed under inverted microscope, cell hypodiploid percentage was analyzed by flow cytometry and cell ultrastructure was observed through electron microscopy. RESULTS: Compared with ASODN group (20 ?mol/L) from (0 h) to 96 h, Gal-PEI-ASODN complex (with ASODN 0.75 ?mol/L) significantly suppressed Bel-7402 cells proliferation, the ASODN concentration within Gal-PEI-ASODN complex and time course acquired were significantly lower and shorter, respectively. Incubated with pure ASODN at different concentrations for 72 hours, cell proliferation was inhibited and IC_(50) was 20.9 ?mol/L; while mediated with galactose receptor for 48 hours, ASODN significantly inhibited cell proliferation and IC_(50) was only 0.294 ?mol/L, the inhibitory efficacy of ASODN enhanced 70.9 folds. While Bel-7402 cells were incubated with Gal-PEI-ASODN complex for 48 hours, cell hypodiploid percentage was much higher than ASODN groups and cell apoptosis was seen under electron microscopy. CONCLUSIONS: Galactose receptor mediated ASODN delivery may significantly increase proliferation inhibition efficacy on Bel-7402 cells. [
3.Optimization of processing parameters for extraction of chlorogenic acid in Fule Granula using Box-Behnken experimental design

Objective To optimize the processing parameters for the extraction of chlorogenic acid in Fule Granula by Box-Behnken experimental design.Methods The content of chlorogenic acid in Fule Granula was determination.The separation was performed on a Agilent ZORBAX Eclipse XDB C18 column(4.6 mm ×250 mm, 5 μm), and column temperature 30 ℃.Aetonitrile-0.2% H3PO4 was used as the mobile phase.The detection wavelength was at 327 nm.A three-factor and three-level Box-Behnken experimental design was employed to investigate effects of extraction time, solid-liquid ratio and extraction times on composite score of extracting amount of chlorogenic acid.Results Optimum process conditions were as follows:12 times the amount of water extracted three times, each time 60 min; chlorogenic acid extraction rate of 3.379%.Conclusion This optimized extraction technology has good predictability by Box-Behnken experimental design, and overall desirability, it provides a reference for application of Fule Granula.
4.Effect of Biyankang Tablets on quality of life and recurrence rate in patients with chronic rhinosinusitis after functional endoscopic sinus surgery

Objective To investigate the effect of Biyankang Tablets on quality of life and recurrence rate in patients with chronic rhinosinusitis after functional endoscopic sinus surgery (FESS). Methods A total of 78 patients with chronic rhinosinusitis treated in our hospital during October 2014 to November 2016 were selected, and they were randomly divided into the control group and the traditional Chinese medicine (TCM) group according to the order of admission, with 39 cases in each group. Both groups were treated with FESS, and the TCM group was additionally treated with Biyankang Tablets. The clinical efficacy, inflammatory factors [interleukin-4 (IL-4), interleukin-8 (IL-8)], the prognosis [visual analogue scale (VAS) score of rhinosinusitis and the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire (EORTC QLQ-C30)], drug safety and recurrence rates in the two groups were observed. Results After treatment, the total effective rate of treatment in TCM group was significantly higher than that in the control group (89.74% vs 71.79%) (χ2=4.044, P<0.05). After treatment, IL-4, IL-8 and VAS scores in the two groups were significantly decreased, and the scores of EORTC and QLQ-C30 were significantly increased, and the changes of scores in TCM group after treatment were more significant than those in the control group (P<0.05). After treatment, there was no significant difference in the incidence of adverse reactions between the two groups, and the recurrence rate in TCM group was significantly lower than that in the control group in 1 year after treatment (P<0.05). Conclusion TCM Biyankang Tablets is significantly effective in the treatment of patients with chronic rhinosinusitis after FESS. It can significantly inhibit the expression of inflammatory factors, effectively improve the prognosis and quality of life, and significantly reduce the recurrence rate.
5.Imaging Features of Blast Lung Injury

Purpose Blast lung injury is severe, and is the main causes of death in explosion. This paper aims to explore the imaging features of blast lung injury, and to provide reference for clinical diagnosis and care. Materials and Methods The clinical data of 37 patients with primary blast lung injury stricken in the 8.2 Kunshan explosion in 2014 were retrospectively studied, with regard to the degree of burn, area, auxiliary emergency treatment and medical history. The imaging manifestations of X-ray and/or CT were also analyzed. Results ① The X-ray and/or CT findings of chest showed that 6 cases (16.2%) presented no obvious abnormality of lung parenchyma, 14 cases (37.8%) manifested increase of lung markings and spot-like clouding opacity, 6 cases (16.2%) presented ground glass opacity, 7 cases (18.9%) manifested patchy shadow, 4 cases (10.8%) showed diffuse consolidation. 6 cases (16.2%) with pleural effusion, 6 cases (16.2%) complicated with pneumothorax and hydropneumothorax, 4 cases (10.8%) with enlarged podoid and pericardial effusion, 2 cases (5.4%) with mediastinal hematoma, 9 cases (24.3%) with rib fracture. The positive rate of X-ray film inspection was 72.2% (13/18), yet that of CT reached 92.9% (13/14). ② Five patients who had not been seen obvious abnormal in emergency lung examination appeared mottling shadows or flake fuzzy shadows in the examination on the second day; 9 cases with aggravated symptoms and infection signs displayed relieved signs, expanded or changed primary lesions. Conclusion Imaging examination is an important tool in the diagnosis of primary blast lung injury. CT is superior to X-ray film on the detection of lesions; therefore, CT examination on chest should be carried out as soon as possible if conditions are allowed.

respectively)was significantly higher than that for digital anterograde venography (0.684 and 0.587,P =0.026,respectively).Conclusion Diagnostic accuracy and detection rate of residuaI or recurrent varicose veins with CT phlebography are found to be more superior than that with digital anterograde venography which has high guidance value in preoperation and assesses the efficacy of post-surgery treatment.

Objective To investigate the clinical value of endometrial receptivity detected by transvaginal color Doppler flow imaging(TV-CDFI) in patients treated with ovulation induction. Methods Fifty-three anovulation patients were divided into two groups at random, 28 cases (67 cycles) in the combined group treated with traditional Chinese medicine and human menopausal gonadotropin(HMG), and 25 cases(66 cycles) in the HMG group were treated with HMG alone. The endometrial thickness, patterns and the parameters of the uterine artery blood flow were detected by TV-CDFI. Objective To study the effect of Konjac polysaccharide (KP) on motilin (MOT) and somatostatin (SS) of gastrointestinal tissue in mice. Methods The mice were divided into four groups randomly. Normal control group (NC) was given normal feed. High, middle, low dose KP treated groups (KPH,KPM,KPL)were given KP 0.8, 0.4, 0.2g per 100g normal feed. The level of motilin and somatostatin of gastrointestinal. tissue was determined simultaneously by radioimmunassay, and blood glucose was determined by blood glucose analyzer after 20 d. Objective To study the expression of IL 1?,TNF ? and nitric oxide (NO) in the cervical intervertebral disc and their roles in the pathogenesis of cervical spondylotic myelopathy (CSM). Methods Fifteen cervical discs were obtained from patients of CSM undergoing anterior disc surgery. Six cervical discs specimens were obtained in the surgical operation with patients with traumatic cervical injury served as the control. The specimens were analyzed for IL 1?、TNF ? by use of ELISA method, NO was measured by optical density assays. Results The contents of IL 1?,TNF ? and NO in disc of CSM patients increased significantly when compared with the control group. Conclusion The results of the present study indicated that IL 1?,TNF ? and NO may be involved in the pathogenesis of CSM.

Objective To study MSCT findings and clinic value of hepatic abscesses on enhanced double-phase and(or)three-phase(arterial and protal venous phase and delayed phase),especially during arterial phase. Methods The MSCT imaging features of 20 cases with hepatic abscesses proved by operation or biopsy were evaluated on enhanced double-phase and(or) three-phase scan during arterial and portal venous phase and delayed phase ,especially during arterial phase.Results (1)The imaging features of aeterial phase were that the hepatic tissues around the lesions showed strong enhancement,but the lesions showed slight enhancement or not or peripheral enhancement. (2)The imaging features of protal venous phase were:a.circular target sign; b.circular sign; c.flower valve sign; d.multiloculated sign. (3)The imaging features of delayed phase were that the masses changed smaller or not and the belt disappeared or got vague. Conclusion The double-phase or three phase MSCT scan fully reflects the pathologic changes of hepatic abcesses and is very important to early diagnosis and differential diagnosis.
